CRISPR-Cas-based genome editing holds immense promise for advancing plant genomics and crop ***,the challenge of low editing activity complicates the identification of editing *** this study,we introduce multiple single transcript unit surrogate reporter(STU-SR)systems to enhance the selection of genome-edited *** systems use the same single guide RNAs designed for endogenous genes to edit reporter genes,establishing a direct link between reporter gene editing activity and that of endogenous *** strategies are used to restore functional reporter genes after genome editing,including efficient single-strand annealing(SSA)for homologous recombination in STUSR-SSA ***-SR-base editor systems leverage base editing to reinstate the start codon,enriching C-to-T and A-to-G base editing *** results showcase the effectiveness of these STU-SR systems in enhancing genome editing events in the monocot rice,encompassing Cas9 nuclease-based targeted mutagenesis,cytosine base editing,and adenine base *** systems exhibit compatibility with Cas9 variants,such as the PAM-less SpRY,and are shown to boost genome editing in Brassica oleracea,a dicot vegetable *** summary,we have developed highly efficient and versatile STU-SR systems for enrichment of genome-edited plants.

Leakages from subsea oil and gas equipment cause substantial economic losses and damage to marine ecosystem,so it is essential to locate the source of the ***,due to the complexity and variability of the marine environment,the signals collected by hydrophone contain a variety of noises,which makes it challenging to extract useful signals for *** solve this problem,a hydrophone denoising algorithm is proposed based on variational modal decomposition(VMD)with grey wolf ***,the average envelope entropy is used as the fitness function of the grey wolf optimizer to find the optimal solution for the parameters K andα.Afterward,the VMD algorithm decomposes the original signal parameters to obtain the intrinsic mode functions(IMFs).Subsequently,the number of interrelationships between each IMF and the original signal was calculated,the threshold value was set,and the noise signal was removed to calculate the time difference using the valid signal obtained by ***,the arrival time difference is used to locate the origin of the *** localization accuracy of the method in finding leaks is investigated experimentally by constructing a simulated leak test rig,and the effectiveness and feasibility of the method are verified.

Vaccines have been the primary remedy in the global fight against coronavirus disease 2.19（COVID-19）.The receptor-binding domain（RBD） of the spike protein, a critical viral immunogen, is affected by the heterogeneity of its glycan structures and relatively low immunogenicity. Here, we describe a scalable synthetic platform that enables the precise synthesis of homogeneously glycosylated RBD, facilitating the elucidation of carbohydrate structure–function relationships. Five homogeneously glycosylated RBDs bearing biantennary glycans were prepared, three of which were conjugated to T-helper epitope(Tpep) from tetanus toxoid to improve their weak immune response. Relative to natural HEK2.3-derived RBD, synthetic RBDs with biantennary N-glycan elicited a higher level of neutralising antibodies against SARS-CoV-2.in mice. Furthermore, RBDs containing Tpepelicited significant immune responses in transgenic mice expressing human angiotensin-converting enzyme 2. Our collective data suggest that trimming the N-glycans and Tpepconjugation of RBD could potentially serve as an effective strategy for developing subunit vaccines providing efficient protection.

Electrochemical impedance spectroscopy(EIS) is a well-established non-destructive characterization technique for assessing the efficacy of electrochemical energy storage and conversion systems. Electrocatalytic systems based on proton exchange membrane(PEM), including PEM fuel cells and PEM water electrolyzers, play a crucial role in renewable energy conversion through electricity–hydrogen interconversion. EIS, along with its derived analysis methods—equivalent circuit model(ECM), distribution of relaxation time(DRT), and dynamic EIS(DEIS), is widely utilized to extract valuable kinetics and impedance *** acquired information affords critical insights into processes such as mass transfer, charge transfer, and proton transfer within PEM systems. This mini review surveys the role of EIS in optimizing components and investigating operational conditions to enhance the efficiency of PEM systems. In addition, it encapsulates the principles and applications of EIS-based methods like DRT and DEIS, highlighting their potential in the practical application of PEM systems.
