Insertion sequences(ISs)exist widely in bacterial genomes,but their roles in the evolution of bacterial antiphage defense remain to be clarified.here,we report that,under the pressure of phage infection,the IS1o96 transposition of Mycobacterium smegmatis into the Isr2 gene can occur at high frequencies,which endows the mutant mycobacterium with a broad-spectrum antiphage ability.Lsr2 functions as a negative regulator and directly silences expression of a gene island composed of 11 lipid metabolism-related genes.The complete or partial loss of the gene island leads to a significant decrease of bacteriophage adsorption to the mycobacterium,thus defending against phage infection.Strikingly,a phage that has evolved mutations in two tail-filament genes can re-escape from the Isr2 inactivation-triggered host defense.This study uncovered a new signaling pathway for activating antimycobacteriophage immunity by Is transposition and provided insight into the natural evolution of bacterial antiphage defense.

The hot deformation behaviors of sulfur-containing gear steel 20MnCr5 containing three different contents of Nb and B(0,0.021%Nb,and 0.024%Nb-0.0022%B)were investigated.Hot compression and tenssion tests were carried out by Gleeble3800 at the austenite region from 850 to 1150℃and the adverse effects of Nb and B were analyzed by the fracture,microstructure and precipitate observations.Hot compression tests showed that the proportions of instable area in hot processing maps of 0.021%Nb and Nb-B steels were higher and the deformability of Nb free steel was better.The tensile deformation experiments showed that the reduction areas of Nb free,0.021%Nb and Nb-B steels were 92%-99%,84%-98%and 67%-97%,respectively.The addition of Nb or Nb and B inhibited the dynamic recrystallization during hot deformation,and consequently,more deformed grains were then formed in 0.021%Nb and Nb-B steels thus to obtain the microstructure with worse uniformity and then deteriorate the deformability.In addition,the interaction between inclusions and microalloyed elements was also significant.NbC particles of 0.021%Nb and Nb-B steels dynamically precipitated during deformation and precipitated together with MnS thus to worsen the deformability,resulting in the decrease of reduction area.

Antimony-based materials with high capacities and moderate potentials are promising anodes for lithium-/-sodium-ion batteries.However,their tremendous volume expansion and inferior conductivity lead to poor structural stability and sluggish reaction kinetics.herein,a doubleconfined nanoheterostructure Sb/Sb2S3@Ti3C2Tx@C has been fabricated through a solvothermal method followed by low-temperature heat treatment.The dual protection of‘‘MXene’’and‘‘carbon’’can better accommodate the volume expansion of Sb/Sb2S3.The strong covalent bond （Ti–S,Ti–O–Sb,C–O–Sb） can firmly integrate Sb-based material with Ti3C2Txand carbon,which significantly improves the structure stability.In addition,the carbon layer can restrain the oxidation of MXenes,and the nano-Sb/Sb2S3can facilitate electron/ion transport and suppress the restacking of MXenes.The heterogeneous interface between Sb and Sb2S3can further promote interfacial charge transfer.The MXene-Sb/Sb2S3@C-1 with the optimal Sb content shows high specific capacities,comparable rate properties and ultra-stable cycling performances (250 m Ah·g-1after 2500 cycles at 1 A·g-1for sodium-ion batteries).Ex situ X-ray diffractometer （XRD）test reveals the storage mechanism including the conversion and alloying process of MXene-Sb/Sb2S3@C-1.Cyclic voltammetry （CV） test results demonstrate that the pseudocapacitance behavior is dominant in MXene-Sb/Sb2S3@C-1,especially at large current.This design paves the way for exploring high-performance alloy-based/conversion-type anode for energy storage devices.

Microorganisms play an essential role in the glacier carbon cycle;how they transform organic matter in mountain glacial cryoconite remains to be studied.here,we applied ultra-high resolution Fourier transform ion cyclotron resonance mass spectrometry(FT-ICR MS)and deep sequencing of 16S rRNA gene,to investigate the temporal microbial transformation of dissolved organic matter(DOM)of the Tibetan Plateau cryoconite.During the 60-day incubation,we found that DOM in cryoconite underwent a three-stage transformation,with decreasing bioavailability over time.The microbial community did not change much in the first week while degrading DOM molecules that were associated with higher H/C_(wa)and lower O/C_(wa).During days 15-30,DOM composition remained stable while microbial diversity increased.By day 60,the DOM was microbially converted into a higher state of recalcitrance,with higher values of aromatic index,O/C_(wa),and lower H/C_(wa),which contained molecules containing more heteroatoms.Cooperation among various microbial taxa,like Cyanobacteria,Bacteroidota,Gammaproteobacteria,Firmicutes,and Actinobacteriota,drove the DOM transformation in cryoconite.This study sheds light on the in-situ transformation of DOM composition meditated by microbial populations in cryoconite at a temporal scale,providing new insights into understanding the microbial roles in the glacier organic carbon transformation.

In the realm of large-scale machine learning,it is crucial to explore methods for reducing computational complexity and memory demands while maintaining generalization performance.Additionally,since the collected data may contain some sensitive information,it is also of great significance to study privacy-preserving machine learning algorithms.This paper focuses on the performance of the differentially private stochastic gradient descent(SGD)algorithm based on random features.To begin,the algorithm maps the original data into a lowdimensional space,thereby avoiding the traditional kernel method for large-scale data storage requirement.Subsequently,the algorithm iteratively optimizes parameters using the stochastic gradient descent approach.Lastly,the output perturbation mechanism is employed to introduce random noise,ensuring algorithmic privacy.We prove that the proposed algorithm satisfies the differential privacy while achieving fast convergence rates under some mild conditions.

Extracellular vesicles are nano-to micro-scale,membrane-bound particles released by cells into extracellular space,and act as carriers of biomarkers and therapeutics,holding promising potential in translational medicine.However,the challenges remain in handling and detecting extracellular vesicles for disease diagnosis as well as exploring their therapeutic capability for disease treatment.here,we review the recent engineering and technology advances by leveraging the power of sound waves to address the challenges in diagnostic and therapeutic applications of extracellular vesicles and biomimetic nanovesicles.We first introduce the fundamental principles of sound waves for understanding different acoustic-assisted extracellular vesicle technologies.We discuss the acoustic-assisted diagnostic methods including the purification,manipulation,biosensing,and bioimaging of extracellular vesicles.Then,we summarize the recent advances in acoustically enhanced therapeutics using extracellular vesicles and biomimetic nanovesicles.Finally,we provide perspectives into current challenges and future clinical applications of the promising extracellular vesicles and biomimetic nanovesicles powered by sound.

The presence of protein aggregates in numerous human diseases underscores the significance of detecting these aggregates to comprehend disease mechanisms and develop novel therapeutic approaches for combating these disorders.Despite the development of various biosensors and fluorescent probes that selectively target amyloid fibers or amorphous aggregates,there is still a lack of tools capable of simultaneously detecting both types of aggregates.herein,we demonstrate the quantitative discernment of amorphous aggregates by QM-FN-SO3,an aggregationinduced emission(AIE)probe initially designed for detecting amyloid fibers.This probe easily penetrates the membranes of the widely-used prokaryotic model organism Escherichia coli,enabling the visualization of both amorphous aggregates and amyloid fibers through near-infrared fluorescence.Notably,the probe exhibits sensitivity in distinguishing the varying aggregation propensities of proteins,regardless of whether they form amorphous aggregates or amyloid fibers in vivo.These properties contribute to the successful application of the QM-FN-SO3 probe in the subsequent investigation of the antiaggregation activities of two outer membrane protein(OMP)chaperones,both in vitro and in their physiological environment.Overall,our work introduces a near-infrared fluorescent chemical probe that can quantitatively detect amyloid fibers and amorphous aggregates with high sensitivity in vitro and in vivo.Furthermore,it demonstrates the applicability of the probe in chaperone biology and its potential as a high-throughput screening tool for protein aggregation inhibitors and folding factors.

Recently,studies show that deep learning-based automatic speech recognition(ASR)systems are vulnerable to adversarial examples(AEs),which add a small amount of noise to the original audio examples.These AE attacks pose new challenges to deep learning security and have raised significant concerns about deploying ASR systems and devices.The existing defense methods are either limited in application or only defend on results,but not on process.In this work,we propose a novel method to infer the adversary intent and discover audio adversarial examples based on the AEs generation process.The insight of this method is based on the observation:many existing audio AE attacks utilize query-based methods,which means the adversary must send continuous and similar queries to target ASR models during the audio AE generation process.Inspired by this observation,We propose a memory mechanism by adopting audio fingerprint technology to analyze the similarity of the current query with a certain length of memory query.Thus,we can identify when a sequence of queries appears to be suspectable to generate audio AEs.Through extensive evaluation on four state-of-the-art audio AE attacks,we demonstrate that on average our defense identify the adversary’s intent with over 90%accuracy.With careful regard for robustness evaluations,we also analyze our proposed defense and its strength to withstand two adaptive attacks.Finally,our scheme is available out-of-the-box and directly compatible with any ensemble of ASR defense models to uncover audio AE attacks effectively without model retraining.

With the promotion of“dual carbon”strategy,data center(DC)access to high-penetration renewable energy sources(RESs)has become a trend in the industry.However,the uncertainty of RES poses challenges to the safe and stable operation of DCs and power grids.In this paper,a multi-timescale optimal scheduling model is established for interconnected data centers(IDCs)based on model predictive control(MPC),including day-ahead optimization,intraday rolling optimization,and intraday real-time correction.The day-ahead optimization stage aims at the lowest operating cost,the rolling optimization stage aims at the lowest intraday economic cost,and the real-time correction aims at the lowest power fluctuation,eliminating the impact of prediction errors through coordinated multi-timescale optimization.The simulation results show that the economic loss is reduced by 19.6%,and the power fluctuation is decreased by 15.23%.

Stemming from the unique in-plane honeycomb lattice structure and the sp^(2)hybridized carbon atoms bonded by exceptionally strong carbon–carbon bonds,graphene exhibits remarkable anisotropic electrical,mechanical,and thermal properties.To maximize the utilization of graphene’s in-plane properties,pre-constructed and aligned structures,such as oriented aerogels,films,and fibers,have been designed.The unique combination of aligned structure,high surface area,excellent electrical conductivity,mechanical stability,thermal conductivity,and porous nature of highly aligned graphene aerogels allows for tailored and enhanced performance in specific directions,enabling advancements in diverse fields.This review provides a comprehensive overview of recent advances in highly aligned graphene aerogels and their composites.It highlights the fabrication methods of aligned graphene aerogels and the optimization of alignment which can be estimated both qualitatively and quantitatively.The oriented scaffolds endow graphene aerogels and their composites with anisotropic properties,showing enhanced electrical,mechanical,and thermal properties along the alignment at the sacrifice of the perpendicular direction.This review showcases remarkable properties and applications of aligned graphene aerogels and their composites,such as their suitability for electronics,environmental applications,thermal management,and energy storage.Challenges and potential opportunities are proposed to offer new insights into prospects of this material.
