AIM:To find out the role of salvianic acid A(SAA) in the protection of vascular endothehal cells(VEC) and its possible mechanism in ***:Added a series of this ingredient to human umbilical vein endothelial cells(HUVEC) treated with 0.5μM lipopolysaccharide(LPS) for 24 *** morphological changes of cells were observed under inverted phase contrast microscope,me cell viability was quantified using MTT assay. Nuclear fragmentation of cells was observed under laser scanning confocal microcopy after being stained with *** cycle distribution was detected by the flow-cytometric after being stained with propidium iodide(PI).The activities of glutathione peroxidase(GPH-PX) as well as maleic dialdehyde(MDA) level in cells were measured by spectrophotometric methods as described in the assay ***:Apoptotic morphological changes and the decrease of cell viability of these cells were obviously inhibited by SAA in a dose-dependent ***,the abnormal cell cycle distribution,the decrease of GSH-Px activity and the increase of MDA level induced by LPS were markedly ***:SAA produces protective action on VEC induced by LPS via an antioxidative mechanism.

目的:探寻丹参素 (丹参酸 A, SAA)对脂多糖(LPS)处理的血管内皮细胞 (VEC) 的保护作用及可能机制。方法:以 LPS 处理培养的人脐静脉血管内皮细胞, 造成细胞凋亡, 培养液中同时加入系列浓度的 SAA; 倒置相差显微镜观察细胞形态变化; 吖啶橙染色+激光共聚焦显微镜观测凋亡小体; PI 染色+流式细胞仪测定细胞周期分布; MTT 法检测细胞存活率; 生化试剂盒检测细胞内谷胱甘肽脱氢酶(GSH-Px) 活性及丙二醛(MDA)水平。结果:SAA 可明显抑制细胞凋亡, 提高细胞存活率。且该作用具浓度依赖性。对于 LPS 导致的细胞周期异常、GSH-Px 活性下降、MDA 水平升高, SAA 均可明显逆转。结论:丹参酸 A 通过抗氧化作用抑制内毒素诱导的血管内皮细胞凋亡。

通过血清代谢组学分析半夏白术天麻汤(Banxia Baizhu Tianma decoction,BBT)干预正常高值血压(prehypertension,PHT)痰湿壅盛证大鼠血清生物内源性物质的变化,明确BBT的代谢调控途径。复制正常高值血痰湿壅盛证大鼠模型,采集大鼠的血压、行为学指标、血清生化指标,评估BBT的调节血压和血脂的药效,利用超高效液相色谱-四极杆/静电场轨道阱高分辨质谱联用(UPLC-Q-orbitrap-MS)代谢组学技术,测定大鼠血清中内源性小分子物质变化,寻找潜在的生物标志物。结果显示,BBT可以对其中9个代谢物进行调控,主要包括花生四烯酸、胆酸、甘脱氧胆酸、N-腺苷酪氨酸、精氨酸、溶血磷脂酰乙醇胺(20:0/0:0)、溶血磷脂(P-18:0)、溶血磷脂(18:0)、溶血磷脂[22:5(7Z,10Z,13Z,16Z,19Z)]。通过MetaboAnalyst进行代谢通路分析,共有7条代谢通路与大鼠血压的改变密切相关,其中花生四烯酸代谢通路最为关键。模型大鼠体内代谢差异物有向正常水平回调的趋势,从代谢组学角度为BBT的作用机制提供了研究依据。本研究已获得山东中医药大学实验动物福利伦理审查委员会批准(批准号:SDUTCM20211103001)。

本实验旨在从下丘脑代谢组学角度探讨三子养亲汤(Sanzi Yangqin decoction,SYD)干预正常高值血压痰湿壅盛证大鼠的作用机制。将WKY大鼠随机分为正常组、模型组和SYD组。采用高脂+高盐方式制备正常高值血压痰湿壅盛证大鼠模型,不同剂量SYD干预模型大鼠8周,测量各组大鼠饮食量、体重、身长、血脂及血压,利用液质联用技术分析筛选各组大鼠下丘脑中差异代谢物,并进行代谢通路分析。结果显示,与模型组相比较,SYD组大鼠饮食量减少,体重降低,血脂血压显著下降。同时下丘脑14个差异代谢物发生回调,主要与氨基酸代谢、谷胱甘肽代谢、鞘脂代谢和甘油酯代谢有关,提示SYD可能通过调节下丘脑饮食行为,进而影响氨基酸代谢与能量代谢来发挥干预作用。本研究已获得山东中医药大学动物伦理审查委员会批准(批准号:SDUTCM20211103001)。

Objective Coronavirus disease 2019(COVID-19)and tuberculosis(TB)are major public health and social issues *** long-term follow-up of COVID-19 with pulmonary TB(PTB)survivors after discharge is *** study aimed to comprehensively describe clinical outcomes,including sequela and recurrence at 3,12,and 24 months after discharge,among COVID-19 with PTB *** From January 22,2020 to May 6,2022,with a follow-up by August 26,2022,a prospective,multicenter follow-up study was conducted on COVID-19 with PTB survivors after discharge in 13 hospitals from four provinces in *** outcomes,including sequela,recurrence of COVID-19,and PTB survivors,were collected via telephone and face-to-face interviews at 3,12,and 24 months after *** Thirty-two COVID-19 with PTB survivors were *** median age was 52(45,59)years,and 23(71.9%)were *** them,nearly two-thirds(62.5%)of the survivors were moderate,three(9.4%)were severe,and more than half(59.4%)had at least one comorbidity(PTB excluded).The proportion of COVID-19 survivors with at least one sequela symptom decreased from 40.6%at 3 months to 15.8%at 24 months,with anxiety having a higher proportion over a *** and amnesia recovered at the 12-month follow-up,while anxiety,fatigue,and trouble sleeping remained after ***,one(3.1%)case presented two recurrences of PTB and no re-positive COVID-19during the follow-up *** The proportion of long symptoms in COVID-19 with PTB survivors decreased over time,while nearly one in six still experience persistent symptoms with a higher proportion of *** recurrence of PTB and the psychological support of COVID-19 with PTB after discharge require more attention.

Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains *** A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in china. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of *** Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. 目的对五加皮Acanthopanacis Cortex的二萜类化学成分及抗炎活性进行研究。方法通过MCI树脂柱、硅胶柱、薄层色谱、半制备HPLC等方法对五加皮药材中化学成分进行提取分离,采用核磁共振波谱、质谱、红外光谱等方法,对单体化合物的结构进行鉴定。采用Griess法测试脂多糖诱导小鼠单核巨噬细胞RAW264.7细胞释放炎症因子模型,对化合物进行抗炎活性评价。结果从五加皮70%乙醇提取物中分离得到13个二萜类化合物,包括7个对映海松烷型二萜7β-羟基-8(14),15-二烯-19-异海松烷酸(1)、7-酮基-8(14),15-二烯-19-异海松烷酸(2)、7β-甲氧基-8(14),15-二烯-19-异海松烷酸(3)、6,8(14),15-三烯-19-异海松羧酸(4)、14-酮基-8,15-二烯-19-异海松羧酸(7)、14-羟基-16-乙烯基-8,11,13-三烯-17-异海松羧酸(8)、7α-羟基-8(14),15-二烯-19-异海松烷酸(11);以及6个对映贝壳杉烷型二萜16α,17-二羟基-19-异贝壳杉烷羧酸(5)、17-羟基-19-异贝壳杉烷羧酸(6)、异贝壳杉烯酸(9)、17-醛基-19-异贝壳杉烯酸(10)、17-醛基-15-烯-19-异贝壳杉烷酸(12)和17-羟基-15-烯-19-异贝壳杉烷酸(13)。其中化合物8为新化合物,命名为细柱五加酸G1;3和4为首次从细柱五加植物中分离得到。体外抗炎活性研究表明,13个化合物均具有一定的抗炎活性,其中化合物1~3、7的活性较好。海松烷型二萜类化合物抗炎活性较贝壳杉烷型二萜强,海松烷型二萜类化合物7位取代活性强弱顺序为β-OH>β-OCH_(3)>Δ^(6)双键>C=O>α-OH。结论五加皮分离鉴定的化合物及其活性研究为五加皮专属性质控方法建立、药用资源合理开发和利用提供了依据。
