NOVA1 promotes SMN2 exon 7 splicing by binding the UCAC motif and increases SMN protein expression

Spinal muscular atrophy(SMA)is a rare hereditary neuromuscular disease with a high lethality rate in infants.Variants in the homologous genes survival of motor neuron(SMN)1 and SMN2 have been reported to be SMA pathogenic factors.Previous studies showed that a highinclusion rate of SMN2 exon 7 increased SMN expression,which in turn reduced the severity of SMA.The inclusion rate of SMN2 exon 7 was higher in neural tissues than in non-neural tissues.Neuro-oncological ventral antigen(NOVA)is a splicing factor that is specifically and highly expressed in neurons.It plays a key role in nervous system development and in the induction of nervous system diseases.Howeve r,it remains unclear whether this splicing factor affects SMA.In this study,we analyzed the inclusion of SMN2 exon 7 in different tissues in a mouse model of SMA(genotype smn^(-/-)SMN2^(2 tg/0))and litter mate controls(genotype smn^(+/-)SMN2^(2 tg/0)).We found that inclusion level of SMN2 exon 7 was high in the brain and spinal co rd tissue,and that NOVA1 was also highly expressed in nervous system tissues.In addition,SMN2 exon 7 and NOVA1 were expressed synchronously in the central nervous system.We further investigated the effects of NOVA1 on disease and found that the number of neurons in the anterior horn of spinal cord decreased in the mouse model of SMA during postnatal days 1-7,and that NOVA1 expression levels in motor neurons decreased simultaneously as spinal muscular atrophy developed.We also found that in vitro expression of NOVA1 increased the inclusion of SMN2 exon 7 and expression ofthe SMN2 protein in the U87 MG cell line,whereas the opposite was observed when NOVA1 was knocked down.Finally,point mutation and RNA pull-down showed that the UCAC motif in SMN2 exon 7 plays a critical role in NOVA1 binding and promoting the inclusion of exon 7.Moreove r,CA was more essential for the inclusion of exon 7 than the order of Y residues in the motif.Collectively,these findings indicate that NOVA1 intera cts with the UCAC motif in exon 7 of SMN2,there

From basic to clinical:Anatomy of Denonvilliers’fascia and its application in laparoscopic radical resection of rectal cancer

The total mesorectal excision(TME)approach has been established as the gold standard for the surgical treatment of middle and lower rectal cancer.This approach is widely accepted to minimize the risk of local recurrence and increase the long-term survival rate of patients undergoing surgery.However,standardized TME causes urogenital dysfunction in more than half of patients,thus lowering the quality of life of patients.Of note,pelvic autonomic nerve damage during TME is the most pivotal cause of postoperative urogenital dysfunction.The anatomy of the Denonvilliers’fascia(DVF)and its application in surgery have been investigated both nationally and internationally.Nevertheless,controversy exists regarding the basic to clinical anatomy of DVF and its application in surgery.Currently,it is a hotspot of concern and research to improve the postoperative quality of life of patients with rectal cancer through the protection of their urinary and reproductive functions after radical resection.Herein,this study systematically describes the anatomy of DVF and its appli-cation in surgery,thus providing a reference for the selection of surgical treatment modalities and the enhancement of postoperative quality of life in patients with middle and low rectal cancer.

Meta-analysis of robotic and laparoscopic surgery for treatment of rectal cancer

AIM: To conduct a meta-analysis to determine the relative merits of robotic surgery (RS) and laparoscopic surgery (LS) for rectal cancer. METHODS: A literature search was performed to identify comparative studies reporting perioperative outcomes for RS and LS for rectal cancer. Pooled odds ratios and weighted mean differences (WMDs) with 95% confidence intervals (95% CIs) were calculated using either the fixed effects model or random effects model. RESULTS: Eight studies matched the selection criteria and reported on 661 subjects, of whom 268 underwent RS and 393 underwent LS for rectal cancer. Compared the perioperative outcomes of RS with LS, reports of RS indicated favorable outcomes considering conversion(WMD: 0.25; 95% CI: 0.11-0.58; P = 0.001). Meanwhile, operative time (WMD: 27.92, 95% CI: -13.43 to 69.27; P = 0.19); blood loss (WMD: -32.35, 95% CI: -86.19 to 21.50; P = 0.24); days to passing flatus (WMD: -0.18, 95% CI: -0.96 to 0.60; P = 0.65); length of stay (WMD: -0.04; 95% CI: -2.28 to 2.20; P = 0.97); complications (WMD: 1.05; 95% CI: 0.71-1.55; P = 0.82) and pathological details, including lymph nodes harvested (WMD: 0.41, 95% CI: -0.67 to 1.50; P = 0.46), distal resection margin (WMD: -0.35, 95% CI: -1.27 to 0.58; P = 0.46), and positive circumferential resection margin (WMD: 0.54, 95% CI: 0.12-2.39; P = 0.42) were similar between RS and LS. CONCLUSION: RS for rectal cancer is superior to LS in terms of conversion. RS may be an alternative treatment for rectal cancer. Further studies are required.

Effects of Different Serum on in vitro Cultured Human Granulosa Cells

Objective To evaluate the effects of different sera on the growth of human granulosa cells （GCs） cultured in vitro. Methods GCs were obtained from women who underwent follicular aspirates during in vitro fertilization （IVF） program. Five groups were divided according to media supple- mented with different types of sera. Group A, 10% heat-inactivated fetal bovine serum （FBS）; group B, 10% heat-inactivated newborn bovine serum; group C, 10% non heat-inactivated FBS; group D, 10% human serum; group E, bovine serum albumin. Morphological characteristics and viability of GCs measured by trypan blue exclusion assay were evaluated after 24 h of incubation.Results GCs cultured in group A and group B showed multiform morphology compared with other groups. GCs cultured in group D and group E were present with cytoplasmic atrophy and less pseudopodium. Moreover, group A and group B showed a similar level in the viability of GCs （P〉0.05）, which displayed no difference between group D and group E as well （P〉0.05）. Group C had a lower level of viability than group A （P〈0.05） but a higher level than group D （P〈0.01）. Conclusion Heat-inactivated sera can improve the growth of GCs. Different types of sera would have different effects on the growth of GCs cultured in vitro. The pre-culture with different types of sera should be performed to get better efficacy.

False esophageal hiatus hernia caused by a foreign body: A fatal event

Foreign body ingestion is a common complaint in gastrointestinal clinics. It is usually not difficult to diagnose because most of the patients report a definitive history of accidental foreign body ingestion. However, in rare cases, patients do not have a clear history. Thus, the actual condition of the patient is difficult to diagnosis or is misdiagnosed; consequently, treatment is delayed or the wrong treatment is administered, respectively. This report describes a fatal case of esophageal perforation caused by an unknowingly ingested fishbone, which resulted in lower esophageal necrosis, chest cavity infection, posterior mediastinum fester, and significant upper gastrointestinal accumulation of blood. However, his clinical symptoms and imaging data are very similar with esophageal hiatal hernia. Unfortunately, because the patient was too late in consulting a physician, he finally died of chest infection and hemorrhage caused by thoracic aortic rupture. First, this case report underlines the importance of immediate consultation with a physician as soon as symptoms are experienced so as not to delay diagnosis and treatment, and thus avoid a fatal outcome. Second, diagnostic imaging should be performed in the early stage, without interference by clinical judgment. Third, when computed tomography reveals esophageal hiatus hernia with stomach incarceration, posterior mediastinal hematoma, and pneumatosis caused by esophageal, a foreign body should be suspected. Finally, medical professionals are responsible for making people aware of the danger of foreign body ingestion, especially among children, those who abuse alcohol, and those who wear dentures, particularly among the elderly, whose discriminability of foreign bodies is decreased, to avoid dire consequences.

Granular Ionogel Particle Inks for 3D Printed Tough and Stretchable Ionotronics

Ionogels have garnered great attention as promising soft conducting materials for the fabrication of flexible energy storage devices,soft actuators,and ionotronics.However,the leakage of the ionic liquids,weak mechanical strength,and poor manufacturability have greatly limited their reliability and applications.Here,we propose a new ionogel synthesis strategy by utilizing granular zwitterionic microparticles to stabilize ionic liquids.The ionic liquids swell the microparticles and physically crosslink microparticles via either electronic interaction or hydrogen bonding.Further introducing a photocurable acrylic monomer enables the fabrication of double-network(DN)ionogels with high stretchability(>600%)and ultrahigh toughness(fracture energy>10 kJ/m^(2)).The synthesized ionogels exhibit a wide working temperature of−60 to 90℃.By tuning the crosslinking density of microparticles and physical crosslinking strength of ionogels,we synthesize DN ionogel inks and print them into three-dimensional(3D)motifs.Several ionogelbased ionotronics are 3D printed as demonstrations,including strain gauges,humidity sensors,and ionic skins made of capacitive touch sensor arrays.Via covalently linking ionogels with silicone elastomers,we integrate the ionogel sensors onto pneumatic soft actuators and demonstrate their capacities in sensing large deformation.As our last demonstration,multimaterial direct ink writing is harnessed to fabricate highly stretchable and durable alternating-current electroluminescent devices with arbitrary structures.Our printable granular ionogel ink represents a versatile platform for the future manufacturing of ionotronics.

A theoretical model for impact protection of flexible polymer material

The relationship between the protective performance of flexible polymer material and material parameters(elasticmodulus,viscosity coefficient)is explored,an impact collision motion equation between two bodies is establishedfrom the viscoelastic material constitutive,and the relationship between the kinematic response and the materialparameters is obtained.Based on the Kelvin constitutive model,a theoretical model for impact between the pro-tective body and the protected body is established,then the dynamic response is obtained.The feasibility of themodel was verified by drop hammer experiment,and the material parameters(elastic modulus,viscosity coeffi-cient)were obtained by formula.The model is discretized and the relationship between local impact response andmaterial parameters is analyzed.The discussion results on the relationship between the impact response and theprotective material performance indicate that adjusting the elastic modulus,viscosity coefficient,and thicknessof the protective material can effectively improve protective effect.

The hydrosilylation of unsaturated carbon-carbon bonds is one of the most critical reactions in silicone industrial production.The homogeneous Speier's catalyst,Karstedt's catalyst,and other noble metal-based catalysts are widely used.However,simplifying the separation of the homogeneous catalyst from reaction products and reducing the high cost of precious metals is still challenging.This review describes the recent development of heterogeneous catalysts for alkene,alkyne,and allene hydrosilylations,which can effectively solve problems in homogeneous hydrosilylation.

Objective Pannexin-1（Panx1）channels are widely expressed in various cell types.Panx1 proteins can function as channels for adenosine triphosphate（ATP）release and Panxl hemichannels in concert with extracellular ATP and P2X7 receptors are involved in several physiological and pathological conditions such as inflammasome activation and macrophage chemotaxis activation of T cells.Recent evidence suggests ATP-gated P2X7 receptors areinvolved in inflammatory bowel diseases（IBD）.However the role of PANX1 in IBD is unknown.In the present study we investigated the role and the underlying mechanisms of PANX-1 in the development of IBD including its relationship with P2X7R.Methods:In our study animal behavior tests in the normal C57mice and PANX1 wildtypc and knockout mice immunohistochemical staining and western blotting were used to determine the effect and the possible underlying mechanisms of PANXlin mice with DSS-induccd colitis.Results:（1）In DSS-induced chronic colitis in mice the weight loss and DAI score were significantly higherthan that of control.（2）The similar results were observed in PANX1widetypewhile lacked in PANX1 knockoutfemale mice.（3）The expressions of PANX1 and P2X7R in the protein level in colon were significantly higher in colitis mice than that of control mice.（4）Immunohistochemical stainingshowed that P2X7 andPANX1 were both localized in epithelial cells and macrophages in the colon of mice.Conclusion:These results suggest that PANX1may play an important role inDSS-induced chronic colitis in mice with the underlying mechanisms probably related to P2X7R in epithelial cells and macrophages.

Transition metal iron and persistent free radicals(PFRs)both affect the redox properties of biochar,but the electron transfer relationship between them and the coupling reduction mechanism of Cr(Ⅵ)requires further investigation.To untangle the interplay between iron and PFRs in biochar and the infuences on redox properties,FeCl_(3)-modified rice husk biochar(FBCs)was prepared and its reduction mechanism for Cr(Ⅵ)without light was evaluated.The FBCs had higher surface positive charges,oxygen-containing functional groups,and PFRs compared with pristine rice husk biochar(BC).Phenoxyl PFRs with high electrondonating capability formed in biochar.The pronounced electron paramagnetic resonance signals showed that the PFRs preferred to form at lower Fe(Ⅲ)concentrations.While a high concentration of Fe(Ⅲ)would be reduced to Fe(Ⅱ)and consumed the formed PFRs.Adsorption kinetics and X-ray photoelectron spectroscopy analysis indicated that the FBCs effectively enhanced the Cr(Ⅵ)removal efficiency by 1.54-8.20 fold and the Cr(Ⅵ)reduction efficiency by 1.88-9.29 fold compared to those of BC.PFRs quenching and competitive reductant addition experiments revealed that the higher Cr(Ⅵ)reduction performance of FBCs was mainly attributed to the formed PFRs,which could contribute to~74.0%of Cr(Ⅵ)reduction by direct or indirect electron transfer.The PFRs on FBCs surfaces could promote the Fe(Ⅲ)/Fe(Ⅱ)cycle through single electron transfer and synergistically accelerate~52.3%of Cr(Ⅵ)reduction.This study provides an improved understanding of the reduction mechanism of iron-modified biochar PFRs on Cr(Ⅵ)in environments.
