Dendrobium species regulate energy homeostasis in neurodegenerative diseases: a review

Brain energy homeostasis is a vital physiological function in maintaining a balanced internal metabolic environment.The impairment of energy homeostasis is recognized as a key pathophysiological basis for brain metabolic disorders and related neurodegenerative diseases.Dendrobium species(‘Shihu’in Chinese)such as D.officinale,D.huoshanense,D.nobile,D.chrysanthum,D.loddigesii,D.moniliforme,D.gratiosissimum,D.candidum and D.caulis are widely used as traditional Chinese medicines/nutraceuticals to control and treat neurodegenerative disorders.These dietary herbs and their derived compounds possess a variety of biological properties,such as suppression of oxidative stress and neuroinflammation,regulation of energy homeostasis mainly through improving brain mitochondria function,insulin signaling and lipid metabolism.Furthermore,they reduce neurotoxicity,alleviate brain injury and neuropathy,and prevent neurodegenerative conditions including stroke,Alzheimer’s disease,Parkinson’s disease,and Huntington’s disease in humans and/or rodents.Moreover,the nutraceuticals from Dendrobium species promote gut health and aid digestion,which appear to be associated with beneficial effects on brain energy homeostasis.Based on the above-mentioned health benefits associated with Dendrobium species,this work reviews their nutraceutical role in neurodegenerative disorders and further suggests the need to elucidate mechanisms of the underlying molecular actions.

印顺法师对大乘佛法判为三系,即性空唯名、虚妄唯识和真常唯心。本文提出会通三系是印顺法师佛学思想的一个重要特征,并从两个方面展开论述。首先,以八不中道、三法印、缘起性空、如来藏、现证法空性等教理教义阐述印顺法师会通三系深刻内涵;其次,指出他在会通三系时所表现出来的理论特征以及注意分辨的几个问题:一是强调三系的差别;二是容忍对三系的异议;三是以空宗兼摄唯识与真常。总的来说,他认为三系各有其圆缺、优劣,必须做到取长补短、参证会通。

This paper deals with a novel local arc length estimator for curves in gray-scale images.The method first estimates a cubic spline curve fit for the boundary points using the gray-level information of the nearby pixels,and then computes the sum of the spline segments’lengths.In this model,the second derivatives and y coordinates at the knots are required in the computation;the spline polynomial coefficients need not be computed explicitly.We provide the algorithm pseudo code for estimation and preprocessing,both taking linear time.Implementation shows that the proposed model gains a smaller relative error than other state-of-the-art methods.

Hepatic fibrosis is a common pathological process that occurs in the development of various chronic liver diseases into cirrhosis and liver cancer,characterized by excessive deposition of the extracellular matrix.In the past,hepatic fibrosis was thought to be a static and irreversible pathological process.In recent years,with the rapid development of molecular biology and the continuous in-depth study of the liver at the microscopic level,more and more evidence has shown that hepatic fibrosis is a dynamic and reversible process.Therefore,it is particularly important to find an effective,simple,and inexpensive method for its prevention and treatment.Traditional Chinese medicine(TCM)occupies an important position in the treatment of hepatic fibrosis due to its advantages of low adverse reactions,low cost,and multi-target effectiveness.A large number of research results have shown that TCM monomers,single herbal extracts,and TCM formulas play important roles in the prevention and treatment of hepatic fibrosis.Oxidative stress(OS)is one of the key factors in the occurrence and development of hepatic fibrosis.Therefore,this article reviews the progress in the understanding of the mechanisms of TCM monomers,single herbal extracts,and TCM formulas in preventing and treating hepatic fibrosis by inhibiting OS in recent years,in order to provide a reference and basis for drug therapy of hepatic fibrosis.

There are many people in China who suffered from physical and mental diseases and need physical and psychological rehabilitation. Traditional treatments can work in rehabilitation, but will take much money and labor. In recent years, virtual reality(VR) technology has become a new direction to innovate rehabilitation. This paper summarizes the research results of VR technology in rehabilitation for stroke, anxiety, depression and autism. Based on these results, we propose a framework for VR rehabilitation system with virtual agents. A prototype system is developed to corroborate the proposed design guidelines. With the prototype rehabilitation system, autistic children can train their life skills in different scenarios. Preliminary test results show the proposed framework can push users to take the rehabilitation training actively and can be a new method for rehabilitation training.

The production of enantiopure D-lysine from L-lysine by a two-strain coupled system

The microbial production of D-lysine to replace chemical approach has gained great interest with the rising concerns over the environment.Here,we employed recombinant E.coli strain BL21-LYR with lysine racemase and strain BL-22A-RB-YB with L-lysine monooxygenase and 5-aminovaleramide amidohydrolase to establish a two-strain coupling whole-cell bioconversion system for D-lysine production from L-lysine.To improve the optical purity of D-lysine,the optimal reaction condition for resolution of DL-lysine after the racemization was investigated.The specificity of BL-22A-RB-YB for L^-lysine and the effects of reaction condition on bioconversion efficiency of whole-cell were accordingly determined.Under the optimal condition,a maximum 53.5 g·L^-1 D-lysine and 48.2 g·L^-15-AVA were obtained with yield of 47.4%and 42.3%,respectively,by the microbial racemization and asymmetric degradation process.The final D-lysine enantiomeric excess was over 99%.Meanwhile,a valuable compound 5-aminovaleric acid was synthesized with the production of D-lysine,indicating the economic feasibility of the two-strain coupling system.

Exploring Mechanism of Key Chinese Herbal Medicine on Breast Cancer by Data Mining and Network Pharmacology Methods

Objective To screen the key Chinese Herbal Medicines(KCHMs)against breast cancer by data mining,and analyze the potential mechanism of KCHMs using network pharmacology method.Methods Clinical prescriptions consisted of CHMs for treating breast cancer were screened,and then Traditional Chinese Medicine Inheritance Support System(TCMISS)was applied to obtain the KCHMs.Subsequently,active ingredients and corresponding target genes of KCHMs were searched by Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)database,and target genes of breast cancer were collected using OMIM and MalaCards.After that,the overlapping target genes of KCHMs and breast cancer were screened,and the protein-protein interaction(PPI)network was built.In addition,a network of“KCHMs-active ingredients-breast cancer-targets”was constructed by Cytoscape 3.7.1.Finally,Kyoto Encyclopedia of Genes and Genomes(KEGG)and Gene Ontology(GO)analysis were performed with Database for Annotation,Visualization and Integrated Discovery(DAVID)database to reveal the action mechanism of KCHMs.Results A total of 7 KCHMs were identified,whose active ingredients include quercetin,luteolin,nobiletin,kaempferol,isorhamnetin,naringenin,and be-ta-sitosterol,etc.Based on protein-protein interaction analysis,core targets were ESR1,MYC,CCND1,EGFR,CASP3,ERBB2,etc.Several KEGG pathways(e.g,PI3K-Akt,p53,ErbB,and HIF-1 signaling pathways)were found.Conclusion Based on the combination of the data mining method and network pharmacology approach,the therapeutic effect of KCHMs on breast cancer may be realized by acting on target genes and signaling pathways related to the formation and progression of breast cancer.

The development of amplification strategies is one of the central challenges for detection of lowabundance targets. One-to-many(1:M) amplification strategies in which one target lights many signal probes, has improved the detection sensitivity in bulk solution, but with discounted contrast in cell imaging, because the lighted probes are dissociative and dispersible. In this work, a one-to-large(1:L) signaling mechanism, in which the lighted probes were orderly connected to each other, was conceptually proposed to enhance the contrast in cell imaging by avoiding signal dispersion in amplification. Accordingly,target-triggered hairpin-free chain-branching assembly(HFCBA) holds great potential to implement the1:L mechanism, but using it in cell imaging has yet to be demonstrated. As a proof of concept, a group of probes were first programmed to implement mi RNA-21-triggered HFCBA. After transfection of probes,gradually-growing signal flares in cells were monitored along with the growth of DNA dendrimers;and the in situ fluorescence accumulation in HFCBA resulted in highly-enhanced contrast to the surrounding by avoiding signal dispersion in amplification. The contrast-enhanced imaging with signal amplification is significant for biological analysis and molecular medicine. We expect the 1:L mechanism will provide a new thought for high-performance imaging of biomarkers in cells.
