Objective: To access the toxicity of forsythin from Forsythia suspensa leaves and evaluate its safety,Methods: Acute toxicity was determined by oral administration of a single dose of 18 100 mg/kg forsythin in NIH mice,Sub-chronic toxicity was evaluated by oral administration of several doses of forsythin for 30 days at does of 0,540,1 620,and 6 480 mg/kg in SD ***: In the acute toxicity study,mortality was not observed after 14 days,In addition,clinically relevant adverse effects,or variations in body weight or food consumption were not observed,Similarly,after 30 days in the sub-chronic toxicity study,no mortality or significant toxicological effects such as decreased food consumption,body weight,biochemical parameters and vital organs etc,were noticed,Conclusion: The results revealed that the forsythin from Forsythia suspensa leaves has low or no toxicity via oral administration,and therefore is suitable for further development and applications.

SARS-CoV-2,the culprit pathogen of COVID-19,elicits prominent immune responses and cytokine *** Cl^(−)is a crucial regulator of host defense,whereas the role of Cl^(−)signaling pathway in modulating pulmonary inflammation associated with SARS-CoV-2 infection remains *** using human respiratory epithelial cell lines,primary cultured human airway epithelial cells,and murine models of viral structural protein stimulation and SARS-CoV-2 direct challenge,we demonstrated that SARS-CoV-2 nucleocapsid(N)protein could interact with Smad3,which downregulated cystic fibrosis transmembrane conductance regulator(CFTR)expression via *** intracellular Cl^(−)concentration([Cl^(−)]i)was raised,resulting in phosphorylation of serum glucocorticoid regulated kinase 1(SGK1)and robust inflammatory *** or knockout of SGK1 abrogated the N protein-elicited airway ***,N protein promoted a sustained elevation of[Cl^(−)]i by depleting intracellular cAMP via upregulation of phosphodiesterase 4(PDE4).Rolipram,a selective PDE4 inhibitor,countered airway inflammation by reducing[Cl^(−)]*** findings suggested that Cl^(−)acted as the crucial pathological second messenger mediating the inflammatory responses after SARS-CoV-2 *** the Cl^(−)signaling pathway might be a novel therapeutic strategy for COVID-19.

Trypanosomes are haemoflagellates parasiting various *** represent a group of early branched eukaryotic,with numbers of diverged characteristics from mammalian *** most obvious one is that they contain a specialized mitochondrial genome,termed a *** kinetoplast forms a disc shape of interlocking DNA circles of two types,maxicircle and *** is generally over 20 kb,and encoding some typical and more encrypted mitochondrial genes,which are decrypted only using hundreds of keys from *** this structure,there are some very interesting questions to be answered.1)How do they maintain this complex mitochondrial genome?2) How does this complex mitochondrial genome work?3) How does this complex mitochondrial genome evolve from and to?To address these questions,the authors investigate the model organisms of Trypanosome brucei,Trypanosome evansi,Trypanosome equiperdum and Trypanosome *** authors confirmed the maintenance of kinetoplast via basal body,and identified novel protein of *** authors revealed the transcriptome patterns of kinetoplast,and identified new structure of *** authors revealed the distinct feathers of various kinetoplasts,and proposed evolutionary scenery of ***,kinetoplast is a distinct and complex structural mitochondrial genome,which is also an excellent examples to show the diversity of eukaryotic cells and stupidity of designer if presents.

Optimal spacing for vertical wells can be effectively predicted with several published methods,but methods suitable for assessing the proper horizontal well spacing are *** work proposes a method for calculating the optimal horizontal well spacing for an ultra-low permeability reservoir e the Yongjin reservoir in the Juggar Basin,northwestern *** result shows that a spacing of 640m is the most economical for the development of the *** better develop the reservoir,simulation approaches are used and a new model is built based on the calculated well *** the reservoir has an ultralow permeability,gas injection is regarded as the preferred enhanced oil recovery(EOR)*** of different gases including carbon dioxide,methane,nitrogen and mixed gas are *** results show that carbon dioxide injection is the most efficient and economical for the development of the ***,if the reservoir produces enough methane,reinjecting methane is even better than injecting carbon dioxide.

Learning Sequential and Structural Dependencies Between Nucleotides for RNA N6-Methyladenosine Site Identification

N6-methyladenosine(m6A)is an important RNA methylation modification involved in regulating diverse biological processes across multiple ***,the identification of m6A modification sites provides valuable insight into the biological mechanisms of complex diseases at the post-transcriptional *** a variety of identification algorithms have been proposed recently,most of them capture the features of m6A modification sites by focusing on the sequential dependencies of nucleotides at different positions in RNA sequences,while ignoring the structural dependencies of nucleotides in their threedimensional *** overcome this issue,we propose a cross-species end-to-end deep learning model,namely CR-NSSD,which conduct a cross-domain representation learning process integrating nucleotide structural and sequential dependencies for RNA m6A site ***,CR-NSSD first obtains the pre-coded representations of RNA sequences by incorporating the position information into single-nucleotide states with chaos game representation *** then constructs a crossdomain reconstruction encoder to learn the sequential and structural dependencies between *** minimizing the reconstruction and binary cross-entropy losses,CR-NSSD is trained to complete the task of m6A site *** experiments have demonstrated the promising performance of CR-NSSD by comparing it with several state-of-the-art m6A identification ***,the results of cross-species prediction indicate that the integration of sequential and structural dependencies allows CR-NSSD to capture general features of m6A modification sites among different species,thus improving the accuracy of cross-species identification.

Segmentally assembled bridges are increasinglyﬁnding engineering applications in recent years due to their unique advantages,especially as urban *** loads are one of the most important variable loads acting on bridge ***,the inﬂuence of overloaded vehicles on existing assembled bridge structures is an urgent concern at *** paper establishes theﬁnite element model of the segmentally assembled bridge based on ABAQUS software and analyzes the inﬂuence of vehicle overload on an assembled girder bridge ***,aﬁnite element model corresponding to the target bridge is established based on ABAQUS software,and the load is controlled to simulate vehicle movement in each area of the traveling zone at different ***-ond,the key cross-sections of segmental girder bridges are monitored in real time based on the force character-istics of continuous girder bridges,and they are compared with the simulation ***,a material damage ontology model is introduced,and the structural damage caused by different overloading rates is compared and *** show that theﬁnite element modeling method is accurate by comparing with on-site measured data,and it is suitable for the numerical simulation of segmental girder bridges;Dynamic sensors installed at 1/4L,1/2L,and 3/4L of the segmental girder main beams could be used to identify the dynamic response of segmental girder bridges;The bottom plate of the segmental girder bridge is mostly damaged at the position where the length of the precast beam section changes and the midspan *** the increase in load,damage in the direction of the bridge develops faster than that in the direction of the transverse ***ﬁndings of this study can guide maintenance departments in the management and maintenance of bridges and vehicles.
