Overcoming adaptive resistance in AML by synergistically targeting FOXO3A-GNG7-mTOR axis with FOXO3A inhibitor Gardenoside and rapamycin

Therapeutic targeting FOxO3A(a forkhead transcription factor)represents a prom-ising strategy to suppress acute myeloid leukemia(AML).However,the effective inhibitors that target FOXO3A are lacking and the adaptive response signaling weakens the cytotoxic effect of FOxO3A depletion on AML cells.Here,we show that FOxO3A deficiency induces a compensa-tory response involved in the reactive activation of mTOR that leads to signaling rebound and adaptive resistance.Mitochondrial metabolism acts downstream of mTOR to provoke activa-tion of JNK/c-JUN via reactive oxygen species(ROS).At the molecular level,FOXO3A directly binds to the promoter of G protein gamma subunit 7(GNG7)and preserves its expression,while GNG7 interacts with mTOR and restricts phosphorylated activation of mTOR.Consequently,combinatorial inhibition of FOXO3A and mTOR show a synergistic cytotoxic effect on AML cells and prolongs survival in a mouse model of AML.Through a structure-based virtual screening,we report one potent small-molecule FOxO3A inhibitor(Gardenoside)that exhibits a strong effect of anti-FOXO3A DNA binding.Gardenoside synergizes with rapamycin to substantially reduce tumor burden and extend survival in AML patient-derived xenograft model.These results demonstrate that mTOR can mediate adaptive resistance to FOxO3A inhibition and validate a combinatorial approach for treating AML.

Bovine and Goat Milk Fat Globule Membrane Supplementation Alleviates Autism-like Behavior by Modulating Lipid Metabolism and Gut Microbiota

ASD (autism spectrum disorder) is a neurodevelopmental disorder influenced by genes and the environment. This study investigated the protective effects of bovine MFGM (BMFGM) and goat MFGM (GMFGM) supplementation on ASD model mice. Analysis of phospholipid composition showed higher levels of phosphatidylcholine in BMFGM and phosphatidylethanolamine in GMFGM. Behavioral results indicated MFGMs ameliorated social deficits， with GMFGM being more effective. MFGMs mitigated neuroinflammation by suppressing microglial overactivation and proinflammatory cytokines expression， meanwhile GMFGM increased the anti-inflammatory factor IL-10. MFGMs also altered gut microbiota composition and maintained gut barrier integrity. Uniquely， GMFGM increased butyrate production. Correlation analysis revealed positive associations between social behavior， levels of phosphatidylcholine， sphingomyelin， and the abundance of Allobaculum， Clostridium＿sensu＿stricto， and Turicibacter. Overall， these findings revealed the protective effects of MFGMs on neurodevelopment in ASD animal models and the underlying mechanism could be partly explained by their regulation of gut microbiota by the phospholipid components in MFGMs.

Spectral element modeling based structure piezoelectric impedance computation and damage identification

This paper presents a numerical simulation study on electromechanical impedance technique for structural damage identification.The basic principle of impedance based damage detection is structural impedance will vary with the occurrence and development of structural damage,which can be measured from electromechanical admittance curves acquired from PZT patches.Therefore,structure damage can be identified from the electromechanical admittance measurements.In this study,a model based method that can identify both location and severity of structural damage through the minimization of the deviations between structural impedance curves and numerically computed response is developed.The numerical model is set up using the spectral element method,which is promised to be of high numerical efficiency and computational accuracy in the high frequency range.An optimization procedure is then formulated to estimate the property change of structural elements from the electric admittance measurement of PZT patches.A case study on a pin-pin bar is conducted to investigate the feasibility of the proposed method.The results show that the presented method can accurately identify bar damage location and severity even when the measurements are polluted by 5%noise.

Sources and characteristics of fine particles over the Yellow Sea and Bohai Sea using online single particle aerosol mass spectrometer

Marine aerosols over the East China Seas are heavily polluted by continental sources. During the Chinese Comprehensive Ocean Experiment in November 2012, size and mass spectra of individual atmospheric particles in the size range from 0.2 to 2.0 μm were measured on board by a single particle aerosol mass spectrometer （SPAMS）. The average hourly particle number （PN） was around 4560 ＋ 3240 in the South Yellow Sea （SYS）, 2900 ± 3970 in the North Yellow Sea （NYS）, and 1700 ± 2220 in the Bohai Sea （BS）. PN in NYS and BS varied greatly over 3 orders of magnitude, while that in SYS varied shghtly. The size distributions were fitted with two log-normal modes. Accumulation mode dominated in NYS and BS, especially during episodic periods. Coarse mode particles played an important role in SYS. Particles were dassifled using an adaptive resonance theory based neural network algorithm （ART-2a）. Six particle types were identified with secondary-containing, aged sea-salt, soot-like, biomass burning, fresh sea-salt, and lead-containing particles accounting for 32%, 21%, 18%, 16%, 4%, and 3% of total PN, respectively. Aerosols in BS were relatively enriched in particles from anthropogenic sources compared to SYS, probably due to emissions from more developed upwind regions and indicating stronger influence of continental outflow on marine environment. Variation of source types depended mainly on origins of transported air masses. This study examined rapid changes in PN, size distribution and source types of fine particles in marine atmospheres. It also demonstrated the effectiveness of hi,h-time-resolution source apportionment by ART-2a.

基于PCA-DBSCAN聚类的焊缝提取研究

针对焊接之后的焊缝提取误差大、不易提取的问题,文章提出了一种DBSCAN聚类(densitybased spatial clustering of applications with noise)与改进主成分分析(principal component analysis,PCA)算法融合的焊缝提取算法。首先对焊缝图像进行灰度化、自适应中值滤波等预处理;其次对图像应用Canny边缘检测算法提取焊缝边缘,并使用DBSCAN密度聚类算法聚类焊缝边缘;之后依据改进的PCA算法寻找焊缝的主成分,将焊缝向主向量映射统计,根据图像分辨率自动分配一个阈值获取焊缝的左右边界,再将焊缝的左右边界反映射到次主向量获取焊缝的上下边界;最后按照文章提出的算法完成了三组对比实验,分析了算法受分辨率、焊接方式、光照强度等因素的影响。实验证明,文章提出的算法对直缝提取效果良好,提取精度超过了95%。

An Offloading Scheme Leveraging on Neighboring Node Resources for Edge Computing over Fiber-Wireless (FiWi) Access Networks

The computation resources at a single node in Edge Computing(EC)are commonly limited,which cannot execute large scale computation tasks.To face the challenge,an Offloading scheme leveraging on NEighboring node Resources(ONER)for EC over Fiber-Wireless(FiWi)access networks is proposed in this paper.In the ONER scheme,the FiWi network connects edge computing nodes with fiber and converges wireless and fiber connections seamlessly,so that it can support the offloading transmission with low delay and wide bandwidth.Based on the ONER scheme supported by FiWi networks,computation tasks can be offloaded to edge computing nodes in a wider range of area without increasing wireless hops(e.g.,just one wireless hop),which achieves low delay.Additionally,an efficient Computation Resource Scheduling(CRS)algorithm based on the ONER scheme is also proposed to make offloading decision.The results show that more offloading requests can be satisfied and the average completion time of computation tasks decreases significantly with the ONER scheme and the CRS algorithm.Therefore,the ONER scheme and the CRS algorithm can schedule computation resources at neighboring edge computing nodes for offloading to meet the challenge of large scale computation tasks.

信息通讯时代,科学技术的进步极大地降低了地图制图的门槛,大量非专业制图人员涌现到地图制图领域,地图的表现形式呈现出显著的泛化趋势。然而,由于专业知识的缺失,普通大众在专题地图选择时容易产生“制图困惑”,难以确定恰当的专题地图类型。针对上述问题,本文尝试提出一种基于可视化维度理论的泛地图知识推荐方法。首先,面向专题制图需求,建立以空间数据、数据特征、泛地图可视化维度、泛地图可视化形式、应用领域为核心要素的泛地图可视化知识本体,通过知识抽取、知识存储操作,构建泛地图可视化知识图谱。其次,对知识图谱进行知识嵌入,实现地图可视化要素的知识化表达。最后,基于泛地图知识要素建立相似度计算模型,计算用户输入数据与知识图谱内空间数据特征、可视化维度、应用领域等的相似度,得出综合相似度结果,实现泛地图可视化形式的精准推荐。结果表明:面对泛化的专题信息表达,本研究提出的泛地图知识推荐方法能够为用图者的制图选择提供较为准确可视化参考,一定程度上满足普通大众的寻图需求。
