Data driven analysis reveals prognostic genes and immunological targets in human sepsis-associated acute kidney injury

BACKGROUND:The molecular mechanism of sepsis-associated acute kidney injury(SA-AKI)is unclear.We analyzed co-differentially expressed genes(co-DEGs)to elucidate the underlying mechanism and intervention targets of SA-AKI.METHODS:The microarray datasets GSE65682,GSE30718,and GSE174220 were downloaded from the Gene Expression Omnibus(GEO)database.We identified the co-DEGs and constructed a gene co-expression network to screen the hub genes.We analyzed immune correlations and disease correlations and performed functional annotation of the hub genes.We also performed single-cell and microenvironment analyses and investigated the enrichment pathways and the main transcription factors.Finally,we conducted a correlation analysis to evaluate the role of the hub genes.RESULTS:Interleukin 32(IL32)was identified as the hub gene in SA-AKI,and the main enriched signaling pathways were associated with hemopoiesis,cellular response to cytokine stimulus,inflammatory response,and regulation of kidney development.Additionally,IL32 was significantly associated with mortality in SA-AKI patients.Monocytes,macrophages,T cells,and NK cells were closely related to IL32 and were involved in the immune microenvironment in SA-AKI patients.IL32 expression increased significantly in the kidney of septic mouse.Toll-like receptor 2(TLR2)was significantly and negatively correlated with IL32.CONCLUSION:IL32 is the key gene involved in SA-AKI and is significantly associated with prognosis.TLR2 and relevant immune cells are closely related to key genes.

Detection of endogenous foreign bodies in Chinese hickory nuts by hyperspectral spectral imaging at the pixel level

It is difficult to differentiate small,but harmful,shell fragments of Chinese hickory nuts from their kernels since they are very similar in color.Including shell fragments of Chinese hickory nuts by mistake may create safety hazards for consumers.Therefore,there is a need to develop an effective method to differentiate the shells from the kernels of Chinese hickory nuts.In this study,a deep learning approach based on a two-dimensional convolutional neural network(2D CNN)and long short-term memory(LSTM)integrated with hyperspectral imaging for distinguishing the shells and kernels of Chinese hickory nuts at the pixel level was proposed.Two classical classification methods,principal component analysis-K-nearest neighbors(PCA-KNN)and the support vector machine(SVM),were employed to establish identification models for comparison.The results showed that the 2D CNN-LSTM model achieved the best performance with an overall classification accuracy of 99.0%.Moreover,the shells in mixtures of shells and kernels were detected based on the proposed deep learning method and visualized for subsequent operations for the removal of foreign bodies.

Proteomic profiling of cerebrospinal fluid in pediatric myelin oligodendrocyte glycoprotein antibody-associated disease

Background Myelin oligodendrocyte glycoprotein(MOG)antibody-associated disease(MOGAD)is an autoimmune demy-elinating disorder of the central nervous system.Methods Extracted proteins from 34 cerebrospinal fluid(CSF)samples[patients with MOGAD(MOG group,n=12);healthy controls(HC group,n=12);patients with MOG seronegative and metagenomics next-generation sequencing-negative inflammatory neurological diseases(IND group,n=10)]were processed and subjected to label-free quantitative proteomics.Supervised partial least squares-discriminant analysis(PLS-DA)and orthogonal PLS-DA(O-PLS-DA)models were also performed based on proteomics data.Functional analysis of differentially expressed proteins(DEPs)was performed using Gene Ontology,InterPro,and Kyoto Encyclopedia Genes and Genomes.An enzyme-linked immunosorbent assay was used to determine the complement levels in serum from patients with MOGAD.Results Four hundred and twenty-nine DEPs(149 upregulated and 280 downregulated proteins)were identified in the MOG group compared to the HC group according to the P value and fold change(FC).Using the O-PLS-DA model,872 differentially abundant proteins were identified with variable importance projection(VIP)scores>1.Five proteins(gamma-glutamyl hydrolase,cathepsin F,interalpha-trypsin inhibitor heavy chain 5,latent transforming growth factor beta-binding protein 4 and leukocyte-associated immunoglobulin-like receptor 1)overlapping between the top 30 DEPs with top-ranked P value and FC and top 30 proteins in PLS-DA VIP lists were acquired.Functional analysis revealed that the dysregulated proteins in the MOG group were primarily involved in complement and coagulation cascades,cell adhesion,axon guidance,and glycosphingolipid biosynthesis compared to the HC group.Conclusion The proteomic alterations in CSF samples from children with MOGAD identified in the current study might provide opportunities for developing novel biomarker candidates.

Flexible MXene/sodium alginate composite fabric with high structural stability and oxidation resistance for electromagnetic interference shielding

Wearable electromagnetic interference(EMI)shielding fabrics with excellent electromagnetic shielding performance,oxidation resistance,and structural stability are highly demanded for the rapid development of electronic devices and wireless communication.MXenes are metallic conductive materials with exceptional EMI shielding properties,but they are prone to oxidation in air and have poor structural stability and durability on fabric substrates.Herein,we present a one-step assembly method to fabricate fabrics coated with MXenes and polymeric sodium alginate(SA)composite(MXene-SA).SA protects MXenes from oxidation and forms a stable interlayer structure by bonding to MXenes.The MXene-SA coated fabrics are breathable and flexible,and have a low sheet resistance of 2.12±0.08Ω/sq and a high EMI shielding performance of 37.05 dB at X-band,which is comparable to the best 42.31 dB.Moreover,the MXene-SA coated fabrics exhibit high structural stability and oxidation resistance under various conditions of sonication disintegration,mechanical abuse,chemical corrosion,and humidity,compared to pure MXenes coated fabrics.We believe that the wearable and high-performance MXene-SA fabrics have great potential for the next generation of ultra-portable and wearable EMI shielding products.

Detection of apple firmness with a novel loudspeaker-based excitation device

Firmness is one of the important indices to evaluate the internal quality of fruit.In this study,a noncontact loudspeaker-based detection system was developed to evaluate apple firmness.The structural parameters of the excitation device were modified in the single-factor experiments,and the best combination of structural parameters was that the inner diameter of the gasket was 40 mm;the distance between fruit surface and loudspeaker was 95 mm.Besides,the proper posture style was that the apple was placed with its stem upward.After the modification of the Laser Doppler Vibrometer(LDV)method,the vibration response signals of 48 apples were measured to establish the firmness prediction model.The results showed that the better prediction performance of stiffness was obtained in multiple models.The Back Propagation Neural Network(BPNN)model had the best prediction performance by using parameters of elasticity index(EI),the peak value at the second resonance frequency f_(2)(A_(2)),and peak area S,with a correlation coefficient of prediction(r_(p))of 0.914;root mean square error of prediction(RMSEP)of 0.491 N/mm.Therefore,the proposed detection system is feasible to nondestructively detect apple firmness,which has the potential to be applied in online detection.

Aggregation-induced emission(AIE)is a photophysical phenomenon that a certain group of luminescent materials that become highly luminous when aggregated in a bad solvent or solid state.This year is the 20th anniversary since the AIE concept firstly proposed in 2001.Many advanced applications were gradually being explored,covering optics,electronics,energy,and bioscience and so on.At present,bibliometrics can enlighten the researchers with comprehensive sights of the achievements and trends of a specific field,which is critical for academic investigations.Herein,we presented a general bibliometric overview of AIE covering 20 years of evolution.With the assistance of Web of Science Core Collection database and several bibliometric software tools,the annual publication and citation,most influential countries/regions,most contributing authors,journals and institutions,second near-infrared(NIR-Ⅱ)related hotspots,as well as the forecast of frontiers were demonstrated and systematically analyzed.This study summarizes the current research status in AIE research field and provides a reference for future research directions.

Background:Although laparoscopic technology has achieved rapid development in the surgical field,it has not been applied to liver transplantation,primarily because of difficulties associated with laparoscopic vascular anastomosis.In this study,we introduced a new magnetic-assisted vascular anastomosis technique and explored its application in laparoscopic liver transplantation in pigs.Methods:Two sets of magnetic vascular anastomosis rings(MVARs)with different diameters were developed.One set was used for anastomosis of the suprahepatic vena cava(SHVC)and the other set was used for anastomosis of the infrahepatic vena cava(IHVC)and portal vein(PV).Six laparoscopic orthotopic liver transplantations were performed in pigs.Donor liver was obtained via open surgery.Hepatectomy was performed in the recipients through laparoscopic surgery.Anastomosis of the SHVC was performed using hand-assisted magnetic anastomosis,and the anastomosis of the IHVC and PV was performed by magnetic anastomosis with or without hand assistance.Results:Liver transplants were successfully performed in five of the six cases.Postoperative ultrasonographic examination showed that the portal inflow was smooth.However,PV bending and blood flow obstruction occurred in one case because the MVARs were attached to each other.The durations of loading of MVAR in the laparoscope group and manual assistance group for IHVC and PV were 13±5 vs.5±1 min(P<0.01)and 10±2 vs.4±1 min(P<0.05),respectively.The durations of MVAR anastomosis in the laparoscope group and manual assistance group for IHVC and PV were 5±1 vs.1±1 min(P<0.01),and 5±1 vs.1±1 min(P<0.01),respectively.The anhepatic phase was 43±4 min in the laparoscope group and 23±2 min in the manual assistance group(P<0.01).Conclusions:Our study showed that magnetic-assisted laparoscopic liver transplantation can be successfully carried out in pigs.

In situ preparation of g-C3N4/Bi4O5I2 complex and its elevated photoactivity in Methyl Orange degradation under visible light

A graphite carbon nitride(g-C3N4)modified Bi4O5I2 composite was successfully prepared insitu via the thermal treatment of a g-C3N4/Bi OI precursor at 400°C for 3 hr.The as-prepared g-C3N4/Bi4O5I2 showed high photocatalytic performance in Methyl Orange(MO)degradation under visible light.The best sample presented a degradation rate of 0.164 min^-1,which is 3.2 and 82 times as high as that of Bi4O5I2 and g-C3N4,respectively.The g-C3N4/Bi4O5I2 was characterized by X-ray powder diffractometer(XRD),scanning electron microscopy(SEM),transmission electron microscopy(TEM),Raman,X-ray photoelectron spectroscopy(XPS),ultraviolet-visible diffuse reflectance spectra(DRS),electrochemical impedance spectroscopy(EIS)and transient photocurrent response in order to explain the enhanced photoactivity.Results indicated that the decoration with a small amount of g-C3N4 influenced the specific surface area only slightly.Nevertheless,the capability for absorbing visible light was improved measurably,which was beneficial to the MO degradation.On top of that,a strong interaction between g-C3N4 and Bi4O5I2 was detected.This interplay promoted the formation of a favorable heterojunction structure and thereby enhanced the charge separation.Thus,the g-C3N4/Bi4O5I2 composite presented greater charge separation efficiency and much better photocatalytic performance than Bi4O5I2.Additionally,g-C3N4/Bi4O5I2 also presented high stability.·O2^- and holes were verified to be the main reactive species.

Comparison Study on the Semiconductive and Dissolution Behaviour of 316L and Alloy 625 in Hydrochloric Acid Solution

The corrosion behaviour of 316L and Alloy 625 was investigated using cyclic polarization,electrochemical impedance spectroscopy,X-ray photoelectron spectroscopy,Auger electron spectroscopy and induced coupled plasma-optical emis sion spectrometer.The results indicated that Alloy 625 showed better corrosion resistance than 316L and the prolonging immersion time could enhance corrosion resistance of the two alloys.The passive film formed on the surface of 316L exhibited an electronic structure of p-p heterojunction,with Fe3O4 and Cr2O3 enriched in the outer and inner layers,respectively.However,Alloy 625 presented the electronic structure of n-p heterojunction dominated by the outer Fe2O3/NiFe2O4 and inner Cr2O3.This resulted in the opposite semiconductive properties of the passive films formed on the two materials.In the acid solutions,Fe and Mo suffered from selective dissolution while Cr and Ni were relatively stable.The corrosion rates were mainly dominated by the dissolution of iron.Alloy 625 presented better corrosion resistance than 316L due to the obviously lower content of Fe and the higher content of Cr and Ni in the passive film.The continuously selective dissolution of iron resulted in the increase in Cr/Fe ratio in the passive film,which was responsible for the enhancement in corrosion resistance.
