Origin of soft-sediment deformation structures in Nihewan Basin

The Nihewan Basin is a rift basin at the junction of northern Shanxi Province and northwestern Hebei Province in north China.The basin is known for its rich paleontological fossils and ancient human remains.There are also abundant soft-sediment deformation structures(SSDS)in the thick lacustrine sediments.Previously,most SSDS have been interpreted as ice-edge features or ignored entirely.Recently,the authors have carried out several field surveys in the Nihewan Basin and found that many SSDS are sandwiched between normal lacustrine strata at multiple sections.In the excavation pit at the 10th Locality of Maliang Site(ML10),10 horizontal SSDS layers and two vertically developed geological features have been identified.Based on genesis analysis and related criteria,these features are divided into two categories:cryoturbation-triggered SSDS and earthquake-triggered SSDS.Among them,a special type of ancient ice-wedge pseudomorph(SSDS-8)of ML10 is recognized in the basin for the first time.The other 9 horizontal SSDS are mainly caused by earthquake-triggered liquefaction and slumping.They can be further divided into 14 seismic event layers.These findings indicate that the tectonic activity in the Nihewan Basin is very strong and frequent,and there were cold periods in the geological history of the basin.At the same time,the SSDS with distinct morphological characteristics and stable horizontal distribution in the basin can be used as an important indicator of stratigraphic correlation.

Expression and predictive value of miR-489 and miR-21 in melanoma metastasis

BACKGROUND Melanoma is a highly malignant skin tumour, and is one of the most rapidly growing malignant tumors in recent years. According to statistics, the morbidity of cancer increases with age, accounting for 1.6% of new cancer cases and 0.6% of deaths worldwide. Melanoma has a serious impact on society and families, thus it is of great significance to find biological markers related to the diagnosis and treatment of melanoma.AIM To explore the expression and predictive value of mir-489 and mir-21 in melanoma metastasis.METHODS A total of 60 patients with malignant melanoma treated at our hospital from June2017 to December 2018 were selected as a research group, while 40 healthy subjects were selected as a control group. qRT-PCR technique was used to detect miR-489 and miR-21 in serum of the two groups. ROC curve was drawn to evaluate the predictive value and diagnostic efficiency. Spearman test was used for correlation analysis. Logistic single-and multiple-factor analyses were performed to identify the risk factors related to melanoma metastasis.RESULTS The expression of miR-489 in the research group was significantly lower than that in the control group(P < 0.001). However, the expression of miR-21 in the research group was significantly higher than that in the control group(P < 0.001).The expression of miR-489 and miR-21 was related to TNM stage and metastasis(P < 0.001). In the diagnosis of melanoma patients, the sensitivity, specificity, and AUC of miR-489 alone were 75.56%, 80.00%, and 0.852, respectively. The sensitivity, specificity, and AUC of miR-21 alone were 77.78%, 82.22%, and 0.844,respectively. MiR-489 was negatively correlated with TNM stage of melanoma(r=-0.612, P < 0.001), while miR-21 was positively correlated with TNM stage(r =0.609, P < 0.001). Logistic single-and multiple-factor regression analyses showed that TNM stage, miR-489, and mir-21 were independent risk factors for malignant melanoma metastasis.CONCLUSION MiR-489 and miR-21 may participate in the process of melanoma occurrence,d

A comparison of perioperative outcomes between extraperitoneal robotic single-port and multiport radical prostatectomy with the da Vinci Si Surgical System

To evaluate outcomes between extraperitoneal robotic single-port radical prostatectomy(epR-spRP)and extraperitoneal robotic multiport radical prostatectomy(epR-mpRP)performed with the da Vinci Si Surgical System,comparison was performed between 30 single-port(SP group)and 26 multiport(MP group)cases.Comparisons included operative time,estimated blood loss(EBL),hospital stay,peritoneal violation,pain scores,scar satisfaction,continence,and erectile function.The median operation time and EBL were not different between the two groups.In the SP group,the median operation time of the first 10 patients was obviously longer than that of the latter 20 patients(P<0.001).The median postoperative hospital stay in the SP group was shorter than that in the MP group(P<0.001).The rate of peritoneal damage in the SP group was less than that in the MP group(P=0.017).The pain score and overall need for pain medications in the SP group were lower than those in the MP group(P<0.001 and P=0.015,respectively).Patients in the SP group were more satisfied with their scars than those in the MP group 3 months postoperatively(P=0.007).At 3 months,the cancer control,recovery of erectile function,and urinary continence rates were similar between the two groups.It is safe and feasible to perform epR-spRP using the da Vinci Si surgical system.Therefore,epR-spRP can be a treatment option for localized prostate cancer.Although epR-spRP still has a learning curve,it has advantages for postoperative pain and self-assessed cosmesis.In the absence of the single-port robotic surgery platform,we can still provide minimally invasive surgery for patients.

An interlaminar damage shell model for typical composite structures

Using the plate/shell elements in commercial software,accurate analysis of interlaminar initial damage in typical composite structures is still a challenging issue.To propose an accurate and efficient model for analysis of interlaminar initial damage,the following work is carried out:(A)A higher-order theory is firstly proposed by introducing the local Legendre polynomials,and then a novel shell element containing initial damage prediction is developed,which can directly predict transverse shear stresses without any postprocessing methods.Unknown variables at each node are independent of number of layers,so the proposed model is more efficient than the 3D-FEM.(B)Compression experiment is carried out to verify the capability of the proposed model.The results obtained from the proposed model are in good agreement with experimental data.(C)Several examples have been analyzed to further assess the capability of the proposed model by comparing to the 3D-FEM results.Moreover,accuracy and efficiency have been evaluated in different damage criterion by comparing with the selected models.The numerical results show that the proposed model can well predict the initial interlaminar damage as well as other damage.Finally,the model is implemented with UEL subroutine,so that the present approach can be readily utilized to analyze the initial damage in typical composite structures.

Climatic impacts induced by winter wheat irrigation over North China simulated by the nonhydrostatic RegCM4.7

Quantification of the impact of winter wheat irrigation on the climate and the occurrence of extreme climatic events over North China is crucial for regional adaptation planning.Previous related studies mainly focused on the impact on surface processes;however,few focused on the effects of extreme events using high-resolution nonhydrostatic regional climate models.Here,the 9-km-resolution nonhydrostatic RegCM4.7 was coupled with a crop irrigation scheme and an updated winter wheat irrigation dataset to better simulate irrigation effects.Two experiments were conducted with and without winter wheat irrigation to isolate the effects of irrigation.Results showed that irrigation simulation reduces the model biases in temperature,precipitation,latent heat flux,soil moisture,sensitive heat flux,and top-layer soil moisture.Moreover,it also reduces the bias and increases the correlation with observations obtained in irrigated areas,especially in summer,indicating better representation of irrigation schemes.Winter wheat irrigation tends to cause substantial cooling of the local surface maximum,minimum,and mean air temperatures(by-1.68,-0.34,and-0.79℃,respectively)over irrigated areas of North China,with the largest changes observed in relation to maximum temperature.Additionally,precipitation is found to increase during spring and summer,which is strongly related to water vapor transport in the lower levels of the atmosphere.Further analyses indicated that the number of annual mean hot days decrease(-13.9 d),whereas the number of both comfort days(+10.2 d)and rainy days(days with total precipitation greater than 1 mm:+6.6 d)increase over irrigated areas,demonstrating beneficial feedback to human perception and agriculture.Fortunately,although the heat wave risk increases(number of annual mean heat wave days:+5.8 d),the impact is limited to small areas within irrigated region.Additionally,no notable change was found in terms of heavy rainfall events and precipitation intensity,which might be an undereastimation caused by th

BACKGROUND In recent years,confocal laser endomicroscopy(CLE)has become a new endoscopic imaging technology at the microscopic level,which is extensively performed for real-time in vivo histological examination.CLE can be performed to distinguish benign from malignant lesions.In this study,we diagnosed using CLE an asymptomatic patient with poorly differentiated gastric adenocarcinoma.CASE SUMMARY A 63-year-old woman was diagnosed with gastric mucosal lesions,which may be gastric cancer,in the small curvature of the stomach by gastroscopy.She consented to undergo CLE for morphological observation of the gastric mucosa.Through the combination of CLE diagnosis and postoperative pathology,the intraoperative CLE diagnosis was considered to be reliable.According to our experience,CLE can be performed as the first choice for the diagnosis of gastric cancer.CONCLUSION CLE has several advantages over pathological diagnosis.We believe that CLE has great potential in the diagnosis of benign and malignant gastric lesions.

The limited infiltration and persistence of chimeric antigen receptor(CAR)-T cells is primarily responsible for their treatment deficits in solid tumors. Here, we present a three-dimensional scaffold, inspired by the physiological process of T-cell proliferation in lymph nodes. This scaffold gathers the function of loading,delivery, activation and expansion for CAR-T cells to enhance their therapeutic effects on solid tumors. This porous device is made from poly(lactic-co-glycolic acid) by a microfluidic technique with the modification of T-cell stimulatory signals, including anti-CD3, anti-CD28 antibodies, as well as cytokines. This scaffold fosters a 50-fold CAR-T cell expansion in vitro and a 15-fold cell expansion in vivo. Particularly, it maintains long-lasting expansion of CAR-T cells for up to 30 days in a cervical tumor model and significantly inhibits the tumor growth. This biomimetic delivery strategy provides a versatile platform of cell delivery and activation for CAR-T cells in treating solid tumors.

HSP110 aggravates ischemia-reperfusion injury after liver transplantation by promoting NF-κB pathway

Background:Ischemia-reperfusion injury(IRI)poses a significant challenge to liver transplantation(LT).The underlying mechanism primarily involves overactivation of the immune system.Heat shock protein 110(HSP110)functions as a molecular chaperone that helps stabilize protein structures.Methods:An IRI model was established by performing LT on Sprague-Dawley rats,and HSP110 was silenced using siRNA.Hematoxylin-eosin staining,TUNEL,immunohistochemistry,ELISA and liver enzyme analysis were performed to assess IRI following LT.Western blotting and quantitative reverse transcription-polymerase chain reaction were conducted to investigate the pertinent molecular changes.Results:Our findings revealed a significant increase in the expression of HSP110 at both the mRNA and protein levels in the rat liver following LT(P<0.05).However,when rats were injected with siRNAHSP110,IRI subsequent to LT was notably reduced(P<0.05).Additionally,the levels of liver enzymes and inflammatory chemokines in rat serum were significantly reduced(P<0.05).Silencing HSP110 with siRNA resulted in a marked decrease in M1-type polarization of Kupffer cells in the liver and downregulated the NF-κB pathway in the liver(P<0.05).Conclusions:HSP110 in the liver promotes IRI after LT in rats by activating the NF-κB pathway and inducing M1-type polarization of Kupffer cells.Targeting HSP110 to prevent IRI after LT may represent a promising new approach for the treatment of LT-associated IRI.

Super-Resolution Fluorescence Microscopy for Cilia Investigation and Ciliopathy Diagnosis(Invited)

The last two decades have witnessed the invention and development of super-resolution microscopy(SRM)that breaks the diffraction limit of light and pushes the fluorescence microscopy resolution to several nanometers.While SRM is widely used in biological studies,such as resolving subdiffraction structures,molecular mapping,tracking single molecules,probing protein-protein interactions,and observing organelle dynamics,its direct application in translational medicine,such as disease diagnosis,is still preliminary.Despite their small size,cilia play a crucial role as organelles in cell signaling and motility,with defects in cilia leading to ciliopathy.Similar to other miniature organelles and macromolecular complexes,cilia are ideal for super-resolution imaging.In this review,we will 1)introduce cilia and ciliopathy,2)show how SRM extends our knowledge of cilia,and 3)focus on how SRM improves the diagnosis of motile ciliopathies.

Sum-Rate Maximization in Active RIS-Assisted Multi-Antenna WPCN

In this paper,we propose an active reconfigurable intelligent surface(RIS)enabled hybrid relaying scheme for a multi-antenna wireless powered communication network(WPCN),where the active RIS is employed to assist both wireless energy transfer(WET)from the power station(PS)to energyconstrained users and wireless information transmission(WIT)from users to the receiving station(RS).For further performance enhancement,we propose to employ both transmit beamforming at the PS and receive beamforming at the RS.We formulate a sumrate maximization problem by jointly optimizing the RIS phase shifts and amplitude reflection coefficients for both the WET and the WIT,transmit and receive beamforming vectors,and network resource allocation.To solve this non-convex problem,we propose an efficient alternating optimization algorithm with the linear minimum mean squared error criterion,semidefinite relaxation(SDR)and successive convex approximation techniques.Specifically,the tightness of applying the SDR is proved.Simulation results demonstrate that our proposed scheme with 10 reflecting elements(REs)and 4 antennas can achieve 17.78%and 415.48%performance gains compared to the single-antenna scheme with 10 REs and passive RIS scheme with 100 REs,respectively.
