Intestinal dysbiosis in pediatric Crohn's disease patients with IL10RA mutations

BACKGROUND Several studies have employed animal models to explore the association between microbiota and interleukin(IL) 10 signaling;however,limited information is available about the human *** To characterize the microbiome in patients with IL10 RA mutations and to explore the association between gut dysbiosis and disease *** Fecal samples were collected from patients who were diagnosed with loss-offunction mutations in the IL10 RA gene between January 2017 and July 2018 at the Children’s Hospital of Fudan ***-matched volunteer children were recruited as healthy *** with Crohn’s disease(CD) were used as disease controls to standardize the antibiotic *** DNA was extracted from the fecal *** analyses were based on the 16 S rRNA gene sequencing *** Seventeen patients with IL10 RA mutations(IL10 RA group),17 patients with pediatric CD, and 26 healthy children were *** patients with IL10 RA mutations and those with CD exhibited a reduced diversity of gut microbiome with increased *** relative abundance of Firmicutes was substantially increased in the IL10 RA group(P=0.02).On further comparison of the relative abundance of taxa between patients with IL10 RA mutations and healthy children,13 taxa showed significant *** IL10 RA-specific dysbiosis indices exhibited a significant positive correlation with weighted pediatric CD activity index and simple endoscopic score for *** In patients with IL10 RA mutations and early onset inflammatory bowel disease,gut dysbiosis shows a moderate association with disease severity.

AIM: To investigate multiple polyps in a Chinese PeutzJeghers syndrome(PJS) infant. METHODS: A nine-month-old PJS infant was admitted to our hospital for recurrent prolapsed rectal polyps for one month. The clinical characteristics, a colonoscopic image, the pathological characteristics of the polyps and X-ray images of the intestinal perforation were obtained. Serine threonine-protein kinase 11(STK11) gene analysis was also performed using a DNA sample from this ***: Here we describe the youngest known Chinese infant with PJS. Five polyps, including a giant polyp of approximately 4 cm × 2 cm in size, were removed from the infant's intestine. Laparotomy was performed to repair a perforation caused by pneumoperitoneum. The pathological results showed that this child had PJS. Molecular analysis of the STK11 gene further revealed a novel frameshift mutation(c.64_65het_del AT) in exon 1 in this PJS ***: The appropriate treatment method for multiple polyps in an infant must be carefully considered. Our results also show that the STK11 gene mutation is the primary cause of PJS.

Quantifying the Tension between Cosmological Models and JWST Red Candidate Massive Galaxies

We develop a Python tool to estimate the tail distribution of the number of dark matter halos beyond a mass threshold and in a given volume in a *** code is based on the extended Press-Schechter model and is computationally efficient,typically taking a few seconds on a personal laptop for a given set of cosmological *** high efficiency of the code allows a quick estimation of the tension between cosmological models and the red candidate massive galaxies released by the James Webb Space Telescope,as well as scanning the theory space with the Markov Chain Monte Carlo *** an example application,we use the tool to study the cosmological implication of the candidate galaxies presented in Labbéet *** standard Λcold dark matter(ΛCDM)model is well consistent with the data if the star formation efficiency can reach~0.3 at high *** a low star formation efficiency ε~0.1,theΛCDM model is disfavored at~2σ-3σconfidence level.

AIEgens can serve as an effective platform for the construction of photosensitizer-based immunogenic cell death(ICD)*** date,several mitochondria or endoplasmic reticulum(ER)-targeted aggregationinduced emission(AIE)molecules have been developed and have evoked massive ICD in ***,due to the complex physicochemical environment in cells,these small AIE molecules cannot maintain a stable aggregate state,which not only affects the fluorescence intensity of the photosensitizer but also decreases the generation of reactive oxygen species(ROS),and thus reducing the effect of the photosensitizer to elicit ***-based nanomicelles,which maintain a stable micellar structure,can prevent defects of AIE molecules in photodynamic therapy(PDT)***,in this study,a mitochondria-targeted AIE nanophotosensitizer was synthesized and used as a highly potent ICD inducer for vaccine preparation and tumor prevention.

Lung cancer is a prevalent malignancy,and fatalities of the disease exceed 400,000 cases *** squamous cell carcinoma(LUSC)has been recognized as the most common pathological form of lung *** comprehensive understanding of molecular features related to LUSC progression has great significance in LUSC prognosis assessment and clinical *** this study,we aim to identify a panel of signature genes closely associated with LUSC,which can provide novel insights into the progression of *** expression profiles were retrieved from public resources including gene expression omnibus(GEO)and the cancer genome atlas(TCGA)*** expressed genes(DEGs)between LUSC specimens and normal lung tissues were identified by bioinformatics analyses.A total of 66 DEGs were identified based on two cohorts of *** plugin of Cytoscape software was utilized for the further analyses of the top 10 candidate hub genes including OGN,ABI3BP,MAMDC2,FGF7,FAM107A,SPARCL1,DCN,COL14A1,and MFAP4 and CHRDL1,which showed significant downregulation in *** LUSC cell lines were used to validate the functions of CHRDL1 and FAM107A through overexpression ***,our data revealed novel candidate tumor-suppressor genes in LUSC,suggesting previously unappreciated mechanisms in the progression of LUSC.

Solar-powered interfacial evaporation is an energy-efficient solution for water *** requires solar absorbers to facilitate upward water transport and limit the heat to the surface for efficient ***,downward salt ion transport is also desired to prevent salt ***,achieving simultaneously fast water uptake,downward salt transport,and heat localization is challenging due to highly coupled water,mass,and thermal ***,we develop a structurally graded aerogel inspired by tree transport systems to collectively optimize water,salt,and thermal *** arched aerogel features root-like,fan-shaped microchannels for rapid water uptake and downward salt diffusion,and horizontally aligned pores near the surface for heat localization through maximizing solar absorption and minimizing conductive heat *** structural characteristics gave rise to consistent evaporation rates of 2.09 kg m^(-2) h^(-1) under one-sun illumination in a 3.5 wt%NaCl solution for 7 days without *** in a high-salinity solution of 20 wt%NaCl,the evaporation rates maintained stable at 1.94 kg m^(-2) h^(-1) for 8 h without salt crystal *** work offers a novel microstructural design to address the complex interplay of water,salt,and thermal transport.

Delayed and nonhealing of diabetic wounds imposes substantial economic burdens and physical pain on *** stem cells(MSCs)promote diabetic wound *** when MSCs aggregate into multicellular spheroids,their therapeutic effect is ***,traditional culture platforms are inadequate for the efficient preparation and delivery of MSC spheroids,resulting in inefficiencies and inconveniences in MSC spheroid *** this study,a three-dimensional porous nanofibrous dressing(NFD)is prepared using a combination of electrospinning and homogeneous *** thermal crosslinking,the NFD not only achieves satisfactory elasticity but also maintains notable *** the design of its structure and chemical composition,the NFD allows MSCs to spontaneously form MSC spheroids with controllable sizes,serving as MSC spheroid delivery systems for diabetic wound *** importantly,MSC spheroids cultured on the NFD exhibit improved secretion of vascular endothelial growth factor,basic fibroblast growth factor,and hepatocyte growth factor,thereby accelerating diabetic wound *** NFD provides a competitive strategy for MSC spheroid formation and delivery to promote diabetic wound healing.

Let(Zn)be a supercritical branching process with immigration in an independent and identically distributed random *** necessary moment conditions,we show the exact convergence rate in the central limit theorem on log Zn and establish the corresponding local limit theorem by using the moments of the natural submartingale and the convergence rates of its *** similar approach and with the help of a change of measure,we also present the so-called integrolocal theorem and integral large deviation theorem to characterize the precise asymptotics of the upper large deviations.

Cosmological Constraints on Neutrino Masses in Light of JWST Red and Massive Candidate Galaxies

The overabundance of the red and massive candidate galaxies observed by the James Webb Space Telescope(JWST)implies efficient structure formation or large star formation efficiency at high redshift z~*** the scenario of a low or moderate star formation efficiency,because massive neutrinos tend to suppress the growth of structure of the universe,the JWST observation tightens the upper bound of the neutrino *** A cold dark matter cosmology and a star formation efficiency∈[0.05,0.3](flat prior),we perform joint analyses of Planck+JWST and Planck+BAO+JWST,and obtain improved constraints∑m_(ν)<0.196 eV and ∑m_(ν)+<0.111 eV at 95% confidence level,*** on the above assumptions,the inverted mass ordering,which implies ∑m_(ν)≥0.1 eV,is excluded by Planck+BAO+JWST at 92.7% confidence level.
