De novo assembly of a Tibetan genome and identification of novel structural variants associated with high-altitude adaptation

Structural variants(SVs) may play important roles in human adaptation to extreme environments such as high altitude but have been under-investigated. Here, combining long-read sequencing with multiple scaffolding techniques, we assembled a high-quality Tibetan genome(ZF1), with a contig N50 length of 24.57 mega-base pairs(Mb) and a scaffold N50 length of 58.80 Mb. The ZF1 assembly filled 80 remaining N-gaps(0.25 Mb in total length) in the reference human genome(GRCh38). Markedly, we detected17 900 SVs, among which the ZF1-specific SVs are enriched in GTPase activity that is required for activation of the hypoxic pathway. Further population analysis uncovered a 163-bp intronic deletion in the MKL1 gene showing large divergence between highland Tibetans and lowland Han Chinese. This deletion is significantly associated with lower systolic pulmonary arterial pressure, one of the key adaptive physiological traits in Tibetans. Moreover, with the use of the high-quality de novo assembly, we observed a much higher rate of genome-wide archaic hominid(Altai Neanderthal and Denisovan) shared non-reference sequences in ZF1(1.32%–1.53%) compared to other East Asian genomes(0.70%–0.98%),reflecting a unique genomic composition of Tibetans. One such archaic hominid shared sequence—a662-bp intronic insertion in the SCUBE2 gene—is enriched and associated with better lung function(the FEV1/FVC ratio) in Tibetans. Collectively, we generated the first high-resolution Tibetan reference genome, and the identified SVs may serve as valuable resources for future evolutionary and medical studies.

The maneuverability and stealth of aerial-aquatic vehicles(AAVs)is of significant importance for future integrated air-sea combat *** improve the maneuverability and stealth of AAVs near the water surface,this paper proposed a high-maneuverability skipping motion strategy for the tandem twin-rotor AAV,inspired by the motion behavior of the flying fish to avoid aquatic and aerial predators near the water *** novel tandem twin-rotor AAV was employed as the research subject and a strategybased ADRC control method for validation,comparing it with a strategy-based PID control *** results indicate that both control methods enable the designed AAV to achieve high stealth and maneuverability near the water surface with robust control *** strategy-based ADRC control method exhibits a certain advantage in controlling height,pitch angle,and reducing impact *** motion strategy will offer an inspiring approach for the practical application of AAVs to some extent.

Improving precision base editing of the zebrafish genome by Rad51DBD-incorporated single-base editors

A warpage-free Si3N4 slurry strategy for vat photopolymerization

Si3N4 ceramics are promising wave-transparent materials with excellent mechanical and dielectric properties. Vat photopolymerization (VPP) three-dimensional (3D) printing provides a strategy for preparing ceramics with controllable complex structures. However, the difficulty in solidifying the slurry due to partial ultraviolet (UV) light absorption and the high refractive index of Si3N4 particles during the VPP process severely hinder the molding of Si3N4 ceramics. A higher laser power must be used to increase the curing depth, which generates large internal stresses and warps the samples. This study presents a method to solve the warpage problem during VPP-3D printing using tributyl citrate as a plasticizer. The plasticizer can weaken the force between polymer molecular chains and reduce the internal stress of the green body. Warpage decreases gradually with increasing tributyl citrate content, and the warpage decreases to 0% when the plasticizer content reaches 30 wt% at high laser powers from 600 to 750 mW. Samples with different layer thicknesses were printed, and the optimum thickness of 40 mu m was obtained, at which the sintered Si3N4 samples possessed a unique combination of mechanical properties, including a bending strength of 338.29 +/- 12.08 MPa and a fracture toughness of 6.94 +/- 0.11 MPam1/2 for the loading direction perpendicular to the build surface and 5.37 +/- 0.99 MPam1/2 for the loading direction parallel to the build surface. The dielectric constant of all the samples is maintained in the range of 5.462-6.414. This work is expected to guide vat photopolymerization and the preparation of complex Si3N4 ceramic components.

碳化物超高温陶瓷具有高熔点(>3000℃)、高硬度、低热导率、优异的耐高温性和良好的化学稳定性等优点,是高超声速飞行器热防护系统的理想涂层材料。本文概述了碳化物超高温陶瓷(TiC、ZrC、HfC、NbC、TaC)的结构与性质,总结了化学气相沉积法、等离子喷涂法和固相反应法制备碳化物超高温陶瓷涂层的研究进展,分析了涂层微观结构、组分、结构设计以及热流密度对烧蚀行为的影响。研究表明,添加第二相形成多元复合涂层和采用多层结构设计,可以有效提升碳化物超高温陶瓷涂层的抗烧蚀性能。添加第二相形成复杂氧化物,可使烧蚀后的氧化层适度烧结,从而获得良好的结构完整性和阻氧性能。采用梯度分层和多层功能结构设计,有效缓解了涂层热应力,抑制了裂纹扩展,并促进了不同层间的协同增强作用。最后,结合研究现状,对碳化物超高温陶瓷抗烧蚀涂层发展面临的挑战与机遇进行了展望。

致密砂岩产水气井井下节流携液机理及预测新模型

致密砂岩气是目前中国开发规模最大的非常规天然气,其普遍采用井下节流工艺进行生产,但目前尚未认清产水后井下节流器对井筒携液的影响规律,导致气井积液加速,气井生命周期骤减,极大程度上制约了单井天然气估算最终采收率(EUR)的提升。为此,以国内典型致密砂岩产水气井为研究对象,基于气液两相井下节流可视化实验,分析了节流器上、下游气液两相流动和分布特征,揭示了井下节流气井携液机理,提出了节流器影响下的液滴夹带率计算新方法,最后建立了井下节流气井携液临界气流速预测机理模型。研究结果表明:(1)井下节流器下游管段中液膜反转是气井积液的起始点,当上游为搅动流时,节流器在一定程度上改善了下游压力的波动,对维持气井稳定生产起到了积极作用;(2)井下节流器下游额外液滴夹带率与面积比(嘴径/管径)成反比,临界携液流速随面积比的减小呈现线性递减特征;(3)建立的新模型预测精度(Erms=8.4,R2=0.94)远高于Wallis、Luo和Belfroid等模型经过现场35口实际井验证,积液误判率仅为8.6%。结论认为,建立的携液临界气流速新模型可以对井下节流的致密砂岩产水气井积液初始条件进行准确预测,并为排水采气工艺的介入时机提供了理论依据,同时对诊断气井积液状况、维持气井稳定生产、延长气井生命周期具有重要意义。

In order to characterize the plastic state of a deformed material, an indentation method to determine the plastic equation of state(PES) was developed. The work-hardening coefficient and the strain rate sensitivity coefficient of the plastic mechanic equation of state were determined by two kinds of indentation tests respectively. Therefore, the PES of materials under deformation can be obtained, and the plastic state of materials can be determined.

针对某企业发动机缸体表面缺陷检测的工业视觉应用中，由于缺陷样本数据集较少导致的识别准确率低，检测速度慢的问题，设计了一套基于深度学习的机器视觉表面缺陷检测系统。采用生成对抗网络pix2pix模型增强缺陷图片数据集，结合灰度增强、带通滤波等传统图像预处理方法突出表面缺陷特征，利用YOLOv5深度学习识别网络进行缺陷识别算法开发，并根据上述框架搭建了缸体表面缺陷检测软硬件系统。将所搭建的检测系统在企业生产线进行应用验证，结果显示，缺陷总体识别准确率高达98.4%，单张图片识别耗时小于0.5 s。所提方法解决了原有小样本数据集难以支撑深度学习训练的难题，提高了缸体表面缺陷的识别精度和识别效率，在工业检测的应用中具有巨大的潜力。

Prediction model for corrosion rate of low-alloy steels under atmospheric conditions using machine learning algorithms

This work constructed a machine learning(ML)model to predict the atmospheric corrosion rate of low-alloy steels(LAS).The material properties of LAS,environmental factors,and exposure time were used as the input,while the corrosion rate as the output.6 dif-ferent ML algorithms were used to construct the proposed *** optimization and filtering,the eXtreme gradient boosting(XG-Boost)model exhibited good corrosion rate prediction *** features of material properties were then transformed into atomic and physical features using the proposed property transformation approach,and the dominant descriptors that affected the corrosion rate were filtered using the recursive feature elimination(RFE)as well as XGBoost *** established ML models exhibited better predic-tion performance and generalization ability via property transformation *** addition,the SHapley additive exPlanations(SHAP)method was applied to analyze the relationship between the descriptors and corrosion *** results showed that the property transformation model could effectively help with analyzing the corrosion behavior,thereby significantly improving the generalization ability of corrosion rate prediction models.
