Bone morphogenetic protein 4(BMP4)can induce the formation of chicken primordial germ cells(PGCs)in vitro;however,its regulatory mechanism in poultry remains unknown.This study aimed to use RNA-seq to analyze PGCs in chicken embryos and iPGCs induced by BMP4 in vitro,clarify the internal regulatory factors of PGCs,analyze the mechanism of the formation of PGCs,and lay a theoretical foundation for the further optimization of PGCs induction systems.Embryonic stem cells(ESCs),PGCs and iPGCs induced by BMP4 in vitro were collected.The transcriptional maps of the three cell types were studied using RNA-seq.The results showed 6,142 genes differentially expressed between PGCs and iPGCs,of which 2,728 were upregulated in iPGCs and 3,414 were downregulated in iPGCs.Compared to that in ESCs,BMP4 was significantly upregulated in PGCs and iPGCs.KEGG results showed that both the TGF-βand Wnt signaling pathways were activated during the formation of PGCs in vitro and in vivo,and the activation was more significant during iPGCs induced by BMP4.The expression of Nodal,an inhibitory factor of TGF-βsignaling,was significantly decreased in PGCs and iPGCs,but was not expressed in iPGCs,which further supports our conclusion.Additionally,the Lysosome and PI3K-AKT signaling pathways were significantly enriched in PGCs and iPGCs,respectively.Further,transmission electron microscopy(TEM)results showed that the number of autolysosomes was significantly higher after the addition of BMP4,which is consistent with the KEGG results.Furthermore,the number of PGCs was significantly reduced after ATG14 was interfered in vivo and in vitro.In conclusion,this study screened out the key signaling pathways during the formation of PGCs,aiming to provide help for enriching the mechanism network regulating PGCs formation in chicken and laying a theoretical foundation for further improving the efficiency of inducing PGCs in vitro.

Dead time is necessary for the coupled power switches to prevent shoot-through,especially in the modular multilevel converters(MMCs)with a large number of power switches.This paper proposes a dead-time effect suppression strategy for MMCs with nearest level modulation.The operational principles of MMCs are first analyzed.According to the operational features of MMCs,the method that removes a switching signal from the coupled switches and the reduced switching frequency voltage balancing algorithms(RSFVBAs)are mixed in the proposed method.In the intervals that are furthest away from the zerocrossing points(ZCP)of arm currents,the single switching signal method can completely eliminate the dead-time effect(DTE).Alternatively,the DTE is suppressed by the RSFVBA in intervals that are close to the ZCP.By the combination of the two methods,the dependence of the DTE suppression method on currents is reduced and the influences of ZCP are also released without degrading the normal operation performance of MMCs.Moreover,the output performance of MMCs is improved and the voltage stress on the arm inductor dramatically decreases.Finally,the validation of the method is verified by the simulation results with the professional tool Matlab/Simulink.

Background Febrile seizure （FS） is the most common seizure disorders. Approximately one third of children with a febrile seizure have recurrent events. The mechanism of FS remains unclear. Heme oxygenase-1 （HO-1） is a member of the heat shock proteins family and can be induced in the brain by various stresses, including hyperthemia and seizure. This study aimed at investigating the changes of HO-1 in the cortex of rats after recurrent FS. Methods FS in rats was induced ten times, once every 2 days. In a bath of warm water, developing rats were randomly divided into two groups： control group （n=16） and warm water-treated group （n=50）. The latter group was subdivided into hyperthermia group （n=19） and FS group （n=23）. The expression and content of HO-1 mRNA in cortex were observed using in situ hybridization and quantitative reverse transcription-polymerase chain reaction （RT-PCR）. The content of HO- 1 protein in cortex was measured using Western blotting. Results HO-1 mRNA expression of cortex neurons in FS group was markedly increased in comparison with those in hyperthermia and control groups （P=0.00）, however, there was no statistic difference bftween hyperthermia group and control group （P=0.16）. The relative amount of HO- 1 mRNA in cortex in FS group was increased by 53.13% and 96% in comparison with those in hyperthermia group and control group respectively （P=0.00）, but there was no obvious difference between the later two groups （P=0.051）. Western blotting analysis showed that the HO-1 protein content in cortex in FS group was increased by 198% and 246% in comparison with those in hyperthermia group and control group respectively （P=0.00）. There was no obvious difference in HO-1 protein content between the later two groups （P=0.09）. Conclusions Recurrent FS in rats can cause the increase of HO-1 mRNA and protein in cortex which may be involved in the mechanism of FS. The short-time recurrent hyperthermia can not induce the increase of HO-1 mRNA and protein.

Rapid and accurate determination of compressor characteristic maps is essential for the initial design of centrifugal compressors in aircraft power systems. The accuracy of existing methodologies, which rely on combinations of loss models, varies significantly depending on the compressor's geometry and operational range. This variance necessitates substantial experimental or Computational Fluid Dynamics(CFD) data for coefficient calibration. To address this challenge, this study presents an axisymmetric characteristic model for compressor performance assessment. This model incorporates the factors of blade angle, meridional passage area, and the radial deflection angle of meridional streamlines of the compressor. These factors are derived from fundamental aerodynamic equations encompassing mass, momentum, and energy conservation of the compressor. In contrast to conventional one-dimensional approaches, the proposed method reduces the number of loss coefficients and more effectively accounts for the impact of geometric alterations on centrifugal compressor properties. Furthermore, the model reduces dependence on experimental and CFD data. Efficacy of the model is validated using experimental data from four distinct types of centrifugal compressors. Correlation analysis reveals that the model's coefficients can be expressed as functions of the ratio of the Reynolds number to the impeller tip speed. This ratio serves as a characteristic parameter for the design and optimization of centrifugal compressors. Consequently, the proposed method offers an efficient and accurate means for the quick computation of centrifugal compressor characteristics. This is of great significance for improving the efficiency of centrifugal compressors and reducing energy consumption.

BACKGROUND Spontaneous coronary artery rupture(SCAR)is a rare and life-threatening complication after lung cancer surgery.We present a case of SCAR following left upper lobectomy,successfully managed through emergency thoracotomy and coronary artery ligation.CASE SUMMARY A 61-year-old male patient underwent left upper lobectomy and mediastinal lymph node dissection for lung cancer.The surgery was performed using singleport video-assisted thoracoscopic surgery,and there were no observed complications during the procedure.However,19 h after surgery,the patient experienced chest discomfort and subsequently developed severe symptoms,including nausea,vomiting,and a drop in blood pressure.Urgent measures were taken,leading to the diagnosis of SCAR.The patient underwent emergency thoracotomy and coronary artery ligation,successfully stopping the bleeding and stabilizing the condition.Despite postoperative complications,the patient made a successful recovery and was discharged from the hospital.CONCLUSION SCAR is a rare but life-threatening complication following lung cancer surgery.Immediate thoracotomy has been shown to be a life-saving measure,while stenting is not the preferred initial approach.

BACKGROUND Gastric cancer(GC)is the fifth most common and the fourth most lethal malignant tumour in the world.Most patients are already in the advanced stage when they are diagnosed,which also leads to poor overall survival.The effect of posto-perative adjuvant chemotherapy for advanced GC is unsatisfactory with a high rate of distant metastasis and local recurrence.AIM To investigate the safety and efficacy of a programmed cell death 1(PD-1)inhibitor combined with oxaliplatin and S-1(SOX)in the treatment of Borrmann large type III and IV GCs.METHODS A retrospective analysis(IRB-2022-371)was performed on 89 patients with Borrmann large type III and IV GCs who received neoadjuvant therapy(NAT)from January 2020 to December 2021.According to the different neoadjuvant treatment regimens,the patients were divided into the SOX group(61 patients)and the PD-1+SOX(P-SOX)group(28 patients).RESULTS The pathological response(tumor regression grade 0/1)in the P-SOX group was significantly higher than that in the SOX group(42.86%vs 18.03%,P=0.013).The incidence of ypN0 in the P-SOX group was higher than that in the SOX group(39.29%vs 19.67%,P=0.05).The use of PD-1 inhibitors was an independent factor affecting tumor regression grade.Meanwhile,the use of PD-1 did not increase postoperative complications or the adverse effects of NAT.CONCLUSION A PD-1 inhibitor combined with SOX could significantly improve the rate of tumour regression during NAT for patients with Borrmann large type III and IV GCs.

Degradation of Rhodamine B by MnFe-LDH/PMS/O_(3) three-phase catalytic system: performance, mechanism and ecotoxicity studies

This study developed a novel MnFe-LDH/PMS/O3 three-phase catalytic system to degrade the organic dye RhB, which was used to address the drawbacks of persulfate oxidation and ozonation techniques. The structure, ionic and elemental composition, specific surface area, and magnetic properties of the LDHs were investigated using a variety of physicochemical characterization tools. The results showed that MnFe-LDH had a large specific surface area, a rich crystalline phase composition, and a functional group structure. The RhB degradation rate of MnFe-LDH/PMS/O3 was 0.34 min−1, which was much higher than that of other comparative systems. RhB could be completely degraded in 10 min after optimization and had a significant effect on TOC removal. The system was found to be effective over a wide pH range. Common anions were largely unaffected and humic acid acted as an inhibitor. At the same time, the system had generally effective degradation performance for different dyes. Combined with quenching experiments and EPR, it was found that SO4•−, •OH, O_(2)•−, and 1O_(2) all participated in the reaction, and •OH contributed more. The degradation pathway of RhB was derived by LC-MS, and the T.E.S.T. evaluation found that the toxicity of the intermediate product was significantly reduced. Finally, the stability and availability of LDHs were verified using cycling experiments and metal ion leaching. This work provides a theoretical basis and data support for the synergistic catalysis of PMS/O3 and the deep treatment of dye wastewater.

Dear Editor,The choice of surgical intervention for nanophthalmos patients with secondary angle-closure glaucoma poses a challenging decision for ophthalmologists.Because nanophthalmos patients who have undergone filtration surgery or other intraocular procedures are more susceptible to severe complications,including choroidal effusion,malignant glaucoma,and explosive choroidal hemorrhage,all of which can lead to vision loss[1].This dilemma is particularly pronounced in patients with shorter axial lengths.Micropulse transscleral laser therapy(M-TLT),also known as micropulse transscleral cyclophotocoagulation(MP-TSCPC),is a nonincisional laser therapy surgery for glaucoma[2].In 2015,Golan and Kurtz[3]were the first to report four patients with secondary angle-closure glaucoma in nanophthalmos with axial lengths ranging between 17 and 19 mm(mean 18 mm)who were successfully treated with M-TLT,and choroidal detachment was observed in all patients(recovery spontaneously in two patients and recovery after systemic steroid treatment in the other two),of which two had a slight decrease in visual acuity.

Metal surfaces play a crucial role in numerous applications,from self-cleaning and anti-icing to anti-fogging and oil-water separation.The regulation of their wettability is essential to enhance their performance in these areas.This paper proposes a multi-state regulation method for metal surface wettability,leveraging nanosecond laser ablation.By creating non-uniform microstructures on a metal surface,the contact area between the solid and liquid phases can be increased,resulting in the attainment of superhydrophilic properties(contact angle(CA),ranging from 4.6°to 8.5°).Conversely,the construction of uniform microstructures leads to a decreased solid-liquid contact area,thereby rendering the metal surface hydrophilic(CA=12.2°–53°).Furthermore,through heat treatment on a surface with uniform microstructures,organic matter adsorption can be promoted while simultaneously reducing surface energy.This process results in the metal surface acquiring hydrophobic properties(CA=92.1°–133.5°),facilitated by the“air cushion effect.”Building on the hydrophobic surface,stearic acid modification can further reduce surface energy,ultimately bestowing the metal surface with superhydrophobic properties(CA=150.1°–152.7°,and sliding angle=3.8°).Performance testing has validated the durability and self-cleaning effectiveness of the fabricated superhydrophobic surface while also highlighting the excellent anti-fog performance of the superhydrophilic surface.These findings strongly indicate the immense potential of these surfaces in various engineering applications.
