Combining metal-microbe and microbe-microbe dual direct electron transfer on Fe(0)-cathode of bio-electrochemical system to enhance anaerobic digestion of cellulose wastewater

Considering that cathode of microbial electrochemical system(MES)is a good electrons source for methane production via direct/indirect electron transfer to electroactive microorganisms,and that Fe(0)is also a confirmed electron donor for some electroactive microorganisms through metal-microbe direct electron transfer(DET),Fe(0)-cathode was equipped into an MES digester to enhance cathodic methane production.The results of this study indicated that the potential DET participator,Clostridium possibly obtained electrons directly from Fe(0)-cathode via metal-microbe electrons transfer,then transferred electrons directly to the definite DET participators,Methanosarcina/Methanothrix via microbemicrobe electrons transfer for CH_(4)production.In addition,Methanobacterium is another specially enriched methanogen on Fe(0)-cathode,which might obtain electrons directly from Fe(0)-cathode to produce CH_(4) via metal/electrode-microbe DET.The increment of conductivity of cathodic sludge in Fe(0)-cathode MES digester(R1)further confirmed the enrichment of electroactive microorganisms participating in DET process.As a consequence,a higher CH_(4) production(1205–1508 m L/d)and chemical oxygen demand(COD)removal(79.0%-93.8%)were achieved in R1 compared with graphite-cathode MES digester(R2,720–1090 m L/d and 63.6%-85.6%)and the conventional anaerobic digester(R3,384–428 m L/d and 35.2%-41.0%).In addition,energy efficiency calculated indicated that the output energy of CH_(4) production was 8.16 folds of electricity input in Fe(0)-cathode MES digester.

On tuning of ADRC with competing design indices:a quantitative study

Controller optimization has mostly been done by minimizing a certain single cost function.In practice,however,engineers must contend with multiple and conflicting considerations,denoted as design indices(DIs)in this paper.Failure to account for such complexity and nuances is detrimental to the applications of any advanced control methods.This paper addresses this challenge heads on,in the context of active disturbance rejection controller(ADRC)and with four competing DIs:stability margins,tracking,disturbance rejection,and noise suppression.To this end,the lower bound for the bandwidth of the extended state observer is first established for guaranteed closed-loop stability.Then,one by one,the mathematical formula is meticulously derived,connecting each DI to the set of controller parameters.To our best knowledge,this has not been done in the context of ADRC.Such formulas allow engineers to see quantitatively how the change of each tuning parameter would impact all of the DIs,thus making the guesswork obsolete.An example is given to show how such analytical methods can help engineers quickly determine controller parameters in a practical scenario.

Salvianolic acid B promotes the invasion and migration of H_(2)O_(2)-induced HTR-8/Svneo trophoblast cells by upregulating matrix metalloproteinase-9 via the phosphatidylinositol-4,5-bisphosphate 3-kinase/protein kinase B pathway

OBJECTIVE:To elucidate the regulatory effects of salvianolic acid B(Sal B)on trophoblast cells in preeclampsia(PE).METHODS:3-(4,5-dimethylthiazol-2-yl)-2,5 diphenyltetrazolium bromide(MTT)assays were used to detect the viability of human extravillous trophoblast HTR-8/Svneo cells induced by H_(2)O_(2)following treatment with different concentrations of Sal B.The levels of oxidative stressrelated molecules,including superoxide dismutase,glutathione-Px and malondialdehyde were detected using corresponding kits.Cell apoptosis was detected using a Terminal deoxynucleotidyl transferase(Td T)d UTP NickEnd Labeling(TUNEL)assay,and the expression of apoptosis-related proteins was detected using western blot analysis.In the present study,wound healing and Transwell assays were performed to measure the levels of cell invasion and migration.Western blot analysis was also used to detect the expression levels of epithelialmesenchymal transition-related proteins.The mechanisms underlying Sal B were further investigated using reverse transcription-quantitative real-time polymerase chain reaction(RT-q PCR)and western blot analysis,to determine the expression levels of matrix metallopeptidase 9(MMP-9)and phosphatidylinositol-4,5-bisphosphate 3-kinase(PI3K)/protein kinase B(Akt).RESULTS:Sal B increased the activity of HTR-8/Svneo cells,inhibited oxidative damage and promoted the invasion and migration of trophoblast cells induced by H_(2)O_(2).Furthermore,the expression levels of MMP-9 and members of the PI3K/Akt signaling pathway were significantly decreased.The pathway agonist,LY294002,and MMP-9 inhibitor,GM6001,reversed the effects of Sal B on H_(2)O_(2)-induced cells.CONCLUSIONS:Sal B promoted the invasion and migration of H_(2)O_(2)-induced HTR-8/Svneo trophoblast cells by upregulating MMP-9 via the PI3K/Akt signaling pathway.

A strategy for enhancing anaerobic digestion of waste activated sludge: Driving anodic oxidation by adding nitrate into microbial electrolysis cell

Cathodic reduction of CO_2 and anodic oxidation of organic matters are crucial to methaneproducing microbial electrolysis cell(MEC) applied in anaerobic digestion of waste activated sludge. However, cathodic CO_2 reduction is usually restrained by slow metabolism rates of H_2-utilizing methanogens and low electron-capturing capacity of CO_2, which consequently slows down the anodic oxidation that participates to sludge disintegration. Herein, a strategy with adding nitrate as electron acceptor to foster electronic transfer between the anode and cathode was proposed to improve anodic oxidation. Results showed that the average efficiency of anodic oxidation in the nitrate-added MEC increased by 55.9%. Accordingly,volatile suspended solid removal efficiency in the nitrate-added MEC was 21.9% higher than that of control MEC. Although the initial cumulative methane production in the nitrateadded MEC was lower than that of control MEC, the cumulative methane production in 24 days was 8.9% higher. Fourier transform infrared spectroscopy analysis indicated that anodic oxidation of MEC with nitrate accelerated the disintegration of sludge flocs and cell walls. Calculation on current signal further revealed that anodic oxidation driven by cathodic nitrate reduction was the main mechanism responsible for the improved sludge digestion.

As a promising new micro-physiological system,organ-on-a-chip has been widely utilized for in vitro pharmaceutical study and tissues engineering based on the three-dimensional constructions of tissues/organs and delicate replication of in vivo-like microenvironment.To better observe the biological processes,a variety of sensors have been integrated to realize in-situ,realtime,and sensitive monitoring of critical signals for organs development and disease modeling.Herein,we discuss the recent research advances made with respect to sensors-integrated organ-on-a-chip in this overall review.Firstly,we briefly explore the underlying fabrication procedures of sensors within microfluidic platforms and several classifications of sensory principles.Then,emphasis is put on the highlighted applications of different types of organ-on-a-chip incorporated with various sensors.Last but not least,perspective on the remaining challenges and future development of sensors-integrated organ-on-a-chip are presented.

Effect of layered joints on rockburst in deep tunnels

The existence of joints in the surrounding rock mass has a considerable efect on tunnel rockbursts.Herein,we studied the efect of layered joints with diferent inclination angles and spacings on rockburst in deep tunnels and investigated the failure area,deformation process of the surrounding rock mass,stress change inside the surrounding rock mass,velocity of the failed rock,and the kinetic energy of the failure.The failure type of the surrounding rock mass can thus be determined.The results showed that the intensity of rockburst increases as rock quality designation(RQD)decreases,while the deformation rate of the surrounding rock mass frst increases and then decreases.The deformation rate exhibits a turning point between RQD=50 and 70,below which the deformation rate of the surrounding rock mass gradually decreases,ultimately ceasing to be a rockburst.Rockburst always occurs perpendicular to the direction of the joint.Whenσ_(x)=σ_(y),as the joint inclination angle changes from 45°to 90°,the intensity of a rockburst frst decreases(from 45°to 60°),and then increases(from 60°to 90°).When combined with the evolution law of stress and strain energy,the rockburst process can be divided into four stages.

Analytical solutions for characteristic radii of circular roadway surrounding rock plastic zone and their application

In the non-uniform stress field, the surrounding rock plastic zone of the circular roadway shows different shapes under the different confining pressure conditions. Based on the boundary shape characteristics of the plastic zone, the characteristic radii of the plastic zone were proposed, namely the horizontal,longitudinal and medial axis radii, which could reflect the plastic zone shapes characteristics and classify the sizes of the key parts. On the theoretical basis of elastic-plastic mechanics, analytical solutions for the characteristic radii were obtained by theoretical deduction, and the relationships between the characteristic radii and key influencing factors were analyzed. Finally, the evaluation criterion of the circular roadway surrounding rock plastic zone shapes, evaluation criterion of the location of potential hazards caused by the roadway surrounding rock and evaluation critical points of roadway dynamic disasters based on characteristic radii were proposed. This work could provide a theoretical basis for stability analysis of the surrounding rock, support design, and guide the prevention and control of dynamic roadway disasters.

1 Introduction The main idea of recommender system is how to learn accurate users’embeddings from behavior data[1].Each dimension of users’embeddings can reflect the interests of users in different potential aspects.In real-world scenarios,users’interests are drifting over time,which brings a challenge to learn accurate dynamic users’embeddings.Recently,various time-aware recommendation methods have been proposed to tackle this problem by modeling the evolution process of users’interests[2−4].However,they usually assume that users’embeddings drift with the same range on all dimensions.In practice,users’embeddings should change diversely on different dimensions over time.Specifically,for the rapidly changing interests of the users,the corresponding dimensions should change significantly.On the contrary,the dimensions representing stable interests may change slightly.

Correlations between phytohemagglutinin response and leukocyte profile,and bactericidal capacity in a wild rodent

phytohemagglutinin(PHA)-induced swelling is widely used to investigate cell-mediated and innate immunity across different vertebrate taxa.However,its physiological mechanism is still an open question due to the complexity of the involved immune components.In the present study,we measured the synchronous variations of PHA response,the proportion of different subtypes of leukocytes,as well as serum bactericidal capacity in circulation blood at 6,12 and 24 h after PHA versus PBS injection in striped hamster,Cricetulus barabensis.First,the results showed that PHA responses reached a peak at 6 h postinjection,then sharply declined at 12 h and 24 h postinjection.Serum bactericidal capacity was higher at 6 h and 12 h than at 24 h.The proportion of different subtypes of leukocytes,as well as the ratio of neutrophils to lymphocytes did not display significant changes across different time points.Second,PHA response was positively correlated with the proportion of neutrophils and serum bactericidal capacity.The proportion of monocytes was negatively correlated with that of eosinophils and neutrophils.The proportion of basophils was negatively correlated with that of lymphocytes.Our results indicate that earlier enhanced PHA response is important for the striped hamster to cope with changing environmental conditions due to its small body mass,and the increased components of innate immunity in circulation blood may contribute to the enhancement of PHA swelling response.

Role of 5-hydroxytryptamine type 3 receptors in the regulation of anxiety reactions

5-Hydroxytryptamine(5-HT)type 3 receptor(5-HT_(3)R)is the only type of ligand-gated ion channel in the 5-HT receptor family.Through the high permeability of Na+,K+,and Ca2+and activation of subsequent voltage-gated calcium channels(VGCCs),5-HT_(3)R induces a rapid increase of neuronal excitability or the release of neurotransmitters from axon terminals in the central nervous system(CNS).5-HT_(3)Rs are widely expressed in the medial prefrontal cortex(mPFC),amygdala(AMYG),hippocampus(HIP),periaqueductal gray(PAG),and other brain regions closely associated with anxiety reactions.They have a bidirectional regulatory effect on anxiety reactions by acting on different types of cells in different brain regions.5-HT_(3)Rs mediate the activation of the cholecystokinin(CCK)system in the AMYG,and theγ-aminobutyric acid(GABA)“disinhibition”mechanism in the prelimbic area of the mPFC promotes anxiety by the activation of GABAergic intermediate inhibitory neurons(IINs).In contrast,a 5-HT_(3)R-induced GABA“disinhibition”mechanism in the infralimbic area of the mPFC and the ventral HIP produces anxiolytic effects.5-HT_(2)R-mediated regulation of anxiety reactions are also activated by 5-HT_(3)R-activated 5-HT release in the HIP and PAG.This provides a theoretical basis for the treatment of anxiety disorders or the production of anxiolytic drugs by targeting 5-HT_(3)Rs.However,given the circuit specific modulation of 5-HT_(3)Rs on emotion,systemic use of 5-HT_(3)R agonism or antagonism alone seems unlikely to remedy anxiety,which deeply hinders the current clinical application of 5-HT_(3)R drugs.Therefore,the exploitation of circuit targeting methods or a combined drug strategy might be a useful developmental approach in the future.
