Recent advances and remaining challenges in lung cancer therapy

Lung cancer remains the most common cause of cancer death.Given the continued research into new drugs and combination therapies,outcomes in lung cancer have been improved,and clinical benefits have been expanded to a broader patient population.However,the overall cure and survival rates for lung cancer patients remain low,especially in metastatic cases.Among the available lung cancer treatment options,such as surgery,radiation therapy,chemotherapy,targeted therapies,and alternative therapies,immunotherapy has shown to be the most promising.The exponential progress in immuno-oncology research and recent advancements made in the field of immunotherapy will further increase the survival and quality of life for lung cancer patients.Substantial progress has been made in targeted therapies using tyrosine kinase inhibitors and monoclonal antibody immune checkpoint inhibitors with many US Food And Drug Administration(FDA)-approved drugs targeting the programmed cell death ligand-1 protein(e.g.,durvalumab,atezolizumab),the programmed cell death-1 receptor(e.g.,nivolumab,pembrolizumab),and cytotoxic T-lymphocyte-associated antigen 4(e.g.,tremelimumab,ipilimumab).Cytokines,cancer vaccines,adoptive T cell therapies,and Natural killer cell mono-and combinational therapies are rapidly being studied,yet to date,there are currently none that are FDA-approved for the treatment of lung cancer.In this review,we discuss the current lung cancer therapies with an emphasis on immunotherapy,including the challenges for future research and clinical applications.

Effects of microenvironment and biological behavior on the paracrine function of stem cells

Mesenchymal stem cells(MSCs),the most well-studied cell type in the field of stem cell therapy,have multi-lineage differentiation and self-renewal potential.MsC-based thera-pies have been used to treat diverse diseases because of their ability to potently repair tissue and locally restore function.An increasing body of evidence demonstrates that paracrine func-tion is central to the effects of MsC-based therapy.Growth factors,cytokines,chemokines,extracellular matrix components,and extracellular vehicles all contribute to the beneficial ef-fects of MSCs on tissue regeneration and repair.The paracrine substances secreted by MSCs change depending on the tissue microenvironment and biological behavior.In this review,we discuss the bioactive substances secreted by MsCs depending on the microenvironment and biological behavior and their regulatory mechanisms,which explain their potential to treat human diseases,to provide new ideas for further research and clinical cell-free therapy.

Learning Scalable Task Assignment with Imperative-Priori Conflict Resolution in Multi-UAV Adversarial Swarm Defense Problem

The multi-UAV adversary swarm defense(MUASD)problem is to defend a static base against an adversary UAV swarm by a defensive UAV swarm.Decomposing the problem into task assignment and low-level interception strategies is a widely used approach.Learning-based approaches for task assignment are a promising direction.Existing studies on learning-based methods generally assume decentralized decision-making architecture,which is not beneficial for conflict resolution.In contrast,centralized decision-making architecture is beneficial for conflict resolution while it is often detrimental to scalability.To achieve scalability and conflict resolution simultaneously,inspired by a self-attention-based task assignment method for sensor target coverage problem,a scalable centralized assignment method based on self-attention mechanism together with a defender-attacker pairwise observation preprocessing(DAP-SelfAtt)is proposed.Then,an imperative-priori conflict resolution(IPCR)mechanism is proposed to achieve conflict-free assignment.Further,the IPCR mechanism is parallelized to enable efficient training.To validate the algorithm,a variant of proximal policy optimization algorithm(PPO)is employed for training in scenarios of various scales.The experimental results show that the proposed algorithm not only achieves conflict-free task assignment but also maintains scalability,and significantly improve the success rate of defense.

The humanoid robot head plays an important role in the emotional expression of human-robot interaction(HRI).They are emerging in industrial manufacturing,business reception,entertainment,teaching assistance,and tour guides.In recent years,significant progress has been made in the field of humanoid robots.Nevertheless,there is still a lack of humanoid robots that can interact with humans naturally and comfortably.This review comprises a comprehensive survey of state-of-the-art technologies for humanoid robot heads over the last three decades,which covers the aspects of mechanical structures,actuators and sensors,anthropomorphic behavior control,emotional expression,and human-robot interaction.Finally,the current challenges and possible future directions are discussed.

Efficient Polytelluride Anchoring for Ultralong-life Potassium Storage: Combined Physical Barrier and Chemisorption in Nanogrid-in-Nanofiber

Metal tellurides(MTes) are highly attractive as promising anodes for high-performance potassium-ion batteries. The capacity attenuation of most reported MTe anodes is attributed to their poor electrical conductivity and large volume variation. The evolution mechanisms, dissolution properties, and corresponding manipulation strategies of intermediates(K-polytellurides, K-pTe_(x)) are rarely mentioned. Herein,we propose a novel structural engineering strategy to confine ultrafine CoTe_(2) nanodots in hierarchical nanogrid-in-nanofiber carbon substrates(CoTe_(2)@NC@NSPCNFs) for smooth immobilization of K-pTe_(x) and highly reversible conversion of CoTe_(2) by manipulating the intense electrochemical reaction process. Various in situ/ex situ techniques and density functional theory calculations have been performed to clarify the formation, transformation, and dissolution of K-pTe_(x)(K_(5)Te_(3) and K_(2)Te), as well as verifying the robust physical barrier and the strong chemisorption of K_(5)Te_(3) and K_(2)Te on S, N co-doped dual-type carbon substrates. Additionally, the hierarchical nanogrid-in-nanofiber nanostructure increases the chemical anchoring sites for K-pTe_(x), provides sufficient volume buffer space, and constructs highly interconnected conductive microcircuits, further propelling the battery reaction to new heights(3500 cycles at 2.0 A g^(-1)). Furthermore, the full cells further demonstrate the potential for practical applications. This work provides new insights into manipulating K-pTe_(x) in the design of ultralong-cycling MTe anodes for advanced PIBs.

Antimony(Sb)-ba sed anode materials are feasible candidates for sodium-ion batteries(SIBs) due to their high theoretical specific capacity and excellent electrical conductivity.However,they still suffer from volume distortion,structural collapse,and ionic conduction interruption upon cycling.Herein,a hierarchical array-like nanofiber structure was designed to address these limitations by combining architecture engineering and anion tuning strategy,in which SbPO_(4-x) with oxygen vacancy nanosheet arrays are anchored on the surface of interwoven carbon nanofibers(SbPO_(4-x)@CNFs).In particular,bulky PO_(4)^(3-) anions mitigate the large volume distortion and generate Na_(3)PO_(4) with high ionic conductivity,collectively improving cyclic stability and ionic transport efficiency.The abundant oxygen vacancies substantially boost the intrinsic electronic conductivity of SbPO_4,further accelerating the reaction dynamics.In addition,hierarchical fibrous structures provide abundant active sites,construct efficient conducting networks,and enhance the electron/ion transport capacity.Benefiting from the advanced structural design,the SbPO_(4-x)@CNFs electrodes exhibit outstanding cycling stability(1000 cycles at 1.0 A g^(-1) with capacity decay of 0.05% per cycle) and rapid sodium storage performance(293.8 mA h g^(-1) at 5.0 A g^(-1)).Importantly,systematic in-/ex-situ techniques have revealed the "multi-step conversion-alloying" reaction process and the "battery-capacitor dual-mode" sodium-storage mechanism.This work provides valuable insights into the design of anode materials for advanced SIBs with elevated stability and superior rate performance.

Efficient multi-scale community search method based on spectral graph wavelet

Community search is an important problem in network analysis,which has attracted much attention in recent years.As a query-oriented variant of community detection problem,community search starts with some given nodes,pays more attention to local network structures,and gets personalized resultant communities quickly.The existing community search method typically returns a single target community containing query nodes by default.This is a strict requirement and does not allow much flexibility.In many realworld applications,however,query nodes are expected to be located in multiple communities with different semantics.To address this limitation of existing methods,an efficient spectral-based Multi-Scale Community Search method(MSCS)is proposed,which can simultaneously identify the multi-scale target local communities to which query node belong.In MSCS,each node is equipped with a graph Fourier multiplier operator.The access of the graph Fourier multiplier operator helps nodes to obtain feature representations at various community scales.In addition,an efficient algorithm is proposed for avoiding the large number of matrix operations due to spectral methods.Comprehensive experimental evaluations on a variety of real-world datasets demonstrate the effectiveness and efficiency of the proposed method.

IL-13/IL-13RA2 signaling promotes colorectal cancer stem cell tumorigenesis by inducing ubiquitinated degradation of p53

Cancer stem cells(CSCs)are considered tumor-initiating cells and the main drivers of disease progression.Targeting these rare cancer cells,however,remains challenging with respect to therapeutic benefit.Here,we report the up-regulation of IL-13RA2 expression in colorectal cancer(CRC)tissues and spheroid cells.The expression of IL-13RA2 was positively correlated with canonical stemness markers in CRC.We further demonstrated that the level of IL-13 was up-regulated in the serum of CRC patients.Biologically,recombinant IL-13(rIL13)stimulation promoted the sphere formation,proliferation,and migration of CRC cells in vitro and enhanced tumorigenesis in vivo.This phenotype could be reversed by knocking down IL-13RA2.Mechanistically,IL-13 activated autophagy by inducing LC3I/LC3II transformation in CRC-CSCs,which was crucial for the biological functions of IL-13.We further demonstrated that IL-13RA2 acted as a modular link of the E3 ligase UBE3C and the substrate p53 protein,enhancing the interaction of UBE3C and p53,thereby inducing the K48-linked ubiquitination of p53.In conclusion,the IL-13/IL-13RA2 signaling cascade promotes CRC-CSC self-renewal and tumorigenesis by inducing p53 ubiquitination,adding an important layer to the connection between IL-13 and p53,which can be translated into novel targeted therapies.

Effect of blank quenching on shear spinning forming precision of 2219 aluminum alloy complex thin-walled components

The quenching-spinning(Q-S)process,i.e.,shear spinning after blank quenching,has been increasingly utilized to form 2219 aluminum alloy complex thin-walled components.However,the changes in material property,shape and stress of the blanks after quenching will affect the spin-ning forming precision.In this study,the rules and mechanisms of these effects are investigated based on a combined finite element(FE)model including blank quenching and component spinning process.The results indicate that the increase of material strength and the existence of distortion of the quenched blank lead to a notable increase in the non-uniformity of the circumferential compres-sive stress in the spinning area and the increase of the flange swing height during spinning.These changes result in an increase in the wall thickness and component-mandrel gap of the components.The quenching residual stress has little effect on wall thickness and roundness but can noticeably reduce the component-mandrel gap.This is because that the existence of quenching residual stress of the blank can lead to the decrease of the maximum circumferential compressive stress of the workpiece in spinning and an obvious drop in the maximum compressive stress after reaching the stress peak.Quenching distortion is the main factor affecting the roundness.Moreover,the opti-mized installation way of the blank for spinning is obtained.

Optical remote sensing image characteristics of large amplitude convex mode-2 internal solitary waves:an experimental study

A series of experiments are designed to propose a new method to study the characteristics of convex mode-2internal solitary waves(ISWs)in optical remote sensing images using a laboratory-based optical remote sensing simulation platform.The corresponding wave parameters of large-amplitude convex mode-2 ISWs under smooth surfaces are investigated along with the optical remote sensing characteristic parameters.The mode-2 ISWs in the experimentally obtained optical remote sensing image are produced by their overall modulation effect on the water surface,and the extreme points of the gray value of the profile curve of bright-dark stripes appear at the same location as the real optical remote sensing image.The present data extend to a larger range than previous studies,and for the characteristics of large amplitude convex mode-2 ISWs,the experimental results show a second-order dependence of wavelength on amplitude.There is a close relationship between optical remote sensing characteristic parameters and wave parameters of mode-2 ISWs,in which there is a positive linear relationship between the bright-dark spacing and wavelength and a nonlinear relationship with the amplitude,especially when the amplitude is very large,there is a significant increase in bright-dark spacing.
