Nef Mutations in Long-term Non-progressors from Former Plasma Donors Infected with HIV-1 Subtype B in China

Objective To study the specific amino acid variation in Nef that may be related to disease progression after infection with HIV-1 subtype B, a predominant strain circulating in China, and to determine whether changes in Nef secondary structure may influence different stages of AIDS development based on the concept that the Nef gene of HIV infection dramatically alter the severity of viral infection and virus replication and disease progression, and that long-term non-progressors （LTNP） of HIV infection are commonly associated with either a deletion of the Nef gene or the defective Nef alleles. Methods The study subjects were divided into LTNPI（n=14）, LTNP2 （n=16） and slow progressor （SP, n=19） groups for mutational analysis of the Nef sequence. The data were obtained by using Bioedit, MEGA, Anthewin and SAS software. Results Residues in Nef TA48/49 and K151 occurred more frequently in the LTNP group while AA48/49 was more frequently observed in the SP group. Of the differences observed in the secondary structure comparison using Nef consensus sequences of these three groups, one was roughly corresponding to the Nef48/49 mutation site. Conclusion TA48/49, Kiss, and AA48/49 in the Nef gene might be associated with the different stages of HIV infection, and there may be a link between the Nef secondary structure and the progression of HIV- 1 infection.

Although antiretroviral therapy(ART)is effective at suppressing the human immunodeficiency virus type 1(HIV-1)replication,HIV-1 infection is still a global public health ***-1 accessory protein viral protein R(Vpr)is a multifunctional protein with a primary role in regulating cellular apoptosis.[1]The amino acid(aa)positions 52-96 in the C-terminus of Vpr were identified as the apoptosis-regulating domain.[2,3]In this study,we found a natural Vpr variant truncated at the 54 aa position(*54Vpr)from HIV patients in the acquired immune deficiency syndrome(AIDS)phase that mediated both pro-and antiapoptotic cellular effects by interacting with distinct adenine nucleotide translocator(ANT)isoforms.A novel apoptosis-regulating domain was further identified in the 23-37 aa position in the N-terminus of Vpr.

BACKGROUND Simulated microgravity environment can lead to gastrointestinal motility *** pathogenesis of gastrointestinal motility disorders is closely related to the stem cell factor(SCF)/c-kit signaling pathway associated with intestinal flora and Cajal stromal ***,intestinal flora can also affect the regulation of SCF/c-kit signaling pathway,thus affecting the expression of Cajal stromal *** cells are the pacemakers of gastrointestinal *** To investigate the effects of Bifidobacterium lactis(***)BLa80 on the intestinal flora of rats in simulated microgravity and on the gastrointestinal motility-related SCF/c-kit *** The internationally recognized tail suspension animal model was used to simulate the microgravity environment,and 30 rats were randomly divided into control group,tail suspension group and drug administration tail suspension group with 10 rats in each group for a total of 28 *** tail group was given *** BLa80 by intragastric administration,and the other two groups were given water intragastric administration,the concentration of intragastric administration was 0.1 g/mL,and each rat was 1 mL/***&eosin staining was used to observe the histopathological changes in each segment of the intestine of each group,and the expression levels of SCF,c-kit,extracellular signal-regulated kinase(ERK)and p-ERK in the gastric antrum of each group were detected by Western blotting and *** fecal flora and mucosal flora of rats in each group were detected by 16S *** Simulated microgravity resulted in severe exfoliation of villi of duodenum,jejunum and ileum in rats,marked damage,increased space between villi,loose arrangement,shortened columnar epithelium of colon,less folds,narrower mucosal thickness,reduced goblet cell number and crypts,and significant improvement after probiotic *** microgravity reduced the expressions of SCF and c-kit,and increased the express

Parkinson’s disease is a common neurodegenerative disease with movement disorders associated with the intracytoplasmic deposition of aggregate proteins such asα-synuclein in *** one of the major intracellular degradation pathways,the autophagy-lysosome pathway plays an important role in eliminating these *** evidence has shown that upregulation of the autophagy-lysosome pathway may contribute to the clearance ofα-synuclein aggregates and protect against degeneration of dopaminergic neurons in Parkinson’s ***,multiple genes associated with the pathogenesis of Parkinson’s disease are intimately linked to alterations in the autophagy-lysosome ***,this pathway appears to be a promising therapeutic target for treatment of Parkinson’s *** this review,we briefly introduce the machinery of ***,we provide a description of the effects of Parkinson’s disease–related genes on the autophagy-lysosome ***,we highlight the potential chemical and genetic therapeutic strategies targeting the autophagy–lysosome pathway and their applications in Parkinson’s disease.

Three-channel CMOS transimpedance amplifier for LiDAR sensor receiver

For time-of-flight(TOF)light detection and ranging(LiDAR),a three-channel high-performance transimpedance amplifier(TIA)with high immunity to input load capacitance is presented.A regulated cascade(RGC)as the input stage is at the core of the complementary metal oxide semiconductor(CMOS)circuit chip,giving it more immunity to input photodiode detectors.A simple smart output interface acting as a feedback structure,which is rarely found in other designs,reduces the chip size and power consumption *** circuit is designed using a 0.5μm CMOS process technology to achieve low *** device delivers a 33.87 dB?transimpedance gain at 350 *** a higher input load capacitance,it shows a-3 dB bandwidth of 461 MHz,indicating a better detector tolerance at the front end of the *** a 3.3 V supply voltage,the device consumes 5.2 mW,and the total chip area with three channels is 402.8×597.0μm2(including the test pads).

最近一种高质量的二维(Two-dimensional,2D)半导体材料MoSi_(2)N_(4)(MSN)在实验上被成功合成,具有优异的电气和机械性能.尽管最近有大量的研究致力于揭示MSN的材料特性,但到目前为止,对MSN的电接触物理特性的探索还比较少.在这项工作中,构建了金属-半导体NbSe_(2)/MSN肖特基结并使用第一性原理密度泛函理论计算研究了该肖特基结的材料特性.发现NbSe_(2)/MSN接触具有超低肖特基势垒高度(Schottky barrier height,SBH),这有利于纳米电子学应用.SBH可以通过施加双轴应变的方式进行有效的调控.当施加拉伸应变时能实现NbSe_(2)/MSN肖特基结由p型肖特基接触转变为p型欧姆接触,而当施加较大的压缩应变时能实现p型肖特基接触和n型肖特基接触之间的转换.我们的研究结果为MSN的2D电触点的物理特性提供了见解,并将为设计基于MSN的2D纳米器件的高性能电触点提供关键的一步.
