Spatial transcriptomics reveals that metabolic characteristics define the tumor immunosuppression microenvironment via iCAF transformation in oral squamous cell carcinoma

Tumor progression is closely related to tumor tissue metabolism and reshaping of the microenvironment. Oral squamous cell carcinoma (OSCC), a representative hypoxic tumor, has a heterogeneous internal metabolic environment. To clarify the relationship between different metabolic regions and the tumor immune microenvironment (TME) in OSCC, Single cell (SC) and spatial transcriptomics (ST) sequencing of OSCC tissues were performed. The proportion of TME in the ST data was obtained through SPOTlight deconvolution using SC and GSE103322 data. The metabolic activity of each spot was calculated using scMetabolism,and k-means clustering was used to classify all spots into hyper-, normal-, or hypometabolic regions. CD4T cell infiltration and TGF-βexpression is higher in the hypermetabolic regions than in the others. Through CellPhoneDB and NicheNet cell-cell communication analysis, it was found that in the hypermetabolic region, fibroblasts can utilize the lactate produced by glycolysis of epithelial cells to transform into inflammatory cancer-associated fibroblasts (iCAFs), and the increased expression of HIF1A in iCAFs promotes the transcriptional expression of CXCL12. The secretion of CXCL12 recruits regulatory T cells (Tregs), leading to Treg infiltration and increased TGF-β secretion in the microenvironment and promotes the formation of a tumor immunosuppressive microenvironment. This study delineates the coordinate work axis of epithelial cells-iCAFs-Tregs in OSCC using SC, ST and TCGA bulk data, and highlights potential targets for therapy.

A Transformer-Assisted Cascade Learning Network for Choroidal Vessel Segmentation

As a highly vascular eye part,the choroid is crucial in various eye disease ***,limited research has focused on the inner structure of the choroid due to the challenges in obtaining sufficient accurate label data,particularly for the choroidal ***,the existing direct choroidal vessel segmentation methods for the intelligent diagnosis of vascular assisted ophthalmic diseases are still unsatisfactory due to noise data,while the synergistic segmentation methods compromise vessel segmentation performance for the choroid layer segmentation *** cascaded structures grapple with error propagation during *** address these challenges,we propose a cascade learning segmentation method for the inner vessel structures of the choroid in this ***,we propose TransformerAssisted Cascade Learning Network(TACLNet)for choroidal vessel segmentation,which comprises a two-stage training strategy:pre-training for choroid layer segmentation and joint training for choroid layer and choroidal vessel *** also enhance the skip connection structures by introducing a multi-scale subtraction connection module designated as MSC,capturing differential and detailed information ***,we implement an auxiliary Transformer branch named ATB to integrate global features into the segmentation *** results exhibit that our method achieves the state-of-the-art performance for choroidal vessel ***,we further validate the significant superiority of the proposed method for retinal fluid segmentation in optical coherence tomography(OCT)scans on a publicly available *** these fully prove that our TACLNet contributes to the advancement of choroidal vessel segmentation and is of great significance for ophthalmic research and clinical application.

　本文对道德博弈和生存博弈在中国传统社会的超稳定演进过程中的作用进行了分析。我们认为,传统社会中的被统治者利用起义等变革手段,通过道德博弈对统治者在生存博弈中破坏了的社会结构进行重建,从而使得中国传统社会表现出超稳定系统的特征。新古典的生存博弈不会因为分配过程的公平而改变传统社会结构,并回复到道德重建的原初状态。道德教化的成功有助于缓和生存博弈的激烈程度。

Endometrial Collagen Fibril Hyperplasia is Associated with Implantation Failure in Women Undergoing IVF-ET

Objective To analyse the effects of controlled ovarian hyperstimulation （ COH） on the endometrial expression of collagen fibril （CF） during the peri-implantation period in patients undergoing IVF, and its relation to endometrial receptivity （ER） in repeated implantation failure（RIF）. Methods Peripheral blood and endometrial biopsies were obtained from 45 infertile women on days 5, 7 or 9 after oocytes retrieval or ovulation in a stimulated cycle （SC） and natural cycle （NC） respectively. CF was assayed by transmission electron microscope and quantified by modified Masson dyeing. The outcome of subsequent embryo transfer（ET） was observed. Results Levels of both E2 and progesterone were higher in the peripheral blood in SC than in NC. Also the expression of CF in the stroma in each secretory phase was increased significantly in SC （P 〈O.05）. After embryo transferring, expression levels of CF in the pregnancy group dropped between the mid- and late-secretory phase, but no change in the non-pregnancy group. In the same term, all patients undergone endometrial curettage had higher pregnancy rate than those without. Conclusion Imbalance of production and degradation of endometrial CF in the secretory phase resulting from COH may be the cause of defective ER and implantation failure in some RIF patients. Endometrial curettage may improve implantation rate by inducing appropriate CF hyperplasia and degradation.

Broadband electromagnetic(EM)wave absorption materials play an important role in military stealth and health ***,metal–organic frameworks(MOFs)-derived magnetic-carbon CoNiM@C(M=Cu,Zn,Fe,Mn)microspheres are fabricated,which exhibit flower-like nano–microstructure with tunable EM response *** on the MOFs-derived CoNi@C microsphere,the adjacent third element is introduced into magnetic CoNi alloy to enhance EM wave absorption *** term of broadband absorption,the order of efficient absorption bandwidth(EAB)value is Mn>Fe=Zn>Cu in the CoNiM@C ***,MOFs-derived flower-like CoNiMn@C microspheres hold outstanding broadband absorption and the EAB can reach up to 5.8 GHz(covering 12.2–18 GHz at 2.0 mm thickness).Besides,off-axis electron holography and computational simulations are applied to elucidate the inherent dielectric dissipation and magnetic *** heterointerfaces in CoNiMn@C promote the aggregation of the negative/positive charges at the contacting region,forming interfacial *** graphitized carbon layer catalyzed by the magnetic CoNiMn core offered the electron mobility path,boosting the conductive *** importantly,magnetic coupling is observed in the CoNiMn@C to strengthen the magnetic responding *** study provides a new guide to build broadband EM absorption by regulating the ternary magnetic alloy.

A chimeric construct consisting of the double CaMV35S promoter fused to the Myc-vhb gene encoding Vitreoscilla (bacterial) hemoglobin(VHb) was introduced into Arabidopsis thaliana and transgenic plants were cultured under various growth and stress conditions with the aim of studying the generality *** analysis indicated that the exogenous vhb was stably integrated into the Arabidopsis genome and successfully *** vhb expression improved the germination rate of transgenic *** submergence conditions,in vitro and in vivo bioassays revealed that the vhb expression protected transgenic plants against prolonged *** discs from the transgenic plants maintained in standard growth conditions reveal increased tolerance to nitrosative stress by sodium nitroprusside(SNP) *** differences in chlorophyll a,b,total protein content and conductivity contents were observed among the transgenic,wild-type(WT) and blank transformation(CK) *** significant differences were observed between the tested VHb,WT and CK lines in the presence of oxidative stresses by paraquat *** expression also affected the levels of some Arabidopsis endogenesis genes that are involved in oxygen metabolism and antioxidants biosynthesis;up-regulating their expression at transcriptional ***-MS revealed that VHb expression shifted plant secondary metabolism,which may reflect the availability of extra metabolite production for possible enhanced protection *** transgenic plants were more tolerant to photo-oxidative damage due to the increased antioxidants,such as *** results suggested that vhb gene might have the potential use in molecular breeding for the improvement of stress resistance and fine chemicals production in plants.

Understanding the foraging behavior is essential for investigating seabird ecology and conservation,as well as monitoring the well-being of the marine *** seabirds adopt diverse foraging strategies to maximize energy gains and cope with the intensified challenges of parenting and *** tradeoff may stem from the heterogeneity of food resources and the constraints of central place ***,abundant marine productivity could alleviate the energy limitation for seabirds,resulting in a consistent foraging ***,we investigated the foraging strategy during the breeding season of a cryptic small-sized seabird,Swinhoe’s Storm-petrel(Hydrobates monorhis),in the Yellow Sea,a productive marginal sea of the Northwest *** GPS tracking,we evaluated habitat preference,quantified the foraging strategy,and tested if environmental conditions and individual traits influence foraging *** found that Swinhoe’s Storm-petrels preferred nearshore areas with shallow water and engaged in primarily short foraging *** southeastward and southwestward strategies emerged when combining trip metrics,including foraging direction,duration,and maximum *** bathymetry,proximity to the coastline,and sea surface temperature differed in two foraging *** strategies exhibited flexibility between individuals,potentially explained by wing morphology,in which longer-winged birds are more likely to embark on longer-distance foraging *** findings highlight the impact of environmental factors and individual traits on seabirds’foraging decisions in productive marginal sea *** study also provides valuable insights into the foraging ecology of this Asian endemic storm-petrel.
