Genetic Engineered Ultrasound-Triggered Injectable Hydrogels for Promoting Bone Reconstruction

Genetic engineering technology can achieve specific gene therapy for a variety of diseases, but the current strategy still has some flaws, such as a complex system, single treatment, and large implantation trauma. Herein, the genetic engineering injectable hydrogels were constructed by ultrasonic technology for the first time to realize in vivo ultrasound-triggered in situ cross-linking and cell gene transfection, and finally complete in situ gene therapy to promote bone reconstruction. First, ultrasound-triggered calcium release was used to activate transglutaminase and catalyze the transamidation between fibrinogen. Simultaneously, liposome loaded with Zinc-finger E-box-binding homeobox 1 (ZEB1) gene plasmid (Lip-ZEB1) was combined to construct an ultrasound-triggered in situ cross-linked hydrogels that can deliver Lip-ZEB1. Second, ultrasound-triggered injectable hydrogel introduced ZEB1 gene plasmid into endothelial cell genome through Lip-ZEB1 sustained release, and then acted on the ZEB1/Notch signal pathway of cells, promoting angiogenesis and local bone reconstruction of osteoporosis through genetic engineering. Overall, this strategy provides an advanced gene delivery system through genetic engineered ultrasound-triggered injectable hydrogels.

Trapped ancient microorganisms in halite fluid inclusions are of special interest to the understanding of biology and ecology in salt lake *** the integration of petrologic,microthermometric,and Raman spectroscopic analyses,this study utilizes fluid inclusions from Chaka Salt Lake,eastern Qaidam Basin,NW China,to assess the possibility of microorganism-trapping by fluid ***,we report that the solid phase of some primary fluid inclusions contains carotenoids,which is interpreted as evidence of Dunaliella algae,and that the coexisting liquid phase comprises SO_(4)^(2-).The homogenization temperatures of single-phase primary fluid inclusions indicate that the precipitation temperature of the Holocene halite in Chaka Salt Lake ranges from 13.5℃ to 36.4℃.This suggests that fluid inclusions in halite are a good medium for trapping and preserving ancient microorganisms and organic matter in salt lakes,and that Raman spectroscopy has good potential to identify halophilic archaea.

Characterization of SARS-CoV-2-specific humoral immunity and its potential applications and therapeutic prospects

Coronavirus disease 2019(COVID-19),caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),is an ongoing pandemic that poses a great threat to human health *** the humoral immune response plays essential roles in disease occurrence and development,understanding the dynamics and characteristics of virus-specific humoral immunity in SARS-CoV-2-infected patients is of great importance for controlling this *** this review,we summarize the characteristics of the humoral immune response after SARS-CoV-2 infection and further emphasize the potential applications and therapeutic prospects of SARSCoV-2-specific humoral immunity and the critical role of this immunity in vaccine ***,serological antibody testing based on the humoral immune response can guide public health measures and control strategies;however,it is not recommended for population surveys in areas with very low *** evidence suggests that asymptomatic individuals have a weaker immune response to SARS-CoV-2 infection,whereas SARS-CoV-2-infected children have a more effective humoral immune response than *** correlations between antibody(especially neutralizing antibody)titers and protection against SARS-CoV-2 reinfection should be further *** addition,the emergence of cross-reactions among different coronavirus antigens in the development of screening technology and the risk of antibody-dependent enhancement related to SARS-CoV-2 vaccination should be given further attention.

The high-frequency(HF)modeling of induction motors plays a key role in predicting the motor terminal overvoltage and conducted emissions in a motor drive *** this study,a physics informed neural network-based HF modeling method,which has the merits of high accuracy,good versatility,and simple parameterization,is *** proposed model of the induction motor consists of a three-phase equivalent circuit with eighteen circuit elements per phase to ensure model *** per phase circuit structure is symmetric concerning its phase-start and phase-end *** symmetry enables the proposed model to be applicable for both star-and delta-connected induction motors without having to recalculate the circuit element values when changing the motor connection from star to delta and vice *** physics knowledge,namely per-phase impedances,are used in the artificial neural network to obtain the values of the circuit *** parameterization can be easily implemented within a few minutes using a common personal computer(PC).Case studies verify the effectiveness of the proposed HF modeling method.

The storage and utilization of carbohydrates in response to elevation mediated by tree organs in subtropical evergreen broad-leaved forests

Global climate change can affect tree growth and carbon sink function by influencing plant carbohydrate synthesis and utilization,while elevation can be used as an ideal setting under natural conditions to simulate climate change *** effect of elevation on tree growth may depend on organ ***,the allocation patterns of nonstructural and structural carbohydrates(NSCs and SCs,respectively)in different tree organs and their response to elevation remain *** selected four dominant tree species,Schima superba,Castanopsis eyrei,Castanopsis fargesii and Michelia maudiae,along an elevation gradient from 609 to 1,207 m in subtropical evergreen broad-leaved forests and analyzed leaf,trunk,and fine root NSCs,carbon(C),nitrogen(N)and phosphorus(P)concentrations and the relative abundance of *** NSCs increased initially and then decreased,and trunk NSCs increased with increasing ***,root NSCs decreased with increasing *** relative abundance of SCs in leaves and trunks decreased,while the relative abundance of root SCs increased with increasing *** significant correlations between SCs and NSCs in leaves were detected,while there were negative correlations between SCs and NSCs in trunks,roots,and all *** partitioning analysis indicated that plant C/N and C/P were the main predictors of changes in SCs and *** results suggest that tree organs have divergent responses to elevation and that increasing elevation will inhibit the aboveground part growth and enhance the root growth of trees.A tradeoff between the C distribution used for growth and storage was confirmed along the elevation gradient,which is mainly manifested in the"sink"organs of *** results provide insight into tree growth in the context of global climate change scenarios in subtropical forest ecosystems.

Geological and Geochemical Characteristics and Exploration Prospect of Coal-Derived Tight Sandstone Gas in China： Case Study of the Ordos, Sichuan, and Tarim Basins

This work extensively investigated global tight sandstone gas, and geologically and geochemically analyzed the tight sandstone gas in China＇s Ordos, Sichuan, and Tarim basins. We compared typical tight sandstone gas in China with that in North America. We proposed six conditions for the formation of China＇s tight sandstone gas, and illustrated the geological characteristics of tight sandstone gas. In China, gas-bearing tight sandstones were mainly deposited in continental lake deltas and marine-terrigenous facies basin environments, associated with coal-measure strata, and were mostly buried deeper than 2000 in under a formation pressure of 20-30 MPa, with pressure coefficients varying from overpressure to negative pressure. In other countries, tight gas bearing sandstones were dominantly deposited in marine to marine-terrigenous facies environments, occurred in coal-measure strata, and were mostly buried shallower than 2000 m in low-pressure systems. We systematically analyzed tight sandstone gas in the Ordos, Sichuan, and Tarim basins in terms of chemical compositions, geochemical characteristics of carbon isotopes, origins, and sources. Tight sandstone gas in China usually has a hydrocarbon content of 〉95%, with CH4 content 〉90%, and a generally higher dry coefficient. In the three above-mentioned large tight sandstone gas regions,δ13C1 and δJ3C2 mainly ranges from -42%o to -28%o and from -28%o to -21%o, respectively. Type III coal-measure source rocks that closely coexist with tight reservoirs are developed extensively in these gas regions. The organic petrology of source rocks and the carbon isotope compositions of gas indicate that tight sandstone gas in China is dominantly coal-derived gas generated by coal-measure strata. Our analysis of carbon isotope series shows that local isotope reversals are mainly caused by the mixing of gases of different maturities and that were generated at different stages. Autophagy,a conserved“self-eating”process for cellular homeostasis,plays a crucial role in the hepatitis B virus(HBV)life cycle,including viral assembly,envelopment,release,and degradation.1 Autophagy can also be hijacked by HBV for its persistence and ***,the association between HBV mutations and autophagy remains ***,we found that an HBV surface antigen(HBsAg)mutation involving sL13H substitution impaired HBsAg expression and caused its abnormal distribution in vitro and in a hydrodynamic injection(HI)-based mouse model for acute HBV *** sL13H mutation also decreased autophagosome formation and inhibited autophagic *** mechanistic analysis revealed that sL13H suppresses HBV-induced autophagy initiation by inhibiting the eukaryotic translation initiation factor 2 alpha kinase 3(EIF2AK3/PERK)eeukaryotic translation initiation factor 2 subunit alpha(EIF2S1/eIF2a)eautophagy related(ATG)5/12 ***,our findings reveal a potential role for HBV mutations in manipulating host autophagic flux for virus persistence and pathogenesis in chronic hepatitis B.
