pAPN基因敲除的IPEC-J2介导的TGEV感染特征分析

旨在探究猪氨基肽酶N(porcine aminopeptidase N,pAPN)基因敲除的猪空肠上皮细胞系(intestinal porcine epithelial cell line J2,IPEC-J2)介导猪传染性胃肠炎病毒(transmissible gastroenteritis virus,TGEV)感染的特征,为深入了解pAPN基因在TGEV感染过程中的作用机制提供理论依据。研究分为pAPN基因敲除IPEC-J2组(IPEC-J2-KO组)、野生型IPEC-J2组(IPEC-J2-WT组)和未接种TGEV的野生型IPEC-J2组(Mock组)3组,每组设置3个重复。首先通过实时荧光定量PCR(quantitative real-time PCR,qPCR)确定IPEC-J2接种TGEV毒株后收取细胞样品的最佳时间节点;其次,对IPEC-J2-KO进行了脱靶效应检测;然后,通过qPCR、蛋白免疫印迹(western blot,WB)、间接免疫荧光分析(indirect immunofluorescence assay,IFA)和50%组织细胞感染量(50%tissue culture infective dose,TCID_(50))对接种TGEV的IPEC-J2-KO、IPEC-J2-WT及Mock进行感染特征分析;最后,通过WB检测IPEC-J2-KO、IPEC-J2-WT和Mock中NF-κB p65及其磷酸化蛋白pp65的表达情况。qPCR结果显示,接毒24 h是收集细胞样本以评估TGEV对IPEC-J2影响的最佳时间节点;脱靶分析结果显示,在IPEC-J2-KO中未检测到脱靶效应;病毒感染特征分析结果显示,与IPEC-J2-WT相比,IPEC-J2-KO内病毒拷贝数、病毒滴度均极显著降低(P0.05),且IPEC-J2-KO内未检测到TGEV-N蛋白的表达;此外,与Mock相比,接种TGEV后,IPEC-J2-WT组NF-κB p65的磷酸化水平极显著上调(P0.05)。本研究表明,IPEC-J2-KO可有效抵抗TGEV的感染,接种TGEV未影响IPEC-J2-KO中先天免疫相关信号通路中转录因子NF-κB的活性。该研究为IPEC-J2作为TGEV感染特征研究的细胞模型提供了理论依据,为阐明pAPN基因在TGEV入侵宿主细胞的机制及抗病猪新品种的研究奠定了基础。

Continuous carbon fiber reinforced lithium aluminosilicate glass ceramics matrix composites(C_f/LAS composites) are joined to Ti60 alloy vacuum brazed using Ti-Zr-Ni-Cu brazing alloy. The effects of the brazing temperature on the interfacial microstructure and mechanical properties of brazed joints are investigated in details. The interfacial microstructure varies apparently with an increase of the brazing temperature. The thicknesses of the banded Ti solid solution(Ti(s, s)) and the reactive layer between Cf/LAS composites and the interlayer grow gradually. The mechanical properties of brazed joints increase firstly and then decrease with an increasing temperature. In addition, a joint that is brazed at 980 °C for 10 min shows the highest shear strength of$38.13 MPa. At the same time, the fracture paths of brazed joints also change as the temperature increases. When the brazing temperature is 950 °C, the fracture position is in the TiC + ZrC +Ti_2O + ZrSi_2+ Ti_5Si_3 layer on the composite side. When the brazing temperature is 980 °C, the fracture position is on the side of the braze seam(Ti, Zr)2(Ni, Cu), Ti_2O + ZrSi_2+ Ti_5Si_3 layer,and carbon fiber in the composite material. When the brazing temperature is 990 °C, the fracture position is in the Ti_2O + ZrSi_2+ Ti_5Si_3 layer on the composite side and the carbon fiber in the composite material.

Improving Chinese Word Representation with Conceptual Semantics

The meaning of a word includes a conceptual meaning and a distributive *** embedding based on distribution suffers from insufficient conceptual semantic representation caused by data sparsity,especially for low-frequency *** knowledge bases,manually annotated semantic knowledge is stable and the essential attributes of words are accurately *** this paper,we propose a Conceptual Semantics Enhanced Word Representation(CEWR)model,computing the synset embedding and hypernym embedding of Chinese words based on the Tongyici Cilin thesaurus,and aggregating it with distributed word representation to have both distributed information and the conceptual meaning encoded in the representation of *** evaluate the CEWR model on two tasks:word similarity computation and short text *** Spearman correlation between model results and human judgement are improved to 64.71%,81.84%,and 85.16%on Wordsim297,MC30,and RG65,***,CEWR improves the F1 score by 3%in the short text classification *** experimental results show that CEWR can represent words in a more informative approach than distributed word *** proves that conceptual semantics,especially hypernymous information,is a good complement to distributed word representation.

Oxide-supported metal single-atom catalysts(SACs)have exhibited excellent catalytic performance for water-gas shift(WGS)***,we report the single-atom catalyst Pt1/FeOx exhibits excellent medium temperature catalytic performance for WGS reactions by the density functional theory(DFT)calculations and experimental *** calculations indicate that H_(2)O molecules are easily dissociated at oxygen vacancies,and the formed*OH and*O are adsorbed on Pt1 single atoms and the adjacent O atoms,*** studying four possible reaction mechanisms,it is found that the optimal WGS reaction pathway is proceeded along the carboxyl mechanism(pathway III),in which the formation of*COOH intermediates can promote the stability of Pt1/FeOx SAC and the easier occurrence of WGS *** energy barrier of the rate-determining step during the entire reaction cycle is only 1.16 eV,showing the high activity for the medium temperature WGS reaction on Pt1/FeOx SAC,which was verified by experimental ***,the calculated turnover frequencies(TOFs)of CO_(2)and H_(2)formation on Pt1/FeOx at 610 K(337℃)can reach up to 1.14×10^(-3)s^(-1)·site^(-1)through carboxyl *** this work,we further expand the application potential of Pt1/FeOx SAC in WGS reaction.

Establishing a quantitative relationship between modern pollen and vegetation remains a pivotal but intricate pursuit within the field of Quaternary *** R-value,a well-established and traditional metric characterized by its simplicity and direct applicability,serves to calibrate the nonlinear disparities between surface pollen and modern *** study entails the construction of a comprehensive pollen R-value dataset for China derived from R-values published between 1987 and *** dataset,compiled after rigorous screening,calibration,and standardization processes,encompasses 898 modern pollen sampling sites and 2115 pollen R-value data entries,encapsulating 152 pollen taxa spanning 65 families and 113 *** the extracted R-values yielded the following insights:(1)A macrospatial analysis validates previously established knowledge at a site-specific ***,pollen R-value variations across China are statistically *** two-thirds of pollen taxa exhibit underrepresentation(e.g.,Taxodiaceae,Corylus,Nitraria,Tamarix,Cyperaceae,Poaceae,and Fabaceae),while the remaining one-third display overrepresentation(e.g.,Pinus,Picea,Betula,Ephedra,Chenopodiaceae,and Artemisia).The degree of underrepresentation surpasses that of overrepresentation,and the representation patterns of Castanea,Quercus,Polygonaceae,and Asteraceae are contingent upon vegetation types.(2)Pollen R-values follow distinct spatial distribution patterns within *** the woody vegetation region of eastern China,R-values progressively decline from north to south,correlating with rising temperatures and ***,in herbaceous vegetation regions of northern and western China,R-values decrease from east to west and from northeast to southwest,corresponding to increased ***,pollen R-values manifest variation contingent on pollen taxa,vegetation types,and climatic regions,even differing for the same taxa across varying

In this study,we systematically investigated the effect of proton concentration on the kinetics of the oxygen reduction reaction(ORR)on Pt(111)in acidic *** results demonstrate a rectangular hyperbolic relationship,i.e.,the ORR current excluding the effect of other variables increases with proton concentration and then tends to a constant *** consider that this is caused by the limitation of ORR kinetics by the trace oxygen concentration in the solution,which determines the upper limit of ORR kinetics.A model of effective concentration is further proposed for rectangular hyperbolic relationships:when the reactant concentration is high enough to reach a critical saturation concentration,the effective reactant concentration will become a constant *** could be due to the limited concentration of a certain reactant for reactions involving more than one reactant or the limited number of active sites available on the *** study provides new insights into the kinetics of electrocatalytic reactions,and it is important for the proper evaluation of catalyst activity and the study of structureperformance relationships.

Asperuloside Promotes Apoptosis of Cervical Cancer Cells through Endoplasmic Reticulum Stress-Mitochondrial Pathway

Objective: To investigate the effects of asperuloside on cervical cancer based on endoplasmic reticulum(ER) stress and mitochondrial pathway. Methods: Different doses(12.5–800 μg/mL) of asperuloside were used to treat cervical cancer cell lines Hela and CaSki to calculate the half maximal inhibitory concentration(IC_(50)) of asperuloside. The cell proliferation was analyzed by clone formation assay. Cell apoptosis, intracellular reactive oxygen species(ROS) and mitochondrial membrane potential were determined by flow cytometry. The protein expressions of cleaved-caspase-3, Bcl-2, Bax, Cyt-c, cleaved-caspase-4 and glucose-regulated protein 78(GRP78) were analyzed by Western blot. And the inhibitor of ER stress, 4-phenyl butyric acid(4-PBA) was used to treat cervical cancer cells to further verify the role of ER stress in the apoptosis of cervical cancer cells induced by asperuloside. Results: Asperuloside of 325, 650, and 1300 μg/mL significantly inhibited the proliferation and promoted apoptosis of Hela and CaSki cells(P<0.01). All doses of asperuloside significantly increased intracellular ROS levels, reduced mitochondrial membrane potential, significantly reduced Bcl-2 protein expression level, and increased Bax, Cyt-c, GRP78 and cleaved-caspase-4 expressions(P<0.01). In addition, 10 mmol/L 4-PBA treatment significantly promoted cell proliferation and reduced apoptosis(P<0.05), and 650 μg/mL asperuloside could reverse 4-PBA-induced increased cell proliferation, decreased apoptosis and cleaved-caspase-3,-4 and GRP78 protein expressions(P<0.05). Conclusion: Our study revealed the role of asperuloside in cervical cancer, suggesting that asperuloside promotes apoptosis of cervical cancer cells through ER stress-mitochondrial pathway.
