Taxonomy of Two New Peritrich Ciliates(Protozoa:Ciliophora)from Inland Saltwaters of China

The inland saltwater lakes harbor exceptional biodiversity.Here,two new species of solitary sessile peritrich ciliates were isolated from Qinghai Lake,the largest inland saltwater lake in China.Their morphology,ciliature,silverline system,and molecular phylogeny were investigated based on live observation,silver staining,and analysis of the small subunit ribosomal DNA(SSU rDNA).Vorticella paraglobosa sp.***.is characterized mainly by its obconical or elongate bell-shaped zooid,C-shaped macronucleus,single ventrally located contractile vacuole,two-rowed infundibular polykinety 3,and 28-38 silverlines between peristome and aboral tro-chal band and 10-15 between aboral trochal band and scopula.Vorticella cotyliformis sp.***.differs from its congeners mainly by its double-layered peristomial lip,cup-shaped zooid,J-shaped macronucleus,single ventrally positioned contractile vacuole,three-rowed infundibular polykinety 3,and 70-85 silverlines between peristome and aboral trochal band and 21-25 between aboral trochal band and scopula.The SSU rDNA sequences of the two new species were obtained,and the subsequent molecular phylogenetic analysis supported their taxonomic classification.

Triaxial mechanical properties and precursors of instability damage in rocks with different laminar dip angles

To investigate the mechanical failure characteristics of volcanic breccia with different bedding dip angles in deep layers,triaxial compression experiments were conducted on specimens taken from volcanic rock layers in the Junggar Basin,Xinjiang.By changing the confining pressure,we compared the mechanical properties,acoustic emission characteristics,and fractal features of the acoustic emission sequence for volcanic breccia with different bedding dip angles.The research results indicate that as the bedding dip angle increases,peak strength,internal friction angle,and cohesion exhibit a“U-shaped”variation pattern.The correlation dimension of the acoustic emission amplitude was calculated using the G-P algorithm,revealing that volcanic breccia exhibits fractal characteristics.Furthermore,a positive correlation between the correlation dimension and the bedding dip angle was observed.When the stress of the test specimen reaches 0.8σ_(c),there is a significant decrease in the fractal dimension,serving as a criterion for predicting the failure of the specimen.By analyzing the changes in fractal dimension at different stress levels,the slope of the fractal dimension△D<-0.5 can be used as a precursor to failure for test specimens with a bedding dip angle of 15°≤β≤45°.Similarly,when the bedding dip angle of the specimen is 0°≤β<15°or 45°<β≤90°,△D<0 can be considered as a precursor to specimen failure.This study provides theoretical references for understanding the failure patterns of volcanic rock.

The angle α between the fault strike and the axial direction of the roadway produces different damage characteristics. In this paper, the research methodology includes theoretical analyses, numerical simulations and field experiments in the context of the Daqiang coal mine located in Shenyang, China. The stability control countermeasure of "pre-splitting cutting roof + NPR anchor cable"(PSCR-NPR) is simultaneously proposed. According to the different deformation characteristics of the roadway, the faults are innovatively classified into three types, with α of type I being 0°-30°, α of type II being 30°-60°, and α of type III being 60°-90°. The full-cycle stress evolution paths during mining roadway traverses across different types of faults are investigated by numerical simulation. Different pinch angles α lead to high stress concentration areas at different locations in the surrounding rock. The non-uniform stress field formed in the shallow surrounding rock is an important reason for the instability of the roadway. The pre-cracked cut top shifted the high stress region to the deep rock mass and formed a low stress region in the shallow rock mass. The high prestressing NPR anchor cable transforms the non-uniform stress field of the shallow surrounding rock into a uniform stress field. PSCR-NPR is applied in the fault-through roadway of Daqiang mine. The low stress area of the surrounding rock was enlarged by 3-7 times, and the cumulative convergence was reduced by 45%-50%. It provides a reference for the stability control of the deep fault-through mining roadway.

Carrimycin(CA),sanctioned by China’s National Medical Products Administration(NMPA)in 2019 for treating acute bronchitis and sinusitis,has recently been observed to exhibit multifaceted biological activities,encompassing anti-inflammatory,antiviral,and anti-tumor properties.Despite these applications,its efficacy in sepsis treatment remains unexplored.This study introduces a novel function of CA,demonstrating its capacity to mitigate sepsis induced by lipopolysaccharide(LPS)and cecal ligation and puncture(CLP)in mice models.Our research employed in vitro assays,real-time quantitative polymerase chain reaction(RT-qPCR),and RNA-seq analysis to establish that CA significantly reduces the levels of pro-inflammatory cytokines,namely tumor necrosis factor-alpha(TNF-α),interleukin 1 beta(IL-1β),and interleukin 6(IL-6),in response to LPS stimulation.Additionally,Western blotting and immunofluorescence assays revealed that CA impedes Nuclear Factor Kappa B(NF-κB)activation in LPS-stimulated RAW264.7 cells.Complementing these findings,in vivo experiments demonstrated that CA effectively alleviates LPS-and CLP-triggered organ inflammation in C57BL/6 mice.Further insights were gained through 16S sequencing,highlighting CA’s pivotal role in enhancing gut microbiota diversity and modulating metabolic pathways,particularly by augmenting the production of short-chain fatty acids in mice subjected to CLP.Notably,a comparative analysis revealed that CA’s anti-inflammatory efficacy surpasses that of equivalent doses of aspirin(ASP)and TIENAM.Collectively,these findings suggest that CA exhibits significant therapeutic potential in sepsis treatment.This discovery provides a foundational theoretical basis for the clinical application of CA in sepsis management.

Failure mechanism and safety control technology of a composite strata roadway in deep and soft rock masses:a case study

The construction of coal mines often encounters deep composite soft rock roadways,which is characterized by significant deformation and poor stability.To deeply study the failure mechanism and large deformation challenges of a composite strata roadway in deep and soft rock masses,a numerical model of 3DEC tetrahedral blocks was established based on the method of rock quality designation(RQD).The results showed that original support cannot prevent asymmetric failure and large deformation due to the adverse geological environment and unsuitable support design.According to the failure characteristics,a coupling support of“NPR bolt/cable+mesh+shotcrete+steel pipe”was proposed to control the stability of the surrounding rock.The excellent mechanical properties of large deformation(approximately 400 mm)and high constant resistance force(bolt with 180 k N;cable with 350 k N)were evaluated by the tensile tests.The numerical results showed that the maximum deformation was minimized to 243 mm,and the bearing capacity of the surrounding rock of the roadway was enhanced.The field test results showed that the maximum deformation of the surrounding rock was 210 mm,and the forces of the NPR bolt and cable were stable at approximately 180 k N and 350 k N,respectively.This demonstrated the effectiveness of the coupling support with the NPR bolt and cable,which could be a guiding significance for the safety control of large deformation and failure in deep composite soft rock roadways.

Fabrication of Conductive Domain Walls in x-cut Congruent Thin-film Lithium Niobate Using an Electrical-field Poling Technique(Invited)

Conductive ferroelectric domain walls have attracted increasing research interest in the field of nanoelectronics, and the fabrication technique for such domain walls is vital. In this study, we investigated in detail the fabrication of conductive domain walls in x-cut congruent thin-film lithium niobate(TFLN) using an electrical-field poling technique. The ferroelectric domain structures can be controlled through the applied electrical field and applied pulse numbers, and the domain inversion process is related to the conduction characteristics of the domain walls. The domain structures in TFLN are revealed using confocal second-harmonic microscopy and piezoresponse force microscopy. The results provide further directions for the development and application of conductive domain walls in TFLN.

Shape memory photonic crystals(SMPCs)are smart composite materials with changeable structural color integrated by shape memory polymer and photonic crystals.SMPC can produce one or more temporary shapes through nanoscale deformation,memorizing current states.SMPC can be recovered to their original shapes or some intermediate states under external stimuli,accompanied by the variation of structural color.As porous carriers with built-in sensing properties,SMPCs promoted the interdisciplinary development of nanophotonic technology in materials science,environmental engineering,biomedicine,chemical engineering,and mechanics.Herein,the recent progress on multifunctional SMPCs and practical applications,including traditional and cold programmable SMPCs,is summarized and discussed.The primary concern is shape programming at the nanoscale that has demonstrated numerous attractive functions,including smart sensing,ink-free printing,solvent detection,reprogrammable gradient wetting,and controllable bubble transportation,under variations of the surface nanostructure.It aims to figure out the nanoscale shape memory effects on structural color conversion and additional performance,inspiring the fabrication of the next generation of SMPCs.Finally,perspectives on future research directions and applications are also presented.It is believed that multifunctional SMPCs are powerful nanophotonic tools for the interdisciplinary development of numerous disciplines in the future.

Controller area networks(CANs),as one of the widely used fieldbuses in the industry,have been extended to the automation field with strict standards for safety and reliability.In practice,factors such as fatigue and insulation wear of the cables can cause intermittent connection(IC)faults to occur frequently in the CAN,which will affect the dynamic behavior and the safety of the system.Hence,quantitatively evaluating the performance of the CAN under the influence of IC faults is crucial to real-time health monitoring of the system.In this paper,a novel methodology is proposed for real-time quantitative evaluation of CAN availability when considering IC faults,with the system availability parameter being calculated based on the network state transition model.First,the causal relationship between IC fault and network error response is constructed,based on which the IC fault arrival rate is estimated.Second,the states of the network considering IC faults are analyzed,and the deterministic and stochastic Petri net(DSPN)model is applied to describe the transition relationship of the states.Then,the parameters of the DSPN model are determined and the availability of the system is calculated based on the probability distribution and physical meaning of markings in the DSPN model.A testbed is constructed and case studies are conducted to verify the proposed methodology under various experimental setups.Experimental results show that the estimation results obtained using the proposed method agree well with the actual values.

Humanized immune system(HIS)mice have been developed and used as a small surrogate model to study human immune function under normal or disease conditions.Although variations are found between models,most HIS mice show robust human T cell responses.However,there has been unsuccessful in constructing HIS mice that produce high-affinity human antibodies,primarily due to defects in terminal B cell differentiation,antibody affinity maturation,and development of primary follicles and germinal centers.In this review,we elaborate on the current knowledge about and previous attempts to improve human B cell development in HIS mice,and propose a potential strategy for constructing HIS mice with improved humoral immunity by transplantation of human follicular dendritic cells(FDCs)to facilitate the development of secondary follicles.
