The mononuclear phagocyte system （MPS）, which con-sists of monocytes, dendritic cells （DCs）, and macro-phages, plays a vital role in the innate immune defense against pathogens. Hepatitis C virus （HCV） is effcient in evading the host immunity, thereby facilitating its devel-opment into chronic infection. Chronic HCV infection is the leading cause of end-stage liver diseases, liver cirrhosis, and hepatocellular carcinoma. Acquired im-mune response was regarded as the key factor to era-dicate HCV. However, innate immunity can regulate the acquired immune response. Innate immunity-derived cytokines shape the adaptive immunity by regulating T-cell differentiation, which determines the outcome of acute HCV infection. Inhibition of HCV-specific T-cell responses is one of the most important strategies for im-mune system evasion. It is meaningful to illustrate the role of innate immune response in HCV infection. With the MPS being the important factor in innate immunity, therefore, understanding the role of the MPS in HCV infection will shed light on the pathophysiology of chronic HCV infection. In this review, we outline the impact of HCV infection on the MPS and cytokine production. We discuss how HCV is detected by the MPS and describe the function and impairment of MPS components in HCV infection.

AIM To investigate the relationship between levels of iron metabolism markers and hepatitis B virus(HBV)-related chronic liver *** This case-control study with 318 participants included 78 cases of chronic hepatitis B, 85 cases of HBV-related liver cirrhosis, 77 cases of HBV-related hepatocellular carcinoma, and 78 healthy controls. Markers of iron metabolism were detected in participants. Hematological and biochemical parameters and HBV-DNA were assessed. Child-Pugh grade and Barcelona Clinic Liver Cancer stage were determined for each hepatocellular carcinoma patient. Perls' staining was performed on liver sections. The SPSS program was used for all statistical analyses, and statistical significance was considered if a P-value turation levels increased proportionally to the extent of liver cirrhosis and poorer Child-Pugh scores(P turation levels were significantly correlated with a smaller hepatocellular carcinoma tumor burden according to Barcelona Clinic Liver Cancer staging. Liver histology showed a clearly increasing trend in iron deposition in the liver tissues with increased fibrosis, which became prominent at stages 3(severe liver fibrosis) and 4(cirrhosis). CONCLUSION Iron metabolism disorders occur in patients with HBVrelated liver diseases. The serum markers of iron metabolism disorders vary in different stages of HBV-related liver diseases.

The conventional perception of astrocytes as mere supportive cells within the brain has recently been called into question by empirical evidence, which has revealed their active involvement in regulating brain function and encoding behaviors associated with ***, astrocytes in the basolateral amygdala have been found to play a role in the modulation of anxiety-like behaviors triggered by chronic stress. Nevertheless, the precise molecular mechanisms by which basolateral amygdala astrocytes regulate chronic stress–induced anxiety-like behaviors remain to be fully elucidated. In this study, we found that in a mouse model of anxiety triggered by unpredictable chronic mild stress, the expression of excitatory amino acid transporter 2 was upregulated in the basolateral amygdala. Interestingly, our findings indicate that the targeted knockdown of excitatory amino acid transporter 2 specifically within the basolateral amygdala astrocytes was able to rescue the anxiety-like behavior in mice subjected to stress. Furthermore, we found that the overexpression of excitatory amino acid transporter 2 in the basolateral amygdala, whether achieved through intracranial administration of excitatory amino acid transporter 2agonists or through injection of excitatory amino acid transporter 2-overexpressing viruses with GfaABC1D promoters, evoked anxiety-like behavior in mice. Our single-nucleus RNA sequencing analysis further confirmed that chronic stress induced an upregulation of excitatory amino acid transporter 2 specifically in astrocytes in the basolateral amygdala. Moreover, through in vivo calcium signal recordings, we found that the frequency of calcium activity in the basolateral amygdala of mice subjected to chronic stress was higher compared with normal *** knocking down the expression of excitatory amino acid transporter 2 in the basolateral amygdala, the frequency of calcium activity was not significantly increased, and anxiety-like behavior was ob

Al2O3/A356-La alloy composites were fabricated by squeeze casting, and the effects of La on the solidified structure and the solute segregation during alloy solidification were studied. The results indicate that the structure of the matrix alloy becomes fine and small by the addition of La. La has been richened at the interface to help improve the wettability between the fiber and Al alloy, but there are no intermetallic compounds richening La found at the interface yet. There is no special influence of La on the Mg segregation in the matrix alloy. The distribution of Mg and La in the composites has been at the same position--near the interface.

Hot spot temperature prediction and operating parameter estimation of racks in data center using machine learning algorithms based on simulation data

In this paper,models to predict hot spot temperature and to estimate cooling air’s working parameters of racks in data centers were established using machine learning algorithms based on simulation ***,simulation models of typical racks were established in computational fluid dynamics(CFD).The model was validated with field test results and results in literature,error of which was less than 3%.Then,the CFD model was used to simulate thermal environments of a typical rack considering different factors,such as servers’power,which is from 3.3 kW to 20.1 kW,cooling air’s inlet velocity,which is from 1.0 m/s to 3.0 m/s,and cooling air’s inlet temperature,which is from 16℃ to 26℃ The highest temperature in the rack,also called hot spot temperature,was selected for each ***,a prediction model of hot spot temperature was built using machine learning algorithms,with servers’power,cooling air’s inlet velocity and cooling air’s inlet temperature as inputs,and the hot spot temperatures as ***,based on the prediction model,an operating parameters estimation model was established to recommend cooling air’s inlet temperatures and velocities,which can not only keep the hot spot temperature at the safety value,but are also energy saving.

Influenza epidemics and pandemics are constant threats to global public *** strategies including vaccines and antiviral drugs have achieved great advances in controlling influenza virus infection,the efficacy of these strategies is limited by the highly frequent mutations in the viral genome and the emergence of drug-resistant *** previous study indicated that boosting the immunity of human Vγ9Vδ2-T cells with the phosphoantigen pamidronate could be a therapeutic strategy to treat seasonal and avian influenza virus ***,one notable drawback ofγδ-T cell-based immunotherapy is the rapid exhaustion of proliferation and effector responses due to repeated treatments with ***,we found that the expression of CD137 was inducible in Vγ9Vδ2-T cells following antigenic ***137^(+)Vγ9Vδ2-T cells displayed more potent antiviral activity against influenza virus than their CD137−counterparts in vitro and in Rag2^(-/-)γc^(-/-)*** further demonstrated that CD137 costimulation was essential for Vγ9Vδ2-T cell activation,proliferation,survival and effector *** humanized mice reconstituted with human peripheral blood mononuclear cells,CD137 costimulation with a recombinant human CD137L protein boosted the therapeutic effects of pamidronate against influenza *** study provides a novel strategy of targeting CD137 to improve the efficacy of Vγ9Vδ2-T cell-based immunotherapy.
