Alzheimer's disease is characterized by deposition of amyloid-β,which forms extracellular neuritic plaques,and accumulation of hyperphosphorylated tau,which aggregates to form intraneuronal neurofibrillary tangles,in the *** NLRP3 inflammasome may play a role in the transition from amyloid-βdeposition to tau phosphorylation and *** NLRP3 is primarily found in brain microglia,and tau is predominantly located in neurons,it has been suggested that NLRP3 expressed by microglia indirectly triggers tau phosphorylation by upregulating the expression of pro-inflammatory ***,we found that neurons also express NLRP3 in vitro and in vivo,and that neuronal NLRP3 regulates tau *** biochemical methods,we mapped the minimal NLRP3 promoter and identified FUBP3 as a transcription factor regulating NLRP3 expression in *** primary neurons and the neuroblastoma cell line Neuro2A,FUBP3 is required for endogenous NLRP3 expression and tau phosphorylation only when amyloid-βis *** the brains of aged wild-type mice and a mouse model of Alzheimer's disease,FUBP3 expression was markedly increased in cortical *** analysis suggested that FUBP3 plays a role in neuron-mediated immune *** also found that FUBP3 trimmed the 5′end of DNA fragments that it bound,implying that FUBP3 functions in stress-induced *** findings suggest that neuronal NLRP3 may be more directly involved in the amyloid-β-to–phospho-tau transition than microglial NLRP3,and that amyloid-βfundamentally alters the regulatory mechanism of NLRP3 expression in *** that FUBP3 was only expressed at low levels in young wild-type mice and was strongly upregulated in the brains of aged mice and Alzheimer's disease mice,FUBP3 could be a safe therapeutic target for preventing Alzheimer's disease progression.

Development of Gravity Theories in the View of TRAPPIST-1e

Contrary to the solar system, most exoplanet systems detected hitherto are close-in and compact. One typical system is TRAPPIST-1, which has seven nearly co-planar terrestrial planets all within the orbit of Mercury,including three in the habitable zone. To evaluate the differences in developing sophisticated gravity theories from the solar system, we use N-body integrations to simulate ephemeris and reproduce some important astronomy phenomena observed on the potentially habitable planet TRAPPIST-1e. Retrograde motions of other planets last1–2 orders of magnitude shorter than in the solar system, but occur much more frequently. Transit events of all inner planets can be observed steadily. Except for Kepler's first law, which is hard to notice for low eccentricities of planets, the other two laws can then be precisely verified in 102days, because the areas swept by planets vary by?0.01% and the observed semimajor axes and periods result in constants with theoretical and observation accuracies both ?2%. However, the mean motion correlation implies that the Great Inequality is not always apparent between one pair of planets like Jupiter and Saturn. Furthermore, general relativity can hardly be discovered because it gives rise to perihelion precession of inner planets only ～0.1% of gravity precession, dozens of times smaller than Mercury. Our results support the possibility of developing part of gravity theories by potential exo-civilizations in compact systems like TRAPPIST-1.

CiNAC2 positively regulates drought stress tolerance by promoting superoxide dismutase activity in pecan(Carya illinoinensis)

Pecan is an extremely important crop cultivated worldwide for edible nuts and nut *** the changes in precipitation and soil moisture caused by climate change and worsening global water scarcity,it is important to understand the mechanism of pecan response to *** this end,this study investigated the response of pecan to drought stress and rehydration using physiological and transcriptomic *** dismutase(SOD)enzyme activity in leaves was significantly upregulated during drought stress,suggesting that it might play an important role in drought *** gene co-expression network analysis of the transcriptome data was used to screen for a key drought-responsive gene,CiNAC2,which was overexpressed in Arabidopsis thaliana for functional *** analysis of stomatal apertures and the water loss rate in leaves showed that CiNAC2 might respond to drought stress via mediating stomatal aperture *** addition,CiNAC2 could promote root growth under drought ***1 was verified as a direct target gene of CiNAC2 by yeast one-hybrid assay dual-luciferase reporter *** one-hybrid analysis confirmed that CiNAC2 bound to the promoters of *** expression in Nicotiana benthamiana epidermis showed that CiNAC2 upregulated the expression of *** results demonstrated that CiNAC2 enhanced drought stress tolerance via promoting SOD activity in pecan and provided a theoretical basis for breeding drought-resistant varieties in pecan.

Preceding crop rotation systems shape the selection process of wheat root-associated bacterial communities

Wheat–maize (WM) and wheat–soybean (WS) double-cropping rotation systems are predominant in the North China Plain, with implications for national agricultural output and *** rotation systems exert legacy effects on soil health and crop productivity, the role of crop rotation in shaping the root-associated microbiome of the succeeding crops has emerged as a pivotal aspect of crop management ***, the effects of the preceding two cycles of WM and WS rotations on the recruitment and filtering of wheat root-associated bacterial communities across wheat developmental stages were *** results revealed that bacterial community diversity and composition were primarily influenced by compartment and developmental stage, while the preceding rotation systems had a slight but significant effect on wheat root-associated bacterial *** co-occurrence networks under WM were more complex in the wheat rhizosphere and rhizoplane, with the operational taxonomic units (OTUs) related to cellulolysis showing greater *** co-occurrence networks under WS were simple but stable in the rhizosphere and complex in the rhizoplane and endosphere, with the OTUs related to ureolysis and nitrogen fixation showing greater *** both stochastic and deterministic processes contributed to the assembly of wheat root-associated bacterial communities, the contributions of deterministic processes under WS were 19.4–38.5%higher than those under the WM rotation across the root-associated compartments,indicating the substantial impact of a soybean legacy effect on wheat root selection of *** growthpromoting rhizobacteria with the potential to fix nitrogen, produce indole-3-acetic acid, and inhibit diseases such as Betaproteobacteriales, Azospirillales and Dyella sp., were identified within the OTUs that were consistently enriched across all the wheat root-associated compartments and developmental stages, which were also important

Factors influencing vaginal cuff dehiscence after laparoscopic hysterectomy and the psychological state of the patients

BACKGROUND Despite laparoscopic total hysterectomy provided more benefits to patients and has become one of the most commonly used surgical methods for total ***,vaginal cuff dehiscence is a serious complication after laparoscopic total hysterectomy and so far,there are limited studies on vaginal cuff dehiscence after a total *** To investigate the factors influencing vaginal cuff dehiscence after laparoscopic hysterectomy and the psychological state of the *** This study retrospectively,univariately,and multivariately analyzed the clinical data of patients who experienced vaginal cuff dehiscence after laparoscopic hysterectomy in Yuyao People’s Hospital from January 2015 to December *** regression was utilized to analyze the high-risk factors of vaginal cuff *** Among 1459 cases with laparoscopic hysterectomy,9 reported vaginal cuff dehiscence postoperatively,with an incidence of 0.617%.Univariate logistic regression analysis revealed that preoperative hemoglobin value[odds ratio(OR)=5.12,P=0.016],vaginal cuff suture method(OR=0.26,P=0.048),and postoperative first sexual lifetime(OR=15.86,P=0.002)were associated with vaginal cuff dehiscence following laparoscopic *** logistic regression analysis revealed that preoperative hemoglobin value of<90 g/L(OR=5.17,P=0.015)and time interval between postoperative first sexual life and operation of<3 months(OR=54.00,P=0.004)are independent risk factors for vaginal cuff dehiscence after laparoscopic *** Postoperative hemoglobin and the time interval between postoperative first sexual life and operation were the independent factors of developing vaginal cuff dehiscence after laparoscopic hysterectomy.

End-to-end dataflow engineering framework of honey manufacturing from intermediates to process by TAS1R2@AuNPs/SPCE biosensor coupled with quality transfer principle

This study reported an original end-to-end dataflow engineering framework for the quality transfer principle to overcome the quality challenges in real-world honey manufacturing. Firstly, 650 pivotal data points of physical and chemical quality attributes from 65 batches of honey intermediates were characterized through multiple sensors, which included rheological properties, acidity, moisture, and sugars. Furthermore, a hypersensitized TAS1R2@AuNPs/SPCE biosensor was developed to identify biological quality attributes of honey, the powerful affinities between honey intermediates and the TAS1R2 receptor were discovered (KD −8 M), and the abnormal batches of B2, B23 and C23 were diagnosed by TAS1R2@AuNPs/SPCE biosensor and multivariable algorithm. Finally, the end-to-end dataflow containing physical, chemical and biological critical quality attributes was successfully established to interpret the quality transfer principle of honey manufacturing, which revealed that the front-end refining process was relatively unstable and the back-end refining process was a negligible influence on the quality of honey manufacturing. This framework embraces quality management, quality transfer, and biosensor information, which will contribute to discovering the quality transfer principle in industrial innovation for intelligent manufacturing. © 2022

The effect of Ti content on the microstructure and mechanical properties of as-cast light-weight Ti_(x)(AlVCr)_(100−x)medium entropy alloys was studied by compressive tests,X-ray diffraction,scanning electron microscopy and transmission electron *** results suggest that yield strength increases and then decreases with the increment of Ti *** Ti_(60)(AlVCr)_(40)alloy has the best combination of high strength of 1204 MPa and uniform plastic strain of 70%,possessing a high specific yield strength of 255 MPa·cm^(3)/*** enhancement of strength is mainly attributed to the synergic effects of solid-solution and coherent nano-precipitation strengthening,while dislocation motion such as dislocation pinning,entanglement and dislocation cells significantly increases the strain-hardening capacity.
