Uncovering the prominent role of satellite cells in paravertebral muscle development and aging by single-nucleus RNA sequencing

To uncover the role of satellite cells(SCs)in paravertebral muscle development and aging,we constructed a single-nucleus transcriptomic atlas of mouse paravertebral muscle across seven timepoints spanning the embryo(day 16.5)to old(month 24)stages.Eight cell types,including SCs,fast muscle cells,and slow muscle cells,were identified.An energy metabolism-related gene set,TCA CYCLE IN SENESCENCE,was enriched in SCs.Forty-two skeletal muscle disease-related genes were highly expressed in SCs and exhibited similar expression patterns.Among them,Pdha1 was the core gene in the TCA CYCLE IN SENESCENCE;Pgam2,Sod1,and Suclg1 are transcription factors closely associated with skeletal muscle energy metabolism.Transcription factor enrichment analysis of the 42 genes revealed that Myod1 and Mef2a were also highly expressed in SCs,which regulated Pdha1 expression and were associated with skeletal muscle development.These findings hint that energy metabolism may be pivotal in SCs development and aging.Three ligand-receptor pairs of extracellular matrix(ECM)-receptor interactions,Lamc1-Dag1,Lama2-Dag1,and Hspg2-Dag1,may play a vital role in SCs interactions with slow/fast muscle cells and SCs self-renewal.Finally,we built the first database of a skeletal muscle single-cell transcriptome,the Musculoskeletal Cell Atlas(http://www.***.tech),which lists 630,040 skeletal muscle cells and provides interactive visualization,a useful resource for revealing skeletal muscle cellular heterogeneity during development and aging.Our study could provide new targets and ideas for developing drugs to inhibit skeletal muscle aging and treat skeletal muscle diseases.

Low-Cycle Fatigue and Creep-Fatigue Behaviors of a Second-Generation Nickel-Based Single-Crystal Superalloy at 760℃

Low-cycle fatigue(LCF)behaviors of a second-generation nickel-based single-crystal superalloys with[001]orientation at 760℃ have been investigated.Different strain amplitudes were introduced to investigate the creep-fatigue effects.The LCF life of none tensile holding(NTH)was higher than that of the 60-s tensile hold(TH)at any strain amplitude.As the strain amplitude was 0.7%,the stacking and cross-slip dislocations appeared together at the γ/γ’coherent microstructure in both TH and NTH specimens.At the strain amplitude of 0.9%,plenty of the cross-slip dislocations appeared inγchannel and other dislocations were stacking at γ/γ’interfaces.However,the SFs still appeared in γ’phase with 60-s TH which caused cyclic softening.As the strain amplitude increased up to 1.2%,the dislocations are piling up at the γ/γ’interfaces and cutting through the γ’phase in both TH and NTH tests,which caused cyclic hardening.The influences of strain amplitude and holding time were complicated.Different stress response behaviors occurred in different loading conditions.The surface characteristic and fracture mechanism were observed by scanning electron microscopy.This result is helpful for building the relationship of various blade fatigue failure modes,cyclic stress response and microstructure deformation under different strain amplitudes.

A two-stage restoration of distorted underwater images using compressive sensing and image registration

Imaging through a time-varying water surface exhibits severe non-rigid geometric distortions and motion blur.Theoretically,although the water surface possesses smoothness and temporal periodicity,random fluctuations are inevitable in an actual video sequence.Meanwhile,considering the distribution of information,the image structure contributes more to the restoration.In this paper,a new two-stage restoration method for distorted underwater video sequences is presented.During the first stage,salient feature points,which are selected through multiple methods,are tracked across the frames,and the motion fields at all pixels are estimated using a compressive sensing solver to remove the periodic distortions.During the second stage,the combination of a guided filter algorithm and an image registration method is applied to remove the structural-information-oriented residual distortions.Finally,the experiment results show that the method outperforms other state-of-the-art approaches in terms of the recovery effect and time.

Structure and flow calculation of cake layer on microfiltration membranes

Submerged membrane bioreactors （SMBR） are widely used in wastewater treatment. The permeability of a membrane declines rapidly because of the formation of a cake layer on the membrane surface. In this paper, a multiple staining protocol was conducted to probe the four major foulants in the cake layer formed on a filtration membrane. Fluorescent images of the foulants were obtained using a confocal laser scanning microscope （CLSM）. The three dimensional structure of the cake layer was reconstructed, and the internal flow was calculated using computational fluid dynamics （CFD）. Simulation results agreed well with the experimental data on the permeability of the cake layer during filtration and showed better accuracy than the calculation by Kozeny-Carman method. B-D-Glucopyranose polysaccharides and proteins are the two main foulants with relatively large volume fractions, while a-D-glucopyranose polysaccharides and nucleic acids have relatively large specific surface areas. The fast growth of B-D-glucopyranose polysaccharides in the volume fraction is mainly responsible for the increase in cake volume fraction and the decrease in permeability. The specific area, or the aggregation/dispersion of foulants, is less important to its permeability compared to its volume fraction.

Size and Dynamics of Ring Polymers under Different Topological Constraints

The impact of ring polymer length N and the influence of interchain and intrachain interactions on the size and dynamic behaviors of ring polymers,including the structural relaxation time τ_(R) and self-diffusion coefficient D,remain poorly understood at present due to a lack of systematic studies with relatively large N values.This work addressed this issue by applying dynamic Monte Carlo simulations with independently tuned interchain and intrachain interactions to investigate the size and dynamics of the ring melts with chain lengths over a wide range of 0.2N_(e)≤N≤80N_(e)(N_(e) is the entanglement length of corresponding linear chains)under different topological constraints,including all-crossing and intercrossing systems.We found that it was inappropriate to treat the unknotting constraint free energy of the ring chains in the melts as the free energy contributed by the excluded volume interactions of polymers in a good solvent.Scaling exponents of 2.5 and 1.5 reflecting the Ndependence of τ_(R) were obtained for long ring chains in non-crossing and intra-crossing systems,respectively,suggesting that the ring chains behaved as individual clusters and exhibited Zimm-like dynamics in intra-crossing systems.A single scaling exponent of-2 reflecting the Ndependence of D was obtained for ring chains in non-crossing and intra-crossing systems,indicating that the intrachain constraints affected only the value of D,and had little influence on the scaling relationship between D and N.Furthermore,the extended Stokes-Einstein relation broke down for the ring chains in the non-crossing and intra-crossing systems because the structural relaxation and translational diffusion were decoupled for the short ring systems,while both the translational diffusion and rotational relaxations,as well as diffusion at short and long time scales,were decoupled for long ring systems.

A Blockchain-Based Flexible Data Auditing Scheme for the Cloud Service

Nowdays, cloud storage technology has become a hot topic, and an increasing number of users are concerned with the security of their data in the cloud.Many auditing schemes on the cloud are proposed and the introduction of a third-party auditor to assist users in verifying the integrity of cloud data. As a centralized node, the third-party auditor has to communicate with all cloud users and cloud service providers, which becomes the bottleneck of the whole scheme. To solve this problem,we design a blockchain-based flexible cloud data auditing scheme. In our scheme, a decentralized auditing framework is proposed to eliminate the dependency on the thirdparty auditor, which increases the stability, security and performance of the whole scheme. Since the cloud service provider can automatically generates auditing proofs, our scheme can relieve the communication burdens of the cloud service provider. The proposed scheme also adapts the Merkle Hash tree to improve the verification performance.Security analysis and experiments show that the proposed scheme is secure and has better stability and verification efficiency.

A Programmable Inchworm-Inspired Soft Robot Powered by a Rotating Magnetic Field

With the growing demand for miniaturized workspaces,the demand for microrobots has been increasing in robotics research.Compared to traditional rigid robots,soft robots have better robustness and safety.With a flexible structure,soft robots can undergo large deformations and achieve a variety of motion states.Researchers are working to design and fabricate flexible robots based on biomimetic principles,using magnetic fields for cable-free actuation.In this study,we propose an inchworm-shaped soft robot driven by a magnetic field.First,a robot is designed and fabricated and force analysis is performed.Then,factors affecting the soft robot’s motion speed are examined,including the spacing between the magnets and the strength and frequency of the magnetic field.On this basis,the motion characteristics of the robot in different shapes are explored,and its motion modes such as climbing are experimentally investigated.The results show that the motion of the robot can be controlled in a two-dimensional plane,and its movement speed can be controlled by adjusting the strength of the magnetic field and other factors.Our proposed soft robot is expected to find extensive applications in various fields.

Constructing BaTiO_(3)/TiO_(2)@polypyrrole composites with hollow multishelled structure for enhanced electromagnetic wave absorbing properties

BaTiO_(3)/TiO_(2)@polypyrrole(PPy)composites with hollow multishelled structure(HoMS)were constructed to enhance the electromagnetic wave absorbing properties of BaTiO_(3)-based absorbing material.BaTiO_(3)/TiO_(2)HoMSs were prepared by hydrothermal crystallization using TiO_(2)Ho MSs as template.Then,FeCl3 was introduced to initiate the oxidative polymerization of pyrrole monomer,forming BaTiO_(3)/TiO_(2)@PPy HoMSs successfully.The electromagnetic wave absorbing properties of BaTiO_(3)/TiO_(2)HoMSs and BaTiO_(3)/TiO_(2)@PPy Ho MSs with different shell number were investigated using a vector network analyzer.The results indicate that BaTiO_(3)/TiO_(2)@PPy HoMSs exhibit improved microwave absorption compared with BaTiO_(3)/TiO_(2)HoMSs.In particular,tripled-shelled BaTiO_(3)/TiO_(2)@PPy HoMS has the most excellent absorbing performance.The best reflection loss can reach up to-21.80 dB at 13.34 GHz with a corresponding absorber thickness of only 1.3 mm,and the qualified absorption bandwidth of tripled-shelled BaTiO_(3)/TiO_(2)@PPy HoMS is up to 4.2 GHz.This work paves a new way for the development of high-performance composite microwave absorbing materials.

Impact of thymosinα1 as an immunomodulatory therapy on long-term survival of non-small cell lung cancer patients after R0 resection:a propensity score-matched analysis

Background:There is limited information about thymosinα1(Tα1)as adjuvant immunomodulatory therapy,either used alone or combined with other treatments,in patients with non-small cell lung cancer(NSCLC).This study aimed to evaluate the effect of adjuvant Tα1 treatment on long-term survival in margin-free(R0)-resected stage IA–IIIA NSCLC patients.Methods:A total of 5746 patients with pathologic stage IA-IIIA NSCLC who underwent R0 resection were included.The patients were divided into the Tα1 group and the control group according to whether they received Tα1 or not.A propensity score matching(PSM)analysis was performed to reduce bias,resulting in 1027 pairs of patients.Results:After PSM,the baseline clinicopathological characteristics were similar between the two groups.The 5-year disease-free survival(DFS)and overall survival(OS)rates were significantly higher in the Tα1 group compared with the control group.The multivariable analysis showed that Tα1 treatment was independently associated with an improved prognosis.A longer duration of Tα1 treatment was associated with improved OS and DFS.The subgroup analyses showed that Tα1 therapy could improve the DFS and/or OS in all subgroups of age,sex,Charlson Comorbidity Index(CCI),smoking status,and pathological tumor-node-metastasis(TNM)stage,especially for patients with non-squamous cell NSCLC and without targeted therapy.Conclusion:Tα1 as adjuvant immunomodulatory therapy can significantly improve DFS and OS in patients with NSCLC after R0 resection,except for patients with squamous cell carcinoma and those receiving targeted therapy.The duration of Tα1 treatment is recommended to be>24 months.
