Time-course effects of different fiber-rich ingredients on energy values,microbiota composition and SCFA profile in growing pigs

This study was to investigate time-course effects of different types of dietary fiber on the energy values,fecal microbiota and short-chain fatty acid(SCFA)concentration in growing pigs.A total of 24 barrows(initial body weight,19.8±0.5 kg)were assigned to 4 dietary treatments based on body weight(BW)in a completely randomized design,including a basal diet(CON)and 3 fiber-rich diets replacing corn,soybean meal and soybean oil in the CON diet with 20%sugar beet pulp(SBP),defatted rice bran(DFRB)or soybean hull(SBH),respectively.Fresh feces were sampled on d 7,14 and 21,followed by 5 d total feces and urine collections.The results showed that there were no differences in DE and ME between any of the fiber ingredients on d 7,14 or 21.However,fiber inclusion decreased the DE and ME of the diet(P<0.05)regardless of the time effect.Principal coordinate analysis(PCoA)revealed distinctly different microbial communities on the DFRB diet and SBH diet across different times(P<0.05)and the fecal microbiota of the 4 diet groups demonstrated notably distinct clusters at each time point(P<0.05).With adaptation time increased from 7 to 21 d,cellulose-degrading bacteria and SCFA-producing bacteria(e.g.,Ruminococcaceae_UCG-014,Rikenellaceae_RC9_gut_group and Bifidobacterium)increased in the fiber inclusion diets,and pathogenic genera(e.g.,Streptococcus and Selenomonas)were increased in the basal diet(P<0.05).Furthermore,the gut microbiota of growing pigs adapted more easily and quickly to the SBP diet compared to the DFRB diet,as reflected by the concentration of propionate,butyrate,isovalerate and total SCFA which increased with time for growing pigs fed the DFRB diet(P<0.05).Collectively,our results indicated at least 7 d adaptation was required to evaluate the energy values of fiber-rich ingredients,as the hindgut microbiota of growing pigs may need more time to adapt to a high fiber diet,especially for insoluble dietary fiber.

Evaluation of energy values of high-fiber dietary ingredients with different solubility fed to growing pigs using the difference and regression methods

The objective of this study was to compare the energy values of high-fiber dietary ingredients with different solubility(sugar beet pulp[SBP]and defatted rice bran[DFRB])in growing pigs using the difference and the regression methods.A total of 21 barrows(initial BW,40.5±1.2 kg)were assigned to 3 blocks with BW as a blocking factor,and each block was assigned to a 7×2 incomplete Latin square design with 7 diets and two 13-d experimental periods.The 7 experimental diets consisted of a corn-soybean meal basal diet and 6 additional diets containing 10%,20%,or 30%SBP or DFRB in the basal diet,respectively.Each of the exper-imental periods lasted 12 d,with a 7 d dietary adaptation period followed by 5-d total fecal and urine collection.Results showed that the digestible energy(DE)and metabolizable energy(ME)of the SBP determined by the difference method with different inclusion levels(10%,20%,or 30%)were 2,712 and 2,628 kcal/kg,2,683 and 2,580 kcal/kg,and 2,643 and 2,554 kcal/kg DM basis,respectively.The DE and ME in the DFRB evaluated by the difference method with 3 different inclusion levels were 2,407 and 2,243 kcal/kg,2,687 and 2,598 kcal/kg,and 2,630 and 2,544 kcal/kg DM basis,respectively.Different inclusion levels had no effects on the energy values of each test ingredient estimated by the difference method.The DE and ME of the SBP and the DFRB estimated by the regression method were 2,562 and 2,472 kcal/kg and 2,685 and 2,606 kcal/kg DM basis,respectively.The energy values of each ingredient determined by the regression method were similar to the values estimated by the difference method with the 20%or 30%inclusion level.However,the energy values of the SBP and DFRB estimated by the difference method with the 10%inclusion level were inconsistent with the values determined by the regression method(P<0.05).In conclusion,the regression method was a robust indirect method to evaluate the energy values for high-fiber ingredients with different solubility in growing pigs.If the number of experimental animals was lim

Effects of dietary cellulose levels on the estimation of endogenous amino acid losses and amino acid digestibility for growing pigs

Two experiments were conducted to investigate the effects of dietary cellulose levels on the determination of the ileal endogenous losses(IEL) of amino acids(AA), apparent ileal digestibility(AID) and standardized ileal digestibility(SID) of AA in corn-soybean meal diets for growing pigs. In the first experiment, 28 pigs(BW, 45.1 ± 2.0 kg) that were fitted with simple T-cannulas at the distal ileum were fed 4 nitrogen-free diets consisting of 4 dietary cellulose levels(0, 3%, 6% and 9%) in a randomized complete block design. In the second experiment, 28 pigs(BW, 45.6 ± 2.0 kg) fitted with simple Tcannulas at the distal ileum were fed 4 corn-soybean meal diets consisting of 4 dietary cellulose levels(0,3%, 6% and 9%) in a randomized complete block design. There were 7 replicates per diet with 1 pig as a replicate in each treatment. Both experiments consisted of a 7-d adjustment period and a 2-d ileal digesta collection period on d 8 and 9. Chromic oxide was used as an indigestible marker to calculate IEL and digestibility of AA. The results showed that the IEL of AA for growing pigs was not influenced by dietary cellulose supplementation(P > 0.05). The AID of Thr, Ser, Glu, Cys, Ile, Tyr, Phe, Lys and His decreased with increasing cellulose supplementation levels for pigs fed corn-soybean meal diets(P < 0.05). The SID of Thr, Ser, Cys, Val, Ile, Tyr, Phe, Lys and His decreased with increasing cellulose supplementation levels in corn-soybean meal diets(P < 0.05). In summary, dietary cellulose levels had no effect on the estimation of IEL of AA for growing pigs. The AID and SID of most AA in corn-soybean meal diets decreased with increasing levels of dietary cellulose supplementation.

本试验旨在通过构建猪肠道上皮细胞(IPEC-J2细胞)经脂多糖(LPS)刺激后细胞损伤模型,揭示卫矛醇(DUL)对LPS处理后的IPEC-J2细胞屏障受损和炎症损伤的缓解作用及其可能的分子机制。采用细胞计数试剂盒-8(CCK-8)检测IPEC-J2细胞活力,以确定LPS造模浓度(150μg/mL)和最佳DUL处理浓度(200μmol/L)。试验设3个组:对照(CON)组、LPS组和DUL组。IPEC-J2细胞贴壁后,CON组先用完全培养基处理24 h,然后用DMEM培养基处理24 h;LPS组先用完全培养基处理24 h,然后用含有150μg/mL LPS的DMEM培养基处理24 h;DUL组先用含有200μmol/L DUL的完全培养基处理24 h,然后用含有150μg/mL LPS的DMEM培养基处理24 h。观察各组IPEC-J2细胞生长状态,划痕试验评价细胞的迁移和增殖能力,实时荧光定量PCR(RT-qPCR)法和蛋白质免疫印迹法(Western blot)检测屏障和炎症相关基因的mRNA和蛋白相对表达水平。结果表明:1)在IPEC-J2细胞上,确定200μmol/L的DUL为最佳处理浓度,150μg/mL的LPS为有效致炎浓度。2)相较于LPS组,DUL预处理缓解了细胞密度降低、细胞空泡增多、细胞边界不清晰的情况。3)相较于LPS组,DUL预处理显著增加了细胞迁移距离和细胞迁移面积(P<0.05)。4)相较于LPS组,DUL预处理显著升高了屏障相关基因闭锁小带蛋白-1(ZO-1)、闭锁蛋白(OCLN)、封闭蛋白1(CLDN1)的mRNA和蛋白相对表达水平及黏蛋白2(MUC2)、大肿瘤抑制激酶1(LATS1)、YES相关蛋白1(YAP1)的mRNA相对表达水平(P<0.05)。5)相较于LPS组,DUL预处理显著降低了通路相关基因髓样分化因子88(Myd88)、核因子-κB抑制蛋白-α(IκB-α)、核因子-κB(NF-κB)、白细胞介素-1β(IL-1β)的mRNA和蛋白相对表达水平和Toll样受体4(TLR4)的蛋白相对表达水平以及核苷酸结合寡聚化结构域样受体蛋白3(NLRP3)、白细胞介素-18(IL-18)的mRNA表达相对表达水平(P<0.05)。由此可见,DUL通过下调TLR4/Myd88/NF-κB信号通路相关蛋白表达,调节炎症因子分泌,提高紧密连接相关基因表达,缓解LPS引起的IPEC-J2细胞屏障损伤和炎症反应。

Effectiveness and safety of acupuncture in treatment of pregnancyrelated symptoms:a systematic review and Meta-analysis

OBJECTIVE:To systematically evaluate the efficacy and safety of acupuncture(AM)in the treatment of pregnancyrelated symptoms such as acute vomiting during pregnancy.METHODS:We comprehensively searched the available literature up to November 2021,including Pub Med,Embase,Cochrane Library,Chinese Biomedical Literature Database,and China National Knowledge Infrastructure Database,for randomized controlled trials(RCTs)on AM for the treatment of severe vomiting,insomnia,pharyngeal and pelvic pain,mood abnormalities,and dyspepsia during pregnancy.RESULTS:Sixteen RCTs with a cumulative sample size of 1178 cases were included.Of these,964 patients were included in the Meta-analysis.The Meta-analysis results showed that AM was more efficient than Western medicine in treating discomfort during pregnancy[odds ratio(OR)=1.19,95%confidence interval(CI)(1.11,1.28),P<0.01].AM was better than the control group in improving the visual analog scale scores[standard mean difference(SMD)=0.62,95%CI(0.53,0.71),P<0.01].AM was superior to the control group in improving Numerical Rating Scale(NRS)symptom scores[OR=7.31,95%CI(3.36,15.94),P<0.01].There was no significant difference in adverse effects between the AM and sham-AM groups and the analgesic drug group[OR=0.70,95%CI(0.39,1.28),P=0.25],but the treatment and control groups had mild adverse effects with a low incidence.CONCLUSIONS:AM is more effective than other treatments or pharmacotherapy alone in the treatment of pregnancy-related symptoms,and is relatively safe.However,the quality of the included trials was rather poor,and high-quality studies are required to confirm our findings.

Removal of Cr(Ⅵ)by glutaraldehyde-crosslinked chitosan encapsulating microscale zero-valent iron:Synthesis,mechanism,and longevity

Microscale zero-valent iron(mZVI)has shown great potential for groundwater Cr(Ⅵ)remediation.However,low Cr(Ⅵ)removal capacity caused by passivation restricted the wide use of mZVI.We prepared mZVI/GCS by encapsulating mZVI in a porous glutaraldehydecrosslinked chitosan matrix,and the formation of the passivation layer was alleviated by reducing the contact between zero-valent iron particles.The average pore diameter of mZVI/GCS was 8.775 nm,which confirmed the mesoporous characteristic of this material.Results of batch experiments demonstrated that mZVI/GCS exhibited high Cr(Ⅵ)removal efficiency in a wide range of pH(2-10)and temperature(5-35℃).Common groundwater coexisting ions slightly affected m ZVI/GCS.The material showed great reusability,and the average Cr(Ⅵ)removal efficiency was 90.41%during eight cycles.In this study,we also conducted kinetics and isotherms analysis.Pseudo-second-order model was the most matched kinetics model.The Cr(Ⅵ)adsorption process was fitted by both Langmuir and Freundlich isotherms models,and the maximum Langmuir adsorption capacity of m ZVI/GCS reached243.63 mg/g,which is higher than the adsorption capacities of materials reported in most of the previous studies.Notably,the column capacity for Cr(Ⅵ)removal of a mZVI/GCS-packed column was 6.4 times higher than that of a mZVI-packed column in a 50-day experiment.Therefore,mZVI/GCS with a porous structure effectively relieved passivation problems of mZVI and showed practical application prospects as groundwater Cr(Ⅵ)remediation material with practical application prospects.

Frequency Regulation of VSC-MTDC System with Offshore Wind Farms

Frequency regulation of voltage source converter-based multi-terminal high-voltage direct current(VSC-MTDC)system with offshore wind farms enhances the frequency stability by compensating the power for a disturbed AC system.However,it is difficult to reasonably allocate frequency-regulation resources due to a lack of coordination mechanisms between wind farms and the MTDC system.Moreover,it is difficult for the frequency control of the wind farms to manage changes in wind speed;and the risk of wind-turbine stalls is high.Thus,based on the kinetic energy of wind turbines and the power margin of the converters,the frequency-regulation capability of wind turbines is evaluated,and a dynamic frequency-support scheme considering the real-time frequency-support capability of the wind turbines and system frequency evolution is proposed to improve the frequency-support performance.A power adaptation technique at variable wind speeds is developed;the active power in the frequency-support stage and restoration stage is switched according to the wind speed.A hierarchical zoning frequency-regulation scheme is designed to use the frequency-regulation resources of different links in the MTDC system with wind farms.The simulation results show that the novel frequency-regulation strategy maintains frequency stability with wind-speed changes and avoids multiple frequency dips.

Characteristics and sources of peroxyacetyl nitrate(PAN)in the rural North China Plain:Results from1-year continuous observations

Peroxyacetyl nitrate(PAN)is an important photochemical pollutant in the troposphere,whereas long-term measurements are scarce in rural areas in North China Plain(NCP),resulting in unclear seasonal variations and sources of PAN in rural NCP.In this study,we conducted a 1-year observation of PAN during 2021-2022 at the rural NCP site.The average concentrations of PAN were 1.10,0.75,0.65,and 0.88 ppbv in spring,summer,autumn,and winter,respectively,with a 1-year average of 0.81±0.60 ppbv.Calculations indicate that the loss of PAN through thermal decomposition in summer accounts for 43.2% of the total formed PAN,which is an important reason for the low concentration of PAN in summer.We speculate that since the correlation between PAN and O_(3) in winter is significantly lower than that in other seasons,the observed regional transport of PAN cannot be ignored in winter.Through budget analysis,regional transport accounted for 12.8% and 55.9% of the observed PAN on the spring and winter pollution days,respectively,which showed that regional transport played key roles during the photochemical pollution of the rural NCP in winter.The potential source contribution function revealed that the transported PAN mainly comes from southern Hebei in spring.In winter,the transported PAN was mainly from Langfang,Hengshui,and southern Beijing.Our findings may aid in understanding PAN variations in different seasons in rural areas and highlight the impact of regional transport on the PAN budget.

High-performance,stable CoNi LDH@Ni foam composite membrane with innovative peroxymonosulfate activation for 2,4-dichlorophenol destruction

In this study,the cobalt-nickel layered double hydroxides(CoNi LDH)were synthesized with a variety of Co/Ni mass ratio,as CoxNiyLDHs.In comparison,Co1Ni3LDH presented the best peroxymonosulfate(PMS)activation efficiency for 2,4-dichlorophenol removal.Meanwhile,CoNi LDH@Nickel foam(CoNi LDH@NF)composite membrane was constructed for enhancing the stability of catalytic performance.Herein,CoNi LDH@NF-PMS system exerted high degradation efficiency of 99.22%within 90 min for 2,4-DCP when[PMS]_(0)=0.4 g/L,Co^(1)Ni^(3)LDH@NF=2 cm×2 cm(0.2 g/L),reaction temperature=298 K.For the surface morphology and structure of the catalyst,it was demonstrated that the CoNi LDH@NF composite membrane possessed abundant cavity structure,good specific surface area and sufficient active sites.Importantly,·OH,SO_(4)·^(-)and^(1)O_(2)played the primary role in the CoNi LDH@NF-PMS system for 2,4-DCP decomposition,which revealed the PMS activation mechanism in CoNi LDH@NF-PMS system.Hence,this study eliminated the stability and adaptability of CoNi LDH@NF composite membrane,proposing a new theoretical basis of PMS heterogeneous catalysts selection.

A Novel Design of a Pre-separation Panel Structure to Protect Against the Impact of an Explosion in an Aircraft Plenum Chamber

To reduce the damage of the pressurizing panel structure of a fuselage caused by an explosion at the“least risk bomb location”in an aircraft structure,a new pre-separation panel structure was designed to resist blast loading.First,the dynamic strain response and morphology of impact damage of the new pre-separation panel were measured in an impact damage test.Second,the commercial software LS-DYNA was used to calculate the propagation of the blast shock wave,and the results were compared with empirical equations to verify the rationality of the numerical calculation method.Finally,the fluid–structure coupling method was used to calculate the damage process of the pre-separation panel structure under the impact of an explosion wave and an impact block.The calculated results were in good agreement with the test results,which showed the rationality of the calculation method and the model.The residual strength of the damaged pre-separation panel was significantly higher than that of the original damaged panel.The results show that the new pre-separation panel structure is reasonable and has certain significance for guiding the design of plenum chambers with strong resistance to implosion for aircraft fuselages.
