Mitochondrial transplantation confers protection against the effects of ischemic stroke by repressing microglial pyroptosis and promoting neurogenesis

Transferring healthy and functional mitochondria to the lateral ventricles confers neuroprotection in a rat model of ischemia-reperfusion *** mitochondrial transplantation is also beneficial in pediatric patients with cardiac ischemia-reperfusion ***,transplantation of functional exogenous mitochondria may be a promising therapeutic approach for ischemic *** explore the neuroprotective effect of mitochondria transplantation and determine the underlying mechanism in ischemic stroke,in this study we established a photo-thrombosis-induced mouse model of focal ischemia and administered freshly isolated mitochondria via the tail vein or to the injury site(in situ).Animal behavior tests,immunofluorescence staining,2,3,5-triphenyltetrazolium chloride(TTC)staining,mRNA-seq,and western blotting were used to assess mouse anxiety and memory,cortical infarct area,pyroptosis,and neurogenesis,*** bioinformatics analysis,western blotting,co-immunoprecipitation,and mass spectroscopy,we identified S100 calcium binding protein A9(S100A9)as a potential regulator of mitochondrial function and determined its possible interacting *** between exogenous and endogenous mitochondria,as well as the effect of exogenous mitochondria on recipient microglia,were assessed in *** data showed that:(1)mitochondrial transplantation markedly reduced mortality and improved emotional and cognitive function,as well as reducing infarct area,inhibiting pyroptosis,and promoting cortical neurogenesis;(2)microglial expression of S100A9 was markedly increased by ischemic injury and regulated mitochondrial function;(3)in vitro,exogenous mitochondria enhanced mitochondrial function,reduced redox stress,and regulated microglial polarization and pyroptosis by fusing with endogenous mitochondria;and(4)S100A9 promoted internalization of exogenous mitochondria by the microglia,thereby amplifying their pro-proliferation and anti-inflammatory ***

目的探索大鼠心肺复苏(CPR)脑损伤后海马区小胶质细胞的激活和焦亡情况。方法建立CPR脑损伤模型,将30只雄性SD大鼠随机分为假手术组(Sham组)和CPR脑损伤组(CPR组),每组15只。采用旷场试验和Morris水迷宫实验分析大鼠的行为变化,确定CPR后脑损伤对负性情绪和认知功能的影响;通过免疫荧光染色技术、流式细胞术和Western blotting观察海马区小胶质细胞的激活、焦亡及相关蛋白的表达情况。结果与Sham组相比,在旷场和Morris水迷宫实验中,CPR组大鼠出现焦虑、抑郁情绪和认知功能障碍(P<0.05);Western blotting结果显示,CPR组大鼠海马组织中的焦亡相关蛋白表达增多(P<0.05);免疫荧光和流式细胞术结果显示,CPR组大鼠海马区小胶质细胞被大量激活,焦亡效应蛋白GSDMD-N表达上调且与激活的小胶质细胞实现了共定位。结论 CPR脑损伤后,海马区GSDMD-N将介导小胶质细胞的焦亡。

A Rapid Estimation Method for Post-earthquake Building Losses

Rapid estimation of post-earthquake building damage and loss is very important in urgent response *** current approach leaves much room for improvement in estimating ground motion and correctly incorporating the uncertainty and spatial correlation of the *** study proposed a new approach for rapidly estimating post-earthquake building loss with reasonable *** proposed method interpolates ground motion based on the observed ground motion using the Ground Motion Prediction Equation(GMPE)as the *** samples the building seismic loss quantile considering the spatial loss correlation that is expressed by Gaussian copula,and kriging is applied to reduce the dimension of direct sampling for estimation *** proposed approach was validated using three historical earthquake events in Japan with actual loss reports,and was then applied to predict the building loss amount for the March 2022 Fukushima Mw7.3 *** proposed method has high potential in future emergency efforts such as search,rescue,and evacuation planning.

Efficacy of luteolin on the human gastric cancer cell line MKN45 and underlying mechanism

OBJECTIVE:To investigate the antitumour efficacy of luteolin on gastric cancer(GC)and study the mechanism underpinning the ***:Effects of luteolin on cell growth inhibition,apoptosis,and cell cycle arrest in MKN45 cells were investigated using the cell counting kit-8 *** in the mitochondrial membrane potential after luteolin treatment were assessed using 5,5′,6,6’-tetra-chloro-1,1’,3,3’-tetraethy lbenzimidazolcar bocyanineiodi-de(JC-1)*** investigate whether apoptotic effect by luteolin is related to the phosphoinositide 3-kinase/v-akt murine thymoma viral oncogene(PI3K/Akt)pathway,cells were additionally treated with LY294002,a PI3K/Akt pathway ***,the expressions of apoptosis-related proteins,namely B-cell lymphoma 2(Bcl-2),Bcl-2 associated X protein(Bax),Akt,p-Akt,caspase-3,and cytochrome C,were detected after luteolin ***:The study revealed that in MKN45 cells,luteolin could inhibit the cell proliferation in a time-and dose-dependent manner;block the cell cycle in the Sphase;induce apoptosis;reduce the mitochondrial membrane potential;increase the expression of Bax,caspase-3,and cytochrome C;and decrease the expression of Bcl-2 and *** might be involved in the PI3K/Akt signalling pathway,indicating that this pathway could be a therapeutic target for GC ***:Luteolin could inhibit the proliferation of GC cells and block the cell cycle in the *** mechanism of inducing apoptosis in these cells was related to the PI3K/Akt signalling pathway.

Mesenchymal stromal cells modulate unfolded protein response and preserve β-cell mass in type 1 diabetes

Introduction:Transplantation of mesenchymal stromal cells(MSCs)is a promising therapy for type 1 diabetes(T1D).However,whether the infused MSCs affect the endoplasmic reticulum stress or subsequent unfolded protein response inβcells remains ***:To investigate this,we induced early-onset T1D in non-obese diabetic mice using ***,T1D mice were randomly assigned to receive either MSCs or phosphate-buffered *** observed the in vivo homing of MSCs and assessed their effectiveness by analyzing blood glucose levels,body weight,histopathology,pancreatic protein expression,and serum levels of cytokines,proinsulin,and ***:Infused MSCs were found in the lungs,liver,spleen,and pancreas of T1D *** exhibited various effects,including reducing blood glucose levels,regulating immunity,inhibiting inﬂammation,increasingβ-cell areas,and reducing the expression of key proteins in the unfolded protein response *** serum proinsulin and C-peptide levels were signiﬁcantly higher in the MSCs treatment group than in the T1D model ***,there was no signiﬁcant difference in the biomarker ofβ-cell endoplasmic reticulum stress,the ratio of fasting serum proinsulin to C-peptide,between the two ***:Ourﬁndings reveal that MSCs infusion does not alleviate endoplasmic reticulum stress inβcells directly but modulates the unfolded protein response pathway to preserveβ-cell mass and function in T1D mice.

Elucidating the effects of anthropogenic pollutants on the photooxidation of biogenic volatile organic compounds is crucial to understanding the fundamental mechanisms of secondary organic aerosol(SOA)***,the impacts of NO_(2)and SO_(2)on SOA formation from the photooxidation of a representative monoterpene,β-pinene,were investigated by a number of laboratory *** results indicated NO_(2)enhanced the SOA mass concentrations and particle number concentrations under both low and highβ-pinene *** could be rationalized that the increased O_(3)concentrations upon the NO_(x)photolysis was helpful for the generation of more amounts of O_(3)-oxidized products,which accelerated the SOA nucleation and *** with NO_(2),the promotion of the SOA yield by SO_(2)was mainly reflected in the increase of mass concentration,which might be due to the elimination of the newly formed particles by the initially formed *** observed low oxidation degree of SOA might be attributed to the fast growth of SOA,resulting in the uptake of less oxygenated gas-phase species onto the particle *** present findings have important implications for SOA formation affected by anthropogenic–biogenic interactions in the ambient atmosphere.

A new static mixer Cross-over-Disc has been invented to strip off the boundary layer and to make strong radial *** pressure drop of Cross-over-Disc is 12–26 times as large as that of empty pipe with equivalent diameter and *** mixing performance of Cross-over-Disc with 14 elements has been investigated in the viscosity range of 190–250 Pa·s by decoloration method,and the gray analysis of images shows that mixing inhomogeneity is about 7.5%and 9.4%for the mixing ratio of 5∶1 and 10∶1,***,mixing inhomogeneity for a combination of static mixing elements(four from Cross-over-Disc and three pairs from Sulzer-type)can be decreased to 2.1%–3.1%within a reasonable range of pressure drop.

为实现二硫化碳氯化工艺中产生的二硫化碳和四氯化碳的完全回收,利用Aspen Plus软件对二硫化碳四氯化碳混合物的精馏I艺进行了模拟计算。分析了理论塔板数、进料口位置回流比对分离效果的影响,选出最适宜的工艺条件:理论塔板数为21块,进料口位于第9块塔板,回流比为1.9。在该工艺条件下,塔顶温度为46.9℃,二硫化碳质量分数为95.1%,满足氯化合成全氯甲硫醇的含量要求;塔底温度76.5℃,四氯化碳质量分数为99.8%,符合GBT 4119-2008对四氯化碳含量的要求。

A rapid and sensitive liquid chromatography–tandem mass spectrometric method for the determination of hederasaponin B in rat plasma: Application to a pharmacokinetic study

A rapid, simple and sensitive ultra-high performance liquid chromatography–tandem mass spectrometric(UPLC–MS/MS) method was developed and validated for the determination of hederasaponin B, an active triterpenoid saponin widely existed in Hedera helix L. Plasma samples were processed by protein precipitation with acetonitrile and separated on a Thermo Hypersil GOLD C18(2.1 mm × 50 mm,1.9 μm) at flow rate of 0.3 ml/min, with a gradient elution consisting of acetonitrile and water containing 0.1%(v/v) formic acid at 30 ℃ and detected by electrospray ionization mass spectrometry in the positive multiple reaction monitoring(MRM) mode. The linearity was found to be within the concentration range of 0.5–5000 ng/ml with a lower limit of quantification of 0.5 ng/ml. The absolute oral bioavailability of hederasaponin B was 0.24 ± 0.49%. This indicated that the concentration-time course of the hederasaponin B existed a double-peak phenomenon. This method was further applied to the determination of hederasaponin B in rat plasma and showed good practicability, for the first time, after intragastric(25 mg/kg) and intravenous(2 mg/kg) administration in rats.
