pH may act as a crucial signal in both animal and plant cells. It is very difficult to monitor pH signals and this has largely hindered progress in the investigation of pH signaling, particulady systematic pH signaling. Here, we report the development of a confocal technique to monitor leaf apoplastic pH in intact plants, which is particularly suitable for the studies on root to shoot signaling. A variety of different pH indicators and plant species were tested. It was found that different pH indicators, for example, 2＇,7＇-Bis-（2-carboxyethyl）-5-（and-6）-carboxyfluoresce （BCECF）, SNARF-4F 5-（and-6）- carboxylic acid （SNARF） and DM-NERF （NERF）, were of different properties, and to successfully monitor pH at a sub-cellular level, the comparability between the pH indicator and plant species must be involved according to their suitable pH range and loading characteristics. The loading characteristics of different pH indicators differ with different plant species, cell types and their developing stages. No matter what methods were adopted, BCECF and SNARF could not be loaded specifically in the leaf apoplast in sunflower, tomato, and Comelina communis L. In contrast, regardless of the methods adopted, NERF could be loaded efficiently and specifically in the leaf apoplast in C. communis, but not in other plants. In C. communis, the determination coefficient for in vitro and in situ calibration of NERF was very high, which was respectively 0.9951 and 0.991 6, and therefore, the adoption of NERF together with C. communis could construct an ideal experimental system that is suitable for the investigation of pH systematic signaling. Ratio image analysis demonstrated that the leaf apoplastic pH was about 5.5 in non-stressed conditions, and water deficit could trigger an increase in pH by about half a pH unit, which is the first evidence to directly indicate that pH is able to act as a systematic signal under water deficit conditions.

High strength steels exhibit superior mechanical properties due to the unique microstructure,which successfully solves the drawback of the inevitable strength-toughness trade-off that occurs in traditional *** we investigated the effect of matrix and precipitates on mechanical properties of Cr-Ni-Mo-V/Nb steel after water quenching and tempering(150-500℃).The results showed that the microstructure of the present steel is noticeably tuned by changing the tempering *** excellent combination of strength(a yield strength of 1308 MPa with a total elongation of 8.2%)and toughness(Charpy V-notch impact toughness of 40.5 J/cm^(2))is obtained upon tempering at 200℃.This is attributed to the lath martensite containing high dislocation density,the martensite-twin substructure,and the strengthening effects of the precipitated needle-likeε-carbides and spherical VC *** acicularε-carbides are replaced by the rod-shaped Fe_(3)C at the tempering temperature of 350℃,resulting in the remarkable deterioration in strength,hardness,and *** carbides formed at a tempering temperature of 500℃ are beneficial to the enhancement of the elongation and toughness,but the strength decreases due to the matrix softening caused by the recovery of dislocation.

Hydroformylation could convert olefins and syngas （mixture of H2and CO） into aldehydes with a great atomic *** transformation serves as one of the most prominent processes in modern chemical industry, as it provides over 10million tons of aldehydes as feedstock for our daily essential products such as plasticizers, paints, solvents, and pharmaceuticals. This process generally requires efficient yet robust catalyst to realize decent efficacy. Compared with metals such as Co, Ru, and Pd, rhodium （Rh）-based catalysts stand out due to its superior activity even under mild reaction conditions, and dominate over 70%of global hydroformylation processes.

The RING-type E3 ligase OsBBI1 regulates rice resistance against Magnaporthe oryzae through modifying cell wall defenses. In this study, we report the function of an OsBBI1 substrate, eukaryotic translation initiation factor OseIF5A4, in rice immunity. OsBBI1 interacts with OseIF5A4 and other four members of the OseIF5A family. The RING domain in OsBBI1 and the eIF-5a domain in OseIF5A4 are critical for the OsBBI1-OseIF5A4 interaction. OsBBI1 ubiquitinates OseIF5A4 and mediates its degradation in vitro and in vivo. Moreover, the expression of OseIF5A4 was upregulated during early stage of compatible interaction but downregulated in incompatible interaction between rice and M. oryzae. Knockout of OseIF5A4 enhances rice immunity against M. oryzae and Xanthomonas oryzae pv. oryzae, boosts pattern-triggered immune responses, and strengthens pathogen-induced defense responses (e.g., expression of defense genes, accumulation of reactive oxygen species and reinforcement of cell wall). However, overexpression of OseIF5A4 attenuates rice immunity and immune responses. These results demonstrate that OseIF5A4, a substrate of the immunity-associated E3 ligase OsBBI1, negatively regulates rice immunity against M. oryzae and X. oryzae pv. oryzae through modulating pathogen-induced defense responses, highlighting the importance of the protein translational machinery in rice immunity. © 2025 Crop Science Society of China and Institute of Crop Science, CAAS
