Potential of Antibody-Dependent Cellular Cytotoxicity in Acute and Recovery Phases of SARS-CoV-2 Infection

Background:Severe acute respiratory syndrome coronavirus-2(SARS-CoV-2)has caused a global pandemic that has resulted in millions of casualties.Although researchers have reported the existence of neutralizing antibodies and viral T cell immunity against SARS-CoV-2,little is known about the presence of antibody-dependent cellular cytotoxicity(ADCC)and its role in combating SARS-CoV-2 infection.Methods:Nineteen acute COVID-19 patients at the First Affiliated Hospital of Guangzhou Medical University from January to February,2020 and 55 recovery COVID-19 patients at the Second Peoples Hospital of Changde City from February,2020 to February,2021 were recruited in this study.Longitudinal plasma samples were collected.A virus-specific ADCC assay was performed to study the COVID-19 plasma samples.The correlations between ADCC and total IgG titer,including anti-RBD,anti-N,and neutralizing antibody titer were analyzed.Results:A high level of ADCC with 0.86%of IFN-g+CD107a+NK cells induced by anti RBD antibodies and with 0.54%of IFN-g+CD107a+NK cells induced by anti N antibodies was observed.This activity peaked at 3 weeks after disease onset with 1.16%and 0.63%of IFN-g+CD107a+NK cells induced by anti RBD and anti N antibodies respectively,declined to 0.32%and 0.32%of IFN-g+CD107a+NK cells respectively after more than 2 months,and persisted for 12 months after disease onset.The ADCC did not aggravate the severity of COVID-19 in terms of sequential organ failure assessment,although ADCC decreased with the age of COVID-19 patients.Interestingly,ADCC response is not correlated with neutralizing antibody titer or total IgG titers against S protein RBD and N protein in acute patients.ADCC in recovered patients showed a significant correlation with anti RBD IgG titer(R2=0.33,P<0.001).Conclusion:Antibodies from COVID-19 patients against the N protein and S protein RBD domains could stimulate high levels of ADCC response.Our results provide evidence that vaccination should not only focus on neutralizing antibodies but also binding a

Understanding the decay and maintenance of long-term SARS-CoV-2 neutralizing antibodies in infected or vaccinated people and how vaccines protect against other SARS-CoV-2 variants is critical for assessing public vaccination plans.Here,we measured different plasm antibody levels 2 and 12 months after disease onset,including anti-RBD,anti-N,total neutralizing antibodies,and two neutralizing-antibody clusters.We found that total neutralizing antibodies declined more slowly than total anti-RBD and anti-N IgG.

SARS-CoV-2 Omicron variant infection generally gives rise to asymptomatic to moderate COVID-19 in vaccinated people.The immune cells can be reprogrammed or“imprinted”by vaccination and infections to generate protective immunity against subsequent challenges.Considering the immune imprint in Omicron infection is unclear,here we delineate the innate immune landscape of human Omicron infection via single-cell RNA sequencing,surface proteome profiling,and plasma cytokine quantification.We found that monocyte responses predominated in immune imprints of Omicron convalescents,with IL-1β-associated and interferon(IFN)-responsive signatures with mild and moderate symptoms,respectively.Low-density neutrophils increased and exhibited IL-1β-associated and IFN-responsive signatures similarly.Mild convalescents had increased blood IL-1β,CCL4,IL-9 levels and PI3+neutrophils,indicating a bias to IL-1βresponsiveness,while moderate convalescents had increased blood CXCL10 and IFN-responsive monocytes,suggesting durative IFN responses.Therefore,IL-1β-or IFN-responsiveness of myeloid cells may indicate the disease severity of Omicron infection and mediate post-COVID conditions.

目的对比不同手术方式对胃溃疡患者胃肠功能的恢复及安全性。方法选取2020年6月至2023年6月山东省高密市人民医院收治的160例胃溃疡患者,随机数字表法分为传统开腹组、腹腔镜组,传统开腹组予以传统开腹修补术治疗,腹腔镜组予以腹腔镜修补术治疗;比较两组临床疗效,及两组炎症因子水平、胃消化功能相关指标、胃肠动力学相关指标及疼痛评分,比较两组并发症发生情况。结果手术后,腹腔镜组MTL、PG水平和治疗总有效率均高于传统开腹组(均P<0.05);手术后,腹腔镜组IL-6、TNF-α、CCK、G-17水平及VAS评分和并发症发生率均低于传统开腹组(均P<0.05)。结论腹腔镜修补术效果优于传统开腹修补术,可有效改善炎症因子水平、胃功能、胃肠功能,减少并发症的出现,促进术后快速恢复。

Enhanced bifunctional oxygen electrochemical catalytic performance using La-doped CoFe_(2)O_(4)spinel supported by 3D-G for Zn-air batteries

The preparation of bifunctional catalysts for oxygen reduction(ORR)and oxygen evolution(OER)is crucial for Zn-air batteries.Here,we report a La doped CoFe_(2)O_(4) spinel catalyst supported on threedimensional graphene(3D-G),where La can facilitate electron transfer from Co to Fe,leading to increased electron cloud density in Fe and improved catalytic performance.The redshift of the G peak in the Raman spectra indicates the interaction between theπbond of 3D-G and d orbitals in La_(0.2)CoFe_(1.8)O_(4).La_(0.2)CoFe_(1.8)/3D-G exhibits superior ORR performance(E_(1/2)=0.86 V vs.RHE)and OER performance(E_(j=10)=1.55 V vs.RHE)to CoFe_(2)O_(4)/3D-G(E_(1/2)=0.831 V vs.RHE,E_(j=10)=1.603 V vs.RHE).Furthermore,it demonstrates excellent bifunctional oxygen catalytic performance while maintaining high power density and stability in liquid zinc-air batteries(ZABs)and flexible ZABs(F-ZABs).This work presents a viable strategy for utilizing rare earth element doped spinels to enhance oxygen catalyst and ZABs performance.

Anaerobic digestion(AD)of waste activated sludge(WAS)is usually limited by the low generation efficiency of methane.Fe(Ⅲ)-loaded chitosan composite(CTS-Fe)have been reported to effectively enhanced the digestion of WAS,but its role in promoting anaerobic sludge digestion remains unclear.In present study,the effects of CTS-Fe on the hydrolysis and methanogenesis stages of WAS anaerobic digestion were investigated.The addition of CTSFe increased methane production potential by 8%-23%under the tested conditions with the addition of 5-20 g/L CTS-Fe.Besides,the results demonstrate that the addition of CTS-Fe could effectively promote the hydrolysis of WAS,evidenced by lower protein or polysaccharides concentration,higher soluble organic carbon in rector adding CTS-Fe,as well as the increased activity of extracellular hydrolase with higher CTS-Fe concentration.Meanwhile,the enrichment of Clostridia abundance(iron-reducing bacteria(IRBs))was observed in CTS-Fe adding reactor(8.9%-13.8%),which was higher than that in the control reactor(7.9%).The observation further suggesting the acceleration of hydrolysis through dissimilatory iron reduction(DIR)process,thus providing abundant substrates for methanogenesis.However,the presence of CTS-Fe was inhibited the acetoclastic and hydrogenotrophic methanogenesis process,which could be ascribed to the Fe(Ⅲ)act as electron acceptor coupled to methane for anaerobic oxidation.Furthermore,coenzyme F420 activity in the CTS-Fe added reactor was 34.9% lower than in the blank,also abundance of microorganisms involved in hydrogenotrophic methanogenesis was decreased.Results from this study could provide theoretical support for the practical applications of CTS-Fe.

Chimeric antigen receptor(CAR)T-cell therapy has demonstrated clinical response in treating both hematologic malignancies and solid tumors.Although instances of rapid tumor remissions have been observed in animal models and clinical trials,tumor relapses occur with multiple therapeutic resistance mechanisms.Furthermore,while the mechanisms underlying the long-term therapeutic resistance are well-known,short-term adaptation remains less understood.However,more views shed light on short-term adaptation and hold that it provides an opportunity window for long-term resistance.In this study,we explore a previously unreported mechanism in which tumor cells employ trogocytosis to acquire CAR molecules from CAR-T cells,a reversal of previously documented processes.This mechanism results in the depletion of CAR molecules and subsequent CAR-T cell dysfunction,also leading to short-term antigen loss and antigen masking.Such type of intercellular communication is independent of CAR downstream signaling,CAR-T cell condition,target antigen,and tumor cell type.However,it is mainly dependent on antigen density and CAR sensitivity,and is associated with tumor cell cholesterol metabolism.Partial mitigation of this trogocytosis-induced CAR molecule transfer can be achieved by adaptively administering CAR-T cells with antigen density-individualized CAR sensitivities.Together,our study reveals a dynamic process of CAR molecule transfer and refining the framework of clinical CAR-T therapy for solid tumors.

The interactions of phenothiazine such as chlorpromazine hydrochloride (CPZ·HCl) and promethazine hydrochloride (PZ·HCl), with nucleic acids (NA) have been investigated. The CPZ-NA system can produce very intense resonance Rayleigh scattering (RRS), but the RRS intensity of the PZ-NA system is very weak. The energy relationship of the two reaction systems has been calculated by the AM1 and B3LYP of density functional methods for quantum chemistry, the binding site and interacting model of CPZ-NA and PZ-NA are studied. The results show that CPZ has strong binding ability with nucleic acids, but PZ hardly react with nucleic acids. It can be expected that RRS method may be a useful means for molecular recognition of nucleic acids and the molecular design of gene medicine in the future. Moreover, the method is used for the quan-titative determination of nucleic acids by employing chlorpromazine hydrochloride as reagent, which has very high sensitivity and good selectivity. Therefore, a sensitive, simple and rapid new method has been developed for the determination of nucleic acids.

房屋建筑调查数据质量是普查成果的重要组成部分。常规的质量检查方法是人工检查与国家质检软件质检相结合,人工检查在检查内容、效率、成本等方面存在多种不足,国家质检软件只能对字段是否必填及图形拓扑关系等进行检查,缺少字段逻辑关系检查和规范性检查。文章针对房屋建筑调查技术导则对数据质量的要求,从空间、属性、拓扑等方面详细描述了房屋建筑调查数据质检软件的设计内容,围绕属性和拓扑关系规则集的内容组织、检查模型构建等方面阐述了质检软件的设计思路,在关键技术和主要功能等方面提出技术思路,为住建部门开展房屋建筑调查数据质量检查工作提供借鉴。
