Genesis of Highly Fractionated I-Type Granites from Fengshun Complex:Implications to Tectonic Evolutions of South China

The South China Block is characterized by the large-scale emplacement of felsic magmas and giant ore deposits during the Yanshanian. We present zircon Hf isotopic compositions, whole-rock major and trace element compositions of the Fengshun complex, located in eastern Guangdong Province, South China. The Fengshun complex is a multi-stage magmatic intrusion. It is composed of two main units, i.e., the Mantoushan（MTS） syeno-monzogranites, alkali feldspar granites and the Hulutian（HLT） alkali feldspar granites. LA-ICPMS zircon dating shows that the complex emplaced in 166–161 and 139±2 Ma, respectively. Geochemically, the MTS granites show relatively various geochemical compositions with low REE contents（87.76×10-6–249.71×10-6）, Rb/Sr ratios（1.19–58.93）, pronounced Eu negative anomaly（0.01–0.37） and low Nb/Ta ratios（2.40–6.82）. In contrast, the HLT granites exhibit relatively stable geochemical characteristics with high REE contents（147.35×10-6– 282.17×10-6）, Rb/Sr ratios（2.05–10.30） and relatively high Nb/Ta ratios（4.45–13.00）. The isotopic data of the MTS granites display relatively enriched values, with ISr varying from 0.708 2 to 0.709 7, εNd（t） from-7.8 to-6.9 and εHf（t） from-7.4 to-3.2, in comparison with those of the HLT which are ISr=0.703 05–0.704 77, εNd（t）=-5–-3.4 and εHf（t）=-0.7–1.8）. The two-stage model ages of the MTS granites（T2DM（Nd）=1.51–1.59 Ga and T2DM（Hf）=1.26–1.48 Ga） are also higher than those of the HLT granites（T2DM（Nd）=1.21–1.34 Ga and T2DM（Hf）=0.96–1.10 Ga）. Thus the MTS and HLT granites might originate from different sources. The former is more likely derived from partial melting of Meso-Proterozoic basement triggered by upwelling of asthenosphere and/or underplate of the basaltic magma and then extensive fractional crystallisation, similar to the genesis of Early Yanshanian granitoids of the EW-trending tectono-magmatism belt in the Nanling range. Using cDNA microarray hybridization from a human testicular cDNA library, one gene exhibiting ten-fold difference at expression level between adult and embryo human testes was cloned and named NYD-SP9, which was believed to be involved in spermatogenesis. Southern blot hybridization results showed that NYD-SP9 expressed highly in testis but low in ovary. Protein motif analysis of this cDNA sequence revealed a cluster of phosphorylation sites, indicating its potential involvement in signal pathways during spermatogenesis. Furthermore, one transmembrane helix was predicted in N-terminal region, indicating that putative NYD-SP6 may be served as a transmembrane protein. The proximity of these potential phosphorylation sites to each other indicates that there may be interaction among these sites to regulate spermatogenesis. These findings suggested that protein kinase NYD-SP9 might play a role in male germ cell differentiation.

This study investigated the expression and distribution of 2810408A11Rik in mouse testis and sperm, and explored its role in sperrnatogenesis and sperm function. The expression levels of 2810408A11Rik mRNA in multiple tissue samples were analyzed using bioinformatic resources and RT-PCR technique. A specific rabbit polyclonal antibody was prepared by prokaryotic expression of 2810408A11Rik recombinant protein and utilized for animal immunization. Western blotting, immunohistochemistry and immunofluorescence were used to detect the expression and distribution of 2810408A11Rik. The results of the bioinformatic analysi and RT-PCR showed that 2810408A11Rik mRNA was specifically expressed in mouse testis, and 2810408AllRik protein included a protein phosphatase inhibitor domain. Western blotting assays, immunohistochemistry and immunofluorescence confirmed the expression of 2810408A11Rik protein in mouse testis, especially in post-meiosis round and long spermatids, and that it is localized in the acrosome and the post-nucleus area of sperm. Our findings suggest that 2810408A11Rik may play an important role in spermatogenesis, sperm capacitation and fertilization.

Spermatogenesis is a complex process of terminal differentiation by which mature sperms are generated,and it can be divided into three phases：mitosis,meiosis and *** a previous study,we established a series of proteomic profiles for spermatogenesis to understand the regulation of male fertility and ***,we further investigated the localization and the role of flotillin-2 in ***-2 expression was investigated in the testis of male CD1 mice at various developmental stages of spermatogenesis by using Western blotting,immunohistochemistry and ***-2 was knocked down in vivo in three-week-old male mice using intratesticular injection of small inhibitory RNA（siRNA）,and sperm abnormalities were assessed three weeks ***-2 was expressed at high levels in male germ cells during ***-2 immunoreactivity was observed in pachytene spermatocytes as a strong dot-shaped signal and in round spermatids as a sickle-shaped distribution ahead of the *** confirmed flotillin-2 was localized in front of the acrosome in round spermatids,indicating that flotillin-2 was localized to the Golgi *** of flotillin-2 in vivo led to a significant increase in head sperm abnormalities isolated from the cauda epididymis,compared with control siRNA-injected *** study indicates that flotillin-2 is a novel Golgi-related protein involved in sperm acrosome biogenesis.

Non-obstructive azoospermia （NOA） is a severe defect in male reproductive health that occurs in 1% of adult men. In a previous study, we identified that rs7099208 is located within the last intron of FAM160B1 at 10q25.3. In this study, we analysed expression Quantitative Trait Loci （eQTL） of FAM16OB1, ABLIM1 and TRUB1, the three genes surrounding rs7099208. Only the expression level of FAM16OB1 was reduced for the homozygous alternate genotype （GG） of rs7099208, but not for the homozygous reference or heterozygous geno- types. FAM160B1 is predominantly expressed in human testes, where it is found in spermatocytes and round sper- matids. From 17 patients with NOA and five with obstructive azoospermia （OA）, immunohistochemistry revealed that expression of FAM160B1 is reduced, or undetectable in NOA patients, but not in OA cases or normal men. We conclude that rs7099208 is associated with NOA via a reduction in the expression of FAM160B1.

Embryonic stem（ES） cells are pluripotent cells that can give rise to derivatives of all three embryonic germ layers. Due to its characteristics, the patient-specific ES cells are of great potential for transplantation therapies. Several strategies can reprogramme somatic cells back to pluripotent stem cells： nuclear transfer, fusion with ES cells, treatment with cell extract and induction by specific factors. Considering the future clinical use, the differentiation from ES to neurons, cardiomyocytes and many other types of cells currently provide basic cognition and experience to regenerative medicine. This article will review two courses, the reprogramming of differentiated cells and the differentiation of ES cells to specific cell types.

Dietary parboiled rice(PR)has a low risk of disease,but little is known about the contribution of PR to the prevention of *** potential role and underlying mechanisms of PR in hyperlipidemia were evaluated in this *** C57BL/6J mice were fed with a normal diet,high-fat diet(HFD)containing refined rice(HFDRR)or PR(HFDPR).It was found that PR intervention improved lipid accumulation in *** data analysis revealed that 27 genes were up-regulated(mostly involved in lipid breakdown)and 86 genes were down-regulated(mostly involved in inflammatory responses)in the HFDPR group compared to the HFDRR *** 15 differentially expressed genes(DEGs)were validated by quantitative real-time PCR(RT-qPCR),while protein interaction network showed that protein tyrosine phosphatase receptor type C(PTPRC)has a central *** gut microbiota of mice was also altered after different dietary treatments,with higher ratio of Firmicutes and Bacteroidetes,increased abundances of Ruminococcaceae,Lachnospiraceae,Christensenellaceae,Porphyromonadaceae,Rikenellaceae and Prevotellaceae,and decreased abundances of Lactobacillaceae,Peptostreptococcaceae,Erysipelotrichaceae and Actinobacteria in the HFDRR *** addition,it was observed that PPAR signaling pathway may act as a bridge between DEGs and differential gut *** results suggested that PR can prevent hyperlipidemia by modulating liver genes and gut microbiota.

DDX3X regulates cell survival and cell cycle during mouse early embryonic development

DDX3X is a highly conserved DEAD-box RNA helicase that participates in RNA transcription, RNA splicing, and mRNA transport, translation, and nucleo-cytoplasmic transport. It is highly expressed in metaphase II （MII） oocytes and is the predominant DDX3 variant in the ovary and embryo. However, whether it is important in mouse early embryo development remains unknown. In this study, we investigated the function of DDX3X in early embryogenesis by cytoplasmic microinjection with its siRNA in zygotes or single blastomeres of 2-cell embryos. Our results showed that knockdown of Ddx3x in zygote cytoplasm led to dramatically diminished blastocyst formarion, reduced cell numbers, and an increase in the number of apoptotic cells in blastocysts. Meanwhile, there was an accumulation of p53 in RNAi blastocysts. In addition, the ratio of cell cycle arrest during 2-cell to 4-cell transition increased following microinjection of Ddx3x siRNA into single blastomeres of 2-cell embryos compared with control. These results suggest that Ddx3x is an essential gene associated with cell survival and cell cycle control in mouse early embryos, and thus plays key roles in normal embryo development.
