Heavy-atom engineered hypoxia-responsive probes for precisive photoacoustic imaging and cancer therapy

Photoacoustic agents combining photodynamic therapy(PDT) and photothermal therapy(PTT) functions have emerged as potent theranostic agents for combating cancer. The molecular approaches for enhancing the near-infrared(NIR)-absorption and maximizing non-radiative energy transfer are essential for effective photoacoustic imaging(PAI) and therapy applications. In addition, such molecules with high specificity and affinity to cancer cells are urgently needed, which would further decrease the side effect during treatments. In this study, we applied a heavy-atom engineering strategy and introduced p-aminophenol,-thio, and-seleno moieties into NIR heptamethine cyanine(Cy7) skeleton(Cy7-X-NH_(2), X = O, S, Se) to significantly increase photothermal conversion efficiency for PTT and promote intersystem crossing for PDT.Additionally, we designed a series of nitroreductase(NTR)-activated photoacoustic probes(Cy7-X-NO_(2),X = O, S, Se), and target hypoxic tumors with NTR overexpression. Our prostate cancer targeting probe,Cy7-Se-NO_(2)-KUE, exhibited specific tumor photoacoustic signals and effective tumor killing through outstanding synergistic PTT/PDT in vivo. These findings highlighted a versatile strategy for cancer photoacoustic diagnosis and enhanced phototherapy.

The Quaternary strata on the Qinghai-Tibet Plateau contain rich information about the paleoclimate and environmental evolution,record the evolution process of the Quaternary regional tectonics,paleogeography,and geomorphology of the plateau,and are extremely important areas for studying the Quaternary geological events and regional environmental evolution.According to a comprehensive analysis of the regional stratigraphic data and the development and evolution characteristics of the biota,based on the differences in the lithostratigraphic units,sedimentary characteristics,landforms,and drainage systems,the Quaternary strata on the Qinghai-Tibet Plateau and its surrounding areas are divided into five stratigraphic regions:the Tarim region,Loess Plateau region,Qinghai-Tibet Plateau region,Yunnan-Guizhou Plateau region,and India-Ganges region.The Qinghai-Tibet Plateau stratigraphic region is divided into seven stratigraphic sub-regions:the West Kunlun-Karakorum,AltunQilian Mountains,Qaidam-Hehuang,East Kunlun-Bayan Har,Qiangtang,East Xizang-West Yunnan-West Sichuan,and Gangdise-Himalayan sub-regions.This paper briefly describes the lithostratigraphic units of the seven stratigraphic sub-regions of the Qinghai-Tibet Plateau.According to the lithostratigraphic sequence and its sedimentary characteristics,stratigraphic contact relationship,formation age,and evolution of the biota in each stratigraphic sub-region,the Quaternary tectonic paleogeographic evolution of the Qinghai-Tibet Plateau is divided into four stages.(1)The inherited differential uplift stage since the Pliocene(2.6-1.8/1.5 Ma):the regional sedimentary differences were significant,and the stratigraphic distribution was limited,the alluvial-proluvial sandy conglomerate was widely developed along the piedmont,and fluvial and lacustrine deposits were developed in the low-lying areas between the mountains.(2)The mountain range flattening stage(1.8/1.5-1.2/0.8 Ma):the erosion unconformity surfaces around the plateau were widely distributed,large rivers were

Targeting oxidative stress with natural products: A novel strategy for esophageal cancer therapy

Esophageal cancer(ESC)is a malignant tumor that originates from the mucosal epithelium of the esophagus and is part of the digestive tract.Although the exact pathogenesis of ESC has not been fully elucidated,excessive oxidative stress is an important characteristic that leads to the development of many cancers.Abnormal expression of several proteins and transcription factors contributes to oxidative stress in ESCs,which alters the growth and proliferation of ESCs and promotes their metastasis.Natural compounds,including alkaloids,terpenes,polyphenols,and xanthine compounds,can inhibit reactive oxygen species production in ESCs.These compounds reduce oxidative stress levels and subsequently inhibit the oc-currence and progression of ESC through the regulation of targets and pathways such as the cytokine interleukins 6 and 10,superoxide dismutase,the NF-+ACY-kappa+ADs-B/MAPK pathway,and the mammalian Nrf2/ARE target pathway.Thus,targeting tumor oxidative stress has become a key focus in anti-ESC therapy.This review discusses the potential of Natural products(NPs)for treating ESCs and summarizes the application prospects of oxidative stress as a new target for ESC treatment.The findings of this review provide a reference for drug development targeting ESCs.Nonetheless,further high-quality studies will be necessary to determine the clinical efficacy of these various NPs.

Increased Expression of Serum Monocyte Chemoattractant Protein-1, Chemokine (C-C motif) Ligand 3, and Interleukins in Patients With Vitiligo

Objective: Vitiligo is a chronic, acquired depigmentation of the skin, which can be divided into active and stable vitiligo. The stable and active stages are not immutable and often alternate. Identification of internal markers would help to diagnose the active stage of vitiligo, and the purpose of this study was to find potential active markers in the progression of vitiligo.Methods: We compared 30 healthy controls to 60 patients with vitiligo (30 with active disease and 30 with stable disease) regarding the expression of serum monocyte chemoattractant protein (MCP)-1, chemokine (C-C motif) ligand (CCL) 3, CCL5, CCL21, interleukin (IL)-6, and IL-17. Specimens were collected from May 2017 to December 2018. Serological detection was carried out by enzyme-linked immunosorbent assay. The difference between groups was compared by Kruskal-WallisH test and Mann-WhitneyU test.Results: We found that the serum levels of MCP-1, CCL3, IL-6, and IL-17 were significantly higher in patients with vitiligo than in healthy controls (P han in those with stable vitiligo (P < 0.01). After 3months of treatment, the serum IL-17 level of patients with active vitiligo was not significantly different from that before treatment (P = 0.526); after 6months of treatment, however, the serum IL-17 level had significantly decreased (P < 0.01).Conclusion: These findings suggest that the serum IL-17 level can be used as a marker of vitiligo disease activity giving it certain guiding significance for clinical treatment.

Establishment of a pig model with enteric and portal venous drainage of pancreatoduodenal transplantation

AIM: To establish the pig model of pancreatoduodenal transplantation with enteric drainage (ED) and portal venous drainage (PVD).METHODS: Forty-six hybrid Landrace pigs were divided into two groups (donors and recipients) randomly, and pancreatoduodenal allotransplantation was performed.Donors were perfused via abdominal aorta without clamping the portal venous outflow with UW solution at 80-100 cm H2O after heparinization. Whole pancreatoduodenal grafts were harvested with segments of abdominal aorta and portal vein, and shaped under 4 ℃ UW solution. Then, end-to-end anastomosis was performed with the donor iliac artery bifurcation Y graft to the recipient superior mesenteric artery and celiac artery. Furthermore, type Ⅰ diabetes model was made by removal of the recipient pancreas. The venous anastomosis was reconstructed between the donor portal vein and the recipient superior mesentery vein. Meanwhile, end-toside anastomosis was performed with the donor common iliac artery bifurcation Y graft to the recipient abdominal aorta, and side-to-side intestinal anastomosis was performed between the donor duodenum and the recipient jejunum. External jugular vein was intubated for transfusion. Levels of plasma glucose, insulin and glucagon were measured during the operation and on the 1st, 3rd, 5th, and 7th d after operation.RESULTS: Pancreatoduodenal allotransplantation was performed on 23 pigs of which 1 died of complication of anesthesia. The success rate of operation was 95.6%.Complications of operation occurred in two cases in which one was phlebothrombosis with an incidence of 4.6%,and the other was duodenojejunal anastomotic leak with an incidence of 4.6%. The level of plasma glucose decreased within 30 min, after removal of pancreas and recovered on the 2nd d after operation. The level of plasma insulin and glucagon increased within 30 min after removal of pancreas and recovered on the 2nd d after operation.Rejection occurred on the 1st d and reached the worst level on the 7th d after transplantation, without ch

BACKGROUND Jianpi Gushen Huayu Decoction(JPGS)has been used to clinically treat diabetic nephropathy(DN)for many years.However,the protective mechanism of JPGS in treating DN remains unclear.AIM To evaluate the therapeutic effects and the possible mechanism of JPGS on DN.METHODS We first evaluated the therapeutic potential of JPGS on a DN mouse model.We then investigated the effect of JPGS on the renal metabolite levels of DN mice using non-targeted metabolomics.Furthermore,we examined the effects of JPGS on c-Jun N-terminal kinase(JNK)/P38-mediated apoptosis and the inflammatory responses mediated by toll-like receptor 4(TLR4)/nuclear factor-kappa B(NF-κB)/NOD-like receptor family pyrin domain containing 3(NLRP3).RESULTS The ameliorative effects of JPGS on DN mice included the alleviation of renal injury and the control of inflammation and oxidative stress.Untargeted metabolomic analysis revealed that JPGS altered the metabolites of the kidneys in DN mice.A total of 51 differential metabolites were screened.Pathway analysis results indicated that nine pathways significantly changed between the control and model groups,while six pathways significantly altered between the model and JPGS groups.Pathways related to cysteine and methionine metabolism;alanine,tryptophan metabolism;aspartate and glutamate metabolism;and riboflavin metabolism were identified as the key pathways through which JPGS affects DN.Further experimental validation showed that JPGS treatment reduced the expression of TLR4/NF-κB/NLRP3 pathways and JNK/P38 pathway-mediated apoptosis related factors.CONCLUSION JPGS could markedly treat mice with streptozotocin(STZ)-induced DN,which is possibly related to the regulation of several metabolic pathways found in kidneys.Furthermore,JPGS could improve kidney inflammatory responses and ameliorate kidney injuries in DN mice via the TLR4/NF-κB/NLRP3 pathway and inhibit JNK/P38 pathwaymediated apoptosis in DN mice.

Background Respiratory syncytial virus(RSV)is the leading global cause of respiratory infections and is responsible for about 3 million hospitalizations and more than 100,000 deaths annually in children younger than 5 years,representing a major global healthcare burden.There is a great unmet need for new agents and universal strategies to prevent RSV infections in early life.A multidisciplinary consensus development group comprising experts in epidemiology,infectious diseases,respiratory medicine,and methodology aims to develop the current consensus to address clinical issues of RSV infections in children.data sources The evidence searches and reviews were conducted using electronic databases,including PubMed,Embase,Web of Science,and the Cochrane Library,using variations in terms for"respiratory syncytial virus","RSV","lower respiratory tract infection","bronchiolitis","acute","viral pneumonia","neonatal","infant""children",and"pediatric".Results Evidence-based recommendations regarding diagnosis,treatment,and prevention were proposed with a high degree of consensus.Although supportive care remains the cornerstone for the management of RSV infections,new monoclonal antibodies,vaccines,drug therapies,and viral surveillance techniques are being rolled out.Conclusions This consensus,based on international and national scientific evidence,reinforces the current recommendations and integrates the recent advances for optimal care and prevention of RSV infections.Further improvements in the management of RSV infections will require generating the highest quality of evidence through rigorously designed studies that possess little bias and sufficient capacity to identify clinically meaningful end points.

Layered oxide cathodes for sodium-ion batteries:microstructure design,local chemistry and structural unit

Because of the low price and abundant reserves of sodium compared with lithium,the research of sodium-ion batteries(SIBs)in the field of large-scale energy storage has returned to the research spotlight.Layered oxides distinguish themselves from the mains cathode materials of SIBs owing to their advantages such as high specific capacity,simple synthesis route,and environmental benignity.However,the commercial development of the layered oxides is limited by sluggish kinetics,complex phase transition and poor air stability.Based on the research ideas from macro-to micro-scale,this review systematically summarizes the current optimization strategies of sodium-ion layered oxide cathodes(SLOC)from different dimensions:microstructure design,local chemistry regulation and structural unit construction.In the dimension of microstructure design,the various structures such as the microspheres,nanoplates,nanowires and exposed active facets are prepared to improve the slow kinetics and electrochemical performance.Besides,from the view of local chemistry regulation by chemical element substitution,the intrinsic electron/ion properties of SLOC have been enhanced to strengthen the structural stability.Furthermore,the optimization idea of endeavors to regulate the physical and chemical properties of cathode materials essentially is put forward from the dimension of structural unit construction.The opinions and strategies proposed in this review will provide some inspirations for the design of new SLOC in the future.

The high-energy photon source(HEPS)is the first fourth-generation synchrotron light source facility in China.The HEPS injector consists of a linear accelerator(Linac)and a full energy booster.The booster captures the electron beam from the Linac and increases its energy to the value required for the storage ring.The full-energy beam could be injected to the storage ring directly or after“high-energy accumulation.”On November 17,2023,the key booster parameters successfully reached their corresponding target values.These milestone results were achieved based on numerous contributions,including nearly a decade of physical design,years of equipment development and installation,and months of beam commissioning.As measured at the extraction energy of 6 GeV,the averaged beam current and emittance reached 8.57 mA with 5 bunches and 30.37 nm rad with a single-bunch charge of 5.58 nC,compared with the corresponding target values of 6.6 mA and 35 nm rad,respectively.This paper presents the physical design,equipment development,installation,and commissioning process of the HEPS booster.

Based on the unified Hauser–Feshbach and exciton model,which can describe the particle emission processes between discrete energy levels with energy,angular momentum,and parity conservations,a statistical theory of light nucleus reaction(STLN)is developed to calculate the double-differential cross-sections of the outgoing neutron and light charged particles for the proton-induced^(6) Li reaction.A significant difference is observed between the p+^(6) Li and p+^(7) Li reactions owing to the discrepancies in the energy-level structures of the targets.The reaction channels,including sequential and simultaneous emission processes,are analyzed in detail.Taking the double-differential cross-sections of the outgoing proton as an example,the influence of contaminations(such as^(1) H,^(7)Li,^(12)C,and^(16)O)on the target is identified in terms of the kinetic energy of the first emitted particles.The optical potential parameters of the proton are obtained by fitting the elastic scattering differential cross-sections.The calculated total double-differential cross-sections of the outgoing proton and deuteron at E_(p)=14 MeV agree well with the experimental data for different outgoing angles.Simultaneously,the mixed double differential cross-sections of^(3) He andαare in good agreement with the measurements.The agreement between the measured data and calculated results indicates that the two-body and three-body breakup reactions need to be considered,and the pre-equilibrium reaction mechanism dominates the reaction processes.Based on the STLN model,a PLUNF code for the p+^(6) Li reaction is developed to obtain an ENDF-6-formatted file of the double-differential cross-sections of the nucleon and light composite charged particles.
