Quality evaluation of lightweight cellular concrete by an ultrasound-based method

The accuracy of subgrade quality evaluation is important for road safety assessment.Since there is little research work devoted to testing lightweight cellular concrete(LCC)by an ultrasound-based method,the quantitative relation between ultrasonic testing results and the quality of LCC subgrade is not well understood.In this paper,the quality of LCC subgrade was evaluated with respect to compressive strength and crack discrimination.The relation between ultrasonic testing results and LCC quality was explored through indoor tests.Based on the quantitative relation between ultrasonic pulse velocity and compressive strength of LCC,a fitting formula was established.Moreover,after the LCC became cracked,the ultrasonic pulse velocity and ultrasonic pulse amplitude decreased.After determining the lower limiting values of the ultrasonic pulse velocity and ultrasonic pulse amplitude through the statistical data,it could be calculated whether there were cracks in LCC subgrade.The ultrasonic testing results showed that the compressive strength of the LCC subgrade was suitable for purpose and there was no crack in the subgrade.Then core samples were taken from the subgrade.Comparisons between ultrasonic testing results of subgrade and test results of core samples demonstrated a good agreement.

During the construction of lightweight cellular concrete(LCC),material damage frequently occurs,causing the degradation and deterioration of the mechanical performance,durability,and subgrade quality of LCC.The construction-induced damage can be more significant than those from the service environment of LCC,such as freeze-thaw(F-T)action in cold regions.However,the effect of construction-induced damage on LCC during F-T cycles is often ignored and the deterioration mechanisms are not yet clarified.In this study,we investigated the factors causing damage during construction using a sample preparation method established to simulate the damage in the laboratory setting.We conducted F-T cycle tests and microstructural characterization to study the effect of microstructural damage on the overall strength of LCC with different water contents under F-T actions.We established the relationship between the pore-area ratio and F-T cycle times,pore-area ratio,and strength,as well as the F-T cycle times and strength under different damage forms.The damage evolution is provided with the rationality of the damage equation,verified by comparing the measured and predicted damage variables.This study would serve as a guide for the construction and performance of LCC in cold regions.

激光夜视图像分辨率较低,边缘信息难以检测,导致图像分割过程存在效率低、精度较差等问题,为此设计基于Roberts算子的激光夜视图像自动分割方法。使用相机采集夜间场景色彩信息,采用加权平均法重构图像数据,输出完整的激光夜视图像。拟合图像周边相邻像素,利用三次样条插值函数平滑处理图像,运用Roberts算子计算图像梯度,实现激光夜视图像边缘检测,得到图像插值后的平滑信息,提升夜视图像分辨率。通过模糊C均值聚类明确聚类范围,引入混沌粒子群算法实现图像分割。实验结果表明,所提方法分割差异率在3%以下,图像分割Dice相似性系数在0.97以上,图像分割平均耗时为9 s。

为充分利用大量未标注样本、节约人力与时间,提出了基于无监督学习的抽油机井示功图自动聚类与批量标注方法。首先,将抽油机驴头往复运动产生的位移、载荷数据转化为示功图图片样本,其中,示功图的横坐标为位移,纵坐标为载荷;其次,加载在ImageNet上训练过的带有一系列权重参数、具有强特征提取能力的卷积神经网络模型;然后,去除该网络模型的全连接层,利用该网络模型提取示功图图片样本的特征;最后,利用k-means聚类算法对提取到的特征进行聚类分析,将具有相似特征的示功图聚到同一文件夹中。批量的对示功图聚类结果进行快速标注,从而形成抽油机井故障诊断的示功图样本集。实验随机搜集了100口抽油机井的20 000条示功图数据,结果表明,基于无监督学习的抽油机井示功图自动聚类与批量标注方法耗时短、准确率高,为示功图样本集标注提供了一种高效方法,对于充分挖掘油田大数据的应用价值具有示范意义。

Objective: Recent evidence indicates that the increased expression of FOXG1 is associated with tumor genesis. This study was designed to explore the expression and role which FOXG1 plays in human glioma. Methods: We detected the expression of FOXG1 by immunohistochemistry in glioma tissue samples. Following the down-regulation of FOXG1 in glioma cell lines by a specific short hairpin RNA, the function of FOXG1 in proliferation and apoptosis was assessed. Results: Glioma tissues exhibited notably higher expression of FOXG1 compared with control brain tissues and was positively correlated with histological malignancy. The down-regulation of FOXG1 in glioma cells led to a cell apoptosis in vitro. Conclusion: The overexpression of FOXG1 is a novel glioma malignancy marker, and FOXG1 may be used as a new target in therapeutic strategies for human glioma.

目的探究国产碟式离心机在规模生产中酵母发酵液菌体分离纯化工艺中的效果,摸索国产碟式离心机对酵母发酵液菌体分离纯化的适用性。方法检测离心过程中上清液A_(600 nm)值,判断离心工艺分离的效果;检测离心后菌体固含量,评价离心效果是否满足菌体分离的要求;通过计算最终菌体回收率来判断在国产碟式离心机离心过程中菌体可能的损耗;对比国产碟式离心机和大容量离心机离心工艺时间,以菌体固含量和菌体形态检测等指标评价国产碟式离心机在生产规模中的适用性;对比国产碟式离心机离心前后酵母形态的完整性,评价碟式离心机剪切力对酵母分离纯化的影响。结果国产碟式离心机和大容量离心机离心后酵母发酵液的上清液A_(600 nm)值差异无统计学意义(P=0.053 3,P>0.05);国产碟式离心机离心后菌体固含量均>80%,接近大容量离心机离心后菌体固含量;国产碟式离心机和大容量离心机离心后菌体回收率差异也无统计学意义(P=0.066 4,P>0.05);规模生产中相同体积的酵母发酵液用国产碟式离心机比大容量离心机能够大幅度缩短离心工艺时间,差异有统计学意义(P<0.000 1);国产碟式离心机离心前后酵母形态的完整性结果显示,离心剪切力对酵母几乎没有影响。结论在酵母发酵液菌体分离中,国产碟式离心机与大容量离心机离心效果接近,但离心纯化时间较短,在规模生产时较适用于本研究中酵母发酵液菌体分离的应用,后续可用于其他规模化生产工艺研究及应用。

目的评价150型AH100-ATS高压均质机和40型AH100-ATS高压均质机在汉逊酵母细胞破碎中的适用性。方法用2种AH100-ATS高压均质机分别重复破碎3组汉逊酵母,每组细胞破碎3次,光学显微镜下观察并比较2种高压均质机破碎汉逊酵母细胞前后的形态;SDS-PAGE电泳检测破碎后细胞中目的蛋白的释出;血球计数板计数且计算破碎后的细胞破碎率;Bradford法检测及计算蛋白释出度;计时破碎所需要的时间。结果光学显微镜下,2种高压均质机破碎汉逊酵母细胞前后的细胞形态均可见明显差别;150型AH100-ATS高压均质机破碎汉逊酵母细胞后释出蛋白的粒径分布体积比高于40型AH100-ATS高压均质机,差异有统计学意义(P<0.05);SDS-PAGE电泳结果显示,150型AH100-ATS高压均质机破碎汉逊酵母后目的蛋白的灰度值高于40型AH100-ATS高压均质机,差异有统计学意义(P<0.05)。150型AH100-ATS高压均质机破碎汉逊酵母细胞3次后,细胞破碎率分别为24.16%、35.63%、48.67%,总细胞破碎率为65.70%;蛋白释出度分别为41.69%、38.29%、15.80%,破碎完成后总蛋白释出度为62.63%;平均使用时间分别为16.7、16.7、19.0 min,破碎完成后总时间为52.3 min。40型AH100-ATS高压均质机破碎破碎汉逊酵母细胞后细胞破碎率分别为16.10%、11.40%、28.78%,总细胞破碎率为58.41%;蛋白释出度分别为24.72%、21.04%、9.57%,破碎完成后总蛋白释出度为55.48%;平均使用时间分别为61.0、59.7、60.7 min,破碎完成后总时间为181.3 min。每次破碎的细胞破碎率、蛋白释出度、所需时间,以及总细胞破碎率、总蛋白释出度、总破碎时间,2种高压均质机差异均有统计学意义(P均<0.05)。结论 150型AH100-ATS破碎汉逊酵母的释出蛋白的粒径分布体积比、目的蛋白、细胞破碎率和蛋白释出度均高于40型AH100-ATS,150型AH100-ATS破碎所需的时间远少于40型AH100-ATS。150型AH100-ATS高压均质机可以用于中试大规模研究中汉逊酵母细胞的破碎,且较40型AH100B-ATS高压均质机破碎需要更短的时间,且具有更好的破碎效果。

The intimate host-anion interactions will regulate thermodynamics and kinetics in the self-assembly of cationic cages mimicking biological counterparts.Herein,we report construction and transformation of three Pd(Ⅱ)-based metal-organic cages(MOCs)depending on different anions.Stoichiometric conversions of the lantern-shaped MOC-34 into either octahedral MOC-35 or tricapped trigonal prism MOC-36 are induced by BF_(4)^(–)or NO_(3)^(–),respectively.MOC-36 is kinetically favored and can undergo quantitative conversion to the thermodynamically preferred MOC-35 upon heating,accelerated by excess BF_(4)^(–)to motivate dissociative dynamics of Pd-vertices and lower activation barrier of cage transformation.The guest encapsulation behaviors of MOC-35 and MOC-36 have also been tested.These results manifest a significance of host-anion dynamics beyond complementary anion template,shedding light on the understanding of intricate anion recognition in nature.
