PSA controls hepatic lipid metabolism by regulating the NRF2 signaling pathway

The activity of proteinase is reported to correlate with the development and progression of nonalcoholic fatty liver disease(NAFLD).Puromycin-sensitive aminopeptidase(PSA/NPEPPS)is an integral nontransmembrane enzyme that functions to catalyze the cleavage of amino acids near the N-terminus of polypeptides.A previous study suggested that this enzyme acts as a regulator of neuropeptide activity;however,the metabolic function of this enzyme in the liver has not been ***,we identified the novel role of PSA in hepatic lipid ***,PSA expression was lower in fatty livers from NAFLD patients and mice(HFD,ob/ob,and db/db).PSA knockdown in cultured hepatocytes exacerbated diet-induced triglyceride accumulation through enhanced lipogenesis and attenuated fatty acid β-***,PSA mediated activation of the master regulator of antioxidant response,nuclear factor erythroid 2-related factor 2(NRF2),by stabilizing NRF2 protein expression,which further induced downstream antioxidant enzymes to protect the liver from oxidative stress and lipid ***,liver-specific PSA overexpression attenuated hepatic lipid accumulation and steatosis in ob/ob ***,in human liver tissue samples,decreased PSA expression correlated with the progression of ***,our findings suggest that PSA is a pivotal regulator of hepatic lipid metabolism and its antioxidant function occurs by suppressing NRF2 ***,PSA may be a potential biomarker and therapeutic target for treating NAFLD.

Particulate nitrate,a key component of fine particles,forms through the intricate gas-to-particle conversion *** process is regulated by the gas-to-particle conversion coefficient of nitrate(ε(NO_(3)^(-))).The mechanism betweenε(NO_(3)^(-))and its drivers is highly complex and nonlinear,and can be characterized by machine learning ***,conventional machine learning often yields results that lack clear physical meaning and may even contradict established physical/chemical mechanisms due to the influence of ambient *** urgently needs an alternative approach that possesses transparent physical interpretations and provides deeper insights into the impact ofε(NO_(3)^(-)).Here we introduce a supervised machine learning approachdthe multilevel nested random forest guided by theory *** approach robustly identifies NH4 t,SO_(4)^(2-),and temperature as pivotal drivers forε(NO_(3)^(-)).Notably,substantial disparities exist between the outcomes of traditional random forest analysis and the anticipated actual ***,our approach underscores the significance of NH4 t during both daytime(30%)and nighttime(40%)periods,while appropriately downplaying the influence of some less relevant drivers in comparison to conventional random forest *** research underscores the transformative potential of integrating domain knowledge with machine learning in atmospheric studies.

This paper introduces the basic parameters,focal mechanism solutions and earthquake sequence characteristics of the Kalpin M_S5.3 earthquake sequence of December 1,2013,and analyzed seismic activity before the earthquake,the adjacent tectonic features and the precursory anomaly at fixed points within a range of 200 km. Research indicates:(1) The earthquake occurred on Kalpin fault,the source rupture type is thrust faulting with sinistral strike-slip component.( 2) The earthquake sequence is mainshock-aftershock type,with the aftershock distribution attenuating quickly and trending NE.(3) Abnormal seismic activity before the earthquake was characterized by seismically nesting quiescence of MS2. 0-4. 0 earthquakes,seismic quiescence of MS4. 0 earthquakes and seismic belts of MS3. 0 earthquakes in the Kalpin block, abnormal enhancement zone of moderate earthquakes on Puchang fault and seismological parameters.(4) Anomalies of precursory observation data at fixed stations are mainly characterized by mutation. Apart from the borehole tiltmeter in Halajun, the spatial distribution of other abnormal precursors showed a phenomenon of migration from the near field to far field and from the epicenter to the peripheries.

[Objectives]This study was conducted to explore the effects of physical diseases and life events on depression among urban elderly people.[Methods]Five hundred urban elderly people in Shiyan City were selected using a convenient sampling method to investigate and diagnose depression using the Geriatric Mental State Schedule and its accompanying computer diagnostic *** Minimum Data Set(MDS)and related factor survey form were used to investigate physical diseases and life events,and statistical analysis was conducted using methods such as logistic regression analysis and factor analysis.[Results]The prevalence rates of angina,stroke,or partial paralysis,migraine,vision problems,hyperthyroidism or hypothyroidism,as well as the number of physical diseases were significantly higher in elderly depression patients than in objects free of *** or partial paralysis[OR(95%CI)=6.389(1.684,24.237)],emotional trauma events[OR(95%CI)=1.438(1.069,1.934)]and adverse health and economic events[OR(95%CI)=1.652(1.099,2.483)]were risk factors for late life depression(LLD).[Conclusions]Some physical diseases and life events may affect the onset process of depression in urban elderly people,and various measures need to be taken to reduce the risk of physical diseases and life events on depression.

High-performance and cost-effective catalysts for water splitting are key components of hydrogen-based energy technologies. Metal-organic framework(MOF)-derived metal phosphide composites have immense potential as highly active and stable electrocatalysts but suffer from the poor efficacy of available electrode assembly methods. In this study, an MOF-derived nitrogen-doped porous carbon/Co/Co P/carbon paper(NC/Co/Co P/CP) composite electrode was assembled by electrophoretic deposition and post-processing reactions. The binder-free electrode showed good catalytic activity, significantly higher than that of traditional electrodes. The electrode required overpotentials of 208 and 350 m V to achieve a current density of 10 m A/cm^2 for the hydrogen and oxygen evolution reactions, respectively. This facile synthetic method provides a promising route for designing metal-doped and multi-metal phase MOF-derived composite electrodes for energy storage and conversion devices.

Carbon dots(CDs),a type of nanoparticle with excellent optical properties,good biocompatibility,and small size,are finding increasing application across the fields of biology and *** recent years,biomass-derived CDs with pharmacological activity(BP-CDs)derived from herbal medicines(HMs),HMs extracts and other natural products with demonstrated pharmaceutical activity have attracted particular ***,we review recent advances in the development of BP-CDs,covering the selection of biomass precursors,different methods used for the synthesis of BP-CDs from natural sources,and the purification of ***,we summarize the many remarkable properties of BP-CDs including optical properties,biocompatibility and pharmaceutical ***,the antibacterial,antiviral,anticancer,biosensing,bioimaging,and other applications of BP-CDs are ***,we discuss the advantages and disadvantages of BP-CDs and Western drug-derived CDs,highlighting the excellent performance of ***,based on the current state of research on BP-CDs,we suggest several aspects of BP-CDs that urgently need to be addressed and identify directions that should be pursued in the *** comprehensive review on BP-CDs is expected to guide the precise design,preparation,and future development of BP-CDs,thereby advancing the application of BP-CDs in biomedicine.

Near-inertial oscillation is an important physical process transferring surface wind energy into deep *** investigated the near-inertial kinetic energy(NIKE)variability using acoustic Doppler current profiler measurements from a mooring array deployed in the tropical western Pacific Ocean along 130°E at 8.5°N,11°N,12.6°N,15°N,and 17.5°N from September 2015 to January *** features,decay timescales,and significant seasonal variability of the observed NIKE were *** the mooring sites of 17.5°N,15°N,and 12.6°N,the NIKE peaks occurred in boreal autumn and the NIKE troughs were observed in boreal *** contrast,the NIKE at 11°N and 8.5°N showed peaks in winter and troughs in *** cyclones and strong wind events played an important role in the emergence of high-NIKE events and explained the seasonality and latitudinal characteristics of the observed NIKE.

Targeting oncogenic mutant p53 represents an attractive strategy for cancer treatment due to the high frequency of gain-of-function mutations and ectopic expression in various cancer *** extensive efforts,the absence of a druggable active site for small molecules has rendered these mutants therapeutically *** we develop a selective and effective proteolysis-targeting chimera(PROTAC)for p53-R175H,a common hotspot mutant with dominant-negative and oncogenic *** a novel iterative molecular docking-guided post-SELEX(systematic evolution of ligands by exponential enrichment)approach,we rationally engineer a high-performance DNA aptamer with improved affinity and specificity for *** this resulting aptamer as a binder for PROTACs,we successfully developed a selective p53-R175H degrader,named ***53m induces the ubiquitin–protea some-dependent degradation of p53-R175H while sparing wildtype ***,dp53m demonstrates significant antitumor efficacy in p53-R175H-driven cancer cells both in vitro and in vivo,without ***,dp53m significantly and synergistically improves the sensitivity of these cells to cisplatin,a commonly used chemotherapy *** findings provide evidence of the potential therapeutic value of dp53m in p53-R175H-driven cancers.

In this paper, we investigate an integrable high order nonlocal coupled Ablowitz-Kaup-Newell-Segur (AKNS) system for the first time. With the aid of Lax pair of this nonlocal system, Darboux transformation (DT) and new soliton-like solutions are obtained. Different from local equations, Darboux transformation of nonlocal systems needs to meet certain conditions. In this article, under the condition of symmetry reduction, the components of Darboux transformation need to satisfy . In order to study the dynamic information of the solutions, the images of the solutions are given.
