Casual associations between blood metabolites and colon cancer

BACKGROUND Limited knowledge exists regarding the casual associations linking blood metabolites and the risk of developing colorectal cancer.AIM To investigate causal associations between blood metabolites and colon cancer.METHODS The study utilized a two-sample Mendelian randomization(MR)analysis to investigate the causal impact of 486 blood metabolites on colorectal cancer.The primary method of analysis used was the inverse variance weighted model.To further validate the results several sensitivity analyses were performed,including Cochran's Q test,MR-Egger intercept test,and MR robust adjusted profile score.These additional analyses were conducted to ensure the reliability and robustness of the findings.RESULTS After rigorous selection for genetic variation,486 blood metabolites were included in the MR analysis.We found Mannose[odds ratio(OR)=2.09(1.10-3.97),P=0.024],N-acetylglycine[OR=3.14(1.78-5.53),P=7.54×10^(-8)],X-11593-O-methylascorbate[OR=1.68(1.04-2.72),P=0.034],1-arachidonoylglycerophosphocholine[OR=4.23(2.51-7.12),P=6.35×10^(-8)]and 1-arachidonoylglycerophosphoethanolamine 4[OR=3.99(1.17-13.54),P=0.027]were positively causally associated with colorectal cancer,and we also found a negative causal relationship between Tyrosine[OR=0.08(0.01-0.63),P=0.014],Urate[OR=0.25(0.10-0.62),P=0.003],N-acetylglycine[0.73(0.54-0.98),P=0.033],X-12092[OR=0.89(0.81-0.99),P=0.028],Succinylcarnitine[OR=0.48(0.27-0.84),P=0.09]with colorectal cancer.A series of sensitivity analyses were performed to confirm the rigidity of the results.CONCLUSION This study showed a causal relationship between 10 blood metabolites and colorectal cancer,of which 5 blood metabolites were found to be causal for the development of colorectal cancer and were confirmed as risk factors.The other five blood metabolites are protective factors.

Exploration of potential biomarkers and therapeutic targets for trauma-related acute kidney injury

Purpose:Acute kidney injury(AKI)is one of the most common functional injuries observed in trauma patients.However,certain trauma medications may exacerbate renal injury.Therefore,the early detection of trauma-related AKI holds paramount importance in improving trauma prognosis.Methods:Qualified datasets were selected from public databases,and common differentially expressed genes related to trauma-induced AKI and hub genes were identified through enrichment analysis and the establishment of protein-protein interaction(PPI)networks.Additionally,the specificity of these hub genes was investigated using the sepsis dataset and conducted a comprehensive literature review to assess their plausibility.The raw data from both datasets were downloaded using R software(version 4.2.1)and processed with the"affy"package19 for correction and normalization.Results:Our analysis revealed 585 upregulated and 629 downregulated differentially expressed genes in the AKI dataset,along with 586 upregulated and 948 downregulated differentially expressed genes in the trauma dataset.Concurrently,the establishment of the PPI network and subsequent topological analysis highlighted key hub genes,including CD44,CD163,TIMP metallopeptidase inhibitor 1,cytochrome b-245 beta chain,versican,membrane spanning 4-domains A4A,mitogen-activated protein kinase 14,and early growth response 1.Notably,their receiver operating characteristic curves displayed areas exceeding 75%,indicating good diagnostic performance.Moreover,our findings postulated a unique molecular mechanism underlying trauma-related AKI.Conclusion:This study presents an alternative strategy for the early diagnosis and treatment of trauma-related AKI,based on the identification of potential biomarkers and therapeutic targets.Additionally,this study provides theoretical references for elucidating the mechanisms of trauma-related AKI.

Polydatin ameliorates hepatic ischemia-reperfusion injury by modulating macrophage polarization

Background:Polydatin,a glucoside of resveratrol,has shown protective effects against various diseases.However,little is known about its effect on hepatic ischemia-reperfusion(I/R)injury.This study aimed to elucidate whether polydatin protects liver against I/R-induced injury and to explore the underlying mechanism.Methods:After gavage feeding polydatin once daily for a week,mice underwent a partial hepatic I/R procedure.Serum alanine aminotransferase(ALT)/aspartate aminotransferase(AST),hematoxylin-eosin(H&E)and TdT-mediated dUTP nick-end labeling(TUNEL)staining were used to evaluate liver injury.The severity related to the inflammatory response and reactive oxygen species(ROS)production was also investigated.Furthermore,immunofluorescence and Western blotting were used to detect macrophage polarization and the NF-κB signaling pathway in macrophages.Results:Compared with the I/R group,polydatin pretreatment significantly attenuated I/R-induced liver damage and apoptosis.The oxidative stress marker(dihydroethidium fluorescence,malondialdehyde,superoxide dismutase and glutathione peroxidase)and I/R related inflammatory cytokines(interleukin1β,interleukin-10 and tumor necrosis factor-α)were significantly suppressed after polydatin treatment.In addition,the result of immunofluorescence indicated that polydatin reduced the polarization of macrophages toward M1 macrophages both in vivo and in vitro.Western blotting showed that polydatin inhibited the pro-inflammatory function of RAW264.7 via down-regulating the NF-κB signaling pathway.Conclusions:Polydatin protects the liver from I/R injury by remodeling macrophage polarization via NFκB signaling.

The Sequential Pricing of Ride-Hailing System with rental Service in the Context of Fleet Electrification

Surging demand and reduced capacity in the ride-hailing industry have prompted numerous ride-hailing platforms to build their own car-rental services catering to drivers who do not possess private vehicles. However, the trade-off between the ride-hailing service and the car-rental service is an important issue that is still unclear in theory. Moreover, ride-hailing platforms are transitioning towards all-electric fleets in the context of Carbon Neutrality goals and government regulations. This paper considers a ride-hailing system comprising a monopolist ride-hailing platform, an electric vehicle (EV) rental firm, and a gasoline vehicle (GV) rental firm. Furthermore, we build a stylized model to study the sequential pricing of the system. The equilibrium outcomes show the significant impact of the ride-hailing platform’s decision to continue or withdraw offering EV rental services on EV and GV drivers’ net earnings, rental prices, and wages. The ride-hailing platform providing EV rental services increases EV drivers’ net earnings but decreases the GV driver wages and rental prices. However, the EV rental service offered by the ride-hailing platform does not necessarily lead to an increased total profit for the system. The improved profitability of the system by the ride-hailing platform providing EV rental services is contingent upon lower rider prices and higher fuel costs. The ride-hailing platform’s EV rental services provision also effectively fosters the ride-hailing fleet’s electrification. Furthermore, as the number of riders increases, the ride-hailing platform should reduce the commission rate for EV drivers to maintain competitiveness and profitability.

An axisymmetric characteristic model for enhancing prediction of centrifugal compressor characteristics

Rapid and accurate determination of compressor characteristic maps is essential for the initial design of centrifugal compressors in aircraft power systems. The accuracy of existing methodologies, which rely on combinations of loss models, varies significantly depending on the compressor's geometry and operational range. This variance necessitates substantial experimental or Computational Fluid Dynamics(CFD) data for coefficient calibration. To address this challenge, this study presents an axisymmetric characteristic model for compressor performance assessment. This model incorporates the factors of blade angle, meridional passage area, and the radial deflection angle of meridional streamlines of the compressor. These factors are derived from fundamental aerodynamic equations encompassing mass, momentum, and energy conservation of the compressor. In contrast to conventional one-dimensional approaches, the proposed method reduces the number of loss coefficients and more effectively accounts for the impact of geometric alterations on centrifugal compressor properties. Furthermore, the model reduces dependence on experimental and CFD data. Efficacy of the model is validated using experimental data from four distinct types of centrifugal compressors. Correlation analysis reveals that the model's coefficients can be expressed as functions of the ratio of the Reynolds number to the impeller tip speed. This ratio serves as a characteristic parameter for the design and optimization of centrifugal compressors. Consequently, the proposed method offers an efficient and accurate means for the quick computation of centrifugal compressor characteristics. This is of great significance for improving the efficiency of centrifugal compressors and reducing energy consumption.

Lithium-ion batteries are widely recognized as a crucial enabling technology for the advancement of electric vehicles and energy storage systems in the grid.The design of battery state estimation and control algorithms in battery management systems is usually based on battery models,which interpret crucial battery dynamics through the utilization of mathematical functions.Therefore,the investigation of battery dynamics with the purpose of battery system identification has garnered considerable attention in the realm of battery research.Characterization methods in terms of linear and nonlinear response of lithium-ion batteries have emerged as a prominent area of study in this field.This review has undertaken an analysis and discussion of characterization methods,with a particular focus on the motivation of battery system identification.Specifically,this work encompasses the incorporation of frequency domain nonlinear characterization methods and dynamics-based battery electrical models.The aim of this study is to establish a connection between the characterization and identification of battery systems for researchers and engineers specialized in the field of batteries,with the intention of promoting the advancement of efficient battery technology for real-world applications.

In situ plasmonic & electrochemical fiber-optic sensor for multi-metal-ions detection

High precision and high throughput detection of heavy metal ions is essential for water quality monitoring and assessment.Herein,we propose a plasmonic & electrochemical dual-mode fiber sensing probe for label-free and real-time detection of multiple ions(Pb2+and Cu2+as examples).This sensor comprises a multimode fiber-single mode fiber reflection probe,the outer surface of which is coated with a gold nanofilm to excite the surface plasmon resonance(SPR) optically and simultaneously serves as an electrochemical working electrode.In traditional electrochemical detection,the enrichment of ions cannot be detected in real-time.However,by utilizing the plasmonic & electrochemical dual-mode detection method,various kinds of metal ions can be deposited onto the gold nanofilm and selectively oxidized during forward potential scanning,and the entire electrochemical process can be monitored by SPR measurement.We experimentally demonstrate that the sensor can simultaneously detect Pb2+and Cu2+in a mixed solution in real-time,providing a linear response over the ion concentration range from 10-12to 10-7M and offering an excellent detection limit(1.69×10-14–5.49×10-13M).The proposed dual-mode fiber sensor has the benefits of remote sensing,compact footprint,and cost-effectiveness and shows excellent potential for water quality risk management in difficult-to-reach environments.

BACKGROUND Primary liver cancer is a malignant tumor with a high recurrence rate that significantly affects patient prognosis.Postoperative adjuvant external radiation therapy(RT)has been shown to effectively prevent recurrence after liver cancer resection.However,there are multiple RT techniques available,and the differ-ential effects of these techniques in preventing postoperative liver cancer re-currence require further investigation.AIM To assess the advantages and disadvantages of various adjuvant external RT methods after liver resection based on overall survival(OS)and disease-free survival(DFS)and to determine the optimal strategy.METHODS This study involved network meta-analyses and followed the PRISMA guidelines.The data of qualified studies published before July 10,2023,were collected from PubMed,Embase,the Web of Science,and the Cochrane Library.We included relevant studies on postoperative external beam RT after liver resection that had OS and DFS as the primary endpoints.The magnitudes of the effects were determined using risk ratios with 95%confidential intervals.The results were analyzed using R software and STATA software.RESULTS A total of 12 studies,including 1265 patients with hepatocellular carcinoma(HCC)after liver resection,were included in this study.There was no significant heterogeneity in the direct paired comparisons,and there were no significant differences in the inclusion or exclusion criteria,intervention measures,or outcome indicators,meeting the assumptions of heterogeneity and transitivity.OS analysis revealed that patients who underwent stereotactic body radiotherapy(SBRT)after resection had longer OS than those who underwent intensity modulated radiotherapy(IMRT)or 3-dimensional conformal RT(3D-CRT).DFS analysis revealed that patients who underwent 3D-CRT after resection had the longest DFS.Patients who underwent IMRT after resection had longer OS than those who underwent 3D-CRT and longer DFS than those who underwent SBRT.CONCLUSION HCC patients who undergo liver cancer resec

Entropy production by dissipation effects and characteristic vortex evolution in a rocket turbopump

The relationship between entropy production and vortex evolution affects the efficiency and stability of rotating machinery.This study investigated the energy characteristics of a rocket turbopump and revealed the correlated mechanisms of the entropy production rate using the dissipation effects and characteristic vortex evolution.For the first time,direct and turbulent dissipation and rigid and shear vorticity decomposition methods were utilized to analyze the correlation between flow loss and characteristic vorticities in rotating machinery.With an increase in the flow rate,the hydraulic losses of the dissipation effects and wall decreased by 60%and 38.3%,respectively,and the proportions of the input energy decreased(from 13%to 8%)and remained stable(8%),respectively.The local direct dissipative entropy production(DDEP)in the inducer-impeller is strongly related to shear entropy,and the correlated effect of total enstrophy on DDEP is weaker than that of shear vorticity,indicating that rigid enstrophy suppresses direct dissipation.The correlation between turbulent dissipation and rigid enstrophy was significantly weaker in the static flow passage of the turbopump owing to the weak rigid rotational effect.The correlation between the rigid entropy and local turbulent dissipative entropy production(TDEP)gradually increased with increasing flow rate,reaching a medium correlation(the maximal correlated degree in the turbopump)and exhibiting rigid rotation effects on the hydraulic loss.Moreover,the flow rate significantly affected the correlation(except for the diffuser),and the two characteristic vorticities reached a maximum at the designed flow rate owing to optimal efficiency and minimum hydraulic loss.
