Ticks are external parasitic arthropods that can transmit a variety of pathogens by sucking ***-temperature tolerance is essential for ticks to survive during the cold *** the protein regulation mechanism of low-temperature tolerance of Haemaphysalis longicornis could help to explain how ticks survive in *** this study,the quantitative proteomics of several tissues of *** exposed to low temperature were studied by data independent acquisition *** of 3699,3422,and 1958 proteins were identified in the salivary gland,midgut,and ovary,*** proteins involved in energy metabolism,cell signal transduction,protein synthesis and repair,and cytoskeleton synthesis changed under low-temperature *** comprehensive analysis of the protein regulation of multiple tissues of female ticks exposed to low temperature showed that maintaining cell homeostasis,maintaining cell viability,and enhancing cell tolerance were the most important means for ticks to maintain vital signs under low *** expression of proteins involved in and regulating the above cell activities was the key to the survival of ticks under low *** the analysis of a large amount of data,we found that the expression levels of arylamine N-acetyltransferase,inositol polyphosphate multikinase,and dual-specificity phosphatase were up-regulated under low *** speculated that they might have important significance in low-temperature ***,we performed RNA interference on the mRNA of these 3 proteins,and the results showed that the ability of female ticks to tolerate low temperatures decreased significantly.

目的:探讨芹菜素通过调控窖蛋白-1(Caveolin-1)对慢性心力衰竭(CHF)大鼠血管紧张素1-7(Ang1-7)及左室舒张功能的机制研究。方法:按照随机数字表法将大鼠分为假手术(Sham)组、CHF组、芹菜素组、Caveolin-1组及联合组,每组10只,除Sham组外均建立CHF大鼠模型。建模后24 h,芹菜素组大鼠腹腔注射2 mg/kg的芹菜素,Caveolin-1组腹腔注射1.2 mg/kg的Caveolin-1质粒的脂质体溶液,联合组大鼠腹腔注射2 mg/kg的芹菜素及1.2 mg/kg的Caveolin-1质粒的脂质体溶液,Sham组及CHF组大鼠腹腔注射等体积生理盐水。比较各组大鼠心脏收缩及舒张功能指标、血清因子水平;苏木精-伊红(HE)染色检测心肌组织病理形态;电镜观察心房超微结构;免疫组化检测心肌组织血管生成情况;蛋白免疫印迹检测心肌组织Caveolin-1、缺氧诱导因子1(HIF-1)、血管内皮生长因子(VEGF)蛋白水平。结果:CHF组与Sham组比较,大鼠左心室射血分数(LVEF)、左心室短轴缩短率(LVFS)、Ang1-7及Caveolin-1蛋白、HIF-1蛋白、VEGF蛋白降低,左心室舒张末期内径(LVEDD)、左心室收缩末期内径(LVESD)、左心室舒张末期容积(LVEDV)、左心室收缩末期容积(LVESV)、血管紧张素Ⅱ(AngⅡ)、醛固酮(ALD)、肾素活性(PRA)升高,差异有统计学意义(P0.05);联合组与芹菜素组及Caveolin-1组各指标比较,差异均有统计学意义(P<0.05)。结论:芹菜素对慢性心力衰竭大鼠心脏舒张及收缩功能具有改善作用,并可通过上调Ang1-7改善心肌重塑从而保护心脏,其机制可能与激活Caveolin-1表达而增加心肌新生血管相关。

为探究凡纳滨对虾(Litopenaeus vannamei)“滴星病”的发病原因,从濒死的患病对虾肝胰腺中分离纯化得到一株优势菌MRY0520,并对菌株MRY0520进行了形态学观察和生理生化鉴定及分子生物学研究。结果表明:该菌株为美人鱼发光杆菌美人鱼亚种(Photobacterium damselae ***),人工回感健康的凡纳滨对虾可致其出现与自然发病虾相同的症状。该菌对凡纳滨对虾72 h的半数致死量LD50为2.15×10^(5)CFU∕g,药敏试验结果显示MRY0520对新霉素(30μg∕片)、多西环素(30μg∕片)等多种药物敏感。该菌对凡纳滨对虾具有致病性,可致对虾肝胰腺出现明显的病理变化,甚至死亡。

In this study,we identified that a conserved circular RNA(circRNA)DICAR,which was downregulated in diabetic mouse *** had an inhibitory effect on diabetic cardiomyopathy(DCM),as the spontaneous cardiac dysfunction,cardiac cell hypertrophy,and cardiac fibrosis occurred in DICAR deficiency(DICAR+/−)mice,whereas the DCM was alleviated in DICARoverexpressed DICARTg *** the cellular level,we found that overexpression of DICAR inhibited,but knockdown of DICAR enhanced the diabetic cardiomyocyte *** the molecular level,we identified that DICAR-VCP-Med12 degradation could be the underlying molecular mechanism in DICAR-mediated *** synthesized DICAR junction part(DICAR-JP)exhibited a similar effect to the entire *** addition,the expression of DICAR in circulating blood cells and plasma from diabetic patients was lower than that from health controls,which was consistent with the decreased DICAR expression in diabetic *** and the synthesized DICAR-JP may be drug candidates for DCM.

虾肝肠胞虫极管蛋白3的鉴定

为探明极管蛋白在EHP侵染宿主过程中的作用机制及虾肝肠胞虫病的预防和治疗提供新的靶标,通过克隆南美白对虾(Litopenaeus vannamei)肝肠胞虫(Enterocytozoon hepatopenaei,EHP)极管蛋白3基因(Polar tube protein 3,PTP3),并进行生物信息学分析,构建其核心区域部分序列编码的蛋白原核表达载体,诱导表达、纯化,制备多克隆抗体。结果表明,PTP3中最长的开放阅读框片段长3 390 bp,编码1 130个氨基酸,预测蛋白质分子质量为125 k D。该蛋白拥有13个N-糖基化位点和多个O-糖基化位点,未发现信号肽结构域;拥有117个磷酸化位点,其中苏氨酸磷酸化位点最多,为71个;二级结构较为简单,α螺旋占41.77%,是最大量的结构元件;延伸链占12.48%,无规卷曲占39.29%,β转角占6.46%。通过免疫印迹试验验证多抗的特异性,用免疫荧光实验观察极管蛋白在极管弹射时的状态,表明EHPPTP3分布于虾肝肠胞虫弹出的极管上,是新鉴定出的虾肝肠胞虫极管蛋白,确认了EHPPTP3的存在。通过生物信息学分析、序列特征、蛋白表达特征、多抗特异性以及定位特征证实本研究克隆的PTP3即为EHP极管蛋白3(EHPPTP3),是虾肝肠胞虫中新鉴定出的一种极管蛋白。

Larch caterpillars are widely distributed in the Great Xing’an boreal forests;however,the relationship between caterpillar defoliation dynamics and climatic factors is poorly *** aims of this study are to investigate the primary weather conditions that might influence forest defoliation and to identify the most important life stage of the larch caterpillar at which forest defoliation might be mitigated by incorporating more inhibitory influences from climatic *** life cycle of the larch caterpillar was partitioned into four stages and multiple linear regression and mixed effect models were combined with a relative weight analysis approach to evaluate the importance and influence of meteorological variables on defoliation *** results show that warmer temperatures in growing seasons and overwintering periods can increase the defoliation area,while rainy and humid growing seasons decrease the defoliation *** precipitation during the early instar larval period had the greatest power to explain the variance in defoliation dynamics and had a very strong inhibitory effect,followed by the accumulative temperatures of the late instar larval period which had a positive impact,and precipitation during the middle instar larval period which had a negative *** conditions during the early instar larval period had the greatest influence on the area defoliated and accounted for 40%of the explained *** study demonstrates that climatic warming and drying will increase the risk of larch caterpillar outbreaks in the Great Xing’an Mountains.

In this study,perovskite-type La_(0.7)Ca_(0.3)Co_(0.3)Fe_(0.6)M_(0.1)O_(3-δ)(M=Cu,Zn)powders were synthesized using a scalable reverse co-precipitation method,presenting them as novel materials for oxygen transport *** comprehensive study covered various aspects including oxygen permeability,crystal structure,conductivity,morphology,CO_(2) tolerance,and long-term regenerative durability with a focus on phase structure and *** membrane La_(0.7)Ca_(0.3)Co_(0.3)Fe_(0.6)M_(0.1)O_(3-δ)exhibited high oxygen permeation fluxes,reaching up to 0.88 and 0.64 mL·min^(−1)·cm^(−2) under air/He and air/CO_(2) gradients at 1173 K,*** 1600 h of CO_(2) exposure,the perovskite structure remained intact,showcasing superior CO_(2) resistance.A combination of first principles simulations and experimental measurements was employed to deepen the understanding of Cu/Zn substitution effects on the structure,oxygen vacancy formation,and transport behavior of the *** findings underscore the potential of this highly CO_(2)-tolerant membrane for applications in high-temperature oxygen *** enhanced insights into the oxygen transport mechanism contribute to the advancement of next-generation membrane materials.

No well-established biomarkers are available for the clinical diagnosis of major depressive disorder(MDD).Vitamin D-binding protein(VDBP)is altered in plasma and postmortem dorsolateral prefrontal cortex(DLPFC)tissues of MDD ***,the role of VDBP as a potential biomarker of MDD diagnosis was further *** extracellular vesicles(EVs)and brain cell-derived EVs(BCDEVs)were isolated from the plasma of first-episode drug-naïve or drug-free MDD patients and well-matched healthy controls(HCs)in discovery(20 MDD patients and 20 HCs)and validation cohorts(88 MDD patients and 38 HCs).VDBP level in the cerebrospinal fluid(CSF)from chronic glucocorticoid-induced depressed rhesus macaques or prelimbic cortex from lipopolysaccharide(LPS)-induced depressed mice and wild control groups was measured to evaluate its relationship with VDBP in plasma microglia-derived extracellular vesicles(MDEVs).VDBP was significantly decreased in MDD plasma MDEVs compared to HCs,and negatively correlated with HAMD-24 score with the highest diagnostic accuracy among *** in plasma MDEVs was decreased both in depressed rhesus macaques and mice.A positive correlation of VDBP in MDEVs with that in CSF was detected in depressed rhesus *** levels in prelimbic cortex microglia were negatively correlated with those in plasma MDEVs in depressed *** main results suggested that VDBP in plasma MDEVs might serve as a prospective candidate biomarker for MDD diagnosis.
