300 MPa grade highly ductile biodegradable Zn-2Cu-(0.2-0.8)Li alloys with novel ternary phases

Although a few high-strength biodegradable Zn alloys with yield strengths(YSs)over 300 MPa in rolled state have been developed,their elongations(ELs)are generally less than 30%.This study developed rolled Zn-2Cu-x Li(x=0.2 wt.%,0.5 wt.%,0.8 wt.%)alloys with YSs of 316-335 MPa and ELs of 44%-61%.Three-dimensional atom probe(3DAP)and time of flight secondary ion mass spectrometry(TOF-SIMS)were employed to characterize Li distribution.Three kinds of Zn-Cu-Li ternary phases are identified,which are blockyε′-(Cu_(0.5),Li_(0.5))Zn 4,blockyβ′-(Li_(0.9),Cu_(0.1))Zn 4,and small roundγparticles with high Li content in the annealed state.Other identified phases are Zn,β-LiZn 4,andε-CuZn 4 phases.With the increase of Li content in the alloys,ε′phase with 6.50 at.%Cu transforms intoβ′phase with 2.12 at.%Cu,i.e.,the average level in the alloys.Withinε′phase,there exist nano-scale Li clusters andεphase,resulting inε′/εstructure.Dense Zn laths precipitate fromβ′phase,resulting inβ′/Zn lamellar structure.The lamel-lar structure is the matrix of Zn-2Cu-0.8Li and leads to near-isotropic plasticity.Electrochemistry tests show that degradation rates fall in the range of 153-196μm/year,which decrease with Li content.All the alloys exert positive effects on the growth of MC3T3-E1 cells with 10%extract.This research reveals how microstructure evolves in Zn-2Cu-x Li alloys,which lays the foundation for their future applications.

Biomaterials regulate macrophages and promote regeneration function,which is a new hot pot in tissue engineering and regenerative medicine.The research based on macrophage materials biology has appeared happy future,but related research on regulating macrophages and promoting tissue regeneration is still in its infancy.The surface roughness of biomaterials is one of the important factors affecting macrophage behavior.Previous study also found that the surface roughness of many biomaterials regulating macrophage polarization,but not including mineralized collagen(MC).In this study,we designed and fabricated MC with different roughness and investigated the influence of MC with different roughness on macrophages.In the study,we found that on the rough surface of MC,macrophages exhibited M1 phenotype-amoeboid morphology and high-level secretory of inflammatory factor(tumor necrosis factor-a and interleukin-6),while smoother surface exhibited M2 phenotype.These data will be beneficial to understand the mechanism deeply and enrich biomaterials tissue regeneration theory,provide a new train of thought biomaterials inducing tissue regeneration and repair and guide the optimum design of new biomaterials,development and reasonable applications.

Repairing damage in the craniofacial skeleton is challenging.Craniofacial bones require intramembranous ossification to generate tissue-engineered bone grafts via angiogenesis and osteogenesis.Here,we designed a mineralized collagen delivery system for BMP-2 and vascular endothelial growth factor(VEGF)for implantation into animal models of mandibular defects.BMP-2/VEGF were mixed with mineralized collagen which was implanted into the rabbit mandibular.Animals were divided into(i)controls with no growth factors;(ii)BMP-2 alone;or(iii)BMP-2 and VEGF combined.CT and hisomputed tomography and histological staining were performed to assess bone repair.New bone formation was higher in BMP-2 and BMP-2-VEGF groups in which angiogenesis and osteogenesis were enhanced.This highlights the use of mineralized collagen with BMP-2/VEGF as an effective alternative for bone regeneration.

Interaction between systemic iron parameters and left ventricular structure and function in the preserved ejection fraction population:a two-sample bidirectional Mendelian randomization study

BACKGROUND Left ventricular(lv)remodeling and diastolic function in people with heart failure(HF)are correlated with iron status;however,the causality is uncertain.This Mendelian randomization(MR)study investigated the bidirectional causal relationship between systemic iron parameters and lv structure and function in a preserved ejection fraction population.METHODS Transferrin saturation(TSAT),total iron binding capacity(TIBC),and serum iron and ferritin levels were extracted as instrumental variables for iron parameters from meta-analyses of public genome-wide association studies.Individuals without myocardial infarction history,HF,or lv ejection fraction(lvEF)lv end-diastolic volume,lv endsystolic volume,lv mass(lvM),and lvM-to-end-diastolic volume ratio(lvMVR).We used a two-sample bidirectional MR study with inverse variance weighting(IVW)as the primary analysis method and estimation methods using different algorithms to improve the robustness of the results.RESULTS In the IVW analysis,one standard deviation(SD)increased in TSAT significantly correlated with decreased lvMVR(β=-0.1365;95%confidence interval[CI]:-0.2092 to-0.0638;P=0.0002)after Bonferroni adjustment.Conversely,no significant relationships were observed between other iron and lv parameters.After Bonferroni correction,reverse MR analysis showed that one SD increase in lvEF significantly correlated with decreased TSAT(β=-0.0699;95%CI:-0.1087 to-0.0311;P=0.0004).No heterogeneity or pleiotropic effects evidence was observed in the analysis.CONCLUSIONS We demonstrated a causal relationship between TSAT and lv remodeling and function in a preserved ejection fraction population.

Energy consumption optimization for edge computing-supported cellular networks based on optimal transport theory

With advancements in mobile communication technologies, there have been multiple user device(UD) connections to cellular networks. Because of changes in the spatial distribution of UDs and application requirements, the traditional offloading mechanism based on the nearest distance will result in heavy loads for some base stations(BSs). Because there is a high-order relationship between the energy consumption of the BS processing tasks and the number of computing tasks, the traditional offloading mechanism will result in high energy consumption, and UD’s offloading decision must be dynamically adjusted. The offloading decision of UDs is optimized based on detailed information about various parameters associated with a network from the standpoint of distribution. This information includes details regarding the spatial distribution of UDs,application requirements, and the offloading period of computing tasks. Based on optimal transport theory,an energy consumption optimization algorithm is suggested to lower the total amount of energy consumed by the offloading process of UDs’ computing tasks by reasonably planning the offloading BSs of the UDs in the networks. The simulation results show that the proposed offloading mechanism based on energy consumption optimization reduces the total energy consumption of the offloading process by 28.09% when compared to the traditional offloading mechanism, and the traffic managed by each BS is balanced.

Synthesis of Anti-Pancreatic Cancer Natural Product Majusculamide D and Analogues Reveals a Preliminary Structure-Activity Relationships

The total synthesis of majusculamide D(1)was achieved from commercially available materials.In addition,we synthesized eight analogues including three stereoisomers of majusculamide D that differ in the fatty acid chain.Six analogues including a simplified analogue 29 exhibited significant nanomolar-level IC50 values against Panc-1 cells in MTT assays.A preliminary SAR analysis indicated that the hydroxyl group at C10 and C2−C3 unsaturated double bond of majusculamide D were essential in maintaining the high activity against Panc-1 cells and the orientation of C40-Me and C42-Me groups was tolerable.

Ancient dormant virus remnant ERVW-1 drives ferroptosis via degradation of GPX4 and SLC3A2 in schizophrenia

Human endogenous retroviruses(HERVs)are remnants of retroviral infections in human germline cells from millions of years ago.Among these,ERVW-1(also known as HERV-W-ENV,ERVWE1,or ENVW)encodes the envelope protein of the HERV-W family,which contributes to the pathophysiology of schizophrenia.Additionally,neuropathological studies have revealed cell death and disruption of iron homeostasis in the brains of individuals with schizophrenia.Here,our bioinformatics analysis showed that differentially expressed genes in the human prefrontal cortex RNA microarray dataset(GSE53987)were mainly related to ferroptosis and its associated pathways.Clinical data demonstrated significantly lower expression levels of ferroptosis-related genes,particularly Glutathione peroxidase 4(GPX4)and solute carrier family 3 member 2(SLC3A2),in schizophrenia patients compared to normal controls.Further in-depth analyses revealed a significant negative correlation between ERVW-1 expression and the levels of GPX4/SLC3A2 in schizophrenia.Studies indicated that ERVW-1 increased iron levels,malondialdehyde(MDA),and transferrin receptor protein 1(TFR1)expression while decreasing glutathione(GSH)levels and triggering the loss of mitochondrial membrane potential,suggesting that ERVW-1 can induce ferroptosis.Ongoing research has shown that ERVW-1 reduced the expression of GPX4 and SLC3A2 by inhibiting their promoter activities.Moreover,Ferrostatin-1(Fer-1),the ferroptosis inhibitor,reversed the iron accumulation and mitochondrial membrane potential loss,as well as restored the expressions of ferroptosis markers GSH,MDA,and TFR1 induced by ERVW-1.In conclusion,ERVW-1 could promote ferroptosis by downregulating the expression of GPX4 and SLC3A2,revealing a novel mechanism by which ERVW-1 contributes to neuronal cell death in schizophrenia.

As a highly invasive carcinoma,esophageal cancer(EC)was the eighth most prevalent malignancy and the sixth leading cause of cancer-related death worldwide in 2020.Esophageal squamous cell carcinoma(ESCC)is the major histological subtype of EC,and its incidence and mortality rates are decreasing globally.Due to the lack of specific early symptoms,ESCC patients are usually diagnosed with advanced-stage disease with a poor prognosis,and the incidence and mortality rates are still high in many countries,especially in China.Therefore,enormous challenges still exist in the management of ESCC,and novel strategies are urgently needed to further decrease the incidence and mortality rates of ESCC.Although the key molecular mechanisms underlying ESCC pathogenesis have not been fully elucidated,certain promising biomarkers are being investigated to facilitate clinical decision-making.With the advent and advancement of highthroughput technologies,such as genomics,proteomics and metabolomics,valuable biomarkers with high sensitivity,specificity and stability could be identified for ESCC.Herein,we aimed to determine the epidemiological features of ESCC in different regions of the world,especially in China,and focused on novel molecular biomarkers associated with ESCC screening,early diagnosis and prognosis prediction.

An Improved Harris Hawk Optimization Algorithm for Flexible Job Shop Scheduling Problem

Flexible job shop scheduling problem(FJSP)is the core decision-making problem of intelligent manufacturing production management.The Harris hawk optimization(HHO)algorithm,as a typical metaheuristic algorithm,has been widely employed to solve scheduling problems.However,HHO suffers from premature convergence when solving NP-hard problems.Therefore,this paper proposes an improved HHO algorithm(GNHHO)to solve the FJSP.GNHHO introduces an elitism strategy,a chaotic mechanism,a nonlinear escaping energy update strategy,and a Gaussian random walk strategy to prevent premature convergence.A flexible job shop scheduling model is constructed,and the static and dynamic FJSP is investigated to minimize the makespan.This paper chooses a two-segment encoding mode based on the job and the machine of the FJSP.To verify the effectiveness of GNHHO,this study tests it in 23 benchmark functions,10 standard job shop scheduling problems(JSPs),and 5 standard FJSPs.Besides,this study collects data from an agricultural company and uses the GNHHO algorithm to optimize the company’s FJSP.The optimized scheduling scheme demonstrates significant improvements in makespan,with an advancement of 28.16%for static scheduling and 35.63%for dynamic scheduling.Moreover,it achieves an average increase of 21.50%in the on-time order delivery rate.The results demonstrate that the performance of the GNHHO algorithm in solving FJSP is superior to some existing algorithms.

Repurposing diacerein to suppress colorectal cancer growth by inhibiting the DCLK1/STAT3 signaling pathway

Double cortin-like kinase 1(DCLK1)exhibits high expression levels across various cancers,notably in human colorectal cancer(CRC).Diacerein,a clinically approved interleukin(IL)-1βinhibitor for osteoarthritis treatment,was evaluated for its impact on CRC proliferation and migration,alongside its underlying mechanisms,through both in vitro and in vivo analyses.The study employed MTT assay,colony formation,wound healing,transwell assays,flow cytometry,and Hoechst 33342 staining to assess cell proliferation,migration,and apoptosis.Additionally,proteome microarray assay and western blotting analyses were conducted to elucidate diacerein’s specific mechanism of action.Our findings indicate that diacerein significantly inhibits DCLK1-dependent CRC growth in vitro and in vivo.Through high-throughput proteomics microarray and molecular docking studies,we identified that diacerein directly interacts with DCLK1.Mechanistically,the suppression of p-STAT3 expression following DCLK1 inhibition by diacerein or specific DCLK1 siRNA was observed.Furthermore,diacerein effectively disrupted the DCLK1/STAT3 signaling pathway and its downstream targets,including MCL-1,VEGF,and survivin,thereby inhibiting CRC progression in a mouse model,thereby inhibiting CRC progression in a mouse model.
