Ovarian reserve is essential for fertility and influences healthy aging in women.Advanced maternal age correlates with the progressive loss of both the quantity and quality of oocytes.The molecular mechanisms and various contributing factors underlying ovarian aging have been uncovered.In this review,we highlight some of critical factors that impact oocyte quantity and quality during aging.Germ cell and follicle reserve at birth de-termines reproductive lifespan and timing the menopause in female mammals.Accelerated diminishing ovarian reserve leads to premature ovarian aging or insufficiency.Poor oocyte quality with increasing age could result from chromosomal cohesion deterioration and misaligned chromosomes,telomere shortening,DNA damage and associated genetic mutations,oxidative stress,mitochondrial dysfunction and epigenetic alteration.We also discuss the intervention strategies to delay ovarian aging.Both the efficacy of senotherapies by antioxidants against reproductive aging and mitochondrial therapy are discussed.Functional oocytes and ovarioids could be rejuvenated from pluripotent stem cells or somatic cells.We propose directions for future interventions.As couples increasingly begin delaying parenthood in life worldwide,understanding the molecular mechanisms during female reproductive aging and potential intervention strategies could benefit women in making earlier choices about their reproductive health.

Frequent Mutation of rs13281615 and Its Association with PVT1 Expression and Cell Proliferation in Breast Cancer

The q24 band of chromosome 8 （8q24） is frequently amplified in human cancers including breast cancer, and several SNPs （single nucleotide polymorphisms） at 8q24, including rs13281615, have been identified for their association with cancer risks. These SNPs are in a ＂gene desert＂ region, and their functions in cancer development remain to be illustrated, although several of the SNPs appear to influence the genes in the ＂desert＂ in a long-range manner, including the v-myc avian myelocytomatosis viral oncogene homolog （MYC） and the non- protein coding plasmacytoma variant translocation 1 （PVT1）, both of which have been implicated in human cancers. In the current study, we examined rs13281615 for its potential role in breast cancer using normal and cancer tissues from 121 Chinese women with breast cancer. In addition to confirming the association of the GG genotype of rs 13281615 with breast cancer risk, we found that germline GG genotype was significantly associated with estrogen receptor （ER） positivity, higher tumor grade and higher proliferation index. We also found frequent somatic mutations （22/121 or 18.2%） of this SNP in breast cancer. Interestingly, the majority of the mutations （17/22 or 77%） involved a G→ A change, resulting in a decrease in the number of cancers with the GG risk genotype and subsequent loss of GG association with higher tumor grade and proliferation index in cancers. Furthermore, PVT1 expression was increased in cancers, and the increase was associated with the GG genotype of rs13281615. These results suggest that the GG genotype of SNP rs13281615 plays a role in breast cancer likely by influencing PVT1 expression, and that during oncogenesis, ＂protective＂ mutations could occur.

Zfhx3 is essential for progesterone/progesterone receptor signaling to drive ductal side-branching and alveologenesis in mouse mammary glands

Progesterone(Pg)/progesterone receptor(PR) signaling drives mammary gland side-branching and alveologenesis, but the mechanisms through which Pg/PR signaling functions remain to be clarified.Using in vitro and in vivo models and histological and molecular analyses, we determined the role of Zfhx3 transcription factor in mammary gland development driven by Pg/PR signaling. Postnatal deletion of Zfhx3 in mouse mammary epithelial cells attenuated side-branching morphogenesis and alveologenesis. These effects were undetectable in the absence of Pg/PR signaling. During the estrus cycle, Zfhx3 expression corresponded to that of Pg, being at the highest level at the diestrus stage; Zfhx3 deletion inhibited mammary gland branching more potently at diestrus than estrus stage. Loss of Zfhx3 not only attenuated the expansion of stem/progenitor cells driven by Pg/PR signaling, but also impaired the function of Pg/PR signaling in the transcriptional activation of multiple genes. In addition, Pg/PR signaling significantly expanded PR-and Zfhx3-positive epithelial cells, and induced the physical association of ZFHX3 with PR. These findings establish Zfhx3 as an integral transcription factor of Pg/PR signaling in driving side-branching and alveologenesis during mammary gland development.

Amyotrophic lateral sclerosis(ALS)is a fatal neurodegenerative disease characterized by progressive degeneration of motor neurons,and it demonstrates high clinical heterogeneity and complex genetic architecture.A variation within TRMT2B(c.1356G>T;p.K452N)was identified to be associated with ALS in a family comprising two patients with juvenile ALS(JALS).Two missense variations and one splicing variation were identified in 10 patients with ALS in a cohort with 910 patients with ALS,and three more variants were identified in a public ALS database including 3317 patients with ALS.A decreased number of mitochondria,swollen mitochondria,lower expression of ND1,decreased mitochondrial complex I activities,lower mitochondrial aerobic respiration,and a high level of ROS were observed functionally in patient-originated lymphoblastoid cell lines and TRMT2B interfering HEK293 cells.Further,TRMT2B variations overexpression cells also displayed decreased ND1.In conclusion,a novel JALS-associated gene called TRMT2B was identified,thus broadening the clinical and genetic spectrum of ALS.

Nanosized and highly reactive magnesium niobate Mg4Nb2O9 were successfully synthesized by a sol-gel method as potential candidates for use in microwave applications as a low-temperature co-fired ceramic （LTCC）.The Nb-Mg-citric acid solution was evaporated resulting in a dry gel that was calcined in the temperature ranging from 550 ℃ to 850 ℃ for crystallization in air atmosphere.The crystallization process from the gel to the crystalline Mg4Nb2O9 was followed by thermal analysis and during calcinations phase evolution of the powders was studied using X-ray diffraction （XRD） technique.Particle size and morphology were examined by high resolution scanning electron microscopy （HR-SEM）.At 850 ℃ the corundum-like phase of magnesium niobate was obtained in air atmosphere with an average particle diameter of 80 nm approximately calculated from the HR-SEM images.

针对超临界二氧化碳(S-CO_(2))动力机组在启动和停止阶段气体轴承产生非常大的摩擦磨损,以及气体轴承承载力低、刚度低、阻尼小、稳定性较差等问题,设计并改进一种新型动静压S-CO_(2)润滑径向可倾瓦轴承结构。设计并搭建S-CO_(2)润滑轴承实验台,针对于实验台转子刚启动和极低转速工况,对新型S-CO_(2)润滑动静压径向轴承在静压状态下的动态特性进行实验研究,得到轴承的动态刚度和动态阻尼,并分析静压对轴承动态性能的影响。实验结果表明,设计的S-CO_(2)动静压径向可倾瓦轴承在启停阶段,在轴瓦与轴颈之间产生了足够的静压压力,可将二者完全分隔开,从而能减少启停阶段的摩擦磨损;随着静压压力的增大,轴承X、Y方向上的整体刚度、主阻尼都增大,且2个方向的主刚度系数差别不大,而交叉刚度和交叉阻尼都接近于0。研究结果为进一步揭示S-CO_(2)润滑径向轴承动压状态特性提供参考。

Excess de novo likely gene-disruptive and missense variants within dozens of genes have been identified in autism spectrum disorder(ASD)and other neurodevelopmental disorders.However,many rare inherited missense variants of these high-risk genes have not been thoroughly evaluated.In this study,we analyzed the rare missense variant burden of POGZ in a large cohort of ASD patients from the Autism Clinical and Genetic Resources in China(ACGC)and further dissected the functional effect of diseaseassociated missense variants on neuronal development.Our results showed a significant burden of rare missense variants in ASD patients compared to the control population(P=4.6×10-5,OR=3.96),and missense variants in ASD patients showed more severe predicted functional outcomes than those in controls.Furthermore,by leveraging published large-scale sequencing data of neurodevelopmental disorders(NDDs)and sporadic case reports,we identified 8 de novo missense variants of POGZ in NDD patients.Functional analysis revealed that two inherited,but not de novo,missense variants influenced the cellular localization of POGZ and failed to rescue the defects in neurite and dendritic spine development caused by Pogz knockdown in cultured mouse primary cortical neurons.Significantly,L1CAM,an autism candidate risk gene,is differentially expressed in POGZ deficient cell lines.Reduced expression of L1cam was able to partially rescue the neurite length defects caused by Pogz knockdown.Our study showed the important roles of rare inherited missense variants of POGZ in ASD risk and neuronal development and identified the potential downstream targets of POGZ,which are important for further molecular mechanism studies.

Effect of intramuscular electro-acupuncture-mediated VEGF gene transfer on experimental limb ischemia

To investigate the feasibility of intramuscular electro-acupuncture-mediated VEGF gene transfer on experimental limb ischemia, the transcription and expression of VEGF gene have been detected with RT-PCR and immunohistochemistry staining, and the biological effect of VEGF gene inside the nat limb has been observed through vascular angiography. Results from angiography show that the transfer of VEGF gene into the rat with experimental limb ischemia can stimulate the formation of neovessel, establish collateral circulation, augment blood perfusion, and ameliorate distal limb necrosis. These findings reveal that intramuscular electro acupuncture-mediated VEGF gene transfer may present a new therapeutic modality for treating tissue ischemia.
