MGMT is down-regulated independently of promoter DNA methylation in rats with all-trans retinoic acidinduced spina bifida aperta

O6-methylguanine DNA methyltransferase(MGMT), a DNA repair enzyme, has been reported in some congenital malformations, but it is less frequently reported in neural tube defects. This study investigated MGMT mRNA expression and methylation levels in the early embryo and in different embryonic stages, as well as the relationship between MGMT and neural tube defects. Spina bifida aperta was induced in rats by a single intragastric administration of all-trans retinoic acid on embryonic day(E) 10, whereas normal control rats received the same amount of olive oil on the same embryonic day. DNA damage was assessed by detecting γ-H2 A.X in spina bifida aperta rats. Real time-polymerase chain reaction was used to examine mRNA expression of MGMT in normal control and spina bifida aperta rats. In normal controls, the MGMT mRNA expression decreased with increasing embryonic days, and was remarkably reduced from E11 to E14, reaching a minimum at E18. In the spina bifida aperta model, γ-H2 A.X protein expression was increased, and mRNA expression of MGMT was markedly decreased on E14, E16, and E18. Bisulfite sequencing polymerase chain reaction for MGMT promoter methylation demonstrated that almost all CpG sites in the MGMT promoter remained unmethylated in both spina bifida aperta rats and normal controls, and there was no significant difference in methylation level between the two groups on either E14 or E18. Our results show that DNA damage occurs in spina bifida aperta rats. The mRNA expression of MGMT is downregulated, and this downregulation is independent of promoter DNA methylation.

Genome-Wide Screening of Aberrant Methylation Loci for Nonsyndromic Cleft Lip

Background： The pathogenicity of cleft lip （CL） is pretty complicated since it is influenced by the interaction of environment and genetic factors. The purpose of this study was to conduct a genome-wide screening of aberrant methylation loci in partial lesion tissues of patients with nonsyndromic CL （NSCL） and preliminarily validate candidate dysmethylated genes associated with ***： Fifteen healthy and sixteen NSCL fetal lip tissue samples were collected. The Infinium HumanMethylation450 BeadChip was used to screen aberrant methylation loci in three NSCL and three healthy lip tissues. The differential methylation sites and functions of the annotated genes between NSCL and healthy lip tissues were analyzed using minfi package of R software, cluster analysis, Gene Ontology （GO） annotation, and metabolic pathway annotation. Gene expression was assessed in nine differentially methylated genes by real-time polymerase chain reaction （PCR）. The transcriptions mRNA levels of three out of nine candidate genes were downregulated remarkably in NSCL lip tissues, and these three genes＇ abnormal methylation loci were validated by pyrosequencing in 16 NSCL cases and 15 healthy ***： In total, 4879 sites in the genes of NSCL odinopoeia fetuses showed aberrant methylation when compared with normal lip tissue genome. Among these, 3661 sites were hypermethylated and 1218 sites were hypomethylated as compared to methylation levels in healthy specimens. These aberrant methylation sites involved 2849 genes and were widely distributed among the chromosomes. Most differentially methylated sites were located in cytosine-phosphoric acid-guanine islands. Based on GO analysis, aberrantly methylated genes were involved in 11 cellular components, 13 molecular functions, and a variety of biological processes. Repair of left ventricular aneurysm： ten-year experience in Chinese patients

Background A large transmural myocardial infarction often results in a dyskinetic or akinetic left ventricular aneurysm （LVA）. This study aimed to explore the early and long-term clinical outcomes and to identify predictors for survivals and hospital re-admission after the repair of left ventricular aneurysm. Methods We followed up 497 patients who had undergone LVA repair from a single center in China between 1995 and 2005. The perioperative parameters were recorded. Risk factors for early mortality and long-term results were analyzed by multivariate Logistic regression. Cox＇s proportional hazard model was used to calculate risk factors for major adverse cardiac and cerebrovascular events, cause of death and re-admission. Kaplan-Meier curve was employed to analyze long-term survival. Results The operative mortality was 2.0%. The long-term mortality was 11.1% and cardiac causes contributed to 61.8% of the overall long-term mortality. Four hundred and thirty-two patients survived during the follow-up period and 37.5% of them had been re-admitted at least one time. One hundred and five patients experienced major adverse cardiac and cerebrovascular events. Survival analysis exhibited that the probability of survival at 1 and 5 years after operation was 96% and 86% respectively. Previous atrial fibrillation was the independent risk factor for early mortality. Independent risk factors for long-term mortality were poor left ventricular ejection fraction and stroke, and risk factors for cardiac mortality were intraventricular block, stroke and poor left ventricular ejection fraction. Stroke, intraventricular block and advanced age were independent risk factors for major adverse cardiac and cerebrovascular events, and New York Heart Association （NYHA） class Ill-IV was the only risk factor for hospital re-admission. Characterization of size-resolved aerosol hygroscopicity and liquid water content in Nanjing of the Yangtze River Delta

Aerosol hygroscopicity and liquid water content(ALWC)have important influences on the environmental and climate effect of *** this study,we measured the hygroscopic growth factors(GF)of particles with dry diameters of 40,80,150,and 200 nm during the wintertime in *** the GF-derived hygroscopicity parameter(κ_(gf))and ALWC increased with particle size,but displayed differing diurnal variations,withκ_(gf)peaking around the midday,while ALWC peaking in the early ***,ammonium and oxygenated organic aerosols(OOA)were found as the chemical components mostly strongly correlated with ALWC.A closure study suggests that during midday photo-oxidation and nighttime high ALWC periods,theκof organic aerosols(κ_(org))was underestimated when using previous ***,we re-constructed parameterizations forκ_(org)and the oxidation level of organics for these periods,which indicates a higher hygroscopicity of photochemically formed OOA than the aqueous OOA,yet both being much higher than the generally assumed OOA ***,in a typical high ALWC episode,concurrently increased ALWC,nitrate,OOA as well as aerosol surface area and mass concentrations were observed under elevated ambient *** strongly indicates a coupled effect that the hygroscopic secondary aerosols,in particular nitratewith strong hygroscopicity,led to large increase in ALWC,which in turn synergistically boosted nitrate and OOA formation by heterogeneous/aqueous *** interaction may represent an important mechanism contributing to enhanced formation of secondary aerosols and rapid growth of fine particulate matter under relatively high RH conditions.

Dear Editor,This letter presents a joint probabilistic scheduling and resource allocation method(PSRA) for 5G-based wireless networked control systems(WNCSs). As a control-aware optimization method, PSRA minimizes the linear quadratic Gaussian(LQG) control cost of WNCSs by optimizing the activation probability of subsystems, the number of uplink repetitions, and the durations of uplink and downlink phases. Simulation results show that PSRA achieves smaller LQG control costs than existing works.

Numerical simulation of microstructure and microporosity morphology in directional solidification of aluminum-copper alloys:Effect of copper content and withdrawal rate

Microporosity formed in the solidification process of Al alloys is detrimental to the alloy properties.A two-dimensional cellular automaton(CA)model was developed to simulate the microstructure and microporosity formation in Al-Cu alloys,considering variations in Cu content and solidification *** results indicate that the Cu content primarily influences the growth of *** validate the model,directional solidification experiments were conducted on Al-Cu alloys with varing Cu contents and withdrawal *** experimental results of dendrites and microporosity characteristics agree well with the predictions from the developed model,thus confirming the validity of the *** alloy’s liquidus temperature,dendrite morphology,and hydrogen saturation solubility arising from different Cu contents have significant effects on microporosity *** withdrawal rate primarily affects the nucleation of hydrogen microporosity by altering cooling rates and dendritic growth rates,resulting in different microporosity characteristics.
