Molecular quantification of herbs(Herb-Q):a pyrosequencingbased approach and its application in Pinellia ternata

Variations in herb dosage due to species adulteration and dosing inaccuracies can substantially affect clinical safety and *** species quantification remains challenging,as current methods often yield inconsistent *** study introduces a novel pyrosequencing-based technique,termed herb molecular quantification(Herb-Q),designed to precisely quantify herbal *** evaluated its effectiveness using Pinellia ternata and five of its ***,we assessed commonly used DNA barcodes with sequences from a public database,identifying two candidate regions,Maturase K(matK)and internal transcribed spacer 2(ITS2),for screening specific single nucleotide polymorphism(SNP)loci,allowing for species-specific *** loci were validated by amplifying and sequencing genomic material from collected *** validation studies showed that Herb-Q demonstrated excellent linearity,accuracy,repeatability,and detection *** established quantitative standard curves with high R^(2)values(>0.99)to enable precise species quantification,which were combined with external standards to provide clear and accurate visual quantification *** average bias in quantifying the tuber of *** was 2.38%,confirming that Herb-Q can accurately identify and quantify herbal product ***,the entire quantification process took less than 4 *** study presents a novel,rapid method for accurately quantifying species in herbal products and advances the application of DNA barcoding from species identification to quantitative detection.

Photocatalytic CO_(2) reduction driven by solar light is a green approach that can decrease the greenhouse effect induced by high CO_(2) concentration in the atmosphere and generate carbon-based chemicals/fuels as *** this paper,non-metal co-catalysts ZnO/ZnS type-II hetero-junction nanoparticles with a rough surface were prepared through a hydrothermal *** used as a photocatalyst for CO_(2) reduction,the optimal one showed good cycle stability and a higher yield rate of 27.8µmol·g^(−1)·h^(−1) for CO_(2) conversion into *** outstanding catalytic activity originated from i)the rich interfaces between ZnO and ZnS in the nanoscale could significantly reduce the delivery path of carriers and improve the utilization efficiency of photo-excited electron/hole pairs and ii)enriched surface oxygen defects could supply much more reaction active sites for CO_(2) adsorption.

Apatinib and gamabufotalin co-loaded lipid/Prussian blue nanoparticles for synergistic therapy to gastric cancer with metastasis

Histamine in food has attracted widespread attention due to the potential toxicity and associated health ***,its influences on immunological components,especially the function of key immune cells,are still poorly *** this work,we explored the effects of exogenous histamine on the function of key immune cells such as intestinal epithelial cells,dendritic cells,and T *** results showed that histamine could affect the expression of allergy-related genes in CMT93 cells at a high dose of ***,it’s found that histamine could cause an imbalance in the levels of relevant immune factors secreted by dendritic cells and T cells,especially those related to *** the same time,the proportion of MHC class IIpositive dendritic cells and the proportion of T helper 2(Th2)cells in CD4^(+)T cells increased after histamine *** concluded that the presence of a certain level of histamine in food may affect the expression of allergy-related cytokines,disrupt the balance of the immune homeostasis,and potentially lead to adverse immune *** work demonstrated the importance of including the estimation of histamine’s immune safety in aquatic products rather than merely considering the potential risk of food poisoning.

The Wangjiazhuang Cu(-Mo)deposit,located within the Zouping volcanic basin in western Shandong Province,China,is unique in this area for having an economic *** order to expound the metallogenetic characteristics of this porphyry-like hydrothermal deposit,a detailed fluid inclusion study has been conducted,employing the techniques of representative sampling,fluid inclusion petrography,microthermometry,Raman spectroscopy,LA-ICP-MS analysis of single fluid inclusions,as well as cathode fluorescence spectrometer analysis of host mineral *** deposit contains mainly two types of orebodies,***-dissemination-stockwork orebodies in the K-Si alteration zone and pegmatiticquartz sulfide veins above *** addition,minor breccia ore occurs *** types of fluid inclusions in the deposit and altered quartz monzonite are identified:L-type one-or two-phase aqueous inclusions,V-type vapor-rich inclusions with V/L ratios greater than 50%-90%,D-type multiphase fluid inclusions containing daughter minerals or solids and S-type silicate-bearing fluid inclusions containing mainly muscovite and *** petrography and fluid inclusion study has revealed a three-stage mineralization process,driven by magmatic-hydrothermal fluid activity,as ***,a hydrothermal fluid,separated from the parent magma,infiltrated into the quartz monzonite,resulting in its extensive K-Si alteration,as indicated by silicate-bearing fluid inclusions trapped in altered quartz *** is followed by the early mineralization,the formation of quartz veinlets and dissemination-stockwork *** the main mineralization stage,due to the participation and mixing of meteoric groundwater with magmatic-sourced hydrothermal fluid,the cooling and phase separation caused deposition of metals from the hydrothermal *** a result,the pegmatitic-quartz sulfide-vein ores *** the late mineralization stage,decreasing fluid salinity led to the formation of L-type aqueous

The Fagaceae,a plant family with a wide distribution and diverse adaptability,has garnered significant interest as a subject of study in plant speciation and ***,certain Fagaceae species are regarded as highly valuable wood resources due to the exceptional quality of their *** this study,we present two high-quality,chromosome-scale genome sequences for Quercus sichourensis(848.75 Mb)and Quercus rex(883.46 Mb).Comparative genomics analysis reveals that the difference in the number of plant disease resistance genes and the nonsynonymous and synonymous substitution ratio(Ka/Ks)of protein-coding genes among Fagaceae species are related to different environmental ***,most genes related to starch synthesis in the investigated Quercoideae species are located on a single chromosome,as compared to the outgroup species,Fagus ***,resequencing and population analysis of *** and *** reveal that *** has lower genetic diversity and higher deleterious mutations compared to *** high-quality,chromosome-level genomes and the population genomic analysis of the critically endangered *** and *** will provide an invaluable resource as well as insights for future study in these two species,even the genus Quercus,to facilitate their conservation.

Single-cell RNA sequencing analysis of the retina under acute high intraocular pressure

High intraocular pressure causes retinal ganglion cell injury in primary and secondary glaucoma diseases,yet the molecular landscape characteristics of retinal cells under high intraocular pressure remain *** models of acute hypertension ocular pressure were established by injection of cross-linked hyaluronic acid hydrogel(Healaflow■).Single-cell RNA sequencing was then used to describe the cellular composition and molecular profile of the retina following high intraocular *** results identified a total of 12 cell types,namely retinal pigment epithelial cells,rod-photoreceptor cells,bipolar cells,Müller cells,microglia,cone-photoreceptor cells,retinal ganglion cells,endothelial cells,retinal progenitor cells,oligodendrocytes,pericytes,and *** single-cell RNA sequencing analysis of the retina under acute high intraocular pressure revealed obvious changes in the proportions of various retinal cells,with ganglion cells decreased by 23%.Hematoxylin and eosin staining and TUNEL staining confirmed the damage to retinal ganglion cells under high intraocular *** extracted data from retinal ganglion cells and analyzed the retinal ganglion cell cluster with the most distinct *** found upregulation of the B3gat2 gene,which is associated with neuronal migration and adhesion,and downregulation of the Tsc22d gene,which participates in inhibition of *** study is the first to reveal molecular changes and intercellular interactions in the retina under high intraocular *** data contribute to understanding of the molecular mechanism of retinal injury induced by high intraocular pressure and will benefit the development of novel therapies.

Mineral potential mapping using RF， Fuzzy AHP and their ensemble model: A case study of the Daping Gold Deposits， Yunnan Province， China

Analyzing and integrating evidential layers is essential for mineral prospectivity mapping. Remote sensing data from Landsat 8 and PALSAR-2/ALOS-2 were used to extract information about hydrothermal alterations and lineaments in a vegetation-covered study area. After integrating geological data， five evidential layers were identified， associated with gold mineralization， including lithology， hydrothermal alterations， lineaments， quartz veins， and geochemical anomalies. Based on these layers， prospective maps for mineralization were generated using RF and Fuzzy AHP models， as well as their ensemble. The P-A plot was used to evaluate the capability of model prediction. The results indicated that the three models yielded similar predictions， with areas of high mineralization potential located in the central part of the Daping area. The ensemble model demonstrated superior predictive performance.

Highly efficient separation of Sc3+ and Y3+ in acid solution by a graphene oxide membrane with interlayer sieving

Sc and Y are key rare earth elements and are widely used in lamp phosphors,lasers and high-performance ***,highly efficient extraction and separation of Sc3+and Y3+is laborious,harmful,slow,and costly,strongly necessitating more efficient extraction and separation ***,we produced hydrated Sc3+-and hydrated Y3+-controlled graphene oxide（GO） membranes and find that both hydrated cations were completely self-rejected by the *** combining this selfrejection effect of the larger hydrated Y3+-controlled GO membrane and the rapid passage of the membrane through the smaller hydrated Sc3+,we proposed a strategy to separate Sc3+and Y3+by using a hydrated Y3+-controlled GO *** experimental results show that the permeation rate of Sc3+exceeds that of Y3+when the separation factor reaches 4.02,which can be attributed to the interlayer sieving effects of the GO *** finding illustrates the use of a forward osmosis process with a GO membrane for the efficient separation of Sc3+and Y3+by interlayer sieving,which provides a new effective and eco-friendly method for the separation of rare earth elements.
