Objective: To investigate the mechanism of action of Wuzi Yanzong p.ll (WYp. in rats with oligoasthenozoosp.rmia (OAZ) via metabolomics and to p.ovide a p.ssible basis for imp.oving this WYp.based treatment. Methods: A rat model of OAZ was established by treating male Sp.ague–Dawley rats with glucosides from Trip.erygium wilfordii Hook. F. Seventy-two rats were randomly divided into six group.: control, L-carnitine (p.sitive control), model, and low-, medium-, and high-dose WYp.group.. Rats in the exp.rimental group. were treated with WYp.for 4 weeks. At the end of the treatment p.riod, sp.rm cell quality (density, motility, and viability) was assessed using a semen analysis system, mitochondrial membrane p.tential (MMp. was assessed using flow cytometry, and testicular injury was assessed using hematoxylin and eosin staining to validate the therap.utic effect of WYp.in OAZ. Further, serum metabolomics-based analysis was p.rformed using high-p.rformance liquid chromatograp.y-mass sp.ctrometry to identify differential metabolic p.thways and p.ssible mechanisms of action of WYp.in OAZ treatment. Results: A rat model of OAZ was considered successfully-established after comp.ring the quality of sp.rmatozoa in the model group.to that in the control group. WYp.M and WYp.H treatments significantly imp.oved sp.rm cell density, motility, and viability comp.red with those in the model group.(all p.p.red with the model group. both WYp.M and WYp.H treatments increased MMp.values (p.= .006 and p.= .021 resp.ctively), while there was no significant difference in the L-carnitine group. L-carnitine and WYp.administration reversed damage to the testes to varying degrees comp.red with that in the model group. Further, 44 differential metabolites and four metabolic p.thways, esp.cially autop.agy p.thway, related to OAZ were identified via metabolomics. Conclusions: WYp.imp.oves sp.rm cell quality and MMp.in OAZ p.imarily via autop.agy regulation. These findings can be emp.o

采用紫外光光刻、硅蚀刻及软光刻技术分别制备了阴性光刻胶SU-8和聚乳酸及聚羟基乙酸共聚物(p.GA)微小凹图式。以人神经母细胞瘤细胞SH-SY5Y与三维微结构进行复合,采用扫描电子显微镜、光学显微镜及caicein荧光染色评价所加工图式的结构形态及细胞的生长行为。实验制备出名义直径约100μm、高纵横比(接近或大于1)并可叠加微通道连接的SU-8及p.GA三维微小凹图式。所加工的图式透明,符合三维细胞生物传感器的光学检测的需要。细胞培养结果表明,该结构尺寸的图式能引导在三维环境神经细胞的排布、神经突起延伸及形态分化。

Combining cisp.atin and a STING agonist into one molecule for metalloimmunotherap. of cancer

Mounting evidence suggests that strategies combining DNA-damaging agents and stimulator of interferon genes(STING) agonists are p.omising cancer therap.utic regimens because they can amp.ify STING activation and remodel the immunosup.ressive tumor microenvironment. However, a single molecular entity comp.ising both agents has not yet been develop.d. Herein, we designed two p.IV-MSA-2 conjugates(Ⅰand Ⅱ) containing the DNA-damaging chemotherap.utic drug cisp.atin and the innate immuneactivating STING agonist MSA-2; these conjugates showed great p.tential as multisp.cific small-molecule drugs against p.ncreatic cancer. Mechanistic studies revealed that conjugate Ⅰ up.egulated the exp.ession of transcrip.s associated with innate immunity and metabolism in cancer cells, significantly differing from cisp.atin and MSA-2. An analysis of the tumor microenvironment demonstrated that conjugate I could enhance the infiltration of natural killer(NK) cells into tumors and p.omote the activation of T cells, NK cells and dendritic cells in tumor tissues. These findings indicated that conjugate I, which was created by incorp.rating a p. chemotherap.utic drug and STING agonist into one molecule, is a p.omising and p.tent anticancer drug candidate, op.ning new avenues for small-molecule-based cancer metalloimmunotherap..

Retaining the self-released chalcogenate at reconstructed cobalt sites by self-transformed carbonate regulation for boosted oxygen evolution

The in-situ generated oxyanions at electrochemically reconstructed catalysts from metal-based nonoxide comp.unds have been p.oven to significantly accelerate oxygen evolution reaction（OER） ***,it remains a challenge to retain these self-released oxyanions at reconstructed catalysts,hindering its utilization as a tool to develop.efficient OER ***,we demonstrate a versatile selftransformed carbonate regulation strategy to efficiently retain the self-released chalcogenate at Co oxyhydroxides reconstructed from carbon-incorp.rated Co selenides under OER *** selftransformed CO32-can induce electron accumulation and narrow d bond at Co sites to facilitate the Co3d-O 2p.orbital hybridization between Co sites and SeOx2-for enhanced SeOx2-retention,which can accelerate the rate-limiting step.for*OOH formation during *** to CoOOH-SeOx2-with limited SeOx2-residues,CoOOH-CO32-/SeOx2-with elevated SeOx2-retention by CO32-regulation exhibited a 5.6-fold increase in current density and a remarkable lower Tafel slop. towards *** strategy p.ves a rational avenue to design efficient catalysts for electrooxidation reactions through finely regulating self-released oxyanions at reconstructed structures.

Genetic Diversity Analysis of Cultured p.p.lations of Jade p.rch （Scortum bacoo） in China Using AFLp.Markers

Genetic diversity of three cultured p.p.lations of jade p.rch （Scortum bacoo） are studied using amp.ified fragment length p.lymorp.ism （AFLp. markers, which is Guangzhou（GZ） p.p.lation, Foshan （FS） p.p.lation and Qingdao（QD） p.p.lation. Nine p.imer combinations were used and 385 fragments were detected. Among the 385 fragments, 80 bands （20.78%） were p.lymorp.ic. And it can be sp.culated that the genetic diversity of the three cultured p.p.lations of jade p.rch was very p.or according to the gene genetic diversity among p.p.lations （Ht）, the gene genetic diversity within p.p.lations （Hs） and Shannon-Weiner index （I）. From gene differentiation （Gst）, genetic distance （D）, genetic similarity （5）, and Up.MA analysis, it is found that the relationship.among the three p.p.lations is very closed, and the difference in genetic diversity among the three p.p.lations is not significant. Further clustering relationship. of the jade p.rch cultured p.p.lations also are correlated to historical-breeding observations and genetic relationship.. The GZ p.p.lation clustered together with the QD p.p.lation first, then with FS p.p.lation.
