AIM: The aims of this study were to identify theclinicopathological features of Chinese HNPCC families andto evaluate the value of criteria for suspected HNPCC(sHNPCC) in clinical ***: According to the follow-up records, 54 HNPCCfamilies (including 12 TCG-HNPCC families and 42 sHNPCCfamilies) were screened out from patients with colorectal cancers(CRCs), operated upon in 2nd Affiliated hospital of zhejiangUniversity from 1984 to 2001. Clinical data of probands andtumor spectrum in these families were listed and analyzed,RESULTS: (1) Mean age, proportion of colonic cancer, poorlydifferentiated cancer, multiple CRCs and Dukes′ A+B of theprobands in ICG-HNPCC and sHNPCC kindred were 39ys and47.5ys, 75 % and 62 %, 0 and 12.8 %, 16.7 % and 14.3 %,58.3 % and 81%,respectively. Compared with sporadiccolorectal cancers, probands from ICG-HNPCC and sHNPCCfamilies were obviously different at age of onset(P＝0.025and 0.031), tumor location (P＝0.001 and 0.000),differentiation(P＝0.002 and 0.011) and development ofmultiple tumors (P＝0.014 and 0.002). (2) A total of 178malignant neoplasms were found in 54 HNPCC families,including 139 colorectal cancers. Besides of colorectal cancer,extracolonic tumors occurred in stomach, endometrium,hepatobiliary system, and so on (8 gastric cancers, 6 endometrialcancers, 6 hepatobiliary system cancers and 19 others) canalso be seen in Chinese ICG-HNPCC and sHNPCC ***: (1) Chinese HNPCC families have specificclinicopathological features, such as early onset, predilectionfor the involvement of colon, tendency of multiple CRCs,development of extracolonic tumors and well differentiation.(2) The criteria for suspected HNPCC is useful in clinicaldiagnosis and management of HNPCC.

AIM:To investigate the relationship between human papillomavirus (HPV) infection and concurrent esophagus and gastric cardia cancer from the same patient (CC) and examine the significance of P16 INK4A protein ***:Polymerase chain reaction was used to detect the presence of HPV type16 (HPV16).The expression of P16 INK4A protein was detected using ***:Among the CC specimens,HPV16-DNA was found in eight cases of esophageal squamous cell carcinoma (ESCC) and five cases of gastric cardia adenocarcinoma (GCA),respectively (47% vs 29%),and two of both ESCC and GCA.P16 INK4A was highly expressed in both ESCC and *** the HPV-associated positive CC,higher P16 INK4A expression was observed in the GCA than in the ESCC (75% vs 25%,P < 0.05).CONCLUSION:HPV16 as a correlated risk factor may play an important role in the development of ESCC and GCA.P16 INK4A may be a screening index in the HPVassociated carcinoma of gastric cardia.

This article is to summarize the molecular and functional analysis of the gene “suppression of tumorigenicity 13” (ST13). ST13 is in fact the gene encoding Hsp70 interacting protein (Hip), a co-factor (co-chaperone) of the 70-kDa heat shock proteins (Hsc/Hsp70). By collaborating with other positive co-factors such as Hsp40 and the Hsp70-Hsp90 organizing protein (Hop), or competing with negative co-factors such as Bcl2-associated athanogen 1 (Bag1), Hip facilitates may facilitate the chaperone function of Hsc/Hsp70 in protein folding and repair, and in controlling the activity of regulatory proteins such as steroid receptors and regulators of proliferation or apoptosis. Although the nomenclature of ST13 implies a role in the suppression of tumorigenicity (ST), to date available experimental data are not sufficient to support its role in cancer development, except for the possible down-regulation of ST13 in gastric and colorectal cancers. Further investigation of this gene at the physiological level would benefit our understanding of diseases such as endocrinological disorders, cancer, and neurodegeneration commonly associated with protein misfolding.

AIM: To investigate the association between peroxisome proliferator-activated receptor-γ (PPAR-γ ) gene polymorphism 34 C>G and colorectal cancer (CRC),a meta-analysis review was performed in this ***: A systematic literature search and selection of eligible relevant studies were carried *** independent studies with a total number of 4533 cases and 6483 controls were included in the meta-analysis on the association between polymorphism 34 C>G and ***: There was no evidence for the association between PPAR-γ 34 C>G and CRC if all of the subjects in the nine studies were ***,CG + GG showed a marginally significant difference from CC (OR = 0.84,95% CI: 0.69-1.01,P = 0.07) in random-effect *** meta-analysis indicated that PPAR-γ 34 C>G was associated with colon cancer (OR = 0.8,95% CI: 0.65-0.99,P = 0.04) in random-effect model,and the G allele decreased colon cancer *** significant association was observed between PPAR-γ 34 C>G and rectal ***: PPAR-γ 34 C>G is associated with colon cancer risk,but not associated with CRC and rectal cancer risk.

A human rectal poorly-differentiated adenocarcinoma cell line HR-8348 is established from thesurgically removed tumor mass of a male adult patient. The cell doubling time is 36.05 h. The mitoticindex is 44.5‰. The cells are all of epithelial type and mostly of polygonal shape. Electron microscopicexamination shows malignant nuclei with enlarged nucleoli; there are abundant microvilli and some se-cretory granules. The plating efficiency in soft agar is 12.9%. In PHA (1μg/ml), the cells show obviousagglutination. Chromosomal analysis reveals a mode of 70 chromosomes per cell, with a marker chromo-some. CEA in cells is detected positively both qualitatively and quantitatively. Hetero-transplantation ofthe cells to nude mice shows that the tumor mass has the same morphology as the original mass fromthe patient, with Alcian blue positive material in the cytoplasm. Repeated detection for mycoplasma con-tamination shows negative.

Objective:We aimed to perform a preliminary study of the association between induced pluripotent stem cell(iPS)-related genes and biological behavior of human colorectal cancer(CRC) cells,and the potential for developing anti-cancer drugs targeting these ***:We used real-time reverse transcriptase polymerase chain reaction(RT-PCR) to evaluate the transcript levels of iPS-related genes NANOG,OCT4,SOX2,C-MYC and KLF4 in CRC cell lines and cancer stem cells(CSCs)-enriched tumor *** was knockdowned in CRC cell line SW620 by lentiviral transduction.3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) assays,plate colony formation,and a mouse xenograft model were used to evaluate alterations in biological behavior in NANOG-knockdown SW620 ***,mock-knockdown and NANOG-knockdown cells were treated with 5-fluorouracil(5-FU) and survival rate was measured by MTT assay to evaluate drug ***:A significant difference in the transcript levels of iPS-related genes between tumor spheres and their parental bulky cells was *** knockdown suppressed proliferation,colony formation,and in vivo tumorigenicity but increased the sensitivity to 5-FU of SW620 cells.5-FU treatment greatly inhibited the expression of the major stemness-associated genes NANOG,OCT4,and ***:These results collectively suggest an overlap between iPS-related genes and CSCs in *** a certain gene NANOG may truncate the aggressiveness of CRC cells.
