Outlier-resistant distributed fusion filtering for nonlinear discrete-time singular systems under a dynamic event-triggered scheme

This paper investigates the problem of outlier-resistant distributed fusion filtering(DFF)for a class of multi-sensor nonlinear singular systems(MSNSSs)under a dynamic event-triggered scheme(DETS).To relieve the effect of measurement outliers in data transmission,a self-adaptive saturation function is used.Moreover,to further reduce the energy consumption of each sensor node and improve the efficiency of resource utilization,a DETS is adopted to regulate the frequency of data transmission.For the addressed MSNSSs,our purpose is to construct the local outlier-resistant filter under the effects of the measurement outliers and the DETS;the local upper bound(UB)on the filtering error covariance(FEC)is derived by solving the difference equations and minimized by designing proper filter gains.Furthermore,according to the local filters and their UBs,a DFF algorithm is presented in terms of the inverse covariance intersection fusion rule.As such,the proposed DFF algorithm has the advantages of reducing the frequency of data transmission and the impact of measurement outliers,thereby improving the estimation performance.Moreover,the uniform boundedness of the filtering error is discussed and a corresponding sufficient condition is presented.Finally,the validity of the developed algorithm is checked using a simulation example.

Frustrated Lewis Pairs(FLPs)represent a unique class of interactions in Lewis acid-base chemistry,driven by spatial hindrance or incongruent orbital energy levels that hinder the formation of effective coordination bonds.FLPs have received significant attention for their application in activating small molecules and facilitating organic synthesis reactions.Recent developments have led to the emergence of Frustrated Radical Pairs(FRPs)as an extension of the radical family.FRPs are formed from FLPs through Single Electron Transfer(SET)and exhibit the ability to activate a variety of chemical bonds.While research on FLPs is well-established,investigations into FRPs in organic reactions remain limited.This review highlights the current state of FRPs in organic synthesis,delves into mechanistic insights,explores their potential,and underscores the challenges in this emerging field.

According to the latest consensus,many traditional diseases are considered meta-bolic diseases,such as cancer,type 2 diabetes,obesity,and cardiovascular disease.Currently,metabolic diseases are increasingly prevalent because of the ever-improving living standards and have become the leading threat to human health.Multiple therapy methods have been applied to treat these diseases,which improves the quality of life of many patients,but the overall effect is still unsatisfactory.Therefore,intensive research on the metabolic process and the pathogenesis of metabolic diseases is imperative.N6-methyladenosine(m°A)is an important modification of eukaryotic RNAs.It is a critical regulator of gene expression that is involved in different cellular functions and physiological processes.Many studies have indi-cated that m6A modification regulates the development of many metabolic processes and metabolic diseases.In this review,we summarized recent studies on the role of m6A modifica-tion in different metabolic processes and metabolic diseases.Additionally,we highlighted the potential m6A-targeted therapy for metabolic diseases,expecting to facilitate m6A-targeted strategies in the treatment of metabolic diseases.

Federated learning is a new type of distributed learning framework that allows multiple participants to share training results without revealing their data privacy.As data privacy becomes more important,it becomes difficult to collect data from multiple data owners to make machine learning predictions due to the lack of data security.Data is forced to be stored independently between companies,creating“data silos”.With the goal of safeguarding data privacy and security,the federated learning framework greatly expands the amount of training data,effectively improving the shortcomings of traditional machine learning and deep learning,and bringing AI algorithms closer to our reality.In the context of the current international data security issues,federated learning is developing rapidly and has gradually moved from the theoretical to the applied level.The paper first introduces the federated learning framework,analyzes its advantages,reviews the results of federated learning applications in industries such as communication and healthcare,then analyzes the pitfalls of federated learning and discusses the security issues that should be considered in applications,and finally looks into the future of federated learning and the application layer.

A causal variant rs3769823 in 2q33.1 involved in apoptosis pathway leading to a decreased risk of non-small cell lung cancer

Objective:Although our previous genome-wide association study(GWAS)has identified chromosome 2q33.1 as a susceptibility locus for non-small cell lung cancer(NSCLC),the causal variants remain unclear.The aims of this study were to identify the causal variants in 2q33.1 and to explore their biological functions in NSCLC.Methods:CCK-8,colony formation,EdU incorporation,Transwell,and quantitative real-time polymerase chain reaction assays were applied to examine variant function.The tumor xenograft model was used to examine variant function in vivo.Caspase-8 activity assays,flow cytometry analysis,and co-immunoprecipitation assays were used to explore the molecular mechanism.Results:The missense variant rs3769823(A>G),which caused the substitution of lysine with arginine at amino acid 14 in caspase-8(caspase-8K14R),was identified as a potential causal candidate in 2q33.1.Compared with the wild type caspase-8(caspase8WT)group,the caspase-8K14R group had higher expression of caspase-8 and cleaved caspase-8.Caspase-8K14R inhibited the proliferation and metastasis of human lung cancer cell lines in vitro.Moreover,caspase-8K14R repressed lung cancer cell growth in vivo.Mechanistically,caspase-8K14R was more sensitive than caspase-8WT to tumor necrosis factor-related apoptosis-inducing ligand(TRAIL)-mediated apoptosis and showed higher binding of caspase-8 and FADD.Conclusions:These results suggested that rs3769823 is the causal variant in chromosome 2q33.1 and is involved in an apoptosis pathway,leading to a decreased risk of NSCLC.

The utility of the polygenic risk score(PRS)to identify individuals at higher risk of stroke beyond clinical risk remains unclear,and we clarified this using Chinese population-based prospective cohorts.Cox proportional hazards models were used to estimate the 10-year risk,and Fine and Gray’s models were used for hazard ratios(HRs),their 95%confidence intervals(CIs),and the lifetime risk according to PRS and clinical risk categories.A total of 41,006 individuals aged 30–75 years with a mean follow-up of 9.0 years were included.Comparing the top versus bottom 5%of the PRS,the HR was 3.01(95%CI 2.03–4.45)in the total population,and similar findings were observed within clinical risk strata.Marked gradients in the 10-year and lifetime risk across PRS categories were also found within clinical risk categories.Notably,among individuals with intermediate clinical risk,the 10-year risk for those in the top 5%of the PRS(7.3%,95%CI 7.1%–7.5%)reached the threshold of high clinical risk(≥7.0%)for initiating preventive treatment,and this effect of the PRS on refining risk stratification was evident for ischemic stroke.Even among those in the top 10%and 20%of the PRS,the 10-year risk would also exceed this level when aged≥50 and≥60 years,respectively.Overall,the combination of the PRS with the clinical risk score improved the risk stratification within clinical risk strata and distinguished actual high-risk individuals with intermediate clinical risk.

Background:Most lung cancer risk prediction models were developed in European and North-American cohorts of smokers aged≥55 years,while less is known about risk profiles in Asia,especially for never smokers or individuals aged<50 years.Hence,we aimed to develop and validate a lung cancer risk estimate tool for ever and never smokers across a wide age range.Methods:Based on the China Kadoorie Biobank cohort,we first systematically selected the predictors and explored the nonlinear association of predictors with lung cancer risk using restricted cubic splines.Then,we separately developed risk prediction models to construct a lung cancer risk score(LCRS)in 159,715 ever smokers and 336,526 never smokers.The LCRS was further validated in an independent cohort over a median follow-up of 13.6 years,consisting of 14,153 never smokers and 5,890 ever smokers.Results:A total of 13 and 9 routinely available predictors were identified for ever and never smokers,respectively.Of these predictors,cigarettes per day and quit years showed nonlinear associations with lung cancer risk(Pnon-linear<0.001).The curve of lung cancer incidence increased rapidly above 20 cigarettes per day and then was relatively flat until approximately 30 cigarettes per day.We also observed that lung cancer risk declined sharplywithin the first 5 years of quitting,and then continued to decrease but at a slower rate in the subsequent years.The 6-year area under the receiver operating curve for the ever and never smokers’models were respectively 0.778 and 0.733 in the derivation cohort,and 0.774 and 0.759 in the validation cohort.In the validation cohort,the 10-year cumulative incidence of lung cancerwas 0.39%and 2.57%for ever smokers with low(<166.2)and intermediate-high LCRS(≥166.2),respectively.Never smokers with a high LCRS(≥21.2)had a higher 10-year cumulative incidence rate than those with a low LCRS(<21.2;1.05%vs.0.22%).An online risk evaluation tool(LCKEY;http://***/lckey/web)was developed to facilitate the use of LCRS.Conclusions:The LCRS can be a

Epidemiological and laboratory-based studies have identified infection with one of 15 high-risk human papillo-mavirus （HPV） types as a necessary but not sufficient cause of cervical cancer. The prevalence of genital HPV in-fections is high in young women, but most of the infections regress without interventions. Host genetic variations in genes involved in immune response pathways may be related to HPV clearance, and HPV E6/E7 oncoproteins interacting or downstream genes, both coding and non-coding, may contribute to the outcome of high risk HPV infection and cervical cancer. Of specific interest for this review has been the selection of genetic variants in genes involved in the above-referred pathways with a summary of their applications in association studies. Because the supportive and opposing data have been reported in different populations, well-designed international collabora-tive studies need to be conducted to define the consistency of the associations, paving the way to better define the patients at high risk of developing cervical cancer.
