COVAD: Content-oriented video anomaly detection using a self attention-based deep learning model

Background Video anomaly detection has always been a hot topic and has attracted increasing attention.Many of the existing methods for video anomaly detection depend on processing the entire video rather than considering only the significant context. Method This paper proposes a novel video anomaly detection method called COVAD that mainly focuses on the region of interest in the video instead of the entire video. Our proposed COVAD method is based on an autoencoded convolutional neural network and a coordinated attention mechanism,which can effectively capture meaningful objects in the video and dependencies among different objects. Relying on the existing memory-guided video frame prediction network, our algorithm can significantly predict the future motion and appearance of objects in a video more effectively. Result The proposed algorithm obtained better experimental results on multiple datasets and outperformed the baseline models considered in our analysis. Simultaneously, we provide an improved visual test that can provide pixel-level anomaly explanations.

Deconvolution of potential drug targets of the central nervous system(CNS)is particularly challenging because of the complicated structure and function of the brain.Here,a spatiotemporally resolved metabolomics and isotope tracing strategy was proposed and demonstrated to be powerful for deconvoluting and localizing potential targets of CNS drugs by using ambient mass spectrometry imaging.This strategy can map various substances including exogenous drugs,isotopically labeled metabolites,and various types of endogenous metabolites in the brain tissue sections to illustrate their microregional distribution pattern in the brain and locate drug action-related metabolic nodes and pathways.The strategy revealed that the sedative-hypnotic drug candidate YZG-331 was prominently distributed in the pineal gland and entered the thalamus and hypothalamus in relatively small amounts,and can increase glutamate decarboxylase activity to elevateγ-aminobutyric acid(GABA)levels in the hypothalamus,agonize organic cation transporter 3 to release extracellular histamine into peripheral circulation.These findings emphasize the promising capability of spatiotemporally resolved metabolomics and isotope tracing to help elucidate the multiple targets and the mechanisms of action of CNS drugs.

The only joint effort area of provincial and municipal governments resides in Guangdong Province and Shenzhen City in China's carbon emission trading system(ETS) pilots,which characterize the national carbon ETS plots.The present study on the operating experience from this area has important reference value for the national carbon ETS.Analysis and comparison of the key elements show many differences in coverage,total allowance,allowance allocation,and MRV mechanism between Guangdong and Shenzhen carbon ETS.The present study provides the following explanation:(1)the design characteristics of carbon ETS(e.***.coverage,total quotas,the allocation,and MRV mechanism) depend on the local geographical conditions and policy goals.The differences of economic structure in Guangdong Province and Shenzhen City result in different coverage,which then result in differences in other management elements.(2)The operating state of the carbon market is affected by overall design of carbon ETS:in the case of tighter total allowance,lower proportion of China Certified Emission Reductions,and harsher punishment,the carbon market is relatively active,which intends to produce carbon financial market.Based on deep analysis of operation characteristics of carbon ETS in Guangdong and Shenzhen,the present study suggests that(1)the allowance should be allocated freely at the beginning stage and then gradually transited to the voluntary paid auction;(2)the allowances assigned to companies shall be linked up with their energy-saving objectives;(3)the output fluctuations and economic influence on the allowance allocation should be properly handled to maintain the fairness and consistence of allowance allocation standards;(4)stable public expectation is one of the key elements to maintain the regular operation of carbon ETS;(5)constrained carbon emission behavior outside ETS can contribute to social justice;and(6)the improvement of professional skills of relevant personnel in the enterprise and independent third party can enhance carbon emissions

A new and facile procedure was developed to synthesize novel 5-0- (6′-O-modified)-desosamine 14-membered ketolides by adopting different protective strategies and comparing various glycosylation conditions. Two trichloroacetimidate donors, with Lev or Ac substituent groups at the C-6 position, were synthesized to couple with the erythronolide. Several novel 5-0- (6′-O-modified)-desosamine 14-membered ketolides were obtained to verify the utility of the method.

BACKGROUND： Numerous studies have shown that magnetic resonance imaging （MRI） can detect survival and migration of super paramagnetic iron oxide-labeled stem cells in models of focal cerebral infarction. OBJECTIVE： To observe distribution of bone marrow mesenchymal stem cells （BMSCs） in a rat model of global brain ischemia following cardiac arrest and resuscitation, and to investigate the feasibility of tracing iron oxide-labeled BMSCs using non-invasive MRI. DESIGN, TIME AND SETTING： The randomized, controlled, molecular imaging study was performed at the Linbaixin Medical Research Center, Second Affiliated Hospital, Sun Yat-sen University, and the Institute of Cardiopulmonary Cerebral Resuscitation, Sun Yat-sen University, China from October 2006 to February 2009. MATERIALS： A total of 40 clean, Sprague Dawley rats, aged 6 weeks and of either gender, were supplied by the Experimental Animal Center, Sun Yat-sen University, China, for isolation of BMSCs. Feridex （iron oxide）, Gyroscan Inetra 1.5T MRI system, and cardiopulmonary resuscitation device were used in this study. METHODS： A total of 30 healthy, male Sprague Dawiey rats, aged 6 months, were used to induce ventricular fibrillation using alternating current. After 8 minutes, the rats underwent 6-minute chest compression and mechanical ventilation, followed by electric defibrillation, to establish rat models of global brain ischemia due to cardiac arrest and resuscitation. A total of 24 successful models were randomly assigned to Feridex-labeled and non-labeled groups （n = 12 for each group）. At 2 hours after resuscitation, 5 ×10^8 Feridex-labeled BMSCs, with protamine sulfate as a carrier, and 5 ×10^6 non-labeled BMSCs were respectively transplanted into both groups of rats through the right carotid artery （cells were harvested in 1 mL phosphate buffered saline）. MAIN OUTCOME MEASURES： Feridex-labeled BMSCs were observed by Prussian blue staining and electron microscopy. Signal intensity, celluar viability, and proliferative capacity of BMSCs were measured

Long non-coding RNAs(lncRNAs) belong to a large and complex family of RNAs, which play many important roles in regulating gene expression. However, the mechanism underlying the dynamic expression of lncRNAs is still not very clear. In order to identify lncRNAs and clarify the mechanisms involved, we collected basic information and highlighted the mechanisms underlying lncRNA expression and regulation. Overall, lncRNAs are regulated by several similar transcription factors and protein-coding genes. Epigenetic modification(DNA methylation and histone modification) can also downregulate lncRNA levels in tissues and cells. Moreover, lncRNAs may be degraded or cleaved via interaction with miRNAs and miRNAassociated protein complexes. Furthermore, alternative RNA splicing(AS) may play a significant role in the post-transcriptional regulation of lncRNAs.

Atherosclerosis(AS)is the leading cause of heart attacks,stroke,and peripheral vascular disease.Berberine(BBR),a botanical medicine,has diversified anti-atherosclerotic effects but with poor absorption.The aim of this study was to develop an effective BBR-entrapped nano-system for treating AS in high-fat diet(HFD)-fed Apoe(-/-)mice,and also explore the possible underlying mechanisms involved.Three D-a-tocopherol polyethylene glycol(PEG)succinate(TPGS)analogues with different PEG chain lengths were synthesized to formulate BBR-entrapped micelles.HFD-fed Apoe(-/-)mice were administered with optimized formula(BBR,100 mg/kg/day)orally for 5 months.The artery plaque onset and related metabolic disorders were evaluated,and the underlying mechanisms were studied.Our data showed that,BT1500M increased BBR deposition in liver and adipose by 107.6%and 172.3%,respectively.In the Apoe(-/-)mice,BT1500M ameliorated HFD-induced hyperlipidemia and lipid accumulation in liver and adipose.BT1500M also suppressed HFD-induced chronic inflammation as evidenced by the reduced liver and adipose levels of interleukin-6(IL-6),tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β);and decreased plasma level of TNF-α,IL-6,IL-1β,interferon-γ(IFN-γ),monocyte chemotactic protein(MCP),and macrophage inflammatory factor(MIP).The mechanism study showed that BT1500M changed Ampk and Nf-Kb gene expression,and interrupted a crosstalk process between adipocytes and macrophages.Further investigation proved that BT1500M decreased endothelial lesion and subsequent macrophage activation,cytokines release,as well as cholesteryl ester gathering in the aortic arch,resulting in ameliorated artery plaque build-up.Our results provide a practical strategy for treating AS using a BBR-entrapped nano-system.

目的探讨血清miR-21、miR-30b对乳腺癌患者术后曲妥珠单抗靶向治疗心脏毒性的预测价值。方法选取乳腺癌曲妥珠单抗靶向治疗患者138例为观察组,根据是否发生化疗相关心脏毒性事件分为非心脏毒性组和心脏毒性组2个亚组;另外选取同期体检健康女性150例为健康组。比较各组血清miR-21、miR-30b水平和临床资料。采用ROC评估血清miR-21、miR-30b对发生心脏毒性的预测价值,采用多因素Logistic回归分析发生心脏毒性的影响因素。结果观察组血清miR-21高于健康组,而miR-30b低于健康组;心脏毒性组血清miR-21和高血压占比高于非心脏毒性组,而miR-30b和左室射血分数(LVEF)低于非心脏毒性组(P<0.05)。LVEF、miR-21、miR-30b是乳腺癌术后化疗心脏毒性的影响因素(P<0.05)。血清miR-21、miR-30b联合预测乳腺癌术后化疗心脏毒性的诊断效能高于miR-21、miR-30b单独预测(P<0.05)。结论血清miR-21、miR-30b是乳腺癌术后化疗心脏毒性的影响因素,两者联合预测乳腺癌术后化疗心脏毒性的效能更高。

A high-performance bio-tissue imaging method using air flow-assisted desorption electrospray ionization coupled with a high-resolution mass spectrometer

Mass spectrometry imaging (MSI) technology can simultaneously obtain the spatial distribution of thousands of chemical compounds and has unique advantages compared to other techniques that allow mapping the surface of bio-tissue. Here, we combined an air flow-assisted desorption electrospray ionization (AFADESI) MSI device with a high-resolution mass spectrometer to optimize the system parameters and achieve more accurate spatial distribution characteristics for compounds of interest while investigating bio-tissue sections. The platform set-up, required instrumentation, sample pretreatment, parameter optimization and bio-tissue characterization are described and discussed.Finally, the parameter conditions that can provide optimal ionic intensity and enhanced resolution were confirmed. The reasonable resolution and sensitivity improvements of AFADESI-MSI have been achieved through tandem a high-resolution mass spectrometer system, therefore, it would be a promising technique for the bio-tissue imaging analysis.
