Nausea and vomiting are important defensive responses to cope with pathogens and toxins that invade the body. The nucleus of the solitary tract(NTS) is important for initiating these responses. However, the molecular heterogeneities and cellular diversities of the NTS occlude a better understanding of these defensive responses. Here, we constructed the single-nucleus transcriptomic atlas of NTS cells and found multiple populations of NTS neurons that may be involved in these defensive responses. Among these, we identified Calbindin1-positive(Calb1+) NTS neurons that are molecularly distinct from Tac1+neurons.These Calb1+neurons are critical for nausea and retching induced by cereulide; an emetic toxin secreted by Bacillus Cereus. Strikingly, we found that cereulide can directly modulate vagal sensory neurons that innervate Calb1+NTS neurons, a novel mechanism distinct from that for nausea and retching induced by Staphylococcal enterotoxin A. Together, our transcriptomic atlas of NTS neurons and the functional analyses revealed the neural mechanism for cereulide-induced retching-like behavior. These results demonstrate the molecular and cellular complexities in the brain that underlie defensive responses to the diversities of pathogens and toxins.

Emerging and recurrent infectious diseases caused by human coronaviruses(HCoVs)continue to pose a significant threat to global public health security.In light of this ongoing threat,the development of a broad-spectrum drug to combat HCoVs is an urgently priority.Herein,we report a series of anti-pan-coronavirus ssDNA aptamers screened using Systematic Evolution of Ligands by Exponential Enrichment(SELEX).These aptamers have nanomolar affinity with the nucleocapsid protein(NP)of Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)and also show excellent binding efficiency to the N proteins of both SARS,MERS,HCoV-OC43 and-NL63 with affinity KD values of 1.31 to 135.36 nM.Such aptamer-based therapeutics exhibited potent antiviral activity against both the authentic SARS-CoV-2 prototype strain and the Omicron variant(BA.5)with EC50 values at 2.00 nM and 41.08 nM,respectively.The protein docking analysis also evidenced that these aptamers exhibit strong affinities for N proteins of pan-coronavirus and other HCoVs(−229E and-HKU1).In conclusion,we have identified six aptamers with a high pan-coronavirus antiviral activity,which could potentially serve as an effective strategy for preventing infections by unknown coronaviruses and addressing the ongoing global health threat.

Application of heat-activated peroxydisulfate process for the chemical cleaning of fouled ultrafiltration membranes

Na Cl O has been widely used to restore membrane flux in practical membrane cleaning processes,which would induce the formation of toxic halogenated byproducts.In this study,we proposed a novel heatactivated peroxydisulfate(heat/PDS)process to clean the membrane fouling derived from humic acid(HA).The results show that the combination of heat and PDS can achieve almost 100%recovery of permeate flux after soaking the HA-fouled membrane in 1 mmol/L PDS solution at 50℃ for 2 h,which is attributed to the changes of HA structure and enhanced detachment of foulants from membranes.The properties of different treated membranes are characterized by scanning electron microscopy(SEM),atomic force microscope(AFM),attenuated total reflection Fourier transform infrared spectroscopy(ATRFTIR),and X-ray photoelectron spectroscopy(XPS),demonstrating that the reversible and irreversible foulants could be effectively removed by heat/PDS cleaning.The filtration process and fouling mechanism of the cleaned membrane were close to that of the virgin membrane,illustrating the good reusability of the cleaned membrane.Additionally,heat/PDS which can avoid the generation of halogenated byproducts shows comparable performance to Na Cl O on membrane cleaning and high performance for the removal of fouling caused by sodium alginate(SA),HA-bovine serum albumin(BSA)-SA mixture and algae,further suggesting that heat/PDS would be a potential alternative for membrane cleaning in practical application.

本文用Lévy过程为保险公司的盈余水平建模.假设保险公司在固定的时间对盈余水平进行观测,并在每次观测后做出决策.如果观察到的盈余水平小于给定的临界水平,且非负时,不足的部分将被一次性注入,使得盈余水平恢复到临界水平.如果观察到的盈余水平为负,就立即宣布破产.我们利用傅里叶余弦级数展开方法,提出了计算破产前有限时间期望折现注资总成本和有限时间期望折现罚函数的数值方法.通过误差分析和数值实例,证明了该方法的准确性和有效性,并研究了各个参数对结果的影响.

为了探究有效扩大轴流泵稳定运行范围的方法,针对轴流泵进行自循环机匣处理的单通道定常数值模拟,分析周向覆盖比例、抽吸口位置和喉部高度对水泵扩稳性能的影响规律,并揭示自循环机匣对轴流泵扩稳的改善机理.结果表明,抽吸口位置保持不变,扩大周向覆盖比例和喉部高度都可以增大流量裕度,但设计点效率降低;喉部高度为1.8 mm的自循环机匣流量裕度随着抽吸口位置的后移先增加后降低,效率先降低后增加,说明在叶顶上方存在一个最佳位置.在研究的范围内,流量裕度最大达到5.93%,设计点的效率最低降低1.12%.通过自循环机匣处理,使叶顶泄漏流与主流在叶顶上方相互作用形成的堵塞区面积有所减小,从而可以扩大轴流泵流量范围和提升小流量工况效率.该研究可为轴流泵在自循环机匣的端壁处理方式下扩稳提供参考.

背景:有研究发现局部应用阿仑膦酸钠能够促进成骨,但少见其在牵张成骨过程中的尝试及探讨。目的:观察阿仑膦酸钠对快速兔下颌骨牵张成骨的促进作用并探讨其机制。方法:36只新西兰雄性白兔在经过3 d的滞留期后以1.5 mm/12 h(3 d)的牵张速率进行快速牵张,随后实验动物被随机分为A、B和C组,每组12只。在固定期第1,3和7天,A组动物牵张间隙注射200μg/kg阿仑膦酸钠,B组动物牵张间隙注射100μg/kg阿仑膦酸钠,C组动物作为对照组。在固定期第4,8周,进行CT和双能量X射线骨密度测量。在第4周完成核素扫描后处死部分动物收集标本进行Western Blotting及抗酒石酸酸性磷酸酶染色;在第8周处死剩余动物后进行三点弯曲力学试验。结果与结论:①CT结果发现B组兔牵张间隙新生骨质明显优于A、C组;②在第4周时B组的骨密度计数为(0.092±0.010)g/cm^(2),是A组的1.26倍(P<0.001)、C组的1.28倍(P<0.001);在第8周时B组的骨密度为(0.175±0.029)g/cm^(2),是A组的1.38倍(P<0.001)、C组的1.45倍(P<0.001);③抗酒石酸酸性磷酸酶染色发现C组切片破骨样细胞计数是A组的2.83倍(P<0.001)、B组的2.21倍(P<0.001);④C组牵张间隙核浓集强度高于A、B组;⑤Western Blotting结果发现B组成骨信号蛋白Runx2表达明显强于A、C组;⑥B组牵张间隙的最大力学负载(158.48±23.21)N是A组的1.26倍(P=0.007)、C组的1.31倍(P=0.003);⑦结果表明低浓度阿仑膦酸钠可能通过抑制破骨信号来促进兔下颌骨快速牵张成骨。

The historical logics and geographical patterns of rural-urban governance in China

The advent of a mobile society has led to profound changes in China’s traditional rural-urban pattern and called for new strategies for urban and rural governance.Based on a macro perspective of temporal and spatial evolution,this study analyzes the historical logics of rural-urban governance,explores the geographical patterns of challenges in China’s rural-urban governance,and finally puts forward targeted strategies for rural-urban governance toward integrated and sustainable development in China.From the historical perspective,the urban originates from the rural,and the connotation of rural,that is,a regional-scale outlook,rural-urban relations,and sociocultural interaction,forms the traditional approach to rural-urban governance.China’s rural-urban governance has evolved from antagonism toward integration.In terms of theoretical development,the turn toward mobility is an important driving factor shaping and promoting the transformation of research focus on rural-urban governance.The mobility of urban and rural factors has especially in recent decades brought extensive challenges for governance,which are highlighted by the disintegration of the stability of rural and urban structures,growing regional disparities in rural education levels,and serious aging and hollowing crises in rural areas.We therefore propose that rural-urban governance should give sufficient consideration to the convergence of wider interests;realize social potential through institutional,cultural and spatial restructuring;and attach importance to collaborative development and governance.And the transfer of rural problems to urban areas should be paid attention to by rural-urban governance.This research enriches the knowledge regarding the logics and patterns of China’s rural-urban governance from an interdisciplinary perspective.It is also helpful in the promotion of rural-urban integration and sustainable development,especially at a time when the mobility of social factors between rural areas and urban areas increasingly challenges tradit

为了改变当前实验室安全教育资源、内容、考核问题所导致的安全知识无法有效内化的现状,促进实验室安全教育效果提升,研究从记忆的原理出发,以建构主义、认知负荷、知识可视化等理论为指导,针对资源形式、内容组织、评价考核提出了系统性的解决方案,设计“通识—专业”模块化的安全教育体系,提出层级与网状相结合的安全教育知识地图构建方法,构建基于知识地图的问题导向式的安全教育微课,拓宽了安全教育理论范围。实践表明新体系可有效提升学生安全教育满意度及效果,具有一定的推广意义与实践价值。

传统的船闸金属结构二维设计具有绘图算量工作量大、设计周期长、专业协同局限、复杂结构绘图识图难等问题,运用BIM技术可统一金属结构设计路线,基于标准模板完成BIM模型创建、有限元计算、工程图纸导出等,实现金属结构BIM正向设计。BIM技术具有模块化设计、协同设计、参数化设计及可视化设计等特性,利用其开展船闸金属结构设计可缩短设计周期,推进产品标准化、系列化进程;促进专业间协作;实现参数的驱动修改与自动更新;精准表达、传递设计意图。实践表明,与传统的二维设计模式相比,运用BIM技术进行船闸金属结构设计能有效提升设计效率与质量,实现信息的高效传递与集成管理,优势显著。
