The common marmoset(Callithrix jacchus)has emerged as a valuable nonhuman primate model in biomedical research with the recent release of high-quality reference genome *** marmosets have been independently reported in two Asian primate research ***,the population genetics within these primate centers and the specific genetic variants associated with epilepsy in marmosets have not yet been ***,we characterized the genetic relationships and risk variants for epilepsy in 41 samples from two epileptic marmoset pedigrees using whole-genome *** identified 14558184 single nucleotide polymorphisms(SNPs)from the 41 samples and found higher chimerism levels in blood samples than in fingernail *** analysis showed fourth-degree of relatedness among marmosets at the primate *** addition,SNP and copy number variation(CNV)analyses suggested that the WW domain-containing oxidoreductase(WWOX)and Tyrosine-protein phosphatase nonreceptor type 21(PTPN21)genes may be associated with epilepsy in ***,KCTD18-like gene deletion was more common in epileptic marmosets than control *** study provides valuable population genomic resources for marmosets in two Asian primate *** analyses identified a reasonable breeding strategy for genetic diversity maintenance in the two centers,while the case-control study revealed potential risk genes/variants associated with epilepsy in marmosets.

The so-called ＂wild abortive＂（wa） type of cytoplasmic male sterility （CMS） derived from a wild rice species Oryza rufipogon has been extensively used for hybrid rice breeding. However, extensive analysis of the structure of the related mitochondrial genome has not been reported, and the CMS-associated gene（s） remain unknown. In this study, we exploited a mitochondrial genome-wide strategy to examine the structural and expressional variations in the mitochondrial genome conferring the CMS. The entire mitochondriai genomes of a CMS-wa line and two normal fertile rice lines were amplified by Long-polymerase chain reaction into tilling fragments of up to 15.2 kb. Restriction and DNA blotting analyses of these fragments revealed that structural variations occurred in several regions in the wa mitochondrial genome, as compared to those of the fertile lines. All of the amplified fragments covering the entire mitochondrial genome were used as RNA blot probes to examine the mitochondriai expression profile among the CMS-wa and fertile lines. As a result, only two mRNAs were found to be differentially expressed between the CMS-wa and the fertile lines, which were detected by a probe containing the nad5 and orf153 genes and the other having the ribosomal protein gene rpl5, respectively. These mRNAs are proposed to be the candidates for further identification and functional studies of the CMS gene.

Alfalfa(Medicago sativa L.)is the most important legume forage crop worldwide with high nutritional value and *** a long time,the breeding of alfalfa was hampered by lacking reliable information on the autotetraploid genome and molecular markers linked to important agronomic *** herein reported the de novo assembly of the allele-aware chromosome-level genome of Zhongmu-4,a cultivar widely cultivated in China,and a comprehensive database of genomic variations based on resequencing of 220 *** 2.74 gb contigs(N50 of 2.06 Mb),accounting for 88.39%of the estimated genome,were assembled,and 2.56 gb contigs were anchored to 32 pseudo-chromosomes.A total of 34,922 allelic genes were identified from the allele-aware *** observed the expansion of gene families,especially those related to the nitrogen metabolism,and the increase of repetitive elements including transposable elements,which probably resulted in the increase of Zhongmu-4 genome compared with Medicago *** structure analysis revealed that the accessions from Asia and South America had relatively lower genetic diversity than those from Europe,suggesting that geography may influence alfalfa genetic divergence during local ***-wide association studies identified 101 single nucleotide polymorphisms(SNPs)associated with 27 agronomic *** candidate genes were predicted to be correlated with fall dormancy and salt *** believe that the alleleaware chromosome-level genome sequence of Zhongmu-4 combined with the resequencing data of the diverse alfalfa germplasms will facilitate genetic research and genomics-assisted breeding in variety improvement of alfalfa.

Mechanical performances and microstructural characteristics of reactive MgO-carbonated silt subjected to freezing-thawing cycles

The characteristics of reactive magnesia(MgO)-carbonated silt in respect to long-term stability have not been well understood in severely cold climate despite the usage of reactive MgO in enhancing the engineering *** the binder content of 15%and initial water content of 25%,MgO-admixed silt specimens were carbonized for 3 h and 6 h and then subjected to different numbers of freezingthawing(F-T)*** different F-T cycles,the physico-mechanical properties of MgO-carbonated silt were analyzed in comparison with Portland cement(PC)-stabilized silt through physical and unconfined compression ***,a series of micro tests on MgO-carbonated specimens was performed including X-ray diffraction(XRD),scanning electron microscopy(SEM)and mercury intrusion porosimetry(MIP)*** results demonstrate that both mass change ratio and moisture content of carbonated/stabilized silt decrease,and these values of MgO-carbonated silt are significantly lower while the density is higher compared to PC-stabilized *** strengths and moduli of MgO-carbonated silt are still two times higher than those of PC-stabilized specimens and the strength change ratio of keeps above0.8 after F-T *** is no visible transformation between nesquehonite and dypingite/hydromagnesite,although the XRD peaks of nesquehonite decrease and the bonding and filling effects weaken *** 6 and 10 F-T cycles,the pore-size characteristics changed from a unimodal distribution to a three-peak and bimodal distribution,*** total,macro and large pore volumes increase obviously while the medium and small pore volumes decrease except for intra-aggregate *** findings show better F-T durability of MgO-carbonated silt,which would be helpful for facilitating the application of MgO carbonation in the soil treatment.

Immunoglobulin g against myelin oligodendrocyte glycoprotein(MOg-Ig g) is detectable in neuromyelitis optica spectrum disorder(NMOSD) without aquaporin-4 Ig g(AQP4-Ig g), but its pathogenicity remains *** this study, we explored the pathogenic mechanisms of MOg-Ig g in vitro and in vivo and compared them with those of AQP4-Ig g. MOg-Ig g-positive serum induced complement activation and cell death in human embryonic kidney(HEK)-293 T cells transfected with human MOg. In C57 BL/6 mice and Sprague-Dawley rats, MOg-Ig g only caused lesions in the presence of complement. Interestingly, AQP4-Ig g induced astroglial damage, while MOgIg g mainly caused myelin loss. MOg-Ig g also induced astrocyte damage in mouse brains in the presence ofcomplement. Importantly, we also observed ultrastructural changes induced by MOg-Ig g and AQP4-Ig g. These findings suggest that MOg-Ig g directly mediates cell death by activating complement in vitro and producing NMOSDlike lesions in vivo. AQP4-Ig g directly targets astrocytes,while MOg-Ig g mainly damages oligodendrocytes.

Hydrous electrolytes with high electrochemical potentials were obtained by hydrating water molecules into solutes to form high Li:water molar ratio electrolytes(HMRE).Solid polyethylene glycol(PEg) were e mployed to enha nce the molar ratio of Li^(+) to water in the electrolytes while reducing the consumption of *** obtained mole ratio of Li^(+) to wa ter molecules in the hydrous electrolytes was greater than 1:1;however,the mass fraction of Li-salt was reduced to 61%(approximately 5.5 mol/kg,based on water and PEg).Compared with that of water-in-salt electrolytes,the mass fraction of Li-salt could be remarkably reduced by adding solid *** electrochemical stability of the electrolytes improved considerably because of the strong hydration of Li^(+) by the water molecules.A beneficial passivation effect,arising from the decomposition of the electrolyte,at a wide potential window was observed.

浮体式结构作为一种适合于沿海地区及内河水利枢纽工程建设的新型生态构筑物,当在平原水利防洪工程中运行时,由于浮体的阻挡,导致水流发生分流产生水头损失,进而影响过流能力。因此,研究浮体式结构阻力系数的变化规律及因素影响机理具有现实意义。本文采用物理模型试验,基于水动力学理论分析,利用量纲分析得到影响浮体式结构阻力系数的影响因素有水位变化系数、浮体结构长度与浮体结构位置比值(长位比)、相对阻水面积及雷诺数。对影响因素与阻力系数进行相关性研究及敏感性分析,结果表明:1)水位变化系数的增加,导致结构背水面的位置出现回流区,水流质点的紊动强度增加,阻力系数增大;2)浮体式结构沿水流长度方向及位置的变化,一定程度上并不会引起水流能量的损失,对浮体结构的阻力系数的影响较小;3)相对阻水面积系数的增大会导致水体过水面积减小,水体质点相互摩擦和碰撞消耗水流能量,引起浮体式结构的阻力系数增大;4)在试验的雷诺数范围内(3.5相对阻水面积>雷诺数>长位比。通过基于最小二乘法的多元线性拟合,比较得到描述浮体式结构阻力系数的计算公式,并采用数理统计对公式进行检验和误差分析,该公式在可靠性及精确度方面能够满足工程需要且形式简单、通用性强,可为相关工程提供理论依据。

A cross-sectional study was conducted to evaluate the risk factors associated with the Salmonella infection status of dairy herds in Henan and Hubei provinces,*** were assigned a Salmonella status based on the isolation of Salmonella from fecal samples obtained from adult lactating dairy *** on potential biosecurity risk factors was collected using a questionnaire template via a 15-min face-to-face interview with dairy cattle producers from May 2020 to March *** questionnaire consisted of 23 questions across two broad categories of potential biosecurity risk factors:farm and biosecurity managemental factors.A total of 48 farms were *** all(100%,48/48)of the surveyed farms,although sick animals were separated from healthy animals using fencing,there were no strict quarantine protocols for newly introduced *** species rearing was practiced in 35.4%(17/48)of the *** were removed more than once a day in 45.8%(22/48)of the farms,whereas the remainder(54.2%,26/48)only removed animal feces once a day.A total of 29.2%(14/48)of the farms were locatedwas consistently performed on all farms,with a major-ity of the workers on most farms(81.3%,39/48)always disinfecting footwear before entering production areas.A significant association between Salmonella-positive status and the high frequency of the presence of wildlife(birds and rodents in sheds and feed storage places)was recorded(OR:11.9,95%Cl:1.7,84.1,p=0.013).Fortunately,no farms shared farm equipment with other *** study highlights the occurrence of wildlife as a risk factor for the presence of Salmonella in investigated dairy *** is a need to institute appropriate on-farm control measures for wild birds and rodents to control the potential spread of Salmonella in dairy production systems.

Jerusalem artichoke(Helianthus tuberosus L.) not just can be used for bioethanol production but may be potentially used in phytoremediation for the removal of heavy metal *** Jerusalem artichoke cultivars,N2 and N5,were subjected to six cadmium(Cd) concentrations(0,5,25,50,100 and 200 mg L1) to investigate Cd tolerance and *** 21 days of growth,the effects of Cd on growth,chlorophyll content,net photosynthetic rate,intercellular CO2 concentration and malondialdehyde content were *** growth parameters were reduced under Cd *** two Jerusalem artichoke cultivars had relatively high Cd tolerance and accumulation capacity(> 100 mg kg1),with N5 being more tolerant and having higher Cd accumulation than *** accumulated more Cd than stems and *** bioconcentration factors(far higher than 1) and translocation factors(lower than 1) decreased with an increase in Cd *** results suggested that Jerusalem artichoke could be grown at relatively high Cd loads,and N5 could be an excellent candidate for phytoremediation of Cd-contaminated soils.
