BACKGROUND/AIMS:The newly developed generic PEG-G-CSF,a long-acting granulocyte colony-stimulating factor was established in china,the aim of this study was to investigate the safety, pharmacokinetic and pharmacodynamic profiles of PEG-G-CSF in cancer patients with chemotherapy-induced neutropenia. METHODS:This open-label,randomized-sequence,2.way crossover study was conducted at two hospitals, *** Chinese cancer patients(eight patients with nonsmall-cell lung cancer and two patients with lymphoma) who had experienced severe neutropenia were *** received the same chemotherapy regimen in their each *** were randomly assigned at a 1:1 ratio to received a single subcutaneous dose of 60 ug/kg PEG-G-CSF or with daily subcutaneous dose of 5μg/kg filgrastim at the 48 hours after chemotherapy ended in the first chemotherapy cycle and administration of the alternate formulation in the second cycle. Pharmacokinetic,pharmacodynamic and safety analyses were *** samples for PEG-G-CSF concentration measurement were collected before PEG-G-CSF administration and froml to 432.hours after the injection of ***:PEG-G-CSF and filgrastim were similar for all efficacy and safety end *** main pharmacokinetic parameters of PEG-G-CSF are as follow:T:2..0±11.0 h,C:138.4±31.3μg/L,t:64.2.11.0h,CL:0.006±0.002.L/h/kg,AUC(0-t):11105±2.43ug/L/*** concentrations of PEG-G-CSF began to decrease after day 5 at the end of chemotherapy and returned to baseline levels on day 15. CONLUSION:The tolerance,pharmacokinetic and pharmacodynamic characteristics of PEG-G-CSF supported that a single dose of 60ug/kg subcutaneous injection once per treatment cycle for prevention of severe neutropenia.
