ESTABLISHMENT AND CHARACTERIZATION OF A CELL LINE HR-8348 DERIVED FROM HUMAN POORLY DIFFERENTIATED ADENOCARCINOMA OF RECTUM

A human rectal poorly-differentiated adenocarcinoma cell line HR-8348 is established from thesurgically removed tumor mass of a male adult patient. The cell doubling time is 36.05 h. The mitoticindex is 44.5‰. The cells are all of epithelial type and mostly of polygonal shape. Electron microscopicexamination shows malignant nuclei with enlarged nucleoli; there are abundant microvilli and some se-cretory granules. The plating efficiency in soft agar is 12.9%. In PHA (1μg/ml), the cells show obviousagglutination. Chromosomal analysis reveals a mode of 70 chromosomes per cell, with a marker chromo-some. CEA in cells is detected positively both qualitatively and quantitatively. Hetero-transplantation ofthe cells to nude mice shows that the tumor mass has the same morphology as the original mass fromthe patient, with Alcian blue positive material in the cytoplasm. Repeated detection for mycoplasma con-tamination shows negative.

Determination of gemcitabine and its metabolite in human plasma using high-pressure liquid chromatography coupled with a diode array detector

AIM: To establish a high pressure liquid chromatography (HPLC) method for determination of the concentration of gemcitabine (dFdC) and its metabolite(dFdU) in human plasma. METHODS: Plasma 1.0 mL spikedwith floxuridine as an internal standard was extracted with 3.0 mL of methanol-acetonitrile (v/v, 1:9). The supernatant was evapo- rated at 60 oC and the residue was reconstituted with 0.5 mL of the solution used as the mobile phase. After centrifugation, 50 μL of the supernatant was injected into the HPLC system. Separation was achieved on a C18 (4.6 mm×250 mm, 5 μm) column at 25 oC with the flow rate of the mobile phase set to 0.8 mL/min. The compounds were detected at 268 nm. The mobile phase consisted of 40.0 mmol/L acetate ammonium buffer solution (pH 5.5) and acetonitrile (v/v, 97.5:2.5). RESULTS: The linear range was 0.20-10.0 mg/L(r=0.9999) for dFdC and 0.50-50.0 mg/L(r=0.9999) for dFdU. The limit of detection (LOD) was 0.10 mg/L for dFdC and 0.25 mg/L for dFdU, while the limit of quantification (LOQ) was 0.20 mg/L (RSD<10 %) for dFdC and 0.50 mg/L(RSD <3 %) for dFdU. The average recovery of dFdC and dFdU by this method were 103.3 % and 98.7 %, respectively. For intra-day and inter-day, the corresponding standard deviations of the measurements of dFdC and dFdU were both less than 5.5 %. CONCLUSION: An analytical method was established to measure the concentrations of dFdC and dFdU in human plasma and was effectively applied to the dFdC and dFdU pharmacokinetic studies of 8 Chinese patients with malignant tumors.

＜正＞Background:Brain metastases （BM） represent one of the most frequent complications related to non-small lung cancer （NSCLC）,and their treatment continues to *** have evaluated the activity,toxicity and the impact on Quality of Life （QoL） of a concomitant treatment with whole brain radiotherapy （WBRT） and Gefitinib in patients with brain metastases from NSCLC in a prospective ***:21 patients were enrolled and received 40 Gy/20f/4w WBRT with concomitant Gefitinib （250 mg/day）.The primary end points were clinical symptoms and objective ***:4 patients had a complete response,13 patients had a partial response,3 patients had stable disease,while 1 patient had progression *** overall response rate was 81.0%.The most frequent toxicities included rash （23.8%） and diarrhea （28.6%）,and only 3 patients developed a grade 3 *** and the recursive partitioning analysis （RPA） were found to be predictive factors for response in *** were statistically significant changes between pretreatment and post-treatment in all domains but additional concerns. Conclusion:We concluded that this treatment was well tolerated,with an encouraging objective response rate and a significant improvement in quality of *** study found a high level of satisfaction for QoL,this provides useful information to share with patients in discussions regarding molecular targeting treatment of these lesions.

Synovial sarcoma usually occurs in the extremities and is rare in the head and neck. In this study, we analyzed a group of 5 synovial sarcomas arising in the Parapharyngeal space. The series comprised 5 men （100%） and no women （0%）. In common with synovial sarcoma at more conventional sites, this is a disease of youngadults:ages ranged from 22-41 years （median, 35 years）. Treatment, all 5 patients received radical excision and postoperative radiotherapy, 2 patients with pulmonary metastasis received additional *** chemotherapy included both doxorubicin and ifosfamide. Histologically, 2 examples were of monophasic type, 3 were biphasic. The tumor size ranged from 2.0 to 6.6 cm. Follow-up on 5 patients revealed that [2 were alive without disease 3.8 and 4 years respectively. 2 patients died of disease 2.2 and 3.1 years after the diagnosis respectively. 1 patient was alive with local recurrence and pulmonary metastasis]. The prognosis of synovial sarcoma in parapharyngeal space seems relatively good for those patients whose tumor size is less than 5 cm.

Purpose:To investigate the express ion of c-kit in human salivary gland adenoid cystic carcinoma and their relationships with clinicopathological parameters and prognostic value and then to determine mutational status of exons 9, 11, 13 and 17 o f c-k it gene which provide theoretical foundation for predicting prognosis and targeted therapy. Methods:The expression of c-kit protein was detected measured in 54 human salivary gland adenoid cystic carcinoma by immunohistochemistry and its relationship to prognosis was analyzed statistically. The mutation of c-kit in exons 9, 11, 13 and 17 was determined by SSCP. Results:The expression rate of c-kit protein in human salivary gland adenoid cystic carcinoma was 85.2% （ 46/54）. The human salivary gland adenoid cystic carcinoma with higher expression rate of c-kit was prone to recur locally. The survival time after operation of the patients with c-kit higher expression were significantly lower than that of the patients with c-kit lower expression. The mutation in exon 9, 11, 13 and exon 17 were founded in 19, 8, 7, 9 of 54 cases with human salivary gland adenoid cystic carcinoma and DNA sequencing also detect the mutation. Conclusions:The expression of c-kit could serve as a prognostic factor in human salivary gland adenoid cystic carcinoma. The mutation in exon 9, 11, 13 and exon 17 was found in human salivary gland adenoid cystic carcinoma. Therefore, these exons may function as targets for treatment modalities based on the suppression of c-kit tyrosine kinase activity.

Objective:To observe the synergistic effects of hyperthrnia and cisplatin on human lung adenocarcinoma cell line ***:Cells treated with or without hyperthermia at 42℃ for 2 hours were detected cisplatin sensitivity with CCK-8 *** managed with hyperthermia at 42 ℃ for 2 hours,ciplatin 10ug/ml for 24 hours or both of two sides,inhibition ratio was detected with CCK-8 assay and flow cytometry analyzed DNA contents in every phase of cell ***: After hyperthermia,IC5o values of cell to cisplatin decreased from 9.51ug/ml to 6.07ug/***-8 assay showed hyperthermia and ciplatin had synergistic killing effects,and FCM also showed hyperthermia had significant effects on cell cycle with increase of G/G phase population and reduction of S phase *** can significantly increase the sensitivity of H 1299 cell line to cisplatin.
