Measures of bioavailable serum sex hormone levels in aging Chinese by protein chip

The purpose of this study was to develop a protein chip technique based on receptor binding assays to measure bioavailable serum sex hormone levels (BSSHL). 224 aging healthy Chinese were investigated to get the referenced values of BSSHL for the first time. In the assays recombined sex hormone receptor proteins were jointed to polysaccharide coated slides to make protein chip,; the dose-dependence curves of sex hormone on chip were prepared. The data showed that this method had good precision (CV>16%); accuracy (Bias>10%),; the sensitivity could reach 1 pmol/L. From the results, BSSHL of men; women declined with aging, but no significant differences were observed. The BSSHL of aging men were higher than those of women. The bioavailable serum;rogen level of men was 52–112 pmol/L, women’s was 3–70 pmol/L; the whole group was 41.9–81.4 pmol/L. The bioavailable serum estrogen level of men was 0.8–3.0 pmol/L, women’s was 1.2–2.5 pmol/L; the whole group was 0.6–2.64 pmol/L. Based on the assays, BSSHL measurement by protein chip can meet the needs of epidemiological studies in terms of speed, accuracy; sample volume required,; was helpful in quantitative assessment of aging people’s health.

The diagnosis of prostate diseases by magnetic resonance imaging (MRI) in vivo is sometimes difficult for the lower signal to noise ratio (SNR). To increase the SNR of the prostate image, we designed a RF coil that can be inserted into rectum and was named endorectal coil. The properties of RF coil were evaluated using a network analyzer. Moreover the images and spectroscopy of a special phantom were acquired and the results were compared to those of the commercial TORSO coil (G.E. Medical Systems, USA). Our coil gave a significantly higher SNR at the region of interest (ROI). The achieved high local SNR and resulting high spatial resolution would add more anatomic and biochemical information to the diagnosis of prostate diseases.

BACKGROUND:Present studies on the reversal of multi-drug resistance(MDR) are almost entirely limited to in vitro systems,and are seldom carried out in vivo. In order to study MDR under the in vivo situation,a MDR cell strain was used to establish a model in nude mice via orthotopic implantation directed by B-ultrasonography. This model is expected to provide a good platform for evaluating strategies to reverse the phenomenon of primary hepatocellular carcinoma(HCC) MDR. METHODS:An orthotopic MDR1 hepatoma was obtained by injecting the cell lines HepG2 and HepG2/ADM subserosally into the liver of nude mice(10 control and 20 MDR mice). The injections were made under B-ultrasonographic direction. Ultrasonography and laparotomy were used to assess tumor growth,and the long chain PCR technique was applied to sequence the MDR1 gene from multi-drug resistant human HCC cells,HepG2/ADM,and corresponding implanted tumor tissues. Furthermore,MDR1 mRNA and P-gp protein expression were evaluated by reverse transcriptase-polymerase chain reaction(RT-PCR),Western blotting,and immunohistochemical methods. RESULTS:The success rates of tumor implantation and induction of MDR were 100%(30/30) and 95%(19/20),respectively. The 3.8kbp MDR1 gene band was detected from the HepG2/ADM cell line and the correspondingimplanted tumor tissues,and the MDR1 gene sequence coincided with that reported in GenBank. The expressions of MDR1 mRNA and P-gp protein in MDR mice were significantly higher than those in the control group. There was a significant difference in P-gp protein expression between MDR and control mice. CONCLUSION:A MDR model has been successfully established in nude mice via orthotopic implantation of multi-drug resistant human HCC cells directed by ultrasonography.

To investigate whether the expression of exogenous heme oxygenase-1 (HO-1) gene within vascular smooth muscle cells (VSMC) could protect the cells from free radical attack and inhibit cell proliferation, we established an in vitro transfection of human HO-1 gene into rat VSMC mediated by a retroviral vector. The results showed that the profound expression of HO-1 protein as well as HO activity was 1.8- and 2.0-fold increased respectively in the transfected cells compared to the non-transfected ones. The treatment of VSMC with different concentrations of H2O2 led to the remarkable cell damage as indicated by survival rate and LDH leakage. However, the resistance of the HO-1 transfected VSMC against H2O2 was significantly raised. This protective effect was dramatically diminished when the transfected VSMC were pretreated with ZnPP-IX, a specific inhibitor of HO, for 24 h. The expression levels of 3 subtypes (α1A, α1B and α1D) of α1-adrenoceptor have been determined by radioligand binding assay and reverse-transcription polymerase chain reaction (RT-PCR) assay in neonatal rat heart. mRNAs for all the 3 subtypes have been detected in RT-PCR assays. The density of total α1-AR ( Bmax) was (134±4) fmol/mg protein and the Kd value was (164 ± 15) pmol/L. The α1-adrenoceptor subtypes selective antagonists, RS17053, ( + )niguldipine, 5-MU, BMY 7378 and WB4101, competitively inhibited binding of 125I-BE 2254 to α1-adrenoceptor in a concentration-dependent manner. The inhibition curves were better fit for the two-site model. The binding sites for α1A-, α1B- and α1D-adrenoceptor amounted to 25% , 50% and 25% of the total binding sites, respectively.

BACKGROUND: Patients with decompensated hepatitis B vires (HBV)-related cirrhosis tend to have low or undetectable HBV replication. However, some patients continue to have high levels of HBV replication and effective suppression of HBV replication with antiviral agents may potentially decrease hepatic necroinflammation and improve or stabilize liver function. This review was to under stand the efficacy and safety of lamivudine in the treatment of decompensated HBV cirrhosis. DATA SOURCES: An English-language literature search (MEDLINE January 1988-July 2005) was performed, and a total of 52 articles/abstracts relevant to the issue were selected. After review of the selected papers, the meaningful results and conclusions were extracted using scientific crite ria. The papers reviewed pertained mainly to the efficacy and safety profiles of lamivudine treatment for decompensated HBV cirrhosis. RESULTS: The ultimate treatment of decompensated HBV cirrhosis is liver transplantation, but lamivudine treatment may lead to rapid suppression of viral replication and improvement of biochemical and clinical parameters, reduced morbidity and hospitalization for complications of liver disease, increased pre-transplant survival as well as reduced need for transplantation. However, viral resistance can develop after prolonged treatment with lamivudine, and breakthrough hepatitis may be fatal in few patients. Adefovir is effective for lamivudine-resistant HBV mutants. CONCLUSIONS: Antiviral therapy with lamivudine for decompensated HBV cirrhosis can be effective. However, some patients may experience a hepatitis flare with the emergence of YMDD mutants resulting in progressive worsening of liver disease, and should be referred for 'rescue' therapy with other nucleoside/nucleotide analogues such as adefovir dipivoxil.

Retinoblastoma is the most common childhood primary intraocular malignancy, with the majority of cases being diagnosed before 5 years of age. Retinoblastoma in adults is extremely rare. Here, we report the case of a 20-year-old man who presented with a 3 year history of blurred vision in the right eye. Imaging did not reveal the typical presentation of retinoblastoma. After considering Coats’ disease, a diagnosis of late-presenting retinoblastoma was made through cytological analysis. Diagnosis of retinoblastoma should be considered in the presence of uncertain mass lesions in the fundus of an adult.
