GenOrigin: A comprehensive protein-coding gene origination database on the evolutionary timescale of life

The origination of new genes contributes to the biological diversity of life. New genes may quickly build their network, exert important functions, and generate novel phenotypes. Dating gene age and inferring the origination mechanisms of new genes, like primate-specific genes, is the basis for the functional study of the genes. However, no comprehensive resource of gene age estimates across species is available. Here,we systematically date the age of 9,102,113 protein-coding genes from 565 species in the Ensembl and Ensembl Genomes databases, including 82 bacteria, 57 protists, 134 fungi, 58 plants, 56 metazoa, and 178 vertebrates, using a protein-family-based pipeline with Wagner parsimony algorithm. We also collect gene age estimate data from other studies and uniformly distribute the gene age estimates to time ranges in a million years for comparison across studies. All the data are cataloged into Gen Origin(http://***/), a user-friendly new database of gene age estimates, where users can browse gene age estimates by species, age, and gene ontology. In Gen Origin, the information such as gene age estimates,annotation, gene ontology, ortholog, and paralog, as well as detailed gene presence/absence views for gene age inference based on the species tree with evolutionary timescale, is provided to researchers for exploring gene functions.

A novel shear-deformation technique, named 'shear pressing' (SP), was developed for fabrication of plate-shaped fine grained metallic materials. The principle of SP is that a material is subjected to shear deformation by utilizing pressing with inclined plane dies. A micrometer order grain structure was obtained in an Al-Mg-Li alloy at strain of e = -2.3 by utilizing this technique. The grain refinement sequences during pressing were examined by electron backscatter diffraction. The enhancement of grain refinement to the Al-Mg-Li alloy was compared with plane strain compression (PSC) at similar strains. The effect of the shear strain on the accelerated grain refining during compressing has been discussed.

BACKGROUND Primary squamous cell carcinoma(SCC)of the renal parenchyma is extremely rare,with only nine cases *** SUMMARY This study reports a 51-year-old man with primary SCC of the renal *** patient was admitted with recurrent dull pain and discomfort in the right lumbar region,which had worsened over 2 weeks,accompanied by painful gross *** antigen(SCCA)levels were elevated,and imaging revealed a renal mass with associated *** patient underwent laparoscopic unilateral nephrectomy and lymph node *** pathology confirmed highly differentiated SCC with necrosis in the right renal parenchyma,with negative renal pelvis and *** pathological stage was *** months after surgery,the tumor recurred with involvement of the liver,right psoas major muscle,and inferior vena *** patient refused chemotherapy and succumbed to the disease 6 months postoperatively due to disease *** We report a case of primary SCC of the renal parenchyma,a rare renal *** clinical symptoms,laboratory tests,and imaging findings are nonspecific,making accurate and timely diagnosis *** to the literature,for patients with renal calculi accompanied by a renal mass,elevated serum SCCA levels,and magnetic resonance imaging showing cystic or cystic-solid masses within the kidney with pseudocapsules and heterogeneous mild enhancement,the possibility of this disease should be considered.

Morphological alterations in dendritic spines have been linked to changes in functional communication between neurons that affect learning and ***-4 KIF21A helps organize the microtubule-actin network at the cell cortex by interacting with KANK1;however,whether KIF21A modulates dendritic structure and function in neurons remains *** this study,we found that KIF21A was distributed in a subset of dendritic spines,and that these KIF21A-positive spines were larger and more structurally plastic than KIF21A-negative ***,the interaction between KIF21A and KANK1 was found to be critical for dendritic spine morphogenesis and synaptic *** of either KIF21A or KANK1 inhibited dendritic spine morphogenesis and dendritic branching,and these deficits were fully rescued by coexpressing full-length KIF21A or KANK1,but not by proteins with mutations disrupting direct binding between KIF21A and KANK1 or binding between KANK1 and *** down KIF21A in the hippocampus of rats inhibited the amplitudes of long-term potentiation induced by high-frequency stimulation and negatively impacted the animals’cognitive *** together,our findings demonstrate the function of KIF21A in modulating spine morphology and provide insight into its role in synaptic function.

目的建立双酚A(BPA)暴露的昆明雌鼠动物模型,研究暴露后子宫内膜胞饮突的形态学改变以及HOXA-10基因在mRNA水平和蛋白水平表达的改变,探讨BPA对子宫内膜容受性的影响及可能机制。方法将昆明雌鼠随机分成4组:高剂量暴露组、中剂量暴露组、低剂量暴露组和空白对照组。每组以花生油为溶剂,分别按250 mg/(kg·d),50 mg/(kg·d),10 mg/(kg·d),0 mg/(kg·d)进行BPA灌胃染毒,共28 d,第29天腹腔注射人绝经期促性腺激素(HMG)7.5 IU,48 h后腹腔注射h CG 10 IU,于当晚雌雄昆明小鼠1∶1合笼,次日可见阴道栓者为孕0 d,孕5 d后断椎处死,取出子宫,采用扫描电镜观察子宫内膜表面的胞饮突的形态学表现,应用Western Blot、免疫组化和荧光定量PCR测定HOXA-10的表达情况。结果扫描电镜结果提示,各剂量BPA干预组着床期胞饮突的数目均比对照组少,分布松散且不均,形态不一。HE染色显示,BPA干预组子宫宫腔均较对照组大,子宫内膜上皮细胞的高度较低,排列紧密,间质细胞连接紧密,腺体数量较少。荧光定量PCR、免疫组化和Western Blot结果提示,BPA干预组HOXA-10基因在mRNA及蛋白水平的表达量均较对照组降低(P<0.05)。结论通过对昆明雌鼠进行BPA的慢性暴露发现,BPA对小鼠胚胎着床期胞饮突的形成表现为抑制作用,同时抑制HOXA-10基因的表达。BPA可能通过改变子宫内膜容受性干扰胚胎着床。
