A new fluorescent reagent, 7 (8 hydroxy 3, 6 disulfonaphthylazo) 8 hydroxyquinoline 5 sulfonic acid (HDNHQ) for the determination of magnesium has been developed. It reacted with magnesium to form a 1∶1 fluorescent complex with λ ex / λ em =356/495 nm immediately at room temperature in ammonia ammonium chloride buffer (pH 10.7). A linear relationship was obtained in the magnesium concentration range of 0 160 ng·mL -1 with the detection limit of 0.04 ng·mL -1 . The proposed method was simple, rapid and sensitive. It has been successfully applied to the determination of trace magnesium in blood serum with recoveries of 103.75% and 98.16%, respectively.

Mornaphthoate E(MPE)is a prenylated naphthoic acid methyl ester isolated from the roots of a famous Chinese medicinal plant Morinda officinalis and shows remarkable cytotoxicity against several human tumor cell *** the current project,the first total synthesis of(±)-MPE was achieved in seven steps and 5.6%overall *** the in vitro anti-tumor activity of MPE was first assessed for both enantiomers in two breast cancer cells,with the levoisomer exerting slightly better *** in vivo anti-tumor effect was further verified by applying the racemate in an orthotopic autograft mouse ***,MPE exerted promising anti-metastasis activity both in vitro and in vivo and showed no obvious toxicity on mice at the therapeutic *** investigations demonstrated that MPE acted as a tubulin polymerization stabilizer and disturbed the dynamic equilibrium of microtubules via regulating PI3K/Akt *** conclusion,our work has provided a new chemical template for the future design and development of next-generation tubulin-targeting chemotherapies.

Expression of pathogenesis-related genes in cotton roots in response to Verticillium dahliae PAMP molecules

Verticillium wilt disease becomes a major threat to many economically important crops. It is unclear whether and how plant immunity takes place during cotton-Verticillium interaction due to the lack of marker genes. Taking advantage of cotton （Gossypium hirsutum） genome, we discovered pathogenesis-related （PR） gene families, which have been widely used as markers of immune responses in plants. To profile the expression of G. hirsutum PR genes in the process of plant immunity, we treated cotton roots with two immunogenic peptides, fig22 and nlp20 known as pathogen-associated molecular patterns, as well as three Verticillium dahliae-derived peptides, nlp20vd2, nlp23vd3, and nlp23vd4 which are highly identical to nlp20. Quantitative real-time PCR results revealed that 14 G. hirsutum PR gene （GhPR） families were induced or suppressed independently in response to fig22, nip20, nlp20va2, nlp23vd3, and nlp23vd4. Most GhPR genes are expressed highest at 3 h post incubation of immunogenic peptides. Compared to fig22 and nlp20, nlp20vd2 is more effective to trigger up-regulated expression of GhPR genes. Notably, both nlp23vd3 and nlp23vd4 are able to induce GhPR gene up-regulation, although they do not induce necrosis on cotton leaves. Thus, our results provide marker genes and new immunogenic peptides for further investigation of cotton-V, dahliae interaction.

Roles of mushroom polysaccharides in chronic disease management

Chronic diseases have drawn much attention as the primary cause of death and disability. In exploring novel sideeffect-free agents against chronic diseases, significant efforts have been devoted to mushroom polysaccharides due to their diverse biological activities. This work reviewed the structural features, biological performances and molecular mechanisms of mushroom polysaccharides in managing cancers, diabetes mellitus and cardiovascular diseases. The potentials of mushroom polysaccharides against chronic diseases highly depend on their structural features, including monosaccharide composition, molecular weight, the type and configuration of glycosidic bonds, degree of branching, the type of substituent pattern and chain conformation. Regarding their working mechanisms, shared and diseasespecific pathways were found. The three chronic diseases shared the regulation of specific signalling pathways and the adjustment of gut microbiota. In addition, the roles of transcription factors, receptors, enzymes, hormones and other functional proteins involved in the molecular mechanisms of mushroom polysaccharides against chronic diseases are first elaborated herein. The present review describes the state of the art of mushroom polysaccharides in treating chronic diseases and addresses the perspectives, and will further promote research on this topic.

AIM: To construct another growth hormone releasing hormone (GHRH) analog, Pro-Pro-hGHRH(1-44)-Gly-Gly-Cys peptide and to compare its activity with that of Pro-Pro-hGHRH(l-44)OH peptide. METHODS: The pro-pro-hGHRH(1-44)-gly-gly-cys DNA fragment was synthesized by polymerase chain reaction. The recombinant protein was expressed to high levels in Escherichia coli BL21 (DE3). The Pro-Pro-hGHRH (1-44)-Gly-Gly-Cys peptide was purified to homogeneity by cell disruption, washing, ethanol precipitation, acid hydrolysis, and SP-Sephadex C-25 and Sephadex G-25 column chromatography. The peptide molecular mass was determined by electrospray ionization mass spectroscopy (ESI-MS). Its purity was determined by SDS-PAGE. The concentration of growth hormone (GH) stimulated by GHRH and its analogs was determined with antiserum kit against human GH. Somatic mutations of the chromatin remodeling gene ARID2 are observed in ～7% of human lung adenocarcinomas(LUADs). However, the role of ARID2 in the pathogenesis of LUADs remains largely unknown. Here we find that ARID2 expression is decreased during the malignant progression of both human and mice LUADs. Using two KrasG12 D-based genetically engineered murine models, we demonstrate that ARID2 knockout significantly promotes lung cancer malignant progression and shortens overall survival. Consistently, ARID2 knockdown significantly promotes cell proliferation in human and mice lung cancer cells. Through integrative analyses of Ch IP-Seq and RNA-Seq data, we find that Hspa1 a is up-regulated by Arid2 loss. Knockdown of Hspa1 a specifically inhibits malignant progression of Arid2-deficient but not Arid2-wt lung cancers in both cell lines as well as animal models. Treatment with an HSPA1 A inhibitor could significantly inhibit the malignant progression of lung cancer with ARID2 deficiency. Together, our findings establish ARID2 as an important tumor suppressor in LUADs with novel mechanistic insights, and further identify HSPA1 A as a potential therapeutic target in ARID2-deficient LUADs.

为评估玉米白斑病在我国发生流行的风险,对疑似白斑病病样进行分离,采用抗病性鉴定以及气象数据对白斑病病原菌流行进行评估,经形态学特征观察和分子鉴定确定山东滕州玉米白斑病病原菌的分类地位。结果表明,引致山东滕州玉米白斑病的病原菌ST-1与云南白斑病病原菌WS-191同为宽胫附球孢(Epicoccum latusicol⁃lum)。两个菌株最适生长温度均为25~28℃,pH值为5~7。气象数据分析表明,我国8个玉米主产区4-9月平均温度为10~30℃,适合白斑病发生流行。在645个西南玉米区试新品种抗病性鉴定中,中抗-高抗377个,占58.44%;感病-高感品种268个,占41.56%。综上所述,我国主要玉米产区均有白斑病发生条件,存在大面积流行的风险。

Remote tracking the variation of air quality in an effective way will be highly helpful to decrease the health risk of human short-and long-term exposures to air ***,high power consumption and poor sensing performance remain the concerned issues,thereby limiting the scale-up in deploying air quality tracking ***,we report a standalone-like smart device that can remotely track the variation of air pollutants in a power-saving ***,the created smart device demonstrated satisfactory selectivity(against six kinds of representative exhaust gases or air pollutants),desirable response magnitude(164–100 ppm),and acceptable response/recovery rate(52.0/50.5 s),as well as linear response relationship to *** aging for 2 weeks,the created device exhibited relatively stable sensing performance more than 3 ***,a photoluminescence-enhanced light fidelity(Li-Fi)telecommunication technique is proposed and the Li-Fi communication distance is significantly ***,our reported standalone-like smart device would sever as a powerful sensing platform to construct high-performance and low-power consumption air quality wireless sensor networks and to prevent air pollutant-induced diseases via a more effective and low-cost approach.
