Exosome-Transmitted miR-224-5p Promotes Colorectal Cancer Cell Proliferation via Targeting ULK2 in p53-Dependent Manner

Objective To investigate the role and molecular mechanism of exosomal miR-224-5p in colorectal cancer(CRC).Methods The miR-224-5p expression in CRC patient tissues and cell-derived exosomes was measured by laser capture microdissection and qRT-PCR,respectively.Dual-luciferase reporter gene assay was used to determine the target gene of miR-224-5p.The protein expressions of p53 and unc-51 like kinase 2(ULK2)in CRC cells were detected by western blot.Flow cytometry was used to detect cell cycle and apoptosis.Cell proliferation was measured by CCK8 and EdU assay.Results The miR-224-5p expression was upregulated in CRC tissues and increased progressively with the rise of CRC stage.CRC cells secreted extracellular miR-224-5p mainly in an exosome-dependent manner,and then miR-224-5p could be transferred to surrounding tumor cells to regulate cell proliferation in the form of autocrine or paracrine.Moreover,ULK2 was characterized as a direct target of miR-224-5p and was downregulated in CRC tissues.Interestingly,ULK2 inhibited CRC cell proliferation in a p53-dependent manner.Furthermore,exosome-derived miR-224-5p partially reversed the proliferation regulation of ULK2 on CRC cells.Conclusion Our findings demonstrate that exosome-transmitted miR-224-5p promotes p53-dependent cell proliferation by targeting ULK2 in CRC,which may offer promising targets for CRC prevention and therapy.

π-conjugated chromophore functionalized high-nuclearity titanium-oxo clusters containing structural unit of anatase for photocatalytic selective oxidation of sulfides

N-doping has significant influence in manipulating the properties of TiO2,and this has stimulated the development of N-donor-functionalized titanium-oxo clusters（TOCs） as molecular models to study the structure-property relationship.However,the structural type and photoresponsive application are still limited for such TOCs,especially regarding the high-nuclearity TOCs that contain structure unit of TiO2for photocatalysis.Herein,we showed the synthesis of a series of high-nuclearity TOCs 1-3 compounds using π-conjugated 1,10-phenanthroline（phen） as chromophore and N-donor functional ligand.Compound 1 features cocrystal structure composed of one [Ti26]2+and half [Ti22]2+,which renders it as the first cocrystallized TOC containing two positively charged species and phen-functionalized TOC showing the highest nuclearity up to 37 Ti centers.By adjusting the synthetic conditions,the individual {Ti22} and {Ti26} clusters can also be isolated as Compounds 2 and 3,respectively.The core structure of {Ti22} is mainly constructed from four lacunary {Ti4} derived from pentagonal {Ti（Ti）5} unit,while {Ti26} is built from four complete {Ti（Ti）5} unit.Notably,a {Ti8O14} structure unit of anatase TiO2can be identified in {Ti26}.Based on the unique structural features and proper photophysical and photochemical properties of Compounds 1-3,they are applied for photocatalytic sulfoxidation.Owing to the presence of anatase structure unit in {Ti26} and the synergistic effect from {Ti22} and {Ti26},the catalytic performance presents in the order of Compound 1> Compound 3> Compound 2.This work provides excellent models to understand the structureproperty relationship from the perspective of cocrystallization and Ti-O binding model and will further promote the application of TOCs as functional catalysts for organic transformation.

Epigenetic modification in liver fibrosis:Promising therapeutic direction with significant challenges ahead

Liver fibrosis,characterized by scar tissue formation,can ultimately result in liver failure.It’s a major cause of morbidity and mortality globally,often associated with chronic liver diseases like hepatitis or alcoholic and non-alcoholic fatty liver diseases.However,current treatment options are limited,highlighting the urgent need for the development of new therapies.As a reversible regulatory mechanism,epigenetic modification is implicated in many biological processes,including liver fibrosis.Exploring the epigenetic mechanisms involved in liver fibrosis could provide valuable insights into developing new treatments for chronic liver diseases,although the current evidence is still controversial.This review provides a comprehensive summary of the regulatory mechanisms and critical targets of epigenetic modifications,including DNA methylation,histone modification,and RNA modification,in liver fibrotic diseases.The potential cooperation of different epigenetic modifications in promoting fibrogenesis was also highlighted.Finally,available agonists or inhibitors regulating these epigenetic mechanisms and their potential application in preventing liver fibrosis were discussed.In summary,elucidating specific druggable epigenetic targets and developing more selective and specific candidate medicines may represent a promising approach with bright prospects for the treatment of chronic liver diseases.

Integrated multi-omics analysis reveals liver metabolic reprogramming by fish iridovirus and antiviral function of alpha-linolenic acid

Iridovirus poses a substantial threat to global aquaculture due to its high mortality rate;however,the molecular mechanisms underpinning its pathogenesis are not well elucidated.Here,a multi-omics approach was applied to groupers infected with Singapore grouper iridovirus(SGIV),focusing on the roles of key metabolites.Results showed that SGIV induced obvious histopathological damage and changes in metabolic enzymes within the liver.Furthermore,SGIV significantly reduced the contents of lipid droplets,triglycerides,cholesterol,and lipoproteins.Metabolomic analysis indicated that the altered metabolites were enriched in 19 pathways,with a notable down-regulation of lipid metabolites such as glycerophosphates and alpha-linolenic acid(ALA),consistent with disturbed lipid homeostasis in the liver.Integration of transcriptomic and metabolomic data revealed that the top enriched pathways were related to cell growth and death and nucleotide,carbohydrate,amino acid,and lipid metabolism,supporting the conclusion that SGIV infection induced liver metabolic reprogramming.Further integrative transcriptomic and proteomic analysis indicated that SGIV infection activated crucial molecular events in a phagosome-immune depression-metabolism dysregulation-necrosis signaling cascade.Of note,integrative multi-omics analysis demonstrated the consumption of ALA and linoleic acid(LA)metabolites,and the accumulation of L-glutamic acid(GA),accompanied by alterations in immune,inflammation,and cell death-related genes.Further experimental data showed that ALA,but not GA,suppressed SGIV replication by activating antioxidant and anti-inflammatory responses in the host.Collectively,these findings provide a comprehensive resource for understanding host response dynamics during fish iridovirus infection and highlight the antiviral potential of ALA in the prevention and treatment of iridoviral diseases.

With the diversified development of big data,detection and precision guidance technologies,electromagnetic(EM)functional materials and devices serving multiple spectrums have become a hot topic.Exploring the multispectral response of materials is a challenging and meaningful scientific question.In this study,MXene/TiO_(2)hybrids with tunable conduction loss and polarization relaxation are fabricated by in situ atomic reconstruction engineering.More importantly,MXene/TiO_(2)hybrids exhibit adjustable spectral responses in the GHz,infrared and visible spectrums,and several EM devices are constructed based on this.An antenna array provides excellent EM energy harvesting in multiple microwave bands,with|S11|up to−63.2 dB,and can be tuned by the degree of bending.An ultra-wideband bandpass filter realizes a passband of about 5.4 GHz and effectively suppresses the transmission of EM signals in the stopband.An infrared stealth device has an emissivity of less than 0.2 in the infrared spectrum at wavelengths of 6-14μm.This work can provide new inspiration for the design and development of multifunctional,multi-spectrum EM devices.

BACKGROUND As a critical early event in hepatocellular carcinogenesis,telomerase activation might be a promising and critical biomarker for hepatocellular carcinoma(HCC)patients,and its function in the genesis and treatment of HCC has gained much attention over the past two decades.AIM To perform a bibliometric analysis to systematically assess the current state of research on HCC-related telomerase.METHODS The Web of Science Core Collection and PubMed were systematically searched to retrieve publications pertaining to HCC/telomerase limited to“articles”and“reviews”published in English.A total of 873 relevant publications related to HCC and telomerase were identified.We employed the Bibliometrix package in R to extract and analyze the fundamental information of the publications,such as the trends in the publications,citation counts,most prolific or influential writers,and most popular journals;to screen for keywords occurring at high frequency;and to draw collaboration and cluster analysis charts on the basis of coauthorship and co-occurrences.VOSviewer was utilized to compile and visualize the bibliometric data.RESULTS A surge of 51 publications on HCC/telomerase research occurred in 2016,the most productive year from 1996 to 2023,accompanied by the peak citation count recorded in 2016.Up to December 2023,35226 citations were made to all publications,an average of 46.6 citations to each paper.The United States received the most citations(n=13531),followed by China(n=7427)and Japan(n=5754).In terms of national cooperation,China presented the highest centrality,its strongest bonds being to the United States and Japan.Among the 20 academic institutions with the most publications,ten came from China and the rest of Asia,though the University of Paris Cité,Public Assistance-Hospitals of Paris,and the National Institute of Health and Medical Research(INSERM)were the most prolific.As for individual contributions,Hisatomi H,Kaneko S,and Ide T were the three most prolific authors.Kaneko S ranked first by H-index,G-index,

Effect of alumina occurrence form on metallurgical properties of hematite and magnetite pellets

The effect of alumina occurrence form on the metallurgical properties of both hematite and magnetite pellets was investigated at the same Al_(2)O level of 2 wt.%,including reduction index(RI),low-temperature reduction disintegration index(RDI),reduction swelling index(RSI),and high-temperature softening-dripping performance.The mineralogy of fired pellets was also studied to reveal the influence of alumina occurrence form on the phase composition and microstructure.From the results,the alumina occurrence form presents tremendous impacts on the metallurgical perfor-mance of both magnetite and hematite pellets.Addition of all alumina occurrence forms contributes to inferior reducibility of pellets,especially in the case of gibbsite for magnetite pellets with a RI of 58.4%and kaolinite for hematite pellets with a RI of 56.8%.However,addition of all alumina occurrence forms improves the RDI of magnetite pellets,while there is no significant difference among various alumina occurrence forms.In contrast,alumina occurrence forms have little influence on the RDI of hematite pellets.The presence of free alumina,gibbsite,and kaolinite tends to improve the RSI of hematite and magnetite pellets,whereas hercynite gives the opposite trend with a RSI of 25.6%.For softening-dripping performance of magnetite pellets,all alumina occurrence forms contribute to narrower softening-melting interval.Meanwhile,alumina,gibbsite,and kaolinite give narrower softening-dripping interval,at 229,217,and 88℃,respectively,whereas addition of hercynite results in the largest melting range at 276℃ due to its high melting point.Regarding hematite pellets,free alumina,gibbsite,and hercynite tend to enlarge melting range,whereas kaolinite contributes to lower dripping temperature of 1148℃ and narrow softening-dripping interval of 88℃ due to the formation of a greater amount of slag phase at high temperatures.

Thermal conductivity and bending strength of SiC composites reinforced by pitch-based carbon fibers

In this work,pitch-based carbon fibers were utilized to reinforce silicon carbide(SiC)composites via reaction melting infiltration(RMI)method by controlling the reaction temperature and resin carbon content.Thermal conductivities and bending strengths of composites obtained under different preparation conditions were characterized by various analytical methods.Results showed the formation of SiC whiskers(SiC_(w))during RMI process according to vapor–solid(VS)mechanism.SiC_(w) played an important role in toughening the C_(pf)/SiC composites due to crack bridging,crack deflection,and SiC_(w) pull-out.Increase in reaction temperature during RMI process led to an initial increase in thermal conductivity along in-plane and thickness directions of composites,followed by a decline.At reaction temperature of 1600℃,thermal conductivities along the in-plane and thickness directions were estimated to be 203.00 and 39.59 W/(m×K),respectively.Under these conditions,bending strength was recorded as 186.15±3.95 MPa.Increase in resin carbon content before RMI process led to the generation of more SiC matrix.Thermal conductivities along in-plane and thickness directions remained stable with desirable values of 175.79 and 38.86 W/(m×K),respectively.By comparison,optimal bending strength improved to 244.62±3.07 MPa.In sum,these findings look promising for future application of pitch-based carbon fibers for reinforcement of SiC ceramic composites.

Cyanided Covalent Triazine Frameworks with Enhanced Adsorption Capability Toward Chloranil

Porous polymer(pyrrolopyrrole)was successfully prepared via domino-ring-formation reaction.The chemical-physical properties of cyanided covalent triazine frameworks(CTF-CN)were characteriazed by fourier transform infrared spectra(FT-IR),scanning electron microscopy(SEM),nuclear magnetic resonance(NMR),specific surface area analyzer(BET)and thermogravimetric analysis(TGA),respectively.The experimental results of adsorption of chloranil(TCBQ)in aqueous solution indicated that CTF-CN exhibited distinctive adsorption capacity toward TCBQ owing to its large specific surface area.Specifically,the adsorption equilibrium of as-prepared polymer was executed within 5 h and the calculated adsorption capacity was 499.76 mg/g.Furthermore,the adsorption kinetics could be well defined with the linear pseudo-second-order model,which implies that the chemical interaction are relative important in the course of TCBQ removal.Finally,the current studies verify that CTF-CN has unique rigid and nano-porous framework structure,which can be employed for the treatment of a series of harmful aromatic substances.

Beam dynamics optimization of very-high-frequency gun photoinjector

Very-high-frequency(VHF)gun photoinjectors,capable of producing high-brightness and high-repetitionrate electron bunches,are some of the best electron sources for driving MHz-class repetition-rate free-electron lasers.In this study,the beam dynamics optimization of a VHF gun photoinjector for Shanghai HIgh Repetition Rate X-ray Free Electron Laser and Extreme Light Facility(SHINE)is systematically demonstrated using a genetic algorithm.Through the inclusion of the solenoid geometry as an optimization variable into the genetic algorithm,the optimum projected normalized emittance for 100 pC bunches with bunch length of 1 mm rms is reduced to 0.1 mm mrad for 100%of the particles and 0.075 mm mrad for 95%of the particles,proving that sub-100 nm emittance can be achieved in the SHINE injector using a single-cell Tsinghua University(THU)VHF gun.This emittance fulfills the requirements not only of SHINE and Linac Coherent Light Source(LCLS)-II but also of LCLS-II-High Energy(LCLS-II-HE).We demonstrate that the optimal emittance in the VHF gun injector is reduced via the optimization of the solenoid geometry,thereby reducing solenoid spherical aberration.Through the inclusion of high-order(H.O.)energy spread among the optimization objectives,the H.O.energy spread can be reduced by a factor of nearly six using a high-harmonic cavity despite a 38%emittance growth.Finally,the beam dynamics in the SHINE main accelerator show that reducing the H.O.energy spread in the injector is of great significance to improving compression efficiency and reducing bunch current spike.
