Mass spectrometry analysis of intact protein N-glycosylation signatures of cells and sera in pancreatic adenocarcinomas

Pancreatic cancer is among the most malignant cancers,and thus early intervention is the key to better survival outcomes.However,no methods have been derived that can reliably identify early precursors of development into malignancy.Therefore,it is urgent to discover early molecular changes during pancreatic tumorigenesis.As aberrant glycosylation is closely associated with cancer progression,numerous efforts have been made to mine glycosylation changes as biomarkers for diagnosis;however,detailed glycoproteomic information,especially site-specific N-glycosylation changes in pancreatic cancer with and without drug treatment,needs to be further explored.Herein,we used comprehensive solid-phase chemoenzymatic glycoproteomics to analyze glycans,glycosites,and intact glycopeptides in pancreatic cancer cells and patient sera.The profiling of N-glycans in cancer cells revealed an increase in the secreted glycoproteins from the primary tumor of MIA PaCa-2 cells,whereas human sera,which contain many secreted glycoproteins,had significant changes of glycans at their specific glycosites.These results indicated the potential role for tumor-specific glycosylation as disease biomarkers.We also found that AMG-510,a small molecule inhibitor against Kirsten rat sarcoma viral oncogene homolog(KRAS)G12C mutation,profoundly reduced the glycosylation level in MIA PaCa-2 cells,suggesting that KRAS plays a role in the cellular glycosylation process,and thus glycosylation inhibition contributes to the anti-tumor effect of AMG-510.

From strengthening the construction of an innovation system,adhering to the green development model of the industry,advancing the rural revitalization strategy,strengthening the protection and inheritance of cultural heritage,accelerating the development of social undertakings,and expanding the space for regional cooperation,this paper discusses the strategies for the construction of sustainable development experimental zone in Fumian District,Yulin City.It is intended to build Fumian District into a vibrant,economically prosperous,urban and rural integrated,characteristic cultural,safe and harmonious,inclusive and open district.

近年来,随着实景三维中国和数字中国的快速发展,建筑物三维模型重建已成为智慧城市建设的重要环节。本文提出了一种全自动建筑物LOD2模型生成框架,旨在解决城市级实景三维场景中建筑物LOD2模型生成的难题。为了自动提取大场景建筑物对象,首先利用场景正射影像,通过图像分割获取建筑物的边界信息;然后使用改进的增强型平面区域分割方法提取高质量的分割平面,并利用混合线性模型对分割结果进行优化,能够更好地处理城市建筑物的局部破损和重建错误等问题。试验结果表明,本文算法生成的建筑物LOD2模型质量和效率更高,能够应对城市建筑模型的复杂情况,得到更稳健的重建结果。

This paper first analyzes the main opportunities for sustainable development of Fumian District,Yulin City,and then discusses the challenges faced by Fumian District in its sustainable development.Finally,it proposes safeguard measures for sustainable development of Fumian District:(i)strengthening leadership and implementing responsibilities;(ii)enhancing cooperation and gathering forces;(iii)reinforcing supervision and ensuring implementation;(iv)increasing input and integrating resources;(v)strengthening publicity and creating an atmosphere.

Cardiac resident macrophages: Spatiotemporal distribution, development, physiological functions, and their translational potential on cardiac diseases

Cardiac resident macrophages (CRMs) are the main population of cardiac immune cells. The role of these cells in regeneration, functional remodeling, and repair after cardiac injury is always the focus of research. However, in recent years, their dynamic changes and contributions in physiological states have a significant attention. CRMs have specific phenotypes and functions in different cardiac chambers or locations of the heart and at different stages. They further show specific differentiation and development processes. The present review will summarize the new progress about the spatiotemporal distribution, potential developmental regulation, and their roles in cardiac development and aging as well as the translational potential of CRMs on cardiac diseases. Of course, the research tools for CRMs, their respective advantages and disadvantages, and key issues on CRMs will further be discussed.

Effects of Different Application Methods and Application Rates of Biological Nano-Selenium on the Quality of Passion Fruit

[Objectives]To study the effects of different application methods and application rates of biological nano-selenium on the quality of passion fruit.[Methods]In this experiment,passion fruit Tainong 1 was sprayed with biological nano-selenium.The concentration of spraying refers to the recommended application rate of biological nano-selenium manufacturers.Three treatments with different concentrations were carried out:3,4.5 and 6 L/ha biological nano-selenium was sprayed on 100%root,(50%root+50%leaf surface)and 100%leaf surface of passion fruit,respectively.[Results]From the aspects of economic cost and rational utilization of resources,spraying 3 L/ha selenium fertilizer on the leaves of passion fruit played the best role in improving the quality of passion fruit.[Conclusions]To study the selenium-rich production technology of agricultural products and improve the quality of agricultural products is of great significance for improving human health and producing high-quality agricultural products.

Absorption and Transfer of Exogenous Selenium in Longan(Dimocarpus longan Lour.) and Its Effect

Different concentrations of sodium selenite and sodium selenate were applied to leaves and roots of longan( Dimocarpus longan Lour.) to study the absorption and transfer characteristics of exogenous selenium in longan,and the effects of nitrogen and phosphorus fertilizers on the absorption of selenium in longan were discussed. This study provides a theoretical basis and reference for the research and influence on the absorption and transfer mechanism of selenium in longan and the production of selenium-enriched longan.

Genetic Diversity of Jinsha Pomelo and Its Closely-related Germplasms Assessed by SSR Molecular Markers

SSR molecular markers were used to analyze the genetic diversity of 8 Jinsha pomelo germplasm and its closely-related varieties from main planting area in Jiangxi Province. A total of 92 alleles were detected from 26 pairs of selected primers with 53. 68% of polymorphism. The alleles detected per locus were ranged from 2 to 8. On average,3. 54 alleles were amplified from per pair of primers. The similarity coefficients of the 8 varieties were in the range of 0. 729-0. 924.According to the genetic similarities based on UPGMA method,all materials were divided into two groups of Jinlan pomelo and Shatian pomelo at the similarity coefficient of 0. 738.

Joint Modeling of Citation Networks and User Preferences for Academic Tagging Recommender System

In the tag recommendation task on academic platforms,existing methods disregard users’customized preferences in favor of extracting tags based just on the content of the articles.Besides,it uses co-occurrence techniques and tries to combine nodes’textual content for modelling.They still do not,however,directly simulate many interactions in network learning.In order to address these issues,we present a novel system that more thoroughly integrates user preferences and citation networks into article labelling recommendations.Specifically,we first employ path similarity to quantify the degree of similarity between user labelling preferences and articles in the citation network.Then,the Commuting Matrix for massive node pair paths is used to improve computational performance.Finally,the two commonalities mentioned above are combined with the interaction paper labels based on the additivity of Poisson distribution.In addition,we also consider solving the model’s parameters by applying variational inference.Experimental results demonstrate that our suggested framework agrees and significantly outperforms the state-of-the-art baseline on two real datasets by efficiently merging the three relational data.Based on the Area Under Curve(AUC)and Mean Average Precision(MAP)analysis,the performance of the suggested task is evaluated,and it is demonstrated to have a greater solving efficiency than current techniques.
