Evaluation of the intracellular lipid-lowering effect of polyphenols extract from highland barley in HepG2 cells

Active ingredients from highland barley have received considerable attention as natural products for developing treatments and dietary supplements against obesity.In practical application,the research of food combinations is more significant than a specific food component.This study investigated the lipid-lowering effect of highland barley polyphenols via lipase assay in vitro and HepG2 cells induced by oleic acid(OA).Five indexes,triglyceride(TG),total cholesterol(T-CHO),low density lipoprotein-cholesterol(LDL-C),aspartate aminotransferase(AST),and alanine aminotransferase(ALT),were used to evaluate the lipidlowering effect of highland barley extract.We also preliminary studied the lipid-lowering mechanism by Realtime fluorescent quantitative polymerase chain reaction(q PCR).The results indicated that highland barley extract contains many components with lipid-lowering effects,such as hyperoside and scoparone.In vitro,the lipase assay showed an 18.4%lipase inhibition rate when the additive contents of highland barley extract were 100μg/m L.The intracellular lipid-lowering effect of highland barley extract was examined using 0.25 mmol/L OA-induced HepG2 cells.The results showed that intracellular TG,LDL-C,and T-CHO content decreased by 34.4%,51.2%,and 18.4%,respectively.ALT and AST decreased by 51.6%and 20.7%compared with the untreated hyperlipidemic HepG2 cells.q PCR results showed that highland barley polyphenols could up-regulation the expression of lipid metabolism-related genes such as PPARγand Fabp4.

Pricing Binary Variables of System-wide Constraints for Power System Optimization

With the trend of multiple energies or flexible demand in power systems,binary variables appear in systemwide constraints,which are the foundation of marginal pricing currently in markets.An appropriate pricing method incentivizes compliance of market participants;otherwise,compliance can be incentivized by paying discriminatory uplift payments which jeopardize transparency of markets.This paper proposes two theorems to examine whether the binary variables brought by multiple energies and flexible demand will impact compliance under marginal pricing.The first theorem shows sufficient conditions with which marginal pricing with fixed binary variables incentivizes compliance,while the second theorem shows sufficient conditions to require uplift payments.To improve transparency by reducing uplift payments under cases which fall into the second theorem,this paper further proposes a pricing method by combining 1)designed constraints to price binary variables in system-wide constraints,and 2)convex hull pricing to price binary variables in private constraints.Effectiveness of the proposed theorems and pricing method is verified in an electricity-gas case(consisting of the IEEE 30-bus system and the NGS 10-node system)and the IEEE 118-bus test system.

Hydrogen bond-bridged phosphorene flexible film for photodynamic inhibiting Staphylococcus aureus

Antibiotics are a widely used and effective treatment for bacterial infections.However,bacteria can gradually evolve during infection,leading to developing resistance to antibiotics,which renders previously effective treatments ineffective.Finding a useful and convenient manner to treat bacterial infections is a great challenge.Here,we report a flexible hydrogen-bond-bridged phosphorene film with photodynamic antibacterial properties and excellent mechanical properties,fabricated from electrochemical exfoliation of black phosphorus(BP).When illuminated under 700 nm light,the hydrogen bond-bridged phosphorene flexible film is capable of converting ground-state triplet oxygen(O_(2))into excited-state singlet oxygen(^(1)O_(2)),destroying the structure of the membrane of Staphylococcus aureus,and eventually leading to bacterial death,via breaking the C=C of unsaturated fatty acids within the bacterial cell membrane after the reaction between^(1)O_(2)and unsaturated fatty acids,thus realizing a highly efficient antibacterial approach,which is supported by gas chromatography-mass spectrometry(GC-MS)technique.This work establishes an effective phototherapy platform for treating bacterial traumatic infections.

Productivity in the Southern Ocean Antarctic Zone during the Northern Hemisphere Glaciation(NHG)and its link to atmospheric pCO_(2)

A decrease in atmospheric CO_(2)partial pressure(pCO_(2))is considered an important prerequisite for the onset and intensification of Northern Hemisphere Glaciation(NHG).However,how the ocean sequestered missing CO_(2)during the NHG is still uncertain.Changes in surface productivity and deep ventilation in the Southern Ocean(SO)have been proposed to explain the variations in atmospheric pCO_(2)over the last eight glacial cycles,but it is unclear whether these mechanisms contributed to the decrease in atmospheric pCO_(2)during the NHG.Using titanium-normalized contents and mass accumulation rates of biogenic opal and total organic carbon from the International Ocean Discovery Program(IODP)Expedition 374 Site U1524A,we reconstruct the productivity in the Ross Sea,Antarctica,from 3.3 Ma to 2.4 Ma.The productivity records exhibit a long-term decreasing trend and several distinct phased evolutionary features.Specifically,the local productivity fluctuated dramatically during 3.3-3.0 Ma,decreased gradually during 3.0-2.6 Ma,and remained relatively constant during 2.6-2.4 Ma.By comparing productivity with its potential influences,we infer that the phased and long-term evolutions of productivity were mainly controlled by changes in deep ocean ventilation.Sea ice expansion might have decreased productivity during 3.3-3.0 Ma by light attenuation.Changes in eolian dust input have little effect on productivity.Further analysis revealed no coupling linkage between productivity and atmospheric pCO_(2),indicating that the productivity in the SO Antarctic Zone(AZ)was not the main factor controlling the atmospheric CO_(2)decrease during the NHG.To improve our understanding of the role of SO processes in the NHG,further studies should focus on the potential influences of deep ocean ventilation on atmospheric pCO_(2)in the AZ,and similar studies should also be extended to the sea area in the Subantarctic Zone.

Supramolecular complex glycoconjugate vaccine generates self-enhancement effects for carbohydrate antigen delivery

Targeting delivery of tumor-associated carbohydrate antigen(TACA)-based vaccine to antigen-presenting cells(APCs)mediated by endogenous antibodies can improve the immunogenicity of TACA.However,an essential requirement of this approach is to generate high titers of endogenous antibodies in vivo through pre-immunization,which complicates the immunization procedure and may cause side effects.Herein,we report a new generation of APC-targeting TACA-based supramolecular complex vaccine,assembled by sialyl Thomsen-nouveau-bovine serum albumin-adamantine(sTn-BSA-Ada)and heptavalent rhamnose(Rha)-modifiedβ-cyclodextrin(β-CD)via host-guest interaction.The complex vaccine retained anti-Rha antibodies recruiting capability and facilitated the APCs uptake of the vaccine via the interaction of the Fc-domain with the Fc receptors on APCs.We demonstrate that direct immunization of complex vaccine elicited anti-Rha and anti-sTn specific immune response synchronously,generating a novel self-enhancement effect that can improve the antigen delivery to APCs in high efficacy.The structure-activity relationship(SAR)study proved that complex vaccine 4 with polyethylene glycol 6(PEG 6)linker in host molecule provoked a robust and specific sTn immune response comparable to the pre-immunization approach.The antisera induced by complex vaccine,either through direct immunization or pre-immunization,exhibited equal potency of cytotoxicity against the sTn expression cancer cells.This study provides a general platform for TACA-based vaccines with self-enhancement effects without the need for pre-immunization.

Fast thermal responsive separators toward long-life and safe lithium metal batteries

lithium metal batteries(LMBs)have been extensively investigated during the past decades because of their ultrahigh energy densities.With the increasing demand for energy density,however,the safety issue of LMBs has become a significant challenge.In particular,localized areas of increased temperature(namely,hotspots)may be induced and even exacerbated within LMBs by uneven current distribution,internal short circuits,or inadequate heat dissipation,which significantly sacrifices battery safety and cycle life.Here,we report the rational design and fabrication of a fast thermal responsive separator capable of inhibiting the growth of lithium dendrites and mitigating thermal propagation,thereby reducing the risk of thermal runaway.The as-achieved separator comprises both an electrospun membrane using a phase change material with superior thermal-storage ability and a thermally conductive modification layer of hexagonal boron nitride nanosheets with a fast heat-transfer feature.It is demonstrated that such a unique integration of heat conduction and heat storage enables the functional separator with attractive abilities to mitigate hotspots and inhibit the growth of lithium dendrites upon the cycling of LMBs.Moreover,pouch cells with the thermal-responsive separator,as well as numerical simulations,verify much enhanced safety and cycle life of LMBs.This work may offer a new conceptual design of intelligent separators that acts as a functional unit encapsulated within a single cell to boost in-situ thermal management,which will help to develop high-safety and energy-dense LMBs.

Epilepsy,one of the most common neurological disorders,is characterized by spon-taneous recurrent seizures.Temporal lobe epilepsy(TLE)is one of the most common medically intractable seizure disorders.Traf2-and NcK-interacting kinase(TNIK)has recently attracted attention as a critical modulation target of many neurological and psychiatric disorders,but its role in epilepsy remains unclear.In this study,we hypothesized the involvement of TNIK in epilepsy and investigated TNIK expression in patients with intractable TLE and in a pilocarpineinduced rat model of epilepsy by western blotting,immunofluorescence,and immunohistochemistry.A pentylenetetrazole(PTZ)-induced epilepsy rat model was used to determine the effect of the TNIK inhibitor NCB-0846 on behavioral manifestations of epilepsy.Coimmunoprecipitation(Co-IP)/mass spectrometry(MS)was used to identify the potential mechanism.Through Co-IP,we detected and confirmed the main potential TNIK interactors.Subcellular fractionation was used to establish the effect of NCB-0846 on the expression of the main interactors in postsynaptic density(PSD)fractions.We found that TNIK was primarily located in neurons and decreased significantly in epilepsy model rats and TLE patients compared with controls.NCB-0846 delayed kindling progression and decreased seizure severity.Co-IP/MS identified 63 candidate TNIK interactors in rat hippocampi,notably CaMKIl.Co-IP showed that TNIK might correlate with endogenous GRIA1,SYN2,PSD-95,CaMKIV,GABRG1,and GABRG2.In addition,the significant decrease in GRIA1 in hippocampal total lysate and PSDs after NCB-0846 treatment might help modify the progression of PTZ kindling.Our results suggest that TNIK contributes to epileptic pathology and is a potential antiepileptic drug target.

Pumping-induced Well Hydraulics and Groundwater Budget in a Leaky Aquifer System with Vertical Heterogeneity in Aquitard Hydraulic Properties

In groundwater hydrology,aquitard heterogeneity is often less considered compared to aquifers,despite its significant impact on groundwater hydraulics and groundwater resources evaluation.A semi-analytical solution is derived for pumping-induced well hydraulics and groundwater budget with consideration of vertical heterogeneity in aquitard hydraulic conductivity(K)and specific storage(S_(s)).The proposed new solution is innovative in its partitioning of the aquitard into multiple homogeneous sub-layers to enable consideration of various forms of vertically heterogeneous K or S_(s).Two scenarios of analytical investigations are explored:one is the presence of aquitard interlayers with distinct K or S_(s) values,a common field-scale occurrence;another is an exponentially depth-decaying aquitard S_(s),a regional-scale phenomenon supported by statistical analysis.Analytical investigations reveal that a low-K interlayer can significantly increase aquifer drawdown and enhance aquifer/aquitard depletion;a high-S_(s) interlayer can noticeably reduce aquifer drawdown and increase aquitard depletion.Locations of low-K or high-S_(s) interlayers also significantly impact well hydraulics and groundwater budget.In the context of an exponentially depth-decaying aquitard S_(s),a larger decay exponent can enhance aquifer drawdown.When using current models with a vertically homogeneous aquitard,half the sum of the geometric and harmonic means of exponentially depth-decaying aquitard S_(s) should be used to calculate aquitard depletion and unconfined aquifer leakage.

Bioactive chemical constituents from the marine-derived fungus Cladosporium sp.DLT-5

A new isochromanone,cladosporinisochromanone(1),accompanied by 15 known compounds(2–16)were obtained from secondary metabolites produced by marine-derived fungus Cladosporium sp.DLT-5.NMR and HRESIMS spectra elucidation determined the planar structure of 1.Subsequent electronic circular dichroism(ECD)experiment assigned the absolute configuration of 1.Compounds 1,2,4–6,and 10 displayed different degrees of neuroprotective activities on human neuroblastoma cells SH-SY5Y.Five compounds(1,3–5,and 13)emerged resistance to protein tyrosine phosphatase 1B(PTP1B),further kinetic analysis and molecular docking study indicated that the most potent compound 13(IC50value of 10.74±0.61μmol/L)was found as a noncompetitive inhibitor for PTP1B.Surface plasmon resonance(SPR)and molecular docking studies also demonstrated the interaction between compound 12 and Niemann-Pick C1 like 1(NPC1L1),which has been identified as significant therapeutic target for hypercholesteremia.In addition,compounds 3,6,and 14 showed attractive inhibitory activity against the phytopathogenic fungi:Colletotrichum capsici.Therefore,library of Cladosporium metabolites is enriched and new active uses of known compounds are explored.

Fabrication of wide temperature lanthanum and cerium doped Cu/TNU-9 catalyst with excellent NH_(3)-SCR performance and outstanding SO_(2)+H_(2)O tolerance

The effects of La on the catalytic performance,SO_(2)and H2O resistance of Cu-Ce/TNU-9 catalyst were studied in the selective catalytic reduction of NO_(x)via ammonia(NH_(3)-SCR).The results show that the La doped Ce-Cu/TNU-9(CCL/T9)catalyst exhibits better SCR performance than Ce-Cu/TNU-9(CC/T9)and Cu/TNU-9(C/T9)in the wide temperature window(200-450℃)due to La benefiting from enhancing Cu^(+)+Ce^(4+)?Cu^(2+)-+Ce^(3+)to facilitate the electron transfer,which further accelerates the oxidation of NO into NO_(2)that leads to the"Fast SCR".The La can also enhance the surface acidic,redox property and is beneficial to the formation of oxygen vacancy.Meanwhile,the larger co ntent of Ce^(3+) can produce oxygen vacancy,resulting in rising SCR activity.The CCL/T9 has outstanding water and SO_(2) resistance,which may be due to the adso rbed S on the surface of these catalysts mainly existing in the form of a small amount of sulfate and sulfite,the decomposition of ammonium bisulphite under high temperature and the hydrophobicity of TNU-9 zeolite.
